    Inbreeding and selection on sex ratio in the bark beetle Xylosandrus germanus

    Background Local Mate Competition (LMC) theory predicts a female should produce a more female-biased sex ratio if her sons compete with each other for mates. Because it provides quantitative predictions that can be experimentally tested, LMC is a textbook example of the predictive power of evolutionary theory. A limitation of many earlier studies in the field is that the population structure and mating system of the studied species are often estimated only indirectly. Here we use microsatellites to characterize the levels of inbreeding of the bark beetle Xylosandrus germanus, a species where the level of LMC is expected to be high. Results For three populations studied, genetic variation for our genetic markers was very low, indicative of an extremely high level of inbreeding (FIS = 0.88). There was also strong linkage disequilibrium between microsatellite loci and a very strong genetic differentiation between populations. The data suggest that matings among non-siblings are very rare (3%), although sex ratios from X. germanus in both the field and the laboratory have suggested more matings between non-sibs, and so less intense LMC. Conclusions Our results confirm that caution is needed when inferring mating systems from sex ratio data, especially when a lack of biological detail means the use of overly simple forms of the model of interest

    Social cues in the expression of sequential alternative reproductive tactics in young males of the peacock blenny, Solaria pavo

    Phenotypic change in response to variation in environmental cues has been widely documented in fish. Transitions in social dominance, in particular, have been shown to induce a rapid switch in reproductive phenotypes in many species. However, this effect has been mainly studied in adults and focused on behavioural transitions. The way social cues constraint the phenotypic development of juveniles remains poorly studied in fish. We tested the importance of social dominance and density in the phenotypic development of juveniles of the peacock blenny Solaria pavo. This species shows sequential male alternative reproductive tactics. In the first breeding season males can reproduce as nest-holders or as parasitic males (female-mimicking), or postpone reproduction; from the following season afterwards all males reproduce as nest-holders. Parasitic males have relatively larger testes that lack a testicular gland, present in the testes of nest-holders. The testicular gland is the main source of androgens in the testes and accordingly nest-holders have higher circulating androgen levels. In addition, exogenous androgen administration to parasitic males promotes the development of secondary sexual characters (SSC) only present in nest-holders such as a head crest and an anal gland. We raised juveniles under a high or low-density treatment and monitored social interactions for 1 month. No significant effect of density on the development of juvenile males was detected. However, within each replicate, the relative body size of juvenile males at the beginning of the experiment determined their dominance status, with dominant males developing towards the nest-holder morphotype. Dominant males engaged in more nest defence behaviour, showed larger testicular glands, had higher levels of 11-ketotestosterone (11-KT) and testosterone (T) and developed more SSC, as compared to subordinate males. However, these effects of social dominance were moderated by body condition as only dominant males in good body condition developed SSC. The effect of social dominance and of the area of the testicular gland on the development of SSC was mediated by 11-KT and on the expression of nest defence behaviour by T. Interestingly, in spite of the higher androgen levels and more pronounced morphologic development of SSC in dominant individuals, gonadal development was independent of social dominance and most fish still had underdeveloped testis at the end of the experiment. In conclusion, social dominance promoted the development of the testicular gland, an increase in circulating androgen levels and the development of SSC, but did not promote testicular development. This suggests a dissociation of mechanisms underlying sexual maturation and the expression of male reproductive traits. This dissociation seems to be the key for the occurrence of female-mimicking males in this species, which are sexually mature despite lacking the SSC typical of nest-holdets. (C) 2012 Elsevier Inc.     Monitoring the 5'UTR landscape reveals isoform switches to drive translational efficiencies in cancer

