    Ex post analysis of the regional impacts of major infrastructure: the Channel Tunnel 10 years on.

    The regional impact of transport infrastructure investment has achieved considerable attention across Europe in recent years. The construction of major bridges and tunnels to overcome natural barriers and the development of the trans-European networks has led to large volumes of research on the likely economic impacts of such investment on neighbouring regions. Most of these studies are ex ante studies undertaken as part of the decision process into the investment, either on the part of the project promoter or the affected regions. At the same time there has been a continuation of interest in the broader question of the wider economic benefits arising from transport improvements at a more aggregate level; whether and under what circumstances such benefits do arise, and if so how they can be incorporated in any evaluation. As a recent study by Flyvbjerg et al (2003) has shown, the claims made for the returns on major projects have tended to be exaggerated, both in terms of underestimates of likely costs and overestimates of potential direct traffic benefits. This paper provides a relatively rare example of an ex post study of what impacts one of the major recent infrastructure projects has had on the local and regional economy by looking at the experience of the Channel Tunnel after its first ten years of operation. The paper reviews the methodological issues in carrying out an ex post study and assembles evidence related to traffic, labour market, investment and development impacts compared to the ex ante expectations. The main methodological issue is in establishing the counter-factual position of how the regional economy would have developed in the absence of the tunnel. A substantial volume of traffic would have continued to have passed through the region’s ports using the ferries which would have required continuing investment over this period. This has been a period also in which major changes have occurred in the EU economy with the move to completion of a Single Market. Many of the driving forces of the regional economy come from the adjacent London metropolitan region. The approach adopted has been to examine trends in the regional economy relative to those in the wider regional and national economies in both the UK and France. There are two basic questions: to what extent would traffic have continued to grow at the rate experienced in the absence of the tunnel infrastructure; and has tunnel related traffic had a differential impact on the local economy from a similar volume of ferry traffic? The paper examines in detail the volume and structure of traffic and compares these with ex ante traffic forecasts; trends in the local labour markets in terms of the growth of job opportunities, occupational structure and the evolution of unemployment; investment (including foreign investment) in the regional economy; and the development and execution of plans for physical development in the region. The paper concludes that although much has changed in the region following the completion of the tunnel, it is difficult to identify a significant difference in the aggregate performance relative to the wider regional and national experiences. Essentially, the improvement of transport infrastructure has enabled the regions to be more integrated into their wider regions and experience a similar economic performance, but whether this has led to a better performance is more difficult to determine.

    Interaction of Cryptococcus neoformans Rim101 and Protein Kinase A Regulates Capsule

    Cryptococcus neoformans is a prevalent human fungal pathogen that must survive within various tissues in order to establish a human infection. We have identified the C. neoformans Rim101 transcription factor, a highly conserved pH-response regulator in many fungal species. The rim101Δ mutant strain displays growth defects similar to other fungal species in the presence of alkaline pH, increased salt concentrations, and iron limitation. However, the rim101Δ strain is also characterized by a striking defect in capsule, an important virulence-associated phenotype. This capsular defect is likely due to alterations in polysaccharide attachment to the cell surface, not in polysaccharide biosynthesis. In contrast to many other C. neoformans capsule-defective strains, the rim101Δ mutant is hypervirulent in animal models of cryptococcosis. Whereas Rim101 activation in other fungal species occurs through the conserved Rim pathway, we demonstrate that C. neoformans Rim101 is also activated by the cAMP/PKA pathway. We report here that C. neoformans uses PKA and the Rim pathway to regulate the localization, activation, and processing of the Rim101 transcription factor. We also demonstrate specific host-relevant activating conditions for Rim101 cleavage, showing that C. neoformans has co-opted conserved signaling pathways to respond to the specific niche within the infected host. These results establish a novel mechanism for Rim101 activation and the integration of two conserved signaling cascades in response to host environmental conditions

