    Structural Repertoire of HIV-1-Neutralizing Antibodies Targeting the CD4 Supersite in 14 Donors

    The site on the HIV-1 gp120 glycoprotein that binds the CD4 receptor is recognized by broadly reactive antibodies, several of which neutralize over 90% of HIV-1 strains. To understand how antibodies achieve such neutralization, we isolated CD4-binding-site (CD4bs) antibodies and analyzed 16 co-crystal structures –8 determined here– of CD4bs antibodies from 14 donors. The 16 antibodies segregated by recognition mode and developmental ontogeny into two types: CDR H3-dominated and VH-gene-restricted. Both could achieve greater than 80% neutralization breadth, and both could develop in the same donor. Although paratope chemistries differed, all 16 gp120-CD4bs antibody complexes showed geometric similarity, with antibody-neutralization breadth correlating with antibody-angle of approach relative to the most effective antibody of each type. The repertoire for effective recognition of the CD4 supersite thus comprises antibodies with distinct paratopes arrayed about two optimal geometric orientations, one achieved by CDR H3 ontogenies and the other achieved by VH-gene-restricted ontogenies

    Production of He-4 and (4) in Pb-Pb collisions at root(NN)-N-S=2.76 TeV at the LHC

    Results on the production of He-4 and (4) nuclei in Pb-Pb collisions at root(NN)-N-S = 2.76 TeV in the rapidity range vertical bar y vertical bar <1, using the ALICE detector, are presented in this paper. The rapidity densities corresponding to 0-10% central events are found to be dN/dy4(He) = (0.8 +/- 0.4 (stat) +/- 0.3 (syst)) x 10(-6) and dN/dy4 = (1.1 +/- 0.4 (stat) +/- 0.2 (syst)) x 10(-6), respectively. This is in agreement with the statistical thermal model expectation assuming the same chemical freeze-out temperature (T-chem = 156 MeV) as for light hadrons.     Elementanalytik und Isotopenverhältnisbestimmungen an humanbiologischen Materialien

    Ziel der Arbeit war es, festzustellen, inwieweit die Bestimmung der Konzentrationen und der Isotopenverhältnisse verschiedener Elemente in humanbiologischen Materialien dazu geeignet ist, Informationen über Herkunft und Wohnort oder zurückliegende Ortswechsel einer Person aufzuzeigen. Dieses Forschungsprojekt wurde in Zusammenarbeit mit dem Bundeskriminalamt Wiesbaden durchgeführt. Dort werden in dem seit 2008 laufenden Projekt „Isohaar“ vor allem Haare, aber auch Fingernägel, von Personen vor, während und nach einer Auslandsreise gesammelt. An diesen Materialien sollte überprüft werden, ob ein Ortswechsel Einfluss auf die Elementgehalte und/oder Isotopenverhältnisse hat.rnAls elementanalytische Methoden wurden die Neutronenaktivierungsanalyse (NAA) und die induktiv gekoppeltes Plasma-Massenspektrometrie (ICP-MS) an flüssigen Aufschlüssen der Proben, sowie direkt an den Proben mittels Laser Ablation (LA-ICP-MS) erprobt. Die Isotopenverhältnisse der Elemente Kohlenstoff und Stickstoff wurden mittels Isotopenverhältnis-Massenspektrometrie (IRMS) bestimmt, die der schweren Elemente Strontium und Blei mittels Thermionen-Massenspektrometrie (TIMS).rnMittels Flüssig-ICP-MS wurde ein Konzentrationsanstieg für einige Elemente (z.B. Na, Mg Ba, Cu, Sr und Ca) vom Haaransatz (jüngster Haarabschnitt) zur Haarspitze (ältester Haarabschnitt) einer analysierten Strähne gefunden. Die Spender dieser Proben hatten keinen Ortswechsel vollzogen. Bei Haarproben von Personen, die einen Aufenthaltsortswechsel erlebt hatten, zeigten sich markante Konzentrationserhöhungen bei Eisen, Mangan, Titan und Blei in den betreffenden Abschnitten. Diese Elemente scheinen evtl. das Potential zu haben, Ortswechsel anzuzeigen.rnBei den Isotopenverhältnissen von Kohlenstoff und Stickstoff konnten einige Unterschiede zwischen verschiedenen Aufenthaltsorten einer Person gefunden werden. Diese Änderungen werden mit der häufig unausweichlichen unterschiedlichen Ernährungsweise des Haarspenders bei Reisen in Zusammenhang gebracht. Die Isotopenverhältnisse von Strontium und Blei konnten bisher nicht einem bestimmten Land oder einer Region zugeordnet werden. Die Proben zwischen den einzelnen Teilnehmern unterschieden sich jedoch. rnFür die Zukunft sollte versucht werden, die Ergebnisse der Analysenmethoden miteinander zu kombinieren, um aussagekräftigere Hinweise auf einen möglichen Ortswechsel zu erhalten.rnThe aim of the present study was to endeavour whether it is possible to obtain information about the travelling behaviour of a person by determining the elemental concentration and isotope ratios of some special elements in biological materials. This project was realised in association with the Federal Criminal Police Office (BKA, Wiesbaden). Since 2008, a study called “Isohaar” has started there. In this project, strands of hair and some fingernails of different persons are collected before, during and after a foreign trip. One goal of the examinations of these materials is to check if a change of location can influence the elemental concentrations and/or the isotope ratios.rnAs elemental analyses, the following methods are used: neutron activation analysis (NAA), and inductively coupled plasma mass spectrometry (ICP-MS). Dissolved hair samples were analysed by the liquid ICP-MS method, and the untreated strands of hair were analysed by connecting a laser ablation system to the ICP-MS (LA-ICP-MS). The isotope ratios of carbon and nitrogen are determined by isotope ratio mass spectrometry (IRMS). The heavy elements strontium and lead are analysed by thermal ionization mass spectrometry (TIMS).rnBy liquid ICP-MS method, the concentrations of some elements (e.g. Na, Mg, Ba, Cu, Sr, and Ca) rise from the hairline (youngest interval) to the hair-ends (oldest interval) of a strand of hair. The owner of these strands didn’t make a change of location. Other samples of hair, its donor had changed its living space, showed a distinct higher concentration of iron, manganese, titanium, and lead in the relevant intervals. These four elements seem to have the capability of indicating a change of place by changing its concentrations.rnIn addition, differences between samples from different locations were also detected by the isotope ratios of carbon and nitrogen. These changes are frequently correlated to the nutrition of a person. For the isotope ratios of strontium and lead, no assignment to a country or a region could be made. The samples of the single participants showed different results.rnOne goal for the future should be to combine the results of the analyses to obtain more promising clues for a change of location.r

