Inefficient Toll-Like Receptor-4 Stimulation Enables Bordetella parapertussis to Avoid Host Immunity
The recognition of bacterial lipopolysaccharide (LPS) by host Toll-like receptor (TLR)4 is a crucial step in developing protective immunity against several gram negative bacterial pathogens. Bordetella bronchiseptica and B. pertussis stimulate robust TLR4 responses that are required to control the infection, but a close relative, B. parapertussis, poorly stimulates this receptor, and TLR4 deficiency does not affect its course of infection. This led us to hypothesize that inefficient TLR4 stimulation enables B. parapertussis to evade host immunity. In a mouse model of infection, B. parapertussis grew rapidly in the lungs, but no measurable increase in TLR4-mediated cytokine, chemokine, or leukocyte responses were observed over the first few days of infection. Delivery of a TLR4 stimulant in the inoculum resulted in a robust inflammatory response and a 10- to 100-fold reduction of B. parapertussis numbers. As we have previously shown, B. parapertussis grows efficiently during the first week of infection even in animals passively immunized with antibodies. We show that this evasion of antibody-mediated clearance is dependent on the lack of TLR4 stimulation by B. parapertussis as co-inoculation with a TLR4 agonist resulted in 10,000-fold lower B. parapertussis numbers on day 3 in antibody-treated wild type, but not TLR4-deficient, mice. Together, these results indicate that inefficient TLR4 stimulation by B. parapertussis enables it to avoid host immunity and grow to high numbers in the respiratory tract of naïve and immunized hosts
Heat stress increase under climate change twice as large in cities as in rural areas : a study for a densely populated midlatitude maritime region
Urban areas are usually warmer than their surrounding natural areas, an effect known as the urban heat island effect. As such, they are particularly vulnerable to global warming and associated increases in extreme temperatures. Yet ensemble climate-model projections are generally performed on a scale that is too coarse to represent the evolution of temperatures in cities. Here, for the first time, we combine unprecedented long-term (35years) urban climate model integrations at the convection-permitting scale (2.8km resolution) with information from an ensemble of general circulation models to assess temperature-based heat stress for Belgium, a densely populated midlatitude maritime region. We discover that the heat stress increase toward the mid-21st century is twice as large in cities compared to their surrounding rural areas. The exacerbation is driven by the urban heat island itself, its concurrence with heat waves, and urban expansion. Cities experience a heat stress multiplication by a factor 1.4 and 15 depending on the scenario. Remarkably, the future heat stress surpasses everywhere the urban hot spots of today. Our results demonstrate the need to combine information from climate models, acting on different scales, for climate change risk assessment in heterogeneous regions. Moreover, these results highlight the necessity for adaptation to increasing heat stress, especially in urban areas
Enhanced production of multi-strange hadrons in high-multiplicity proton-proton collisions
At sufficiently high temperature and energy density, nuclear matter undergoes a transition to a phase in which quarks and gluons are not confined: the quark-gluon plasma (QGP)(1). Such an exotic state of strongly interacting quantum chromodynamics matter is produced in the laboratory in heavy nuclei high-energy collisions, where an enhanced production of strange hadrons is observed(2-6). Strangeness enhancement, originally proposed as a signature of QGP formation in nuclear collisions(7), is more pronounced for multi-strange baryons. Several effects typical of heavy-ion phenomenology have been observed in high-multiplicity proton-proton (pp) collisions(8,9), but the enhanced production of multi-strange particles has not been reported so far. Here we present the first observation of strangeness enhancement in high-multiplicity proton-proton collisions. We find that the integrated yields of strange and multi-strange particles, relative to pions, increases significantly with the event charged-particle multiplicity. The measurements are in remarkable agreement with the p-Pb collision results(10,11), indicating that the phenomenon is related to the final system created in the collision. In high-multiplicity events strangeness production reaches values similar to those observed in Pb-Pb collisions, where a QGP is formed.Peer reviewe
