Lost Highway Not Forgotten: Satellite Tracking of a Bowhead Whale (Balaena mysticetus) from the Critically Endangered Spitsbergen Stock
The Spitsbergen bowhead whale stock is critically endangered. It is believed to number in the tens. Here we report results from the first satellite transmitter ever deployed on an individual from this stock. A female whale was tagged on 3 April 2010 (at 79Ë54' N, 01Ë03' E), but no locations were transmitted by the tag until 30 April 2010, after which data were received continuously for 86 days. Additionally, three small clusters of locations were transmitted later in the year; the latest was received 20 December 2010 (262 days after deployment). During the 86 days of continuous tracking, the whale initially remained in the middle of the Fram Strait, between 77Ë45' N, 5Ë W and 80Ë10' N, 5Ë E. For a two-week period starting around 10 June 2010, the whale traveled southwest down to 73Ë40' N (at least 950 km). Subsequently it remained at southern latitudes between ~70Ë and 73Ë N until the tag stopped continuous transmissions on 24 July. Movement patterns analyzed using first-passage times (FTP), fitted as functions of various environmental variables using Cox Proportional Hazards models, showed that the whale spent most of its time in waters close to the ice edge with modest ice coverage, over areas where the bottom slope was relatively steep. Winter positions (27 November â 20 December 2010) revealed that the whale was back in the North at about 80Ë N. This information, in combination with recent data from passive acoustic listening devices, suggests that the Spitsbergen bowhead stock overwinters at high-latitude locations. The north-south movements of this whale during summer are consistent with the patterns that early whalers described for bowhead whales in this region in the 16th and 17th centuries.La population de baleines borĂ©ales de Spitzberg est en danger critique dâextinction. Mitogenomics and the genetic differentiation of contemporary <i>Balaena mysticetus</i> (Cetacea) from Svalbard
Full mitochondrial genomes were assembled for 12 recently sampled animals from the Svalbard bowhead whale (Balaena mysticetus) stock via high-throughput sequencing data, facilitating analysis of the demographic history of the population for the first time. The Svalbard population has retained noticeable amounts of mitochondrial genome diversity despite extreme historical harvest levels. Haplotype and nucleotide diversities were similar to those estimated earlier for other bowhead whale populations. The reconstructed demographic history was in accordance with a boomâbust scenario, combining a slight Pleistocene population growth 25 000â35 000 years ago and a Holocene decline. Employing a mutation rate of 3.418 Ă 10â8 substitutions per site per year, the time to the most recent common ancestor for the mitochondrial genomes of the contemporary Svalbard bowhead whales was estimated to be 68 782 (54 353â83 216) years before the present. Based on 370 bp fragments of the D-loop region, significant genetic differentiation was detected between all extant bowhead whale populations across the circumpolar Arctic. Thus, the Svalbard bowhead whales can be regarded as a population with its own genetic legacy
Institutionalising Kant's political philosophy: Foregrounding cosmopolitan right
There exists a longstanding debate over the global institutional implications of Immanuel Kant's political philosophy: does such a philosophy entail a federal world government, or instead only a confederal âleague of nationsâ? However, while the systematic nature of Kant's tripartite âdoctrine of right' is well recognised, this debate has been conducted with all but exclusive focus on âinternational right' in particular. This article, by contrast, brings âcosmopolitan right' firmly into view. It proceeds by way of engagement with the two Kantian arguments made in defence of a âleague of nationsâ in discussion of international right, each of which appeals to aspects of statesâ supposed âpersonhoodâ: the first appeals to statesâ distinctive moral personality; the second to statesâ physical manifestation. The article considers what happens when we assess these arguments not just in light of the demands of international right, but also in light of cosmopolitan right, and thus in light of public right more comprehensively. The answer is that such arguments cannot succeed as full defences of a league of nations. Indeed, when we assess such arguments with cosmopolitan right in view, they point instead â either tentatively or definitively â in the direction of world government
Pretransplant Serum Uromodulin and Its Association with Delayed Graft Function Following Kidney TransplantationâA Prospective Cohort Study
