    A kinematic algorithm to identify gait events during running at different speeds and with different footstrike types

    Although a number of algorithms exist for estimating ground contact events (GCEs) from kinematic data during running, they are typically only applicable to heelstrike running, or have only been evaluated at a single running speed. The purpose of this study was to investigate the accuracy of four kinematics-based algorithms to estimate GCEs over a range of running speeds and footstrike types. Subjects ran over a force platform at a range of speeds; kinetic and kinematic data was captured at 1000 Hz, and kinematic data was downsampled to 250 Hz. A windowing process initially identified reduced time windows containing touchdown and toe-off. Algorithms based on acceleration and jerk signals of the foot markers were used to estimate touchdown (2 algorithms), toe-off (2 algorithms), and ground contact time (GCT) (4 algorithms), and compared to synchronous ‘gold standard’ force platform data. An algorithm utilising the vertical acceleration peak of either the heel or first metatarsal marker (whichever appeared first) for touchdown, and the vertical jerk peak of the hallux marker for toe-off, resulted in the lowest offsets (+3.1 ms, 95% Confidence Interval (CI): -11.8 to +18.1 ms; and +2.1 ms, CI: -8.1 to +12.2 ms respectively). This method also resulted in the smallest offset in GCT (-1.1 ms, CI: -18.6 to +16.4 ms). Offsets in GCE and GCT estimates from all algorithms were typically negatively correlated to running speed, with offsets decreasing as speed increased. Assessing GCEs and GCT using this method may be useful when a force platform is unavailable or impractical

PURPOSE: To determine the relationship between individual and combined kinematic measures of technique with both RE and performance. METHODS: Ninety-seven endurance runners (47 female) of diverse competitive standards performed a discontinuous protocol of incremental treadmill running (4 min stages, 1 km.h increments).     The influence of running speed and sex on running economy is unclear and may have been confounded by measurements of oxygen cost that do not account for known differences in substrate metabolism, across a limited range of speeds, and differences in performance standard. Therefore, this study assessed the energy cost of running over a wide range of speeds in high-level and recreational runners to investigate the effect of speed (considered in absolute and relative terms) and sex (males vs. females of equivalent performance standard) on running economy. 92 healthy runners (high-level males, n=14; high-level females, n=10; recreational males, n=35; recreational females, n=33) completed a discontinuous incremental treadmill test for the determination of the energy cost (kcal·kg 1·km-1) of submaximal running, speed at lactate turnpoint (sLTP) and the maximal rate of oxygen uptake (V̇O2max). There were no sex specific differences in the energy cost of running for the recreational or high-level runners when compared at absolute or relative running speeds (P>0.05). The absolute and relative speed-energy cost relationships for the high-level runners demonstrated a curvilinear inverted “u shape” with a nadir reflecting the most economical speed at 13 km.h-1 or 70% sLTP. The high-level runners were more economical than the recreational runners at all absolute and relative running speeds (P<0.05). These findings demonstrate that there is an optimal speed for economical running; there is no sex-specific difference; and, high-level endurance runners exhibit a better running economy than recreational endurance runners

    From Aristotle to Arendt : a phenomenological exploration of forms of knowledge and practice in the context of child protection social work in the UK

    This paper attempts to explore the relationship between different forms of knowledge and the kinds of activity that arise from them within child protection social work practice. The argument that social work is more than either ‘science’ or ‘art’ but distinctly ‘practice’ is put through a historical description of the development of Aristotle’s views of the forms of knowledge and Hannah Arendt’s later conceptualisations as detailed in The Human Condition (1958). The paper supports Arendt’s privileging of Praxis over Theoria within social work and further draws upon Arendt’s distinctions between Labour, Work and Action to delineate between different forms of social work activity. The author highlights dangers in social work relying too heavily on technical knowledge and the use of theory as a tool in seeking to understand and engage with the people it serves and stresses the importance of a phenomenological approach to research and practice as a valid, embodied form of knowledge. The argument further explores the constructions of service users that potentially arise from different forms of social work activity and cautions against over-prescriptive use of ‘outcomes’ based practice that may reduce the people who use services to products or consumables. The author concludes that social work action inevitably involves trying to understand humans in a complex and dynamic way that requires engagement and to seek new meanings for individual humans

