    Congenital Lipoid Adrenal Hyperplasia: Functional Characterization of Three Novel Mutations in the STAR Gene

    AbstractContext: The steroidogenic acute regulatory protein (StAR) has been shown to be essential for steroidogenesis by mediating cholesterol transfer into mitochondria. Inactivating StAR mutations cause the typical clinical picture of congenital lipoid adrenal hyperplasia.Objective: The objective of the investigation was to study the functional and structural consequences of three novel StAR mutations (p.N148K in an Italian patient; p.P129fs and p.Q128R in a Turkish patient).Methods and Results: Transient in vitro expression of the mutant proteins together with P450 side-chain cleavage enzyme, adrenodoxin, and adrenodoxin reductase yielded severely diminished cholesterol conversion of the p.N148K mutant, the combined p.P129fs and p.Q128R mutant, and the p.P129fs mutant by itself. The p.Q128R mutant led to a higher cholesterol conversion than the wild-type StAR protein. As derived from three-dimensional protein modeling, the residue N148 is lining the ligand cavity of StAR. A positively charged lysine residue at position 148 disturbs the hydrophobic cluster formed by the α4-helix and the sterol binding pocket. The frame shift mutation p.P129fs truncates the StAR protein. Residue p.Q128 is situated at the surface of the molecule and is not part of any functionally characterized region of the protein.Conclusion: The mutations p.N148K and p.P129fs cause adrenal insufficiency in both cases and lead to a disorder of sex development with complete sex reversal in the 46, XY case. The mutation p.Q128R, which is not relevant for the patient's phenotype, is the first reported variant showing a gain of function. We speculate that the substitution of hydrophilic glutamine with basic arginine at the surface of the molecule may accelerate cholesterol transfer

As distance education becomes important everyday, the indispensable elements of teaching and education, laboratories must be reachable via remote connection. Consequently, the education that is going to be given to the students will be more flexible with respect to place and time constraints and students can reach laboratory facilities at any time and anywhere not only in lectures and practical hours. In this study, European Remote Radio Laboratory (ERRL) which is a distance remote Radio Frequency (RF) laboratory designed for electrical-electronics students, is described generally.     Turner syndrome and associated problems in turkish children: A multicenter study

    Objective: Turner syndrome (TS) is a chromosomal disorder caused by complete or partial X chromosome monosomy that manifests various clinical features depending on the karyotype and on the genetic background of affected girls. This study aimed to systematically investigate the key clinical features of TS in relationship to karyotype in a large pediatric Turkish patient population. Methods: Our retrospective study included 842 karyotype-proven TS patients aged 0-18 years who were evaluated in 35 different centers in Turkey in the years 2013-2014. Results: The most common karyotype was 45,X (50.7%), followed by 45,X/46,XX (10.8%), 46,X,i(Xq) (10.1%) and 45,X/46,X,i(Xq) (9.5%). Mean age at diagnosis was 10.2±4.4 years. The most common presenting complaints were short stature and delayed puberty. Among patients diagnosed before age one year, the ratio of karyotype 45,X was significantly higher than that of other karyotype groups. Cardiac defects (bicuspid aortic valve, coarctation of the aorta and aortic stenosi) were the most common congenital anomalies, occurring in 25% of the TS cases. This was followed by urinary system anomalies (horseshoe kidney, double collector duct system and renal rotation) detected in 16.3%. Hashimoto’s thyroiditis was found in 11.1% of patients, gastrointestinal abnormalities in 8.9%, ear nose and throat problems in 22.6%, dermatologic problems in 21.8% and osteoporosis in 15.3%. Learning difficulties and/or psychosocial problems were encountered in 39.1%. Insulin resistance and impaired fasting glucose were detected in 3.4% and 2.2%, respectively. Dyslipidemia prevalence was 11.4%. Conclusion: This comprehensive study systematically evaluated the largest group of karyotype-proven TS girls to date. The karyotype distribution, congenital anomaly and comorbidity profile closely parallel that from other countries and support the need for close medical surveillance of these complex patients throughout their lifespan. © Journal of Clinical Research in Pediatric Endocrinology

    SET-NUP214 fusion in acute myeloid leukemia- and T-cell acute lymphoblastic leukemia-derived cell lines

