    Associations between proinflammatory cytokines in the synovial fluid and radiographic grading and pain-related scores in 47 consecutive patients with osteoarthritis of the knee

    <p>Abstract</p> <p>Background</p> <p>One of the sources of knee pain in osteoarthritis (OA) is believed to be related to local chronic inflammation of the knee joints, which involves the production of inflammatory cytokines such as tumor necrosis factor alpha (TNFα), interleukin (IL)-6, and nerve growth factor (NGF) in the synovial membrane, and these cytokines are believed to promote pathological OA. In the present study, correlations between proinflammatory cytokines in knee synovial fluid and radiographic changes and functional scores and pain scores among OA patients were examined.</p> <p>Methods</p> <p>Synovial fluid was harvested from the knees of 47 consecutive OA patients, and the levels of TNFα, IL-6, and NGF were measured using enzyme-linked immunosorbent assays. Osteoarthritic knees were classified using Kellgren-Lawrence (KL) grading (1-4). The Western Ontario and McMaster University Osteoarthritis Index (WOMAC) was used to assess self-reported physical function, pain, and stiffness.</p> <p>Results</p> <p>TNFα and IL-6 were detectable in knee synovial, whereas NGF was not. TNFα was not correlated with the KL grade, whereas IL-6 had a significantly negative correlation. We observed differences in the correlations between TNFα and IL-6 with WOMAC scores and their subscales (pain, stiffness, and physical function). TNFα exhibited a significant correlation with the total score and its 3 subscales, whereas IL-6 exhibited a moderately significant negative correlation only with the subscale of stiffness.</p> <p>Conclusions</p> <p>The present study demonstrated that the concentrations of proinflammatory cytokines are correlated with KL grades and WOMAC scores in patients with knee OA. Although TNFα did not have a significant correlation with the radiographic grading, it was significantly associated with the WOMAC score. IL-6 had a significant negative correlation with the KL grading, whereas it had only a weakly significant correlation with the subscore of stiffness. The results suggest that these cytokines play a role in the pathogenesis of synovitis in osteoarthritic knees in different ways: TNFα is correlated with pain, whereas IL-6 is correlated with joint function.</p

    Correspondence between otolith microstructual changes and early life history events in Anguilla marmorata leptocephali and glass eels

    To determine the exact correspondence between otolith characteristics and early life history events such as metamorphosis, coastal migration and recruitment to estuaries, both otolith microstructure and microchemistry analyses were applied to a sequential developmental series of samples, e. g. leptocephali, a metamorphosing larva, oceanic glass eels, and coastal glass eels. Total length and age were 10.1-50.7 mm and 22-137 d in leptocephali, 46.3 mm and 147 d in a metamorphosing larva, 47.8, 48.6 mm and 159, 160 d in oceanic glass eels, and 47.9-54.8 mm and 119-168 d in coastal glass eels. Checks at hatching and first feeding were observed in all specimens, but metamorphosis and freshwater checks were observed only in some specimens. It was confirmed that the abrupt drop in otolith Sr : Ca ratios and drastic increases of otolith increment widths in the metamorphosing larval stage correspond to the onset of metamorphosis, and the decrease after the peaks suggested the completion of metamorphosis, because the metamorphosing larva had no decrease in incremental widths. The relatively conserved Sr : Ca ratios decreased sharply in synchrony with the increasing increment widths. This study provides the first direct evidence that these drastic changes in otolith microstructure and microchemistry actually occur during metamorphosis, which has been only hypothesized

    Feasibility Study of Technology Demonstration Mission for Integrated Attitude-Orbit Control of Solar Sail

    We are developing a micro solar sail called PIERIS . The purpose of this project is to demonstrate the following two world-first technologies in Low Earth Orbit. The first technology is to control the external torque with a single gimbal motor. It will be possible to achieve a completely propellant-free Integrated Attitude-Orbit Control. The second technology is a sail structure that guarantees the accuracy of the Pyramid-Shaped sail membrane shape and reduces disturbance torque caused by membrane surface deformation. This project has been selected for the Feasibility-Study phase of the JAXA Small Satellite Rush Program and is scheduled for launch in FY2026 if approved to proceed to the next phase. We are currently conducting mission and system feasibility studies and developing a Bread Board Model

    Promoting neuro-supportive properties of astrocytes with epidermal growth factor hydrogels

    Biomaterials provide novel platforms to deliver stem cell and growth factor therapies for central nervous system (CNS) repair. The majority of these approaches have focused on the promotion of neural progenitor cells and neurogenesis. However, it is now increasingly recognized that glial responses are critical for recovery in the entire neurovascular unit. In this study, we investigated the cellular effects of epidermal growth factor (EGF) containing hydrogels on primary astrocyte cultures. Both EGF alone and EGF‐hydrogel equally promoted astrocyte proliferation, but EGF‐hydrogels further enhanced astrocyte activation, as evidenced by a significantly elevated Glial fibrillary acidic protein (GFAP) gene expression. Thereafter, conditioned media from astrocytes activated by EGF‐hydrogel protected neurons against injury and promoted synaptic plasticity after oxygen–glucose deprivation. Taken together, these findings suggest that EGF‐hydrogels can shift astrocytes into neuro‐supportive phenotypes. Consistent with this idea, quantitative‐polymerase chain reaction (qPCR) demonstrated that EGF‐hydrogels shifted astrocytes in part by downregulating potentially negative A1‐like genes (Fbln5 and Rt1‐S3) and upregulating potentially beneficial A2‐like genes (Clcf1, Tgm1, and Ptgs2). Further studies are warranted to explore the idea of using biomaterials to modify astrocyte behavior and thus indirectly augment neuroprotection and neuroplasticity in the context of stem cell and growth factor therapies for the CNS.     Search for dark matter produced in association with bottom or top quarks in √s = 13 TeV pp collisions with the ATLAS detector

    A search for weakly interacting massive particle dark matter produced in association with bottom or top quarks is presented. Final states containing third-generation quarks and miss- ing transverse momentum are considered. The analysis uses 36.1 fb−1 of proton–proton collision data recorded by the ATLAS experiment at √s = 13 TeV in 2015 and 2016. No significant excess of events above the estimated backgrounds is observed. The results are in- terpreted in the framework of simplified models of spin-0 dark-matter mediators. For colour- neutral spin-0 mediators produced in association with top quarks and decaying into a pair of dark-matter particles, mediator masses below 50 GeV are excluded assuming a dark-matter candidate mass of 1 GeV and unitary couplings. For scalar and pseudoscalar mediators produced in association with bottom quarks, the search sets limits on the production cross- section of 300 times the predicted rate for mediators with masses between 10 and 50 GeV and assuming a dark-matter mass of 1 GeV and unitary coupling. Constraints on colour- charged scalar simplified models are also presented. Assuming a dark-matter particle mass of 35 GeV, mediator particles with mass below 1.1 TeV are excluded for couplings yielding a dark-matter relic density consistent with measurements