    Mineralocorticoid Receptor Blockade Attenuates Chronic Overexpression of the Renin-Angiotensin- Aldosterone System Stimulation of Reduced Nicotinamide Adenine Dinucleotide Phosphate Oxidase and Cardiac Remodeling

    doi: 10.1210/en.2006-1691The renin-angiotensin-aldosterone system contributes to cardiac remodeling, hypertrophy, and left ventricular dysfunction. Angiotensin II and aldosterone (corticosterone in rodents) together generate reactive oxygen species (ROS) via reduced nicotinamide adenine dinucleotide phosphate (NADPH) oxidase, which likely facilitate this hypertrophy and remodeling. This investigation sought to determine whether cardiac oxidative stress and cellular remodeling could be attenuated by in vivo mineralocorticoid receptor (MR) blockade in a rodent model of the chronically elevated tissue renin-angiotensin-aldosterone system, the transgenic TG (mRen2) 27 rat (Ren2). The Ren2 overexpresses the mouse renin transgene with resultant hypertension, insulin resistance, proteinuria, and cardiovascular damage. Young (6- to 7-wk-old) male Ren2 and age-matched Sprague-Dawley rats were treated with spironolactone or placebo for 3 wk. Heart tissue ROS, immunohistochemical analysis of 3-nitrotyrosine,and NADPH oxidase (NOX) subunits (gp91phox recently renamed NOX2, p22phox, Rac1, NOX1, and NOX4) were measured. Structural changes were assessed with cine-magnetic resonance imaging, transmission electron microscopy, and light microscopy. Significant increases in Ren2 septal wall thickness (cine-magnetic resonance imaging) were accompanied by perivascular fibrosis, increased mitochondria, and other ultrastructural changes visible by light microscopy and transmission electron microscopy. Although there was no significant reduction in systolic blood pressure, significant improvements were seen with MR blockade on ROS formation and NOX subunits (each P < 0.05). Collectively, these data suggest that MR blockade, independent of systolic blood pressure reduction, improves cardiac oxidative stress-induced structural and functional changes, which are driven, in part, by angiotensin type 1 receptor-mediated increases in NOX.This research was supported by National Institutes of Health (NIH) Grants R01 HL73101-01A1 (to J.R.S.) and P01 HL-51952 (to C.F.), the Veterans Affairs Merit System (0018) (to J.R.S.), and Advanced Research Career Development (to C.S.).     Young pregnant women's views on the acceptability of screening for chlamydia as part of routine antenatal care

    <p>Abstract</p> <p>Background</p> <p>In pregnancy, untreated chlamydia infection has been associated with adverse outcomes for both mother and infant. Like most women, pregnant women infected with chlamydia do not report genital symptoms, and are therefore unlikely to be aware of their infection. The aim of this study was to determine the acceptability of screening pregnant women aged 16-25 years for chlamydia as part of routine antenatal care.</p> <p>Methods</p> <p>As part of a larger prospective, cross-sectional study of pregnant women aged 16-25 years attending antenatal services across Melbourne, Australia, 100 women were invited to participate in a face-to-face, semi structured interview on the acceptability of screening for chlamydia during pregnancy. Women infected with chlamydia were oversampled (n = 31).</p> <p>Results</p> <p>Women had low levels of awareness of chlamydia before the test, retained relatively little knowledge after the test and commonly had misconceptions around chlamydia transmission, testing and sequelae. Women indicated a high level of acceptance and support for chlamydia screening, expressing their willingness to undertake whatever care was necessary to ensure the health of their baby. There was a strong preference for urine testing over other methods of specimen collection. Women questioned why testing was not already conducted alongside other antenatal STI screening tests, particularly in view of the risks chlamydia poses to the baby. Women who tested positive for chlamydia had mixed reactions, however, most felt relief and gratitude at having had chlamydia detected and reported high levels of partner support.</p> <p>Conclusions</p> <p>Chlamydia screening as part of routine antenatal care was considered highly acceptable among young pregnant women who recognized the benefits of screening and strongly supported its implementation as part of routine antenatal care. The acceptability of screening is important to the uptake of chlamydia screening in future antenatal screening strategies.</p

    A Viral Ubiquitin Ligase Has Substrate Preferential SUMO Targeted Ubiquitin Ligase Activity that Counteracts Intrinsic Antiviral Defence