    <p>Abstract</p> <p>Background</p> <p><it>SET-NUP214 </it>fusion resulting from a recurrent cryptic deletion, del(9)(q34.11q34.13) has recently been described in T-cell acute lymphoblastic leukemia (T-ALL) and in one case of acute myeloid leukemia (AML). The fusion protein appears to promote elevated expression of <it>HOXA </it>cluster genes in T-ALL and may contribute to the pathogenesis of the disease. We screened a panel of ALL and AML cell lines for <it>SET-NUP214 </it>expression to find model systems that might help to elucidate the cellular function of this fusion gene.</p> <p>Results</p> <p>Of 141 human leukemia/lymphoma cell lines tested, only the T-ALL cell line LOUCY and the AML cell line MEGAL expressed the <it>SET(TAF-</it>Iβ)-<it>NUP214 </it>fusion gene transcript. RT-PCR analysis specifically recognizing the alternative first exons of the two <it>TAF-</it>I isoforms revealed that the cell lines also expressed <it>TAF-</it>Iα-<it>NUP214 </it>mRNA. Results of fluorescence in situ hybridization (FISH) and array-based copy number analysis were both consistent with del(9)(q34.11q34.13) as described. Quantitative genomic PCR also confirmed loss of genomic material between <it>SET </it>and <it>NUP214 </it>in both cell lines. Genomic sequencing localized the breakpoints of the <it>SET </it>gene to regions downstream of the stop codon and to <it>NUP214 </it>intron 17/18 in both LOUCY and MEGAL cells. Both cell lines expressed the 140 kDa SET-NUP214 fusion protein.</p> <p>Conclusion</p> <p>Cell lines LOUCY and MEGAL express the recently described <it>SET-NUP214 </it>fusion gene. Of special note is that the formation of the <it>SET </it>exon 7/<it>NUP214 </it>exon 18 gene transcript requires alternative splicing as the <it>SET </it>breakpoint is located downstream of the stop codon in exon 8. The cell lines are promising model systems for <it>SET-NUP214 </it>studies and should facilitate investigating cellular functions of the the SET-NUP214 protein.</p

    Osmia (Ozbekosmia) avosetta.

Includes bibliographical references (p. 21-22).Herein we describe the nests (including construction, closure, orientation, and depth of cells) of the bee Osmia (Ozbekosmia) avosetta Warncke found nesting near Antalya, Turkey, and Sepidan, Iran. Cells are unusual in that they are lined by two layers of colorful flower petals that sandwich a thin middle layer of mud. Analyses of pollen taken from scopal hairs of specimens from the Turkish site were identified as solely from Onobrychis viciifolia Scop. (Fabaceae) whereas those from the Iranian site were from a related plant, Hedysarum elymaiticum Boiss. and Hausskn. These facts coupled with analyses of scopal pollen from 11 other sites in Turkey, Jordan, and Syria strongly suggest that this bee is oligolectic with respect to the plant tribe Hedysareae. The egg and last larval instar of Osmia avosetta are described. The presence of an egg taken from a cell and provisionally identified as belonging to Sapyga pulcherrima Morawitz suggests that this cleptoparasite may have this bee as one of its hosts. In addition, we report new information on and review published accounts concerning the use of whole petals or large petal pieces in the construction of cell walls of osmiine bees. Only Osmia (Ozbekosmia) avosetta and species of Osmia (Tergosmia) have three-layered cell walls with the middle layer made of mud. Recorded also are the similarities and differences exhibited in pollen and petal preferences and nest characteristics of species in these two related subgenera

    Search for New Physics with Jets and Missing Transverse Momentum in pp collisions at sqrt(s) = 7 TeV

    A search for new physics is presented based on an event signature of at least three jets accompanied by large missing transverse momentum, using a data sample corresponding to an integrated luminosity of 36 inverse picobarns collected in proton--proton collisions at sqrt(s)=7 TeV with the CMS detector at the LHC. No excess of events is observed above the expected standard model backgrounds, which are all estimated from the data. Exclusion limits are presented for the constrained minimal supersymmetric extension of the standard model. Cross section limits are also presented using simplified models with new particles decaying to an undetected particle and one or two jets

    Using Barkhausen Noise to Measure Coating Depth of Coated High-Speed Steel

    Coated high-speed steel tools are widely used in machining processes as they offer an excellent tool life to cost ratio, but they quickly need replacing once the coated layer is worn away. It would be therefore useful to be able to measure the tool life remaining non-destructively and cheaply. To achieve this, the work presented here aims to measure the thickness of the coated layer of high-speed cutting tools by using Barkhausen noise (BHN) techniques. Coated high-speed steel specimens coated with two different materials (chromium nitride (CrN), titanium nitride (TiN)) were tested using a cost-effective measuring system developed for this study. Sensory features were extracted from the signal received from a pick-up coil and the signal features, Root mean square, peak count, and signal energy, were successfully correlated with the thickness of the coating layer on high-speed steel (HSS) specimens. The results suggest that the Barkhausen noise measuring system developed in this study can successfully indicate the different thickness of the coating layer on CrN/TiN coated HSS specimens

    Carrageenan-based hydrogels for the controlled delivery of PDGF-BB in bone tissue engineering applications

    One of the major drawbacks found in most bone tissue engineering approaches developed so far consists in the lack of strategies to promote vascularisation. Some studies have addressed different issues that may enhance vascularisation in tissue engineered constructs, most of them involving the use of growth factors (GFs) that are involved in the restitution of the vascularity in a damaged zone. The use of sustained delivery systems might also play an important role in the re-establishment of angiogenesis. In this study, !-carrageenan, a naturally occurring polymer, was used to develop hydrogel beads with the ability to incorporate GFs with the purpose of establishing an effective angiogenesis mechanism. Some processing parameters were studied and their influence on the final bead properties was evaluated. Platelet derived growth factor (PDGF-BB) was selected as the angiogenic factor to incorporate in the developed beads, and the results demonstrate the achievement of an efficient encapsulation and controlled release profile matching those usually required for the development of a fully functional vascular network.
