    Contaminants in Unionid Mussels from the Confluence of the Mississippi and Illinois Rivers

    Unionid mussels were collected from three mussel beds near the confluence of the Mississippi and Illinois rivers in 2003 to evaluate concentrations of selected elements and organic compounds in three abundant species and to preliminarily investigate the relative contribution of these waterways to observed contaminant burdens. Copper (Cu), selenium (Se), and zinc (Zn) concentrations were higher and lead (Pb) concentrations were lower in Amblema plicata collected downstream of the confluence than in those collected upstream. Mean concentrations of nickel (Ni), total mercury (Hg), methylmercury (MeHg), Pb, and Zn varied by species. Concentrations of cadmium (Cd) decreased with age in A. plicata from two of three sites. Tissue concentrations of some elements, e.g., arsenic (As), Cd, Cu, Pb, Se, and Zn, were similar to or higher than those previously reported for unionid mussels from areas of contaminated sediment. Concentrations of Cd, Cu, and Zn in A. plicata were comparable to those collected from the Mississippi River approximately 450 and 900 km upstream from our study sites (Naimo et al. 1992). Although total Hg concentrations we observed were an order of magnitude lower than in that study, MeHg concentrations were above those associated with reductions in soft tissue mass in a study of Elliptio complanata (Salazar et al. 1995). A number of polychlorinated biphenyl (PCB) congeners were detected in A. plicata tissues, with 85% of detections occurring in mussels from downstream of the confluence. Concentrations of individual PCB congeners were ???33 ng/g ww and the maximum summed PCB congener concentration was 100.2 ng/g ww. Although few persistent pesticides were detected, -hexachlorocyclohexane (HCH) was detected in each of the species collected from below the confluence of the two rivers, and in A. plicata collected above it on both the Mississippi and Illinois rivers, at a maximum concentration of 103.5 ng/g ww. Aldrin, ??-HCH and dichlorodiphenyltrichloroethane (DDT) were detected in few of the specimens collected. The findings of this preliminary investigation suggest that unionid mussels from near the confluence of the Mississippi and Illinois rivers may be at risk of negative health effects of elevated exposure to certain environmental contaminants.     Alternativity and reciprocity in the Cayley-Dickson algebra

    We calculate the eigenvalue \rho of the multiplication mapping R on the Cayley-Dickson algebra A_n. If the element in A_n is composed of a pair of alternative elements in A_{n-1}, half the eigenvectors of R in A_n are still eigenvectors in the subspace which is isomorphic to A_{n-1}. The invariant under the reciprocal transformation A_n \times A_{n} \ni (x,y) -> (-y,x) plays a fundamental role in simplifying the functional form of \rho. If some physical field can be identified with the eigenspace of R, with an injective map from the field to a scalar quantity (such as a mass) m, then there is a one-to-one map \pi: m \mapsto \rho. As an example, the electro-weak gauge field can be regarded as the eigenspace of R, where \pi implies that the W-boson mass is less than the Z-boson mass, as in the standard model.Comment: To be published in J.     Turbulence Heating ObserveR – satellite mission proposal

    The Universe is permeated by hot, turbulent, magnetized plasmas. Turbulent plasma is a major constituent of active galactic nuclei, supernova remnants, the intergalactic and interstellar medium, the solar corona, the solar wind and the Earth’s magnetosphere, just to mention a few examples. Energy dissipation of turbulent fluctuations plays a key role in plasma heating and energization, yet we still do not understand the underlying physical mechanisms involved. THOR is a mission designed to answer the questions of how turbulent plasma is heated and particles accelerated, how the dissipated energy is partitioned and how dissipation operates in different regimes of turbulence. THOR is a single-spacecraft mission with an orbit tuned to maximize data return from regions in near-Earth space – magnetosheath, shock, foreshock and pristine solar wind – featuring different kinds of turbulence. Here we summarize the THOR proposal submitted on 15 January 2015 to the ‘Call for a Medium-size mission opportunity in ESAs Science Programme for a launch in 2025 (M4)’.     Assessing the carcinogenic potential of low-dose exposures to chemical mixtures in the environment: the challenge ahead.

