    Summoning the Ghosts: Records as Agents in Community Archives

    Although records have traditionally been defined by their physical form, our research reveals that records documenting marginalized communities disrupt these limiting definitions by surfacing the ways that community members conceive of the agency of records. Based on focus groups we conducted with fifty-four community members at five different Southern California–based community archives, this paper examines how community archives users conceive of records as agents, embodied with the voices of past lives, and capable of facilitating meaning for those who access, activate, and interpret them. In our findings, users of community archives not only surfaced the notion of records as dynamic, sentient, and generative agents, but also of records as haunted, emphasizing that dominant historical narratives are haunted by their absences

    Wearable multi-channel microelectrode membranes for elucidating electrophysiological phenotypes of injured myocardium

    Understanding the regenerative capacity of small vertebrate models has provided new insights into the plasticity of injured myocardium. Here, we demonstrate the application of flexible microelectrode arrays (MEAs) in elucidating electrophysiological phenotypes of zebrafish and neonatal mouse models of heart regeneration. The 4-electrode MEA membranes were designed to detect electrical signals in the aquatic environment. They were micro-fabricated to adhere to the non-planar body surface of zebrafish and neonatal mice. The acquired signals were processed to display an electrocardiogram (ECG) with high signal-to-noise-ratios, and were validated via the use of conventional micro-needle electrodes. The 4-channel MEA provided signal stability and spatial resolution, revealing the site-specific electrical injury currents such as ST-depression in response to ventricular cryo-injury. Thus, our polymer-based and wearable MEA membranes provided electrophysiological insights into long-term conduction phenotypes for small vertebral models of heart injury and regeneration with a translational implication for monitoring cardiac patients

    Challenging Official Propaganda? Public Opinion Leaders on Sina Weibo.

    This article examines the prominence of various user categories as opinion leaders, defined as initiators, agenda setters or disseminators, in 29 corruption cases exposed on SinaWeibo. It finds that ordinary citizens made up the largest category of initiators but that their power of opinion leadership was limited as they had to rely on media organizations to spread news about the cases. News organizations and online media were the main opinion leaders. Government and Party bodies initiated a fair number of cases and, despite not being strong agenda setters or disseminators, were able to dominate public opinion owing to the fact that news organizations and online media mainly published official announcements about the cases. Media organizations also played a secondary role as the voice of the people. While individuals from some other user categories were able to become prominent opinion leaders, news workers are likely to be the most promising user category to challenge official propaganda.Faculty Research Support Scheme of the Faculty of Arts and Social Sciences, the University of Sydney.     Can the Weak Surface Currents of the Cape Verde Frontal Zone Be Measured With Altimetry?

    Three data types are compared in the low-current-velocity regime in the southeastern North Atlantic, between 12-degrees-N and 30-degrees-N, 29-degrees-W and 18-degrees-W: Geosat altimetric sea level and derived surface geostrophic velocities, shallow current meter velocities, and dynamic heights derived from hydrographic data from cruises 4, 6, and 9 of the research vessel Meteor. The four current meter daily time series, at depths around 200 m, were smoothed over 1 month; the altimetric geostrophic velocities were computed from sea surface slopes over 142 km every 17 days. The correlation coefficients between the current meter and altimetric geostrophic velocities range between 0.64 and 0.90 for the moorings near 29-degrees-N but between 0.32 and 0.71 for the two around 21-degrees-N; the associated rms discrepancies between the two measurement types range between 1.5 and 4.4 cm/s, which is 49% to 127% of the rms of the respective current meter time series. Dynamic heights relative to 1950 dbar for the months of November 1986 (d(M4)), November 1987 (d(M6)), and February 1989 (d(M9)) were computed from Meteor cruises 4, 6, and 9. Both dynamic heights and altimetric heights (h(M4), h(M6), h(M9)) were averaged over 1-degrees boxes for the duration of each cruise. Differences d(M4) - d(M6) and d(M9) - d(M6) were computed only at bins where at least one station from both cruises existed, Assuming that dynamic heights d in dynamic centimeters are equivalent to sea level h in centimeters, the standard deviation sigma of the differences ((h(M4) - h(M6)) - (d(M4) - d(M6))) and corresponding M9 - M6 values was 2.1 cm. This value (squared) is only 13% of the (5.8 cm)2 variance of the dynamic height differences and is indistinguishable from the 2.7- to 5.6-cm natural variability of sea level in the area expected between the times when the ship and the satellite sampled the ocean. The areally averaged discrepancy for M9 - M6 was only 0.7 cm, but the corresponding value for M4 - M6 was 5.2 cm. A systematic difference between the water vapor corrections used before and after July 1987 is responsible for the M4 - M6 difference. The average M4 - M6 discrepancy is only 0.1 cm using the Fleet Numerical Oceanography Center correction, with a standard deviation of 3.1 cm. In spite of the underlying differences in sampling and physics, including unknown barotropic components not included in our hydrographic dynamic heights, and in data errors, including water vapor, ionospheric, and orbital effects on the altimetry, consistent interannual changes of the mean sea level from the independently obtained altimetric and hydrographic data sets are obtained, and correlated seasonal changes in surface currents are observed with both altimetry and current meters

