    177Lu-Dendrimer conjugated to folate and bombesin with gold nanoparticles in the dendritic cavity: A potential theranostic radiopharmaceutical

    177Lu-labeled nanoparticles conjugated to biomolecules have been proposed as a new class of theranostic radiopharmaceuticals. The aim of this research was to synthesize 177Lu-dendrimer(PAMAM-G4)-folate-bombesin with gold nanoparticles (AuNPs) in the dendritic cavity and to evaluate the radiopharmaceutical potential for targeted radiotherapy and the simultaneous detection of folate receptors (FRs) and gastrin-releasing peptide receptors (GRPRs) overexpressed in breast cancer cells. p-SCN-Benzyl-DOTA was conjugated in aqueous-basic medium to the dendrimer.The carboxylate groups of Lys1Lys3(DOTA)-bombesin and folic acid were activatedwithHATUand also conjugated to the dendrimer.The conjugate was mixed with 1%HAuCl4 followed by the addition of NaBH4 and purified by ultrafiltration. Elemental analysis (EDS), particle size distribution (DLS), TEM analysis, UV-Vis, and infrared and fluorescence spectroscopies were performed. The conjugate was radiolabeled using 177LuCl3 or 68GaCl3 and analyzed by radio-HPLC. Studies confirmed the dendrimer functionalization with high radiochemical purity (>95%). Fluorescence results demonstrated that the presence ofAuNPs in the dendritic cavity confers useful photophysical properties to the radiopharmaceutical for optical imaging.     Provenance behavior in the cork-oak International network trials fair 202

    The international network of provenance trials in cork-oak was established as a result of the EU Concerted Action FAIR 202, 1995-2000. 34 provenances were selected in the natural range of cork oak and trials were established in 1996/97 in France, Italy, Portugal, Tunisia and Spain. This study compares the behavior of provenances in the trials of Grighini (Sardinia), Monte Fava and Quinta da Nogueira (Portugal), Monfrague (Spain), Les Maures (France) and Tebabe (Tunisia). Observations on total height (Ht) and diameter at beast height (DBH) are used to compare and assess adaptation within sites and among sites. Climate at the provenance site seed collection and at trials sites is used to characterize the behavior of provenances. The results reveal that the provenances, ES 5 (1063 mm), IT16 (910 mm), IT12 (937 mm), 937, TU32 (948 mm), FR2 (958 mm, FR1 (963 mm, MA31 (970 mm), ES8 (993 mm) are able to adapt and show good growth under conditions of decrease of total annual rainfall and strong decrease in case of TU33 (1610mm), MA27 (1280 mm). On the other hand provenances coming from sites of low precipitation not always show relevant performance in sites of higher precipitation as IT14 (448 mm), ES10 (455 mm), MA29 (479 mm). These results are discussed in the impact climate change (CC) may have in adaptation and evolution of cork oak. Our results show that cork oak will not face serious threats if drought increases in the Mediterranean region, since some populations are showing capable to cope successfully with decrease of total precipitation. The ongoing results from the cork-oak international network trials FAIR 202 show the importance of this line of research and the need to establish harmonized criteria on data collection to enhance the comparability. As cork quality is a key issue on cork-oak economic sustainability it is critical to establish harmonized criteria in cork harvesting for further studies to compare cork quality with special reference for the assessment of the genetic control of cork production and heritability.info:eu-repo/semantics/publishedVersio

    High-Throughput Synthesis of Pillared-Layered Magnesium Tetraphosphonate Coordination Polymers: Framework Interconversions and Proton Conductivity Studies

    Novel pillared-layered framework materials were synthesized by high-throughput or microwave-assisted methodology that contain Mg2+ and the zwitterionic linker HDTMP (hexamethylenediamine-N,N,N0,N0-tetrakis(methylenephosphonic acid)). Three compounds were structurally characterized by X-ray powder diffraction. In the compound {Mg[(HO3PCH2)2N(CH2)6N (CH2PO3H2)2](H2O)}n(1), obtained at 140 ºC by hydrothermal or microwave-assisted reaction, the layers are built by isolated Mg2+ octahedra coordinated by oxygen atoms from six different zwitterionic HDTMP ligands. Each amino-bis(methylenephosphonate) moiety links three Mg2+ ions, bridging two of them through one phosphonate group and connecting the third polyhedron in a monodentate fashion. In Compound 2, {Mg[(HO3PCH2)2N(CH2)6N(CH2PO3H2)2]}n, hydrothermally synthesized at 180 C, the layers are composed of bidentate amino-bis(methylenephosphonate) moieties connected to three Mg2+ ions, with one of the phosphonate groups acting as a bridging ligand. Various subtle structural changes are noted for the other two compounds. Thermodiffraction of 1 reveals that a crystalline-to-crystalline phase transformation occurs concomitantly with its dehydration, leading to a new anhydrous phase, namely, {Mg[(HO3PCH2)2N(CH2)6N(CH2PO3H2)2]}n(1deh). This process is fully reversible upon equilibrating the solid at room temperature. The reported compounds can adsorb ammonia and CO2. Compound 1 exhibits a moderate proton conductivity, ~1.5 x 10-5 S·cm-1 at 80 ºC and 95% RH, that increases a half order of magnitude after experiencing a complete dehydration/rehydration process

    Quercus suber geographic variation: preliminary results of the Iberian Península provenance trials

