    Seismological structure of the 1.8 Ga Trans-Hudson Orogen of North America

    Precambrian tectonic processes are debated: what was the nature and scale of orogenic events on the younger, hotter, and more ductile Earth? Northern Hudson Bay records the Paleoproterozoic collision between the Western Churchill and Superior plates—the ∼1.8 Ga Trans-Hudson Orogeny (THO)—and is an ideal locality to study Precambrian tectonic structure. Integrated field, geochronological, and thermobarometric studies suggest that the THO was comparable to the present-day Himalayan-Karakoram-Tibet Orogen (HKTO). However, detailed understanding of the deep crustal architecture of the THO, and how it compares to that of the evolving HKTO, is lacking. The joint inversion of receiver functions and surface wave data provides new Moho depth estimates and shear velocity models for the crust and uppermost mantle of the THO. Most of the Archean crust is relatively thin (∼39 km) and structurally simple, with a sharp Moho; upper-crustal wave speed variations are attributed to postformation events. However, the Quebec-Baffin segment of the THO has a deeper Moho (∼45 km) and a more complex crustal structure. Observations show some similarity to recent models, computed using the same methods, of the HKTO crust. Based on Moho character, present-day crustal thickness, and metamorphic grade, we support the view that southern Baffin Island experienced thickening during the THO of a similar magnitude and width to present-day Tibet. Fast seismic velocities at >10 km below southern Baffin Island may be the result of partial eclogitization of the lower crust during the THO, as is currently thought to be happening in Tibet

    How to ask sensitive questions in conservation: A review of specialized questioning techniques

    Tools for social research are critical for developing an understanding of conservation problems and assessing the feasibility of conservation actions. Social surveys are an essential tool frequently applied in conservation to assess both people’s behaviour and to understand its drivers. However, little attention has been given to the weaknesses and strengths of different survey tools. When topics of conservation concern are illegal or otherwise sensitive, data collected using direct questions are likely to be affected by non-response and social desirability biases, reducing their validity. These sources of bias associated with using direct questions on sensitive topics have long been recognised in the social sciences but have been poorly considered in conservation and natural resource management. We reviewed specialized questioning techniques developed in a number of disciplines specifically for investigating sensitive topics. These methods ensure respondent anonymity, increase willingness to answer, and critically, make it impossible to directly link incriminating data to an individual. We describe each method and report their main characteristics, such as data requirements, possible data outputs, availability of evidence that they can be adapted for use in illiterate communities, and summarize their main advantages and disadvantages. Recommendations for their application in conservation are given. We suggest that the conservation toolbox should be expanded by incorporating specialized questioning techniques, developed specifically to increase response accuracy. By considering the limitations of each survey technique, we will ultimately contribute to more effective evaluations of conservation interventions and more robust policy decisions

    Decay-accelerating factor expression in the rat kidney is restricted to the apical surface of podocytes.BackgroundDecay-accelerating factor (DAF) has inhibitory activity toward complement C3 and C5 convertases. DAF is present in human glomeruli and on cultured human glomerular visceral epithelial cells (GEC). We studied the distribution and function of rat DAF.MethodsFunction-neutralizing antibodies (Abs) were raised against DAF. The distribution of DAF in vivo was determined by immunoelectron microscopy. Functional studies were performed in cultured GEC and following IV injection of anti-DAF Abs into rats.ResultsDAF was present exclusively on the apical surfaces of GEC, and was not present on the basal surfaces of GEC, nor other glomerular or kidney cells. DAF was functionally active on cultured GEC, and served to limit complement activation in concert with CD59, an inhibitor of C5b-9 formation. Upon injection into normal rats, anti-DAF F(ab′)2 Abs bound to GEC in vivo, yet there was no evidence for complement activation and animals did not develop abnormal albuminuria. Anti-megalin complement-activating IgG Abs were “planted” on GEC, which activated complement as evidenced by the presence of C3d on GEC. Attempts to inhibit DAF function with anti-DAF Abs did not affect the quantity of complement activation by these anti-megalin Abs, nor did it lead to development of abnormal albuminuria. In contrast, in the puromycin aminonucleoside model of GEC injury and proteinuria, anti-DAF Abs slowed the recovery from renal failure that occurs in this model.ConclusionIn cultured rat GEC, DAF is an effective complement regulator. In vivo, DAF is present on GEC apical surfaces. Yet, it appears that DAF is not essential to prevent complement activation from occurring under normal circumstances and in those cases in which complement-activating Abs are present on the basal surfaces of GEC in vivo. However, in proteinuric conditions, DAF appears to be protective to GEC