    Transcriptional and translational control are key determinants of gene expression, however, to what extent these two processes can be collectively coordinated is still poorly understood. Here, we use Nanopore long-read sequencing and cap analysis of gene expression (CAGE-seq) to document the landscape of 5' and 3' untranslated region (UTR) isoforms and transcription start sites of epidermal stem cells, wild-type keratinocytes and squamous cell carcinomas. Focusing on squamous cell carcinomas, we show that a small cohort of genes with alternative 5'UTR isoforms exhibit overall increased translational efficiencies and are enriched in ribosomal proteins and splicing factors. By combining polysome fractionations and CAGE-seq, we further characterize two of these UTR isoform genes with identical coding sequences and demonstrate that the underlying transcription start site heterogeneity frequently results in 5' terminal oligopyrimidine (TOP) and pyrimidine-rich translational element (PRTE) motif switches to drive mTORC1-dependent translation of the mRNA. Genome-wide, we show that highly translated squamous cell carcinoma transcripts switch towards increased use of 5'TOP and PRTE motifs, have generally shorter 5'UTRs and expose decreased RNA secondary structures. Notably, we found that the two 5'TOP motif-containing, but not the TOP-less, RPL21 transcript isoforms strongly correlated with overall survival in human head and neck squamous cell carcinoma patients. Our findings warrant isoform-specific analyses in human cancer datasets and suggest that switching between 5'UTR isoforms is an elegant and simple way to alter protein synthesis rates, set their sensitivity to the mTORC1-dependent nutrient-sensing pathway and direct the translational potential of an mRNA by the precise 5'UTR sequence

    Ecological variation drives morphological differentiation in a highly social vertebrate

    Animals may respond to ecological heterogeneity by genetic differentiation or phenotypic plasticity. Responses of organisms to their ecology can include adaptation at various levels of organization, including morphology, behaviour and social structure. Adaptations at one level might constrain or enhance adaptations on other levels, which highlights the importance of understanding their interactions. In highly social animals, understanding the influence of their ecological niche on the evolution and maintenance of complex social organization poses an intriguing challenge. Predation risk and habitat characteristics determine social structure in the cooperatively breeding cichlid Neolamprologus pulcher. Here we examine how varying combinations of these ecological factors across eight distinct populations influence morphological differentiation. We investigated the relationship between body shape and ecological parameters for 137 wild-caught individuals from eight distinct populations. Furthermore, we examined the genetic structure and differentiation among these populations using microsatellites. Finally, to disentangle heritable from plastic responses, we raised two successive generations from six populations in the laboratory under common garden conditions and screened 188 individuals for morphological differentiation. We found that body shape of N. pulcher strongly correlates with the measured ecological parameters. Low predation risk, low habitat structure and small shelter size favoured shallow bodies, whereas at the opposite end of these environmental gradients deep body shapes prevail. These consistent morphological differences persisted over two laboratory-reared generations, revealing a heritable basis. In contrast to the significant effect of local ecology on morphological differentiation between populations, both geographical and genetic distance had little explanatory power, suggesting that morphological differentiation between populations is not a simple by-product of genetic isolation. Remarkably, the largest difference in body depth emerged between the two populations located closest to each other, but differing strongly in their ecological niche. These results highlight that morphological variation is a key component of local adaptation in neighbouring populations of a highly social species. Such morphological differentiation has the potential to influence individual cooperative behaviour, which will eventually feed back on group structure and mediate the evolution and maintenance of complex social systems

    Advice on testing the null hypothesis that a sample is drawn from a Normal distribution.

    The Normal distribution remains the most widely-used statistical model, so it is only natural that researchers will frequently be required to consider whether a sample of data appears to have been drawn from a Normal distribution. Commonly-used statistical packages offer a range of alternative formal statistical tests of the null hypothesis of Normality, with inference being drawn on the basis of a calculated p-value. Here we aim to review the statistical literature on the performance of these tests, and briefly survey current usage of them in recently-published papers, with a view to offering advice on good practice. We find that authors in animal behaviour seem to be using such testing most commonly in situations where it is inadvisable (or at best unnecessary) involving pre-testing to select parametric or not-parametric analyses; and making little use of it in model-fitting situations where it might be of value. Of the many alternative tests, we recommend the routine use of either the Shapiro-Wilk or Chen-Shapiro tests; these are almost always superior to commonly-used alternatives like the Kolmogorov-Smirnov test, often by a substantial margin. We describe how both our recommend tests can be implemented. In contrast to current practice as indicated by our survey, we recommend that the results of these tests are reported in more detail (providing both the calculated sample statistic and the associated p-value). Finally, emphasize that even the higher-performing tests of Normality have low power (generally below 0.5 and often much lower) when sample sizes are less than 50, as is often the case in our field.     Pre- and postcopulatory sexual selection favor aggressive, young males in polyandrous groups of red junglefowl