    Lifestyle factors are responsible for a considerable portion of cancer incidence worldwide, but credible estimates from the World Health Organization and the International Agency for Research on Cancer (IARC) suggest that the fraction of cancers attributable to toxic environmental exposures is between 7% and 19%. To explore the hypothesis that low-dose exposures to mixtures of chemicals in the environment may be combining to contribute to environmental carcinogenesis, we reviewed 11 hallmark phenotypes of cancer, multiple priority target sites for disruption in each area and prototypical chemical disruptors for all targets, this included dose-response characterizations, evidence of low-dose effects and cross-hallmark effects for all targets and chemicals. In total, 85 examples of chemicals were reviewed for actions on key pathways/mechanisms related to carcinogenesis. Only 15% (13/85) were found to have evidence of a dose-response threshold, whereas 59% (50/85) exerted low-dose effects. No dose-response information was found for the remaining 26% (22/85). Our analysis suggests that the cumulative effects of individual (non-carcinogenic) chemicals acting on different pathways, and a variety of related systems, organs, tissues and cells could plausibly conspire to produce carcinogenic synergies. Additional basic research on carcinogenesis and research focused on low-dose effects of chemical mixtures needs to be rigorously pursued before the merits of this hypothesis can be further advanced. However, the structure of the World Health Organization International Programme on Chemical Safety 'Mode of Action' framework should be revisited as it has inherent weaknesses that are not fully aligned with our current understanding of cancer biology

    Rethinking the Assessment of Work Intensity

    : Digitalization and flexible work arrangements have profoundly changed workplaces and are often associated with an increasing work intensity, putting employees’ well-being at risk. To respond to increasing work intensity effectively, it is crucial to assess work intensity thoroughly and regularly. However, there are no specific measures that capture work intensity through a comprehensive and contemporary understanding. In two studies, we confirmed the factorial structure of a newly developed questionnaire and pictorial scales on work intensity, providing evidence for the verbal questionnaire’s convergent and incremental validity. The pictorial scales allow for a complementary methodological approach to the assessment of work intensity as measured by the questionnaire. Overall, the questionnaire allows for a comprehensive assessment of work intensity and the pictorial scales meet the demand for a simple and concise instrument suitable for repeated measures such as in the context of diary studies.Die Erfassung von Arbeitsintensität neu gedacht. In zwei Studien wurde die faktorielle Struktur eines neu entwickelten Fragebogens und von Bilderskalen zur Arbeitsintensität bestätigt sowie die konvergente und inkrementelle Validität des verbalen Fragebogens nachgewiesen. Die Bilderskalen ermöglichen einen ergänzenden methodischen Zugang für eine Einschätzung der Arbeitsintensität, so wie sie mit dem Fragebogen erfasst wird.     The Impact of Small Molecule Binding on the Energy Landscape of the Intrinsically Disordered Protein C-Myc

    Intrinsically disordered proteins are attractive therapeutic targets owing to their prevalence in several diseases. Yet their lack of well-defined structure renders ligand discovery a challenging task. An intriguing example is provided by the oncoprotein c-Myc, a transcription factor that is over expressed in a broad range of cancers. Transcriptional activity of c-Myc is dependent on heterodimerization with partner protein Max. This protein-protein interaction is disrupted by the small molecule 10058-F4 (1), that binds to monomeric and disordered c-Myc. To rationalize the mechanism of inhibition, structural ensembles for the segment of the c-Myc domain that binds to 1 were computed in the absence and presence of the ligand using classical force fields and explicit solvent metadynamics molecular simulations. The accuracy of the computed structural ensembles was assessed by comparison of predicted and measured NMR chemical shifts. The small molecule 1 was found to perturb the composition of the apo equilibrium ensemble and to bind weakly to multiple distinct c-Myc conformations. Comparison of the apo and holo equilibrium ensembles reveals that the c-Myc conformations binding 1 are already partially formed in the apo ensemble, suggesting that 1 binds to c-Myc through an extended conformational selection mechanism. The present results have important implications for rational ligand design efforts targeting intrinsically disordered proteins

    The core aspect of the senescent phenotype is a stable state of cell cycle arrest. However, this is a disguise that conceals a highly active metabolic cell state with diverse functionality. Both the cell-autonomous and the non-cell-autonomous activities of senescent cells create spatiotemporally dynamic and context-dependent tissue reactions. For example, the senescence-associated secretory phenotype (SASP) provokes not only tumour-suppressive but also tumour-promoting responses. Senescence is now increasingly considered to be an integrated and widespread component that is potentially important for tumour development, tumour suppression and the response to therapy.This is the author accepted manuscript.