    Hairpin ribozyme-antisense RNA constructs can act as molecular lassos

    We have developed a novel class of antisense agents, RNA Lassos, which are capable of binding to and circularizing around complementary target RNAs. The RNA Lasso consists of a fixed sequence derived from the hairpin ribozyme and an antisense segment whose size and sequence can be varied to base pair with accessible sites in the target RNA. The ribozyme catalyzes self-processing of the 5′- and 3′-ends of a transcribed Lasso precursor and ligates the processed ends to produce a circular RNA. The circular and linear forms of the self-processed Lasso coexist in an equilibrium that is dependent on both the Lasso sequence and the solution conditions. Lassos form strong, noncovalent complexes with linear target RNAs and form true topological linkages with circular targets. Lasso complexes with linear RNA targets were detected by denaturing gel electrophoresis and were found to be more stable than ordinary RNA duplexes. We show that expression of a fusion mRNA consisting of a sequence from the murine tumor necrosis factor-α (TNF-α) gene linked to luciferase reporter can be specifically and efficiently blocked by an anti-TNF Lasso. We also show in cell culture experiments that Lassos directed against Fas pre-mRNA were able to induce a change in alternative splicing patterns

    Engineering Nano- and Microparticles to Tune Immunity

    The immune system can be a cure or cause of disease, fulfilling a protective role in attacking cancer or pathogenic microbes but also causing tissue destruction in autoimmune disorders. Thus, therapies aimed to amplify or suppress immune reactions are of great interest. However, the complex regulation of the immune system, coupled with the potential systemic side effects associated with traditional systemic drug therapies, has presented a major hurdle for the development of successful immunotherapies. Recent progress in the design of synthetic micro- and nano-particles that can target drugs, deliver imaging agents, or stimulate immune cells directly through their physical and chemical properties is leading to new approaches to deliver vaccines, promote immune responses against tumors, and suppress autoimmunity. In addition, novel strategies, such as the use of particle-laden immune cells as living targeting agents for drugs, are providing exciting new approaches for immunotherapy.     A Family of Helminth Molecules that Modulate Innate Cell Responses via Molecular Mimicry of Host Antimicrobial Peptides

    Over the last decade a significant number of studies have highlighted the central role of host antimicrobial (or defence) peptides in modulating the response of innate immune cells to pathogen-associated ligands. In humans, the most widely studied antimicrobial peptide is LL-37, a 37-residue peptide containing an amphipathic helix that is released via proteolytic cleavage of the precursor protein CAP18. Owing to its ability to protect against lethal endotoxaemia and clinically-relevant bacterial infections, LL-37 and its derivatives are seen as attractive candidates for anti-sepsis therapies. We have identified a novel family of molecules secreted by parasitic helminths (helminth defence molecules; HDMs) that exhibit similar biochemical and functional characteristics to human defence peptides, particularly CAP18. The HDM secreted by Fasciola hepatica (FhHDM-1) adopts a predominantly α-helical structure in solution. Processing of FhHDM-1 by F. hepatica cathepsin L1 releases a 34-residue C-terminal fragment containing a conserved amphipathic helix. This is analogous to the proteolytic processing of CAP18 to release LL-37, which modulates innate cell activation by classical toll-like receptor (TLR) ligands such as lipopolysaccharide (LPS). We show that full-length recombinant FhHDM-1 and a peptide analogue of the amphipathic C-terminus bind directly to LPS in a concentration-dependent manner, reducing its interaction with both LPS-binding protein (LBP) and the surface of macrophages. Furthermore, FhHDM-1 and the amphipathic C-terminal peptide protect mice against LPS-induced inflammation by significantly reducing the release of inflammatory mediators from macrophages. We propose that HDMs, by mimicking the function of host defence peptides, represent a novel family of innate cell modulators with therapeutic potential in anti-sepsis treatments and prevention of inflammation
