    Simulation Studies of the Prepulse-Mainpulse XeCl Discharge Lasers with Magnetic Switching

    A self-consistent model of a self-sustained discharge XeCl laser (Ne/Xe/HCl mixture) with prepulse-mainpulse excitation and magnetic switching which leads to high efficiency operation is described. The validity of the model is confirmed by comparing the results of the calculations with the measured time dependences of discharge voltage, current and lasing pulse for different operation modes as well as by comparing the results with the dependences of the laser output energy and efficiency on the charging voltage and capacitance of the pulse forming network for two different laser heads. The numerical evaluation has shown that our developed laser system operates under optimum conditions

    This article examines the ways in which Jewish personal belongings that have been appropriated by gentiles during, and in the aftermath of, the Holocaust have been identified, demanded back, passed down from generation to generation, and commodified. Focusing on Biłgoraj and Izbica (Poland), and Mir and Iŭje (Belarus), our objective is to determine whether the Jewish identity of personal belongings appropriated by local non-Jewish communities during, or in the aftermath of, ‘Holocaust by bullets,’ survived in the postwar communities in which they have been circulating, and define what role they played for the postwar relations between Jews and non-Jews.Peer Reviewe

    Retrieval methods of effective cloud cover for the GOME instrument: an intercomparison

    International audienceThe radiative scattering by clouds leads to errors in the retrieval of column densities and concentration profiles of atmospheric trace gas species from satellites. Moreover, the presence of clouds changes the UV actinic flux and the photo-dissociation rates of various species significantly. The Global Ozone Monitoring Experiment (GOME) instrument on the ERS-2 satellite, principally designed to retrieve trace gases in the atmosphere is also capable of detecting clouds. Four cloud fraction retrieval methods for GOME data that have been developed are discussed in this paper (the Initial Cloud Fitting Algorithm, the PMD Cloud Retrieval Algorithm, the Optical Cloud Recognition Algorithm and the Fast Retrieval Scheme for Cloud Observables). Their results of cloud fraction retrieval are compared to each-other and also to synoptic surface observations. It is shown that all studied retrieval methods calculate an effective cloud fraction that is related to a cloud with a high optical thickness. Generally, we found ICFA to produce the lowest cloud fractions, followed by OCRA, then FRESCO and PC2K along four processed tracks (+2%, +10% and +15% compared to ICFA respectively). Synoptical surface observations gave the highest absolute cloud fraction when compared with individual PMD sub-pixels of roughly the same size

    Lysosomal Targeting of Epidermal Growth Factor Receptors via a Kinase-dependent Pathway Is Mediated by the Receptor Carboxyl-terminal Residues 1022-1123

    Binding of epidermal growth factor (EGF) to its receptor induces rapid internalization and degradation of both ligand and receptor via the lysosomal pathway. To study the mechanism of intracellular sorting of EGF-EGF receptor complexes to lysosomes, NIH 3T3 cells transfected with wild-type and mutant EGF receptors were employed. The kinetics of 125I-EGF trafficking was analyzed using low concentrations of the ligand to avoid saturation of the specific sorting system. The relative size of the pool of internalized 125I-EGF-receptor complexes that were capable of recycling decreased as receptors traversed the endosomal system. The rate of 125I-EGF sequestration from the recycling pathway correlated with the rate of 125I-EGF transition from early to late endosomes as measured by Percoll gradient fractionation. Deletion of the last 63 amino acids of the EGF receptor cytoplasmic tail did not inhibit the process of sequestration and targeting to the late endosomes and lysosomes. Truncation of the 123 residues, however, resulted in impaired lysosomal targeting and increased recycling of EGF. Receptor mutant in which 165 residues were deleted displayed maximal ability to recycle and a minimal extent of sorting to the late endosomes. The data suggest that two regions of the EGF receptor molecule, residues 1022-1063 and to a lesser extent residues 1063-1123, contribute in the regulation of routing of EGF receptors to the degradation pathway. The kinase-negative receptor mutant recycled EGF more intensively compared with the wild-type receptor, and the transport of this mutant to late endosomes was inhibited. These results support the view that the receptor kinase activity is important for ligand-induced sorting of EGF receptors to the pathway of lysosomal degradation

