    United we stand? - Survey results on the views of French, German and Italian parliamentarians on EU and EMU reforms

    Der europäische Integrationsprozess wird gleich von mehreren Krisen erschüttert – der Euro-Schuldenkrise, der Flüchtlingsfrage und dem Brexit-Konflikt –, was eine umfassende Reform-Debatte ausgelöst hat. Bei dieser Debatte geht es sowohl um die Entwicklung der Institutionen der Europäischen Währungsunion (WWU), die Aufteilung von Kompetenzen zwischen der EU und den Mitgliedstaaten als auch um Reformen bezüglich der Entscheidungsfindung und der Finanzierung innerhalb der EU. Obwohl es zu all diesen Themen eine Fülle innovativer Ideen gibt, sind die Hürden für weitreichende Reformen dennoch hoch, da sie einen Konsens unter allen Veto-Mächten erfordern. Dieser Policy Brief fasst die Erkenntnisse aus einer kürzlich durchgeführten einzigartigen Umfrage zusammen, die unter den nationalen Parlamenten der drei größten Mitgliedstaaten der Post-Brexit-EU vorgenommen wurde. Jede weitreichende WWU- oder EU-Reform muss von den nationalen Parlamenten der Mitgliedsstaaten genehmigt werden. Ein Konsens unter diesen drei Ländern ist daher eine notwendige Bedingung für die politische Umsetzbarkeit einer solchen Reform. Die Umfrage zur Zukunft der europäischen Integration wurde in den nationalen Parlamenten in Frankreich, Deutschland und Italien durchgeführt, darunter in der französischen Nationalversammlung und im Sénat, im Deutschen Bundestag sowie in der italienischen Camera dei deputati und dem Senato della Repubblica. Die Umfrage lief von September 2018 bis Januar 2019.     A Spitzer search for cold dust within globular clusters

    Globular cluster stars evolving off the main sequence are known to lose mass, and it is expected that some of the lost material should remain within the cluster as an intracluster medium (ICM). Most attempts to detect such an ICM have been unsuccessful. The Multiband Imaging Photometer for Spitzer on the Spitzer Space Telescope was used to observe eight Galactic globular clusters in an attempt to detect the thermal emission from ICM dust. Most clusters do not have significant detections at 70 microns; one cluster, NGC 6341, has tentative evidence for the presence of dust, but 90 micron observations do not confirm the detection. Individual 70 micron point sources which appear in several of the cluster images are likely to be background galaxies. The inferred dust mass and upper limits are < 4e-4 solar masses, well below expectations for cluster dust production from mass loss in red and asymptotic giant branch stars. This implies that either globular cluster dust production is less efficient, or that ICM removal or dust destruction is more efficient, than previously believed.     A Spitzer Space Telescope far-infrared spectral atlas of compact sources in the Magellanic Clouds. I. The Large Magellanic Cloud

    [abridged] We present 52-93 micron spectra obtained with Spitzer in the MIPS-SED mode, of a representative sample of luminous compact far-IR sources in the LMC. These include carbon stars, OH/IR AGB stars, post-AGB objects and PNe, RCrB-type star HV2671, OH/IR red supergiants WOHG064 and IRAS05280-6910, B[e] stars IRAS04530-6916, R66 and R126, Wolf-Rayet star Brey3a, Luminous Blue Variable R71, supernova remnant N49, a large number of young stellar objects, compact HII regions and molecular cores, and a background galaxy (z~0.175). We use the spectra to constrain the presence and temperature of cold dust and the excitation conditions and shocks within the neutral and ionized gas, in the circumstellar environments and interfaces with the surrounding ISM. Evolved stars, including LBV R71, lack cold dust except in some cases where we argue that this is swept-up ISM. This leads to an estimate of the duration of the prolific dust-producing phase ("superwind") of several thousand years for both RSGs and massive AGB stars, with a similar fractional mass loss experienced despite the different masses. We tentatively detect line emission from neutral oxygen in the extreme RSG WOHG064, with implications for the wind driving. In N49, the shock between the supernova ejecta and ISM is revealed by its strong [OI] 63-micron emission and possibly water vapour; we estimate that 0.2 Msun of ISM dust was swept up. Some of the compact HII regions display pronounced [OIII] 88-micron emission. The efficiency of photo-electric heating in the interfaces of ionized gas and molecular clouds is estimated at 0.1-0.3%. We confirm earlier indications of a low nitrogen content in the LMC. Evidence for solid state emission features is found in both young and evolved object; some of the YSOs are found to contain crystalline water ice.Comment: Accepted for publication in The Astronomical Journal.     Genetic association study of QT interval highlights role for calcium signaling pathways in myocardial repolarization.

