    Integrating human endogenous retroviruses into transcriptome-wide association studies highlights novel risk factors for major psychiatric conditions

    Human endogenous retroviruses (HERVs) are repetitive elements previously implicated in major psychiatric conditions, but their role in aetiology remains unclear. Here, we perform specialised transcriptome-wide association studies that consider HERV expression quantified to precise genomic locations, using RNA sequencing and genetic data from 792 post-mortem brain samples. In Europeans, we identify 1238 HERVs with expression regulated in cis, of which 26 represent expression signals associated with psychiatric disorders, with ten being conditionally independent from neighbouring expression signals. Of these, five are additionally significant in fine-mapping analyses and thus are considered high confidence risk HERVs. These include two HERV expression signatures specific to schizophrenia risk, one shared between schizophrenia and bipolar disorder, and one specific to major depressive disorder. No robust signatures are identified for autism spectrum conditions or attention deficit hyperactivity disorder in Europeans, or for any psychiatric trait in other ancestries, although this is likely a result of relatively limited statistical power. Ultimately, our study highlights extensive HERV expression and regulation in the adult cortex, including in association with psychiatric disorder risk, therefore providing a rationale for exploring neurological HERV expression in complex neuropsychiatric traits.</p

    Global connectivity of the fronto-parietal cognitive control network is related to depression symptoms in the general population

    We all vary in our mental health, even among people not meeting diagnostic criteria for mental illness. Understanding this individual variability may reveal factors driving the risk for mental illness, as well as factors driving subclinical problems that still adversely affect quality of life. To better understand the large-scale brain network mechanisms underlying this variability, we examined the relationship between mental health symptoms and resting-state functional connectivity patterns in cognitive control systems. One such system is the fronto-parietal cognitive control network (FPN). Changes in FPN connectivity may impact mental health by disrupting the ability to regulate symptoms in a goal-directed manner. Here we test the hypothesis that FPN dysconnectivity relates to mental health symptoms even among individuals who do not meet formal diagnostic criteria but may exhibit meaningful symptom variation. We found that depression symptoms severity negatively correlated with between-network global connectivity (BGC) of the FPN. This suggests that decreased connectivity between the FPN and the rest of the brain is related to increased depression symptoms in the general population. These findings complement previous clinical studies to support the hypothesis that global FPN connectivity contributes to the regulation of mental health symptoms across both health and disease. Understanding how large-scale network interactions in the brain contribute to (or serve a protective role against) mental health symptoms is an important step toward developing more effective mental health treatments. Here we test the hypothesis that cognitive control networks play an important role in mental health by being highly connected to other brain networks and able to serve as a feedback mechanism capable of regulating symptoms in a goal-directed manner. We found that the more well connected the fronto-parietal cognitive control network was to other networks in the brain the less depression symptoms were reported by participants. These results contribute to our understanding of how brain network interactions are related to mental health symptoms, even in individuals who have not been diagnosed with a disorder

    Suspicious binds: Conspiracy thinking and tenuous perceptions of causal connections between co?occurring and spuriously correlated events

    Previous research indicates that conspiracy thinking is informed by the psychological imposition of order and meaning on the environment, including the perception of causal relations between random events. Four studies indicate that conspiracy belief is driven by readiness to draw implausible causal connections even when events are not random, but instead conform to an objective pattern. Study 1 (N = 195) showed that conspiracy belief was related to the causal interpretation of real?life, spurious correlations (e.g., between chocolate consumption and Nobel prizes). In Study 2 (N = 216), this effect held adjusting for correlates including magical and non?analytical thinking. Study 3 (N = 214) showed that preference for conspiracy explanations was associated with the perception that a focal event (e.g., the death of a journalist) was causally connected to similar, recent events. Study 4 (N = 211) showed that conspiracy explanations for human tragedies were favoured when they comprised part of a cluster of similar events (vs. occurring in isolation); crucially, they were independently increased by a manipulation of causal perception. We discuss the implications of these findings for previous, mixed findings in the literature and for the relation between conspiracy thinking and other cognitive processes

    The moral impact of studying science

    Science and religion are most usually compared on epistemic grounds: what do they tell us about the natural world and what methods do they use to determine those truths? The suggestion here is that the two fields should be compared on moral grounds: how do scientific and religious experiences affect the way a person lives his or her life? A hypothesis is presented in this vein: engaging in scientific work or education alters a person’s moral outlook on everyday matters. In this chapter, I articulate and motivate this claim by framing it against both theological and philosophical debate. I explore how it might be tested as a claim in moral psychology. The resulting vision presented here is of science and religion engaged in dialogue—at times necessarily embroiled—not only about the nature of the world, but regarding how best we navigate our way in it

    A Cell/Cilia Cycle Biosensor for Single-Cell Kinetics Reveals Persistence of Cilia after G1/S Transition Is a General Property in Cells and Mice

    The cilia and cell cycles are inextricably linked. Centrioles in the basal body of cilia nucleate the ciliary axoneme and sequester pericentriolar matrix (PCM) at the centrosome to organize the mitotic spindle. Cilia themselves respond to growth signals, prompting cilia resorption and cell cycle re-entry. We describe a fluorescent cilia and cell cycle biosensor allowing live imaging of cell cycle progression and cilia assembly and disassembly kinetics in cells and inducible mice. We define assembly and disassembly in relation to cell cycle stage with single-cell resolution and explore the intercellular heterogeneity in cilia kinetics. In all cells and tissues analyzed, we observed cilia that persist through the G1/S transition and into S/G2/M-phase. We conclude that persistence of cilia after the G1/S transition is a general property. This resource will shed light at an individual cell level on the interplay between the cilia and cell cycles in development, regeneration, and disease. The cilia and cell cycles are fundamental processes coupled through shared machinery.     Understanding the rift, the (still) uneasy bedfellows of History and Organization Studies

    Although the use of History has become increasingly discussed and more widely applied within Organization Studies (OS), its relevance for OS still remains far from centrally accepted. This article historicizes the relationship between Sociology and History as a means of better understanding the tensions, perceived and real, that exist between History and Organization Studies. In particular we analyse three differences of epistemological standpoint (method, objectivity and usefulness) that are commonly seen as the foundation stones to incompatibility. Perhaps surprisingly for an analysis of apparent disciplinary differences, we find that these distinctions in terms of approach, once closely examined, are rarely clear-cut and historians and OS scholars are frequently closer in intention and method than they are distant. However, despite their large intersection of interests, we argue that important distinctions between the two fields should be acknowledged. Our contribution to the debates over the need for more historical approaches within OS therefore centrally rests on abandoning aspirations for fully integrative models of working together, in favour of cooperative modes that concede the fields’ differences. This subtle shift of emphasis will, we believe, greatly benefit OS scholars who hope to include historical perspectives in their work
