    Suckling, male effect and kisspeptin in the reproductive management of ewes in postpartum anestrus

    Objective: To describe the management strategies of controlled suckling and the male effect to reduce postpartum anestrus, and to show the participation of kisspeptin to regulate the effect of both factors. Design/Methodology/Approach: A review of scientific publications was conducted, in order to show the importance of suckling and the male effect as strategies to reduce postpartum anestrus in the ewe, as well as the relation of kisspeptin with both factors. Results: Seasonal anestrus can be avoided with the use of breeds adapted to the local environment, such as Pelibuey. Postpartum anestrus occurs mainly as a result of suckling, since the latter inhibits the pulsating secretion of the gonadotropin-releasing hormone (GnRH) and the luteinizing hormone (LH). The exact path of this inhibition is unknown, although it seems that endogenous opioid peptides and kisspeptin are intermediaries. Controlled suckling and the male effect are management strategies that improve the reproductive behavior of postpartum ewes. Kisspeptin regulates the influence of the male effect through the secretion of GnRH/LH. Study Limitations/Implications: To understand the impacts of suckling and the male effect on the duration of postpartum anestrus, as well as the participation of kisspeptin in the regulation of both effects, will allow designing management strategies to improve the reproductive efficiency of the ewes. Findings/Conclusions: Controlled suckling and the male effect reduce postpartum anestrus and improve the reproductive behavior of the ewes; advancing knowledge of the kisspeptin effect could improve the effectiveness of both techniques

    Niveles de fósforo disponible aparente en pollas Leghorn blanca durante las etapas de iniciación, crecimiento, desarrollo e inicio de postura

    Phosphorus (P) is important for the growth and metabolism of pullets and young laying hens, although the requeriments of this mineral could be overvalued. An experiment was conducted with 750 Hy-line W-36 pullets to evaluate the effects of apparent available phosphorus (aaP) levels during starter (1 d to 6 wk), growing (7 to 12 wk), development (13 to 18 wk), and initial laying period. The analyzed variables were: body weight (BW), feed intake (FI), percentage of P in excreta (PPE) and tibia bone (TBP), tibia breaking strength (TBS), egg mass (EM), and specific gravity (SG). The evaluated aaP levels had no effect (P>0.05) on FI, FP, TBP, RB, or EM in the stages under study. There was a difference (P0.05) in the starter period, the optimum biological levels for growing and finishing stages were 0.28 and 0.26 % of aaP, respectively. It was concluded that the requirements of apparent available phosphorus could be lower than the value suggested by NRC (1994) for the growth stages and the variables measured, without affecting the body condition and bone mineralization of pullets.El fósforo es importante en el metabolismo y desarrollo de las pollitas de reemplazo, aunque las necesidades de este mineral pueden estar sobrevaloradas. Se realizó un experimento con 750 pollas de la lí­nea Hy-line W-36, para evaluar los efectos de niveles de fósforo disponible aparente (Pda) durante las etapas de iniciación (1 dí­a a 6 semanas), crecimiento (7 a 12 semanas), desarrollo (13 a 18 semanas) y de inicio a pico de postura. Las caracterí­sticas estudiadas fueron: peso vivo (PV), consumo de alimento (CA), porcentaje de fósforo total en excretas y tibias (PTE y PT), resistencia a ruptura de tibias (RR), masa de huevo (MH) y gravedad especí­fica (GE). El nivel de Pda no afectó (P>0.05) el CA, PTE, PT, RR y MH en las etapas estudiadas. Hubo diferencias (P0.05), mientras que los niveles óptimos biológicos para PV en crecimiento fueron con 0.28 % y desarrollo 0.26 %. Se concluye que las necesidades de fósforo disponible aparente pueden ser menores a lo sugerido por el NRC (1994), durante las etapas y caracterí­sticas medidas, sin afectar la condición corporal y mineralización ósea de las aves

    Comparison of two phenotypical methods to segregate resistant and susceptible lambs to parasitic nematodes