    The QT interval, an electrocardiographic measure reflecting myocardial repolarization, is a heritable trait. QT prolongation is a risk factor for ventricular arrhythmias and sudden cardiac death (SCD) and could indicate the presence of the potentially lethal mendelian long-QT syndrome (LQTS). Using a genome-wide association and replication study in up to 100,000 individuals, we identified 35 common variant loci associated with QT interval that collectively explain ∼8-10% of QT-interval variation and highlight the importance of calcium regulation in myocardial repolarization. Rare variant analysis of 6 new QT interval-associated loci in 298 unrelated probands with LQTS identified coding variants not found in controls but of uncertain causality and therefore requiring validation. Several newly identified loci encode proteins that physically interact with other recognized repolarization proteins. Our integration of common variant association, expression and orthogonal protein-protein interaction screens provides new insights into cardiac electrophysiology and identifies new candidate genes for ventricular arrhythmias, LQTS and SCD

    Fast relapse and high drop out rate of 48 weeks daily interferon monotherapy in HIV-infected patients with chronic hepatitis C

    BACKGROUND: The standard of care for HCV Hepatitis is the combination of interferon (IFN) plus Ribavirin. In HIV patients the use of this combination therapy may induce drug interactions, and reduces the adherence to HAART. The aim of this study is to evaluate safety and efficacy of a 48 weeks daily dose IFN schedule. METHODS: We evaluated 50 coinfected patients; alpha IFN 2a was administered at a dose of 3 MU daily. The baseline values were the following : CD4+ 515 cells/mmc (mean); HIV-RNA <50 copies/ml in all patients; HCV-RNA 28, 3 × 106 copies/ml. RESULTS: At 48 weeks, 10 patients (20%) achieved a biochemical and virological response according to an intention to treat analysis. Twenty four patients (48%) underwent a drop-out mainly by side effects related to overlapping toxicity of interferon and antiretroviral therapy. All the patients, who responded to the treatment, showed a fast relapse one month after the end of treatment. CONCLUSION: Although our results demonstrated a very poor outcome and a bad tolerance to interferon monotherapy, this approach should not be dropped out, mainly in patients at high risk for side effects and in those with cirrhosis who do not tolerate or are at increased risk for the use of ribavirin

    Safety and Immunogenicity of an HIV-1 Gag DNA Vaccine with or without IL-12 and/or IL-15 Plasmid Cytokine Adjuvant in Healthy, HIV-1 Uninfected Adults

    DNA vaccines are a promising approach to vaccination since they circumvent the problem of vector-induced immunity. DNA plasmid cytokine adjuvants have been shown to augment immune responses in small animals and in macaques.We performed two first in human HIV vaccine trials in the US, Brazil and Thailand of an RNA-optimized truncated HIV-1 gag gene (p37) DNA derived from strain HXB2 administered either alone or in combination with dose-escalation of IL-12 or IL-15 plasmid cytokine adjuvants. Vaccinations with both the HIV immunogen and cytokine adjuvant were generally well-tolerated and no significant vaccine-related adverse events were identified. A small number of subjects developed asymptomatic low titer antibodies to IL-12 or IL-15. Cellular immunogenicity following 3 and 4 vaccinations was poor, with response rates to gag of 4.9%/8.7% among vaccinees receiving gag DNA alone, 0%/11.5% among those receiving gag DNA+IL-15, and no responders among those receiving DNA+high dose (1500 ug) IL-12 DNA. However, after three doses, 44.4% (4/9) of vaccinees receiving gag DNA and intermediate dose (500 ug) of IL-12 DNA demonstrated a detectable cellular immune response.This combination of HIV gag DNA with plasmid cytokine adjuvants was well tolerated. There were minimal responses to HIV gag DNA alone, and no apparent augmentation with either IL-12 or IL-15 plasmid cytokine adjuvants.