    The ubiquity of frequency effects in first language acquisition

    This review article presents evidence for the claim that frequency effects are pervasive in children's first language acquisition, and hence constitute a phenomenon that any successful account must explain. The article is organized around four key domains of research: children's acquisition of single words, inflectional morphology, simple syntactic constructions, and more advanced constructions. In presenting this evidence, we develop five theses. (i) There exist different types of frequency effect, from effects at the level of concrete lexical strings to effects at the level of abstract cues to thematic-role assignment, as well as effects of both token and type, and absolute and relative, frequency. High-frequency forms are (ii) early acquired and (iii) prevent errors in contexts where they are the target, but also (iv) cause errors in contexts in which a competing lower-frequency form is the target. (v) Frequency effects interact with other factors (e.g. serial position, utterance length), and the patterning of these interactions is generally informative with regard to the nature of the learning mechanism. We conclude by arguing that any successful account of language acquisition, from whatever theoretical standpoint, must be frequency sensitive to the extent that it can explain the effects documented in this review, and outline some types of account that do and do not meet this criterion

    Identification of Homogentisate Dioxygenase as a Target for Vitamin E Biofortification in Oilseeds

    Soybean (Glycine max) is a major plant source of protein and oil and produces important secondary metabolites beneficial for human health. As a tool for gene function discovery and improvement of this important crop, a mutant population was generated using fast neutron irradiation. Visual screening of mutagenized seeds identified a mutant line, designated MO12, which produced brown seeds as opposed to the yellow seeds produced by the unmodified Williams 82 parental cultivar. Using forward genetic methods combined with comparative genome hybridization analysis, we were able to establish that deletion of the GmHGO1 gene is the genetic basis of the brown seeded phenotype exhibited by the MO12 mutant line. GmHGO1 encodes a homogentisate dioxygenase (HGO), which catalyzes the committed enzymatic step in homogentisate catabolism. This report describes to our knowledge the first functional characterization of a plant HGO gene, defects of which are linked to the human genetic disease alkaptonuria. We show that reduced homogentisate catabolism in a soybean HGO mutant is an effective strategy for enhancing the production of lipid-soluble antioxidants such as vitamin E, as well as tolerance to herbicides that target pathways associated with homogentisate metabolism. Furthermore, this work demonstrates the utility of fast neutron mutagenesis in identifying novel genes that contribute to soybean agronomic traits

    Genetic associations at 53 loci highlight cell types and biological pathways relevant for kidney function.

    Reduced glomerular filtration rate defines chronic kidney disease and is associated with cardiovascular and all-cause mortality. We conducted a meta-analysis of genome-wide association studies for estimated glomerular filtration rate (eGFR), combining data across 133,413 individuals with replication in up to 42,166 individuals. We identify 24 new and confirm 29 previously identified loci. Of these 53 loci, 19 associate with eGFR among individuals with diabetes. Using bioinformatics, we show that identified genes at eGFR loci are enriched for expression in kidney tissues and in pathways relevant for kidney development and transmembrane transporter activity, kidney structure, and regulation of glucose metabolism. Chromatin state mapping and DNase I hypersensitivity analyses across adult tissues demonstrate preferential mapping of associated variants to regulatory regions in kidney but not extra-renal tissues. These findings suggest that genetic determinants of eGFR are mediated largely through direct effects within the kidney and highlight important cell types and biological pathways

    Surface Reconstruction of Maltese Cisterns Using ROV Sonar Data for Archeological Study

    We present a methodology and algorithm for the reconstruction of three dimensional geometric models of ancient Maltese water storage systems, i.e. cisterns, from sonar data. This project was conducted as a part of a four week expedition on the islands of Malta and Gozo. During this expedition, investigators used underwater robot systems capable of mapping ancient underwater cisterns and tunnels. The mapping included probabilistic algorithms for constructing the maps of the sonar data and computer graphics for surface reconstruction and visualization. This paper presents the general methodology for the data acquisition and the novel application of algorithms from computer graphics for surface reconstruction to this new data setting. In addition to reconstructing the geometry of the cisterns, the visualization system includes methods to enhance the understanding of the data by visualizing water level and texture detail either through the application of real image data via projective textures or by more standard texture mapping techniques. The resulting surface reconstructions and visualizations can be used by archaeologists for educational purposes and to help understand the shape and history of such water receptacles