    The objective of this study was to compare two segregation methods to select resistant and susceptible female Pelibuey lambs infected naturally with gastrointestinal nematodes (GINs) in relation to their haematological and immunological response. For 6 months, faeces and blood samples were taken fortnightly from 40 grazing 5-month-old female lambs. The lambs were classified according to two methods using faecal egg count (FEC) as a phenotypical trait. In the first (reference) method (M3SE, n = 22), resistant (RES) lambs had FEC lower than the mean – 3 standard errors, the susceptible (SUS) lambs levels higher than + 3 standard errors(n = 10) and the intermediate (INT) lambs (n = 8) were categorised by having FECs between the two values. The second method (QUM) divided the population, using quartiles, into resistant (RES; 25%), intermediate (INT; 50%), and susceptible (SUS; 25%) lambs. The agreement between both methods was estimated using the Kappa index. The packed cell volume (PCV), total plasma protein (TPP) and peripheral eosinophils (EOS) were determined for each group. Serum was used to evaluate the IgA levels. PCV and TPP values were higher (P<0.01) in the RES lambs (31.5 ± 3.4 and 6.16 ± 0.5 g/dL by QUM, respectively, and 31.5 ± 3.9 and 6.24 ± 0.49 g/dL by M3SE, respectively) than the SUS lambs (28.1 ± 4.7 and 5.94 ± 0.5 g/d, respectively, by both methods). The EOS and IgA values increased with age. M3SE and QUM were in moderate agreement (Kappa = 0.43). We concluded that the two segregation methods allowed for the identification of the same female SUS lambs, but a greater number of animals were categorised phenotypically as resistant using the M3SE method. PCV and TPP can help to identify phenotypically resistant animals

    Luteolytic failure as the cause of low efficiency in synchronization with prostaglandins in cows under tropical grazing

    Our objective was to determine the cause of the low response in the exclusive use of prostaglandin in synchronization programs in cows under tropical grazing compared to the use of progesterone (CIDR). Thirty-five cows with the presence of CL were randomly distributed: first group (GPG; n=23), synchronized with two doses of PGF2α (25 mg, Dinoprost®), with an interval of 12 days. The second group (GCIDR; n=12) synchronized with an intravaginal device (1.9 g progesterone), 2 mg estradiol benzoate and 50 mg progesterone), at day 7 a dose of PGF2α (pm) was administered and at day 8 the CIDR (am) was withdrawn. CL regression, luteolitic failure, progesterone concentration and CL size were determined. The data were subjected to normality test, then to Mann-Whitney U test or independent t student test and chi square. Finding that only 82.6% (19/23) of GPG cows at the second dose of prostaglandins have functional CL (<1 ng/ml of progesterone). Obtaining only in 43.5% of the cows a regression of the CL in the GPG, vs. 91.7% in GCIDR (P=0.0001). In addition, GPG cows showed luteolytic failure of 39.1% and asynchrony of 17.4%. It is concluded, a low efficiency of prostaglandins on the synchrony and regression of CL (luteolitic failure) in cows fed under tropical grazing, attributed to the low efficiency in synchronization programs.Objective: To determine the reason for the low response to the exclusive use of prostaglandin in synchronization programs in cows under tropical grazing compared with the use of progesterone (CIDR).     Revisiting the epidemiology of bloodstream infections and healthcare-associated episodes: results from a multicentre prospective cohort in Spain (PRO-BAC Study)

    PROBAC REIPI/GEIH-SEIMC/SAEI Group.The epidemiology of bloodstream infections (BSIs) is dynamic as it depends on microbiological, host and healthcare system factors. The aim of this study was to update the information regarding the epidemiology of BSIs in Spain considering the type of acquisition. An observational, prospective cohort study in 26 Spanish hospitals from October 2016 through March 2017 including all episodes of BSI in adults was performed. Bivariate analyses stratified by type of acquisition were performed. Multivariate analyses were performed by logistic regression. Overall, 6345 BSI episodes were included; 2510 (39.8%) were community-acquired (CA), 1661 (26.3%) were healthcare-associated (HCA) and 2056 (32.6%) hospital-acquired (HA). The 30-day mortality rates were 11.6%, 19.5% and 22.0%, respectively. The median age of patients was 71 years (interquartile range 60–81 years) and 3656 (58.3%; 95% confidence interval 57.1–59.6%) occurred in males. The proportions according to patient sex varied according to age strata. Escherichia coli (43.8%), Klebsiella spp. (8.9%), Staphylococcus aureus (8.9%) and coagulase-negative staphylococci (7.4%) were the most frequent pathogens. Multivariate analyses confirmed important differences between CA and HCA episodes, but also between HCA and HA episodes, in demographics, underlying conditions and aetiology. In conclusion, we have updated the epidemiological information regarding patients’ profiles, underlying conditions, frequency of acquisition types and aetiological agents of BSI in Spain.     A Proof-Of-Principle Study of Epigenetic Therapy Added to Neoadjuvant Doxorubicin Cyclophosphamide for Locally Advanced Breast Cancer

