    Exosomes in Cancer Diagnosis and Radiation Therapy

    Exosomes are a subgroup of extracellular vesicles that are released by all types of cells, including tumor cells, and mediate intercellular communication via the transport of various intracellular components, including microRNAs, messenger RNAs, and proteins. Radiation produces reactive oxygen species and induces DNA double-strand break in cancer cells and normal cells. Cancer cells have severe damage and die by irradiation, but normal cells can keep proliferation with their high DNA repair ability. Irradiated cells generate communication signals and cause biological changes in neighboring or distant non-irradiated cells. This review outlines the role of exosomes in radiation therapy. In the tumor microenvironment, exosomes are considered to regulate cell survival, migration, and resistance to therapy by interacting with vascular endothelial cells and various types of immune cells. Nowadays, radiation therapy is typically combined with immunotherapy. Regulation of the activity of exosomes may overcome the problem of resistance to immunotherapy. Furthermore, exosomes can attenuate resistance to chemotherapy by transporting certain types of microRNA. The current evidence suggests that exosomes may be useful in the diagnosis and treatment of cancer in the future

    New drugs and vaccines for drug-resistant Mycobacterium tuberculosis infections

    Tuberculosis remains the most common cause of death due to a single infective organism. Despite the availability of a vaccine and chemotherapeutic options, the global disease burden remains relatively unaffected. The ability of the mycobacterial etiological agents to adopt a semidormant, phenotypically drug-resistant state requires that chemotherapy is both complex and lengthy. The emergence of drug resistance has raised the possibility of virtually untreatable tuberculosis. Furthermore, the currently used bacillus Calmette–Guerin vaccine has had mixed success in protecting susceptible populations. Given this backdrop, the need for novel anti-infectives and more effective vaccines is clearly evident.     Diminished Medial Prefrontal Activity behind Autistic Social Judgments of Incongruent Information

    Individuals with autism spectrum disorders (ASD) tend to make inadequate social judgments, particularly when the nonverbal and verbal emotional expressions of other people are incongruent. Although previous behavioral studies have suggested that ASD individuals have difficulty in using nonverbal cues when presented with incongruent verbal-nonverbal information, the neural mechanisms underlying this symptom of ASD remain unclear. In the present functional magnetic resonance imaging study, we compared brain activity in 15 non-medicated adult males with high-functioning ASD to that of 17 age-, parental-background-, socioeconomic-, and intelligence-quotient-matched typically-developed (TD) male participants. Brain activity was measured while each participant made friend or foe judgments of realistic movies in which professional actors spoke with conflicting nonverbal facial expressions and voice prosody. We found that the ASD group made significantly less judgments primarily based on the nonverbal information than the TD group, and they exhibited significantly less brain activity in the right inferior frontal gyrus, bilateral anterior insula, anterior cingulate cortex/ventral medial prefrontal cortex (ACC/vmPFC), and dorsal medial prefrontal cortex (dmPFC) than the TD group. Among these five regions, the ACC/vmPFC and dmPFC were most involved in nonverbal-information-biased judgments in the TD group. Furthermore, the degree of decrease of the brain activity in these two brain regions predicted the severity of autistic communication deficits. The findings indicate that diminished activity in the ACC/vmPFC and dmPFC underlies the impaired abilities of individuals with ASD to use nonverbal content when making judgments regarding other people based on incongruent social information

    Reconstructing the Inflaton Potential --- an Overview

    We review the relation between the inflationary potential and the spectra of density (scalar) perturbations and gravitational waves (tensor perturbations) produced, with particular emphasis on the possibility of reconstructing the inflaton potential from observations. The spectra provide a potentially powerful test of the inflationary hypothesis; they are not independent but instead are linked by consistency relations reflecting their origin from a single inflationary potential. To lowest-order in a perturbation expansion there is a single, now familiar, relation between the tensor spectral index and the relative amplitude of the spectra. We demonstrate that there is an infinite hierarchy of such consistency equations, though observational difficulties suggest only the first is ever likely to be useful. We also note that since observations are expected to yield much better information on the scalars than on the tensors, it is likely to be the next-order version of this consistency equation which will be appropriate, not the lowest-order one. If inflation passes the consistency test, one can then confidently use the remaining observational information to constrain the inflationary potential, and we survey the general perturbative scheme for carrying out this procedure. Explicit expressions valid to next-lowest order in the expansion are presented. We then briefly assess the prospects for future observations reaching the quality required, and consider a simulated data set that is motivated by this outlook.Comment: 69 pages standard LaTeX plus 4 postscript figures.