    Paramutation is a form of non-Mendelian inheritance in which the expression of a paramutable allele changes when it encounters a paramutagenic allele. This change in expression of the paramutable alleles is stably inherited even after segregation of both alleles. While the discovery of paramutation and studies of its underlying mechanism were made with alleles that change plant pigmentation, paramutation-like phenomena are known to modulate the expression of other traits and in other eukaryotes, and many cases have probably gone undetected. It is likely that epigenetic mechanisms are responsible for the phenomenon, as paramutation forms epialleles, genes with identical sequences but different expression states. This could account for the intergenerational inheritance of the paramutated allele, providing profound evidence that triggered epigenetic changes can be maintained over generations. Here, we use a case of paramutation that affects a transgenic selection reporter gene in tetraploid Arabidopsis thaliana. Our data suggest that different types of small RNA are derived from paramutable and paramutagenic epialleles. In addition, deletion of a repeat within the epiallele changes its paramutability. Further, the temperature during the growth of the epiallelic hybrids determines the degree and timing of the allelic interaction. The data further make it plausible why paramutation in this system becomes evident only in the segregating F2 population of tetraploid plants containing both epialleles. In summary, the results support a model for polyploidy-associated paramutation, with similarities as well as distinctions from other cases of paramutation

    Patients With Multiple Myeloma or Monoclonal Gammopathy of Undetermined Significance

    Background: Multiple myeloma (MM) is a malignant plasma-cell disease that arises on the basis of a so-called monoclonal gammopathy of undetermined significance (MGUS). The median age at disease onset is over 70. In Germany, there are approximately eight new cases per 100 000 inhabitants per year, or about 6000 new patients nationwide each year. Methods: To prepare this clinical practice guideline, a systematic literature review was carried out in medical databases (MEDLINE, CENTRAL), guideline databases (GIN), and the search portal of the German Institute for Quality and Efficiency in Health Care (IQWiG). The recommendations to be issued were based on two international guidelines, 40 dossier evaluations and systematic reviews, 10 randomized controlled trials, and 37 observational studies and finalized in a structured consensus process. Results: Because of its prognostic relevance, the use of the International Staging System (ISS) is recommended to stage MM and related plasma-cell neoplasms. When symptomatic MM is diagnosed, it is recommended to determine the extent of skeletal involvement by whole-body computed tomography. The indications for treatment shall be determined on the basis of the SLiM-CRAB criteria; in all patients with MM it is recommended to include the biological (rather than chronological) age in the decision -making process. In suitable patients, it is recommended that initial treatment includes high-dose therapy, followed by main -tenance treatment. Even without high-dose treatment, a median progression-free survival of more than three years can be achieved with combination therapies. For the treatment of relapse, combinations of three drugs are more effective than doublet regimens with a median progression-free survival ranging from 10 to 45 months, depending on the study and prior therapy. Fol-lowing anti-myeloma therapy, it is recommended to promptly offer physical exercise adapted to individual abilities to all patients who have the potential for rehabilitation, so that their quality of life can be sustained and improved. Conclusion: This new clinical practice guideline addresses, in particular, the modalities of care that can be offered in addition to systemic antineoplastic therapy.