First measurement of Ξc0 production in pp collisions at s=7 TeV
The production of the charm-strange baryon Csi0c is measured for the first time at the LHC via its
semileptonic decay into e+Csi-νe in pp collisions at sqrt(s) = 7 TeV with the ALICE detector. The transverse momentum (pT) differential cross section multiplied by the branching ratio is presented in the interval 1 < pT < 8 GeV/c at mid-rapidity, |y| < 0.5. The transverse momentum dependence of the Csi0c baryon production relative to the D0 meson production is compared to predictions of event generators with various tunes of the hadronisation mechanism, which are found to underestimate the measured cross- section ratio.The production of the charm-strange baryon Csi0c is measured for the first time at the LHC via its

Measurement of Z(0)-boson production at large rapidities in Pb-Pb collisions at root s(NN)=5.02 TeV
The production of Z(0) bosons at large rapidities in Pb-Pb collisions at root s(NN) = 5.02 TeV is reported. Z(0) candidates are reconstructed in the dimuon decay channel (Z(0) -> mu(+) mu(-)), based on muons selected with pseudo-rapidity -4.0 20 GeV/c. The invariant yield and the nuclear modification factor, RAA, are presented as a function of rapidity and collision centrality. The value of R-AA for the 0-20% central Pb-Pb collisions is 0.67 +/- 0.11(stat.) +/- 0.03 (syst.) f 0.06(corr. syst.), exhibiting a deviation of 2.6 sigma from unity. The results are well-described by calculations that include nuclear modifications of the parton distribution functions, while the predictions using vacuum PDFs deviate from data by 2.3 sigma in the 0-90% centrality class and by 3 sigma in the 0-20% central collisions. Longitudinal asymmetry and its effect on pseudorapidity distributions in Pb-Pb collisions at root s(NN)=2.76 TeV
First results on the longitudinal asymmetry and its effect on the pseudorapidity distributions in Pb-Pb collisions at root s(NN) = 2.76 TeV at the Large Hadron Collider are obtained with the ALICE detector. The longitudinal asymmetry arises because of an unequal number of participating nucleons from the two colliding nuclei, and is estimated for each event by measuring the energy in the forward neutron-Zero-Degree-Calorimeters (ZNs). The effect of the longitudinal asymmetry is measured on the pseudorapidity distributions of charged particles in the regions vertical bar eta vertical bar < 0.9, 2.8 < eta < 5.1 and -3.7 < eta < -1.7 by taking the ratio of the pseudorapidity distributions from events corresponding to different regions of asymmetry. The coefficients of a polynomial fit to the ratio characterise the effect of the asymmetry. A Monte Carlo simulation using a Glauber model for the colliding nuclei is tuned to reproduce the spectrum in the ZNs and provides a relation between the measurable longitudinal asymmetry and the shift in the rapidity (y(0)) of the participant zone formed by the unequal number of participating nucleons. The dependence of the coefficient of the linear term in the polynomial expansion, c(1), on the mean value of y(0) is investigated.Peer reviewe
Inclusive J/ψ production at forward and backward rapidity in p-Pb collisions at √sNN=8.16 TeV
Inclusive J/psi production is studied in p-Pb interactions at a centre-of-mass
energy per nucleon-nucleon collision sqrt(s_NN) = 8.16TeV, using the ALICE detector at the
CERN LHC. The J/psi meson is reconstructed, via its decay to a muon pair, in the centre-of-mass
rapidity intervals 2.03 < ycms < 3.53 and -4.46 < ycms < -2.96, where positive
and negative ycms refer to the p-going and Pb-going direction, respectively. The transverse
momentum coverage is pT < 20 GeV/c. In this paper, ycms- and pT-differential cross
sections for inclusive J/psi production are presented, and the corresponding nuclear modification
factors RpPb are shown. Forward results show a suppression of the J/psi yield with
respect to pp collisions, concentrated in the region pT < 5 GeV/c. At backward rapidity
no significant suppression is observed. The results are compared to previous measurements
by ALICE in p-Pb collisions at sqrt(s_NN) = 5.02TeV and to theoretical calculations. Finally,