    A challenge in evolutionary biology is to understand the operation of sexual selection on males in polyandrous groups, where sexual selection occurs before and after mating. Here, we combine fine-grained behavioral information (>41,000 interactions) with molecular parentage data to study sexual selection in replicated, age-structured groups of polyandrous red junglefowl, Gallus gallus. Male reproductive success was determined by the number of females mated (precopulatory sexual selection) and his paternity share, which was driven by the polyandry of his female partners (postcopulatory sexual selection). Pre- and postcopulatory components of male reproductive success covaried positively; males with high mating success also had high paternity share. Two male phenotypes affected male pre- and postcopulatory performance: average aggressiveness toward rival males and age. Aggressive males mated with more females and more often with individual females, resulting in higher sexual exclusivity. Similarly, younger males mated with more females and more often with individual females, suffering less intense sperm competition than older males. Older males had a lower paternity share even allowing for their limited sexual exclusivity, indicating they may produce less competitive ejaculates. These results show that—in these populations—postcopulatory sexual selection reinforces precopulatory sexual selection, consistently promoting younger and more aggressive males

    Aims The 2019 report from the European Society of Cardiology (ESC) Atlas provides a contemporary analysis of cardiovascular disease (CVD) statistics across 56 member countries, with particular emphasis on international inequalities in disease burden and healthcare delivery together with estimates of progress towards meeting 2025 World Health Organization (WHO) non-communicable disease targets. Methods and results In this report, contemporary CVD statistics are presented for member countries of the ESC. The statistics are drawn from the ESC Atlas which is a repository of CVD data from a variety of sources including the WHO, the Institute for Health Metrics and Evaluation, and the World Bank. The Atlas also includes novel ESC sponsored data on human and capital infrastructure and cardiovascular healthcare delivery obtained by annual survey of the national societies of ESC member countries. Across ESC member countries, the prevalence of obesity (body mass index ≥30 kg/m2) and diabetes has increased two- to three-fold during the last 30 years making the WHO 2025 target to halt rises in these risk factors unlikely to be achieved. More encouraging have been variable declines in hypertension, smoking, and alcohol consumption but on current trends only the reduction in smoking from 28% to 21% during the last 20 years appears sufficient for the WHO target to be achieved. The median age-standardized prevalence of major risk factors was higher in middle-income compared with high-income ESC member countries for hypertension {23.8% [interquartile range (IQR) 22.5–23.1%] vs. 15.7% (IQR 14.5–21.1%)}, diabetes [7.7% (IQR 7.1–10.1%) vs. 5.6% (IQR 4.8–7.0%)], and among males smoking [43.8% (IQR 37.4–48.0%) vs. 26.0% (IQR 20.9–31.7%)] although among females smoking was less common in middle-income countries [8.7% (IQR 3.0–10.8) vs. 16.7% (IQR 13.9–19.7%)]. There were associated inequalities in disease burden with disability-adjusted life years per 100 000 people due to CVD over three times as high in middle-income [7160 (IQR 5655–8115)] compared with high-income [2235 (IQR 1896–3602)] countries. Cardiovascular disease mortality was also higher in middle-income countries where it accounted for a greater proportion of potential years of life lost compared with high-income countries in both females (43% vs. 28%) and males (39% vs. 28%). Despite the inequalities in disease burden across ESC member countries, survey data from the National Cardiac Societies of the ESC showed that middle-income member countries remain severely under-resourced compared with high-income countries in terms of cardiological person-power and technological infrastructure. Under-resourcing in middle-income countries is associated with a severe procedural deficit compared with high-income countries in terms of coronary intervention, device implantation and cardiac surgical procedures. Conclusion A seemingly inexorable rise in the prevalence of obesity and diabetes currently provides the greatest challenge to achieving further reductions in CVD burden across ESC member countries. Additional challenges are provided by inequalities in disease burden that now require intensification of policy initiatives in order to reduce population risk and prioritize cardiovascular healthcare delivery, particularly in the middle-income countries of the ESC where need is greatest