    Intrinsic antiviral resistance represents the first line of intracellular defence against virus infection. During herpes simplex virus type-1 (HSV-1) infection this response can lead to the repression of viral gene expression but is counteracted by the viral ubiquitin ligase ICP0. Here we address the mechanisms by which ICP0 overcomes this antiviral response. We report that ICP0 induces the widespread proteasome-dependent degradation of SUMO-conjugated proteins during infection and has properties related to those of cellular SUMO-targeted ubiquitin ligases (STUbLs). Mutation of putative SUMO interaction motifs within ICP0 not only affects its ability to degrade SUMO conjugates, but also its capacity to stimulate HSV-1 lytic infection and reactivation from quiescence. We demonstrate that in the absence of this viral countermeasure the SUMO conjugation pathway plays an important role in mediating intrinsic antiviral resistance and the repression of HSV-1 infection. Using PML as a model substrate, we found that whilst ICP0 preferentially targets SUMO-modified isoforms of PML for degradation, it also induces the degradation of PML isoform I in a SUMO modification-independent manner. PML was degraded by ICP0 more rapidly than the bulk of SUMO-modified proteins in general, implying that the identity of a SUMO-modified protein, as well as the presence of SUMO modification, is involved in ICP0 targeting. We conclude that ICP0 has dual targeting mechanisms involving both SUMO- and substrate-dependent targeting specificities in order to counteract intrinsic antiviral resistance to HSV-1 infection

    Oceanographic and climatic consequences of the tectonic evolution of the southern scotia sea basins, Antarctica

    The Scotia Sea is a complex geological area located in the Southern Ocean which evolution is closely linked to the opening of the Drake Passage. Structural highs of continental nature derived from the former continental bridge between South America and the Antarctic Peninsula surround the abyssal plains of the Scotia Sea, restricting small isolated sedimentary basins along its southern margin. Morpho-structural and seismo-stratigraphic analyses of multichannel seismic reflection profiles, and additional geophysical data available in the region, have been conducted, decoding regional and global implications of the basins' evolution. The main aim of this work is to describe the stratigraphic evolution of the southern Scotia Sea basins, from their opening in the back-arc tectonic context of the Scotia Sea, to the last oceanographic changes which have carried on global climatic implications. The evolution of the south Scotia Sea occurred through two major tectonic stages registered in the sedimentary record of the region: 1) the end of the subduction in the northwest part of the Weddell Sea during the early Miocene, which shortened the back-arc subduction trench generating a major change in the regional tectonic field that determined the evolution of the southern basins towards two different types of passive margins: magma-poor and magma-rich; and 2) the full development of the southern Scotia Sea basins during the middle Miocene, that led to the opening of deep oceanic gateways along the South Scotia Ridge.     Plasticity in bilateral superior temporal cortex: effects of deafness and cochlear implantation on auditory and visual speech processing

    While many individuals can benefit substantially from cochlear implantation, the ability to perceive and understand auditory speech with a cochlear implant (CI) remains highly variable amongst adult recipients. Importantly, auditory performance with a CI cannot be reliably predicted based solely on routinely obtained information regarding clinical characteristics of the CI candidate. This review argues that central factors, notably cortical function and plasticity, should also be considered as important contributors to the observed individual variability in CI outcome. Superior temporal cortex (STC), including auditory association areas, plays a crucial role in the processing of auditory and visual speech information. The current review considers evidence of cortical plasticity within bilateral STC, and how these effects may explain variability in CI outcome. Furthermore, evidence of audio-visual interactions in temporal and occipital cortices is examined, and relation to CI outcome is discussed. To date, longitudinal examination of changes in cortical function and plasticity over the period of rehabilitation with a CI has been restricted by methodological challenges. The application of functional near-infrared spectroscopy (fNIRS) in studying cortical function in CI users is becoming increasingly recognised as a potential solution to these problems. Here we suggest that fNIRS offers a powerful neuroimaging tool to elucidate the relationship between audio-visual interactions, cortical plasticity during deafness and following cochlear implantation, and individual variability in auditory performance with a CI