    BACKGROUND: Aberrant DNA methylation and histone deacetylation participate in cancer development and progression; hence, their reversal by inhibitors of DNA methylation and histone deacetylases (HDACs) is at present undergoing clinical testing in cancer therapy. As epigenetic alterations are common to breast cancer, in this proof-of-concept study demethylating hydralazine, plus the HDAC inhibitor magnesium valproate, were added to neoadjuvant doxorubicin and cyclophosphamide in locally advanced breast cancer to assess their safety and biological efficacy. METHODOLOGY: This was a single-arm interventional trial on breast cancer patients (ClinicalTrials.gov Identifier: NCT00395655). After signing informed consent, patients were typed for acetylator phenotype and then treated with hydralazine at 182 mg for rapid-, or 83 mg for slow-acetylators, and magnesium valproate at 30 mg/kg, starting from day –7 until chemotherapy ended, the latter consisting of four cycles of doxorubicin 60 mg/m(2) and cyclophosphamide 600 mg/m(2) every 21 days. Core-needle biopsies were taken from primary breast tumors at diagnosis and at day 8 of treatment with hydralazine and valproate. MAIN FINDINGS: 16 patients were included and received treatment as planned. All were evaluated for clinical response and toxicity and 15 for pathological response. Treatment was well-tolerated. The most common toxicity was drowsiness grades 1–2. Five (31%) patients had clinical CR and eight (50%) PR for an ORR of 81%. No patient progressed. One of 15 operated patients (6.6%) had pathological CR and 70% had residual disease <3 cm. There was a statistically significant decrease in global 5(m)C content and HDAC activity. Hydralazine and magnesium valproate up- and down-regulated at least 3-fold, 1,091 and 89 genes, respectively. CONCLUSIONS: Hydralazine and magnesium valproate produce DNA demethylation, HDAC inhibition, and gene reactivation in primary tumors. Doxorubicin and cyclophosphamide treatment is safe, well-tolerated, and appears to increase the efficacy of chemotherapy. A randomized phase III study is ongoing to support the efficacy of so-called epigenetic or transcriptional cancer therapy

    The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & Nemésio 2007; Donegan 2008, 2009; Nemésio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016

    Search for new particles in events with energetic jets and large missing transverse momentum in proton-proton collisions at root s=13 TeV

    A search is presented for new particles produced at the LHC in proton-proton collisions at root s = 13 TeV, using events with energetic jets and large missing transverse momentum. The analysis is based on a data sample corresponding to an integrated luminosity of 101 fb(-1), collected in 2017-2018 with the CMS detector. Machine learning techniques are used to define separate categories for events with narrow jets from initial-state radiation and events with large-radius jets consistent with a hadronic decay of a W or Z boson. A statistical combination is made with an earlier search based on a data sample of 36 fb(-1), collected in 2016. No significant excess of events is observed with respect to the standard model background expectation determined from control samples in data. The results are interpreted in terms of limits on the branching fraction of an invisible decay of the Higgs boson, as well as constraints on simplified models of dark matter, on first-generation scalar leptoquarks decaying to quarks and neutrinos, and on models with large extra dimensions. Several of the new limits, specifically for spin-1 dark matter mediators, pseudoscalar mediators, colored mediators, and leptoquarks, are the most restrictive to date.Peer reviewe