    Evaluation of Negation and Uncertainty Detection and its Impact on Precision and Recall in Search

    Radiology reports contain information that can be mined using a search engine for teaching, research, and quality assurance purposes. Current search engines look for exact matches to the search term, but they do not differentiate between reports in which the search term appears in a positive context (i.e., being present) from those in which the search term appears in the context of negation and uncertainty. We describe RadReportMiner, a context-aware search engine, and compare its retrieval performance with a generic search engine, Google Desktop. We created a corpus of 464 radiology reports which described at least one of five findings (appendicitis, hydronephrosis, fracture, optic neuritis, and pneumonia). Each report was classified by a radiologist as positive (finding described to be present) or negative (finding described to be absent or uncertain). The same reports were then classified by RadReportMiner and Google Desktop. RadReportMiner achieved a higher precision (81%), compared with Google Desktop (27%; p < 0.0001). RadReportMiner had a lower recall (72%) compared with Google Desktop (87%; p = 0.006). We conclude that adding negation and uncertainty identification to a word-based radiology report search engine improves the precision of search results over a search engine that does not take this information into account. Our approach may be useful to adopt into current report retrieval systems to help radiologists to more accurately search for radiology reports

    Infection and genotype remodel the entire soybean transcriptome

    <p>Abstract</p> <p>Background</p> <p>High throughput methods, such as high density oligonucleotide microarray measurements of mRNA levels, are popular and critical to genome scale analysis and systems biology. However understanding the results of these analyses and in particular understanding the very wide range of levels of transcriptional changes observed is still a significant challenge. Many researchers still use an arbitrary cut off such as two-fold in order to identify changes that may be biologically significant. We have used a very large-scale microarray experiment involving 72 biological replicates to analyze the response of soybean plants to infection by the pathogen <it>Phytophthora sojae </it>and to analyze transcriptional modulation as a result of genotypic variation.</p> <p>Results</p> <p>With the unprecedented level of statistical sensitivity provided by the high degree of replication, we show unambiguously that almost the entire plant genome (97 to 99% of all detectable genes) undergoes transcriptional modulation in response to infection and genetic variation. The majority of the transcriptional differences are less than two-fold in magnitude. We show that low amplitude modulation of gene expression (less than two-fold changes) is highly statistically significant and consistent across biological replicates, even for modulations of less than 20%. Our results are consistent through two different normalization methods and two different statistical analysis procedures.</p> <p>Conclusion</p> <p>Our findings demonstrate that the entire plant genome undergoes transcriptional modulation in response to infection and genetic variation. The pervasive low-magnitude remodeling of the transcriptome may be an integral component of physiological adaptation in soybean, and in all eukaryotes.</p