    GABA transporter function, oligomerization state, and anchoring: correlates with subcellularly resolved FRET

    The mouse γ-aminobutyric acid (GABA) transporter mGAT1 was expressed in neuroblastoma 2a cells. 19 mGAT1 designs incorporating fluorescent proteins were functionally characterized by [^3H]GABA uptake in assays that responded to several experimental variables, including the mutations and pharmacological manipulation of the cytoskeleton. Oligomerization and subsequent trafficking of mGAT1 were studied in several subcellular regions of live cells using localized fluorescence, acceptor photobleach Förster resonance energy transfer (FRET), and pixel-by-pixel analysis of normalized FRET (NFRET) images. Nine constructs were functionally indistinguishable from wild-type mGAT1 and provided information about normal mGAT1 assembly and trafficking. The remainder had compromised [^3H]GABA uptake due to observable oligomerization and/or trafficking deficits; the data help to determine regions of mGAT1 sequence involved in these processes. Acceptor photobleach FRET detected mGAT1 oligomerization, but richer information was obtained from analyzing the distribution of all-pixel NFRET amplitudes. We also analyzed such distributions restricted to cellular subregions. Distributions were fit to either two or three Gaussian components. Two of the components, present for all mGAT1 constructs that oligomerized, may represent dimers and high-order oligomers (probably tetramers), respectively. Only wild-type functioning constructs displayed three components; the additional component apparently had the highest mean NFRET amplitude. Near the cell periphery, wild-type functioning constructs displayed the highest NFRET. In this subregion, the highest NFRET component represented ~30% of all pixels, similar to the percentage of mGAT1 from the acutely recycling pool resident in the plasma membrane in the basal state. Blocking the mGAT1 C terminus postsynaptic density 95/discs large/zona occludens 1 (PDZ)-interacting domain abolished the highest amplitude component from the NFRET distributions. Disrupting the actin cytoskeleton in cells expressing wild-type functioning transporters moved the highest amplitude component from the cell periphery to perinuclear regions. Thus, pixel-by-pixel NFRET analysis resolved three distinct forms of GAT1: dimers, high-order oligomers, and transporters associated via PDZ-mediated interactions with the actin cytoskeleton and/or with the exocyst

    Radiative forcing in the 21st century due to ozone changes in the troposphere and the lower stratosphere

    Radiative forcing due to changes in ozone is expected for the 21st century. An assessment on changes in the tropospheric oxidative state through a model intercomparison ("OxComp'') was conducted for the IPCC Third Assessment Report (IPCC-TAR). OxComp estimated tropospheric changes in ozone and other oxidants during the 21st century based on the "SRES'' A2p emission scenario. In this study we analyze the results of 11 chemical transport models (CTMs) that participated in OxComp and use them as input for detailed radiative forcing calculations. We also address future ozone recovery in the lower stratosphere and its impact on radiative forcing by applying two models that calculate both tropospheric and stratospheric changes. The results of OxComp suggest an increase in global-mean tropospheric ozone between 11.4 and 20.5 DU for the 21st century, representing the model uncertainty range for the A2p scenario. As the A2p scenario constitutes the worst case proposed in IPCC-TAR we consider these results as an upper estimate. The radiative transfer model yields a positive radiative forcing ranging from 0.40 to 0.78 W m(-2) on a global and annual average. The lower stratosphere contributes an additional 7.5-9.3 DU to the calculated increase in the ozone column, increasing radiative forcing by 0.15-0.17 W m(-2). The modeled radiative forcing depends on the height distribution and geographical pattern of predicted ozone changes and shows a distinct seasonal variation. Despite the large variations between the 11 participating models, the calculated range for normalized radiative forcing is within 25%, indicating the ability to scale radiative forcing to global-mean ozone column change