    Cork oak (Quercus suber L.) has a great social and ecological importance in the Iberian Peninsula. Yet, a large proportion of cork oak stands is old and natural regeneration is often problematic. During the last decade large areas were reforested with this species in the context of the Common Agriculture Policy (CAP) both in Portugal and Spain. Survival rates were often low due to inappropriate nursery and installation techniques, as well as to the use of unsuitable genetic material. In fact, the cork oak adaptability to environmental conditions is poorly understood; moreover climate change scenarios foreseen enhanced water deficits in the Mediterranean region. In order to allocate the most adequate seed sources to each set of climatic conditions in future afforestation activities and considering that an important source of variation in the characteristics related with adaptability may be at the provenance level, we evaluated inter-provenance variability and phenotypic plasticity for growth, survival, morphological structure and drought tolerance five years after plantation, in four test sites located across the Iberian Peninsula and covering a wide range of ecological conditions. Such trials are part of a multi-locality provenance test belonging to a Network that was bring up by Concerted action.     Identification of lptA, lpxE, and lpxO, Three Genes Involved in the Remodeling of Brucella Cell Envelope.

    The brucellae are facultative intracellular bacteria that cause a worldwide extended zoonosis. One of the pathogenicity mechanisms of these bacteria is their ability to avoid rapid recognition by innate immunity because of a reduction of the pathogen-associated molecular pattern (PAMP) of the lipopolysaccharide (LPS), free-lipids, and other envelope molecules. We investigated the Brucella homologs of lptA, lpxE, and lpxO, three genes that in some pathogens encode enzymes that mask the LPS PAMP by upsetting the core-lipid A charge/hydrophobic balance. Brucella lptA, which encodes a putative ethanolamine transferase, carries a frame-shift in B. abortus but not in other Brucella spp. and phylogenetic neighbors like the opportunistic pathogen Ochrobactrum anthropi. Consistent with the genomic evidence, a B. melitensis lptA mutant lacked lipid A-linked ethanolamine and displayed increased sensitivity to polymyxin B (a surrogate of innate immunity bactericidal peptides), while B. abortus carrying B. melitensis lptA displayed increased resistance. Brucella lpxE encodes a putative phosphatase acting on lipid A or on a free-lipid that is highly conserved in all brucellae and O. anthropi. Although we found no evidence of lipid A dephosphorylation, a B. abortus lpxE mutant showed increased polymyxin B sensitivity, suggesting the existence of a hitherto unidentified free-lipid involved in bactericidal peptide resistance. Gene lpxO putatively encoding an acyl hydroxylase carries a frame-shift in all brucellae except B. microti and is intact in O. anthropi. Free-lipid analysis revealed that lpxO corresponded to olsC, the gene coding for the ornithine lipid (OL) acyl hydroxylase active in O. anthropi and B. microti, while B. abortus carrying the olsC of O. anthropi and B. microti synthesized hydroxylated OLs. Interestingly, mutants in lptA, lpxE, or olsC were not attenuated in dendritic cells or mice. This lack of an obvious effect on virulence together with the presence of the intact homolog genes in O. anthropi and B. microti but not in other brucellae suggests that LptA, LpxE, or OL β-hydroxylase do not significantly alter the PAMP properties of Brucella LPS and free-lipids and are therefore not positively selected during the adaptation to intracellular life

    Origin and pathways of the mineral dust transport to two Spanish EARLINET sites: Effect on the observed columnar and range-resolved dust optical properties

    In this paper, is presented a method for estimation of the effect of the transport process to aerosol optical properties. Aerosol optical data retrieved by lidars and sun-photometer measurements, are applied to Saharan dust events observed simultaneously at the two EARLINET/AERONET sites of Barcelona and Granada during the periods of June–September of 2012 and 2013. For this purpose, elastic lidar profiles and sun-photometer columnar retrievals are analyzed together with satellite observations and dust forecast models. Granada presents more than twice Saharan dust outbreaks compared to Barcelona. The scenarios favoring the Saharan dust outbreaks are identified in both places. The mineral dust originating in the Sahara region and arriving at both stations is usually transport wither over the Atlas Mountains or through an Atlantic pathway. Analyses of dust events affecting both stations reveal how differences in the transport process lead to differences in the aerosol optical properties measured at each station. Mean dust related Ångström exponent is 1.8 times higher in Barcelona than in Granada. This difference is a result of the additional contribution of anthropogenic aerosol, mainly in the aerosol fine mode, during the transport of the mineral dust plume over the Iberian Peninsula.Andalusia Regional Government through the project P12-RNM-2409Spanish Ministry of Economy and Competitiveness through the project CGL2013-45410-

    Measurement of the cross-section and charge asymmetry of WW bosons produced in proton-proton collisions at s=8\sqrt{s}=8 TeV with the ATLAS detector

    This paper presents measurements of the W+μ+νW^+ \rightarrow \mu^+\nu and WμνW^- \rightarrow \mu^-\nu cross-sections and the associated charge asymmetry as a function of the absolute pseudorapidity of the decay muon. The data were collected in proton--proton collisions at a centre-of-mass energy of 8 TeV with the ATLAS experiment at the LHC and correspond to a total integrated luminosity of 20.2~\mbox{fb^{-1}}. The precision of the cross-section measurements varies between 0.8% to 1.5% as a function of the pseudorapidity, excluding the 1.9% uncertainty on the integrated luminosity. The charge asymmetry is measured with an uncertainty between 0.002 and 0.003. The results are compared with predictions based on next-to-next-to-leading-order calculations with various parton distribution functions and have the sensitivity to discriminate between them.Comment: 38 pages in total, author list starting page 22, 5 figures, 4 tables, submitted to EPJC.