the ratios RFB between forward- and backward-ycms RpPb values are shown and discussed
Intravenous alteplase for stroke with unknown time of onset guided by advanced imaging: systematic review and meta-analysis of individual patient data
Background: Patients who have had a stroke with unknown time of onset have been previously excluded from thrombolysis. We aimed to establish whether intravenous alteplase is safe and effective in such patients when salvageable tissue has been identified with imaging biomarkers. Methods: We did a systematic review and meta-analysis of individual patient data for trials published before Sept 21, 2020. Randomised trials of intravenous alteplase versus standard of care or placebo in adults with stroke with unknown time of onset with perfusion-diffusion MRI, perfusion CT, or MRI with diffusion weighted imaging-fluid attenuated inversion recovery (DWI-FLAIR) mismatch were eligible. The primary outcome was favourable functional outcome (score of 0–1 on the modified Rankin Scale [mRS]) at 90 days indicating no disability using an unconditional mixed-effect logistic-regression model fitted to estimate the treatment effect. Secondary outcomes were mRS shift towards a better functional outcome and independent outcome (mRS 0–2) at 90 days. Safety outcomes included death, severe disability or death (mRS score 4–6), and symptomatic intracranial haemorrhage. This study is registered with PROSPERO, CRD42020166903. Findings: Of 249 identified abstracts, four trials met our eligibility criteria for inclusion: WAKE-UP, EXTEND, THAWS, and ECASS-4. The four trials provided individual patient data for 843 individuals, of whom 429 (51%) were assigned to alteplase and 414 (49%) to placebo or standard care. A favourable outcome occurred in 199 (47%) of 420 patients with alteplase and in 160 (39%) of 409 patients among controls (adjusted odds ratio [OR] 1·49 [95% CI 1·10–2·03]; p=0·011), with low heterogeneity across studies (I2=27%). Alteplase was associated with a significant shift towards better functional outcome (adjusted common OR 1·38 [95% CI 1·05–1·80]; p=0·019), and a higher odds of independent outcome (adjusted OR 1·50 [1·06–2·12]; p=0·022). In the alteplase group, 90 (21%) patients were severely disabled or died (mRS score 4–6), compared with 102 (25%) patients in the control group (adjusted OR 0·76 [0·52–1·11]; p=0·15). 27 (6%) patients died in the alteplase group and 14 (3%) patients died among controls (adjusted OR 2·06 [1·03–4·09]; p=0·040). The prevalence of symptomatic intracranial haemorrhage was higher in the alteplase group than among controls (11 [3%] vs two [<1%], adjusted OR 5·58 [1·22–25·50]; p=0·024). Interpretation: In patients who have had a stroke with unknown time of onset with a DWI-FLAIR or perfusion mismatch, intravenous alteplase resulted in better functional outcome at 90 days than placebo or standard care. A net benefit was observed for all functional outcomes despite an increased risk of symptomatic intracranial haemorrhage. Although there were more deaths with alteplase than placebo, there were fewer cases of severe disability or death. Funding: None
Constraints on jet quenching in p-Pb collisions at root s(NN)=5.02 TeV measured by the event-activity dependence of semi-inclusive hadron-jet distributions

Jets are reconstructed from charged-particle tracks using the anti-k(T) algorithm with resolution parameter R = 0.2 and 0.4. A data-driven statistical approach is used to correct the uncorrelated background jet yield. Recoil jet distributions are reported for jet transverse momentum 15 < p(T,jet)(ch) < 50 GeV/c and are compared in various intervals of p-Pb event activity, based on charged-particle multiplicity and zero-degree neutral energy in the forward (Pb-going) direction. The semi-inclusive observable is self-normalized and such comparisons do not require the interpretation of p-Pb event activity in terms of collision geometry, in contrast to inclusive jet observables. These measurements provide new constraints on the magnitude of jet quenching in small systems at the LHC. In p-Pb collisions with high event activity, the average medium-induced out-of-cone energy transport for jets with R = 0.4 and 15 < p(T,jet)(ch) < 50 GeV/c is measured to be less than 0.4 GeV/c at 90% confidence, which is over an order of magnitude smaller than a similar measurement for central Pb-Pb collisions at root s(NN) = 2.76 TeV.