Delayed graft function (DGF) following kidney transplantation is associated with increased risk of graft failure, but biomarkers to predict DGF are scarce. We evaluated serum uromodulin (sUMOD), a potential marker for tubular integrity with immunomodulatory capacities, in kidney transplant recipients and its association with DGF. We included 239 kidney transplant recipients and measured sUMOD pretransplant and on postoperative Day 1 (POD1) as independent variables. The primary outcome was DGF, defined as need for dialysis within one week after transplantation. In total, 64 patients (27%) experienced DGF. In multivariable logistic regression analysis adjusting for recipient, donor and transplant associated risk factors each 10 ng/mL higher pretransplant sUMOD was associated with 47% lower odds for DGF (odds ratio (OR) 0.53, 95% confidence interval (95%-CI) 0.30â0.82). When categorizing pretransplant sUMOD into quartiles, the quartile with the lowest values had 4.4-fold higher odds for DGF compared to the highest quartile (OR 4.41, 95%-CI 1.54â13.93). Adding pretransplant sUMOD to a model containing established risk factors for DGF in multivariable receiver-operating-characteristics (ROC) curve analysis, the area-under-the-curve improved from 0.786 [95%-CI 0.723â0.848] to 0.813 [95%-CI 0.755â0.871, p = 0.05]. SUMOD on POD1 was not associated with DGF. In conclusion, higher pretransplant sUMOD was independently associated with lower odds for DGF, potentially serving as a non-invasive marker to stratify patients according to their risk for developing DGF early in the setting of kidney transplantation

Therapeutic Effects of Autologous Tumor-Derived Nanovesicles on Melanoma Growth and Metastasis
Cancer vaccines with optimal tumor-associated antigens show promise for anti-tumor immunotherapy. Recently, nano-sized vesicles, such as exosomes derived from tumors, were suggested as potential antigen candidates, although the total yield of exosomes is not sufficient for clinical applications. In the present study, we developed a new vaccine strategy based on nano-sized vesicles derived from primary autologous tumors. Through homogenization and sonication of tumor tissues, we achieved high yields of vesicle-bound antigens. These nanovesicles were enriched with antigenic membrane targets but lacked nuclear autoantigens. Furthermore, these nanovesicles together with adjuvant activated dendritic cells in vitro, and induced effective anti-tumor immune responses in both primary and metastatic melanoma mouse models. Therefore, autologous tumor-derived nanovesicles may represent a novel source of antigens with high-level immunogenicity for use in acellular vaccines without compromising safety. A Crucial Role for Infected-Cell/Antibody Immune Complexes in the Enhancement of Endogenous Antiviral Immunity by Short Passive Immunotherapy
Antiviral monoclonal antibodies (mAbs) represent promising therapeutics. However, most mAbs-based immunotherapies conducted so far have only considered the blunting of viral propagation and not other possible therapeutic effects independent of virus neutralization, namely the modulation of the endogenous immune response. As induction of long-term antiviral immunity still remains a paramount challenge for treating chronic infections, we have asked here whether neutralizing mAbs can, in addition to blunting viral propagation, exert immunomodulatory effects with protective outcomes. Supporting this idea, we report here that mice infected with the FrCasE murine retrovirus on day 8 after birth die of leukemia within 4â5 months and mount a non-protective immune response, whereas those rapidly subjected to short immunotherapy with a neutralizing mAb survive healthy and mount a long-lasting protective antiviral immunity with strong humoral and cellular immune responses. Interestingly, the administered mAb mediates lysis of infected cells through an antibody-dependent cell cytotoxicity (ADCC) mechanism. In addition, it forms immune complexes (ICs) with infected cells that enhance antiviral CTL responses through FcÎłR-mediated binding to dendritic cells (DCs). Importantly, the endogenous antiviral antibodies generated in mAb-treated mice also display the same properties, allowing containment of viral propagation and enhancement of memory cellular responses after disappearance of the administered mAb. Thus, our data demonstrate that neutralizing antiviral mAbs can act as immunomodulatory agents capable of stimulating a protective immunity lasting long after the end of the treatment. They also show an important role of infected-cells/antibody complexes in the induction and the maintenance of protective immunity through enhancement of both primary and memory antiviral T-cell responses. They also indicate that targeting infected cells, and not just viruses, by antibodies can be crucial for elicitation of efficient, long-lasting antiviral T-cell responses. This must be considered when designing antiviral mAb-based immunotherapies
Strategies to Target Tumor Immunosuppression
The tumor microenvironment is currently in the spotlight of cancer immunology research as a key factor impacting tumor development and progression. While antigen-specific immune responses play a crucial role in tumor rejection, the tumor hampers these immune responses by creating an immunosuppressive microenvironment. Recently, major progress has been achieved in the field of cancer immunotherapy, and several groundbreaking clinical trials demonstrated the potency of such therapeutic interventions in patients. Yet, the responses greatly vary among individuals. This calls for the rational design of more efficacious cancer immunotherapeutic interventions that take into consideration the âimmune signatureâ of the tumor. Multimodality treatment regimens that aim to enhance intratumoral homing and activation of antigen-specific immune effector cells, while simultaneously targeting tumor immunosuppression, are pivotal for potent antitumor immunity