    Studies on the antiobesity effect of zinc-α2-glycoprotein in the ob/ob mouse

    OBJECTIVE: To investigate the mechanism of the lipid depletion by zinc-a(2)-glycoprotein (ZAG). DESIGN: Studies were conducted in the ob/ob mouse, or on isolated adipocytes from these animals or their lean counterparts. RESULTS: Treatment of these animals for 15 days with ZAG (100? µg, intravenously, daily) resulted in a reduction of body weight of 6.55? g compared with phosphate-buffered saline-treated controls, without a change in food or water intake, but with a 0.4?°C rise in rectal temperature. ZAG-treated mice had a 30% reduction in carcass fat mass and a twofold increase in weight of brown adipose tissue. Epididymal adipocytes from ZAG-treated mice showed an increased expression of ZAG and hormone-sensitive lipase (HSL), and this was maintained for a further 3 days in the absence of ZAG. There was an increased lipolytic response to isoproterenol, which was retained for 3 days in vitro in the absence of ZAG. Expression of HSL was also increased in subcutaneous and visceral adipose tissue, as was also adipose triglyceride lipase (ATGL). There was a rapid loss of labelled lipid from epididymal adipose tissue of ZAG-treated mice, but not from the other depots, reflecting the difference in sensitivity to lipolytic stimuli. The increased expression of HSL and ATGL may involve the extracellular signal-regulated kinase (ERK) pathway, as the active (phospho) form was upregulated in all adipose depots after ZAG administration, whereas in vitro studies showed induction of HSL and ATGL by ZAG to be attenuated by PD98059, an inhibitor of the ERK pathway. CONCLUSION: These results suggest that ZAG not only induces direct lipolysis, but also sensitizes adipose tissue to other lipolytic stimuli

    Non local spatial and angular matching : enabling higher spatial resolution diffusion MRI datasets through adaptive denoising

    Diffusion magnetic resonance imaging (MRI) datasets suffer from low Signal-to-Noise Ratio (SNR), especially at high b-values. Acquiring data at high b-values contains relevant information and is now of great interest for microstructural and connectomics studies. High noise levels bias the measurements due to the non-Gaussian nature of the noise, which in turn can lead to a false and biased estimation of the diffusion parameters. Additionally, the usage of in-plane acceleration techniques during the acquisition leads to a spatially varying noise distribution, which depends on the parallel acceleration method implemented on the scanner. This paper proposes a novel diffusion MRI denoising technique that can be used on all existing data, without adding to the scanning time. We first apply a statistical framework to convert both stationary and non stationary Rician and non central Chi distributed noise to Gaussian distributed noise, effectively removing the bias. We then introduce a spatially and angular adaptive denoising technique, the Non Local Spatial and Angular Matching (NLSAM) algorithm. Each volume is first decomposed in small 4D overlapping patches, thus capturing the spatial and angular structure of the diffusion data, and a dictionary of atoms is learned on those patches. A local sparse decomposition is then found by bounding the reconstruction error with the local noise variance. We compare against three other state-of-the-art denoising methods and show quantitative local and connectivity results on a synthetic phantom and on an in-vivo high resolution dataset. Overall, our method restores perceptual information, removes the noise bias in common diffusion metrics, restores the extracted peaks coherence and improves reproducibility of tractography on the synthetic dataset. On the 1.2 mm high resolution in-vivo dataset, our denoising improves the visual quality of the data and reduces the number of spurious tracts when compared to the noisy acquisition. Our work paves the way for higher spatial resolution acquisition of diffusion MRI datasets, which could in turn reveal new anatomical details that are not discernible at the spatial resolution currently used by the diffusion MRI community

    Precambrian Plate Tectonics in Northern Hudson Bay: Evidence From P and S Wave Seismic Tomography and Analysis of Source Side Effects in Relative Arrival-Time Data Sets

    The geology of northern Hudson Bay, Canada, documents more than 2 billion years of history including the assembly of Precambrian and Archean terranes during several Paleoproterozoic orogenies, culminating in the Trans‐Hudson Orogen (THO) ∼1.8 Ga. The THO has been hypothesized to be similar in scale and nature to the ongoing Himalaya‐Karakoram‐Tibetan orogen, but the nature of lithospheric terrane boundaries, including potential plate‐scale underthrusting, is poorly understood. To address this problem, we present new P and S wave tomographic models of the mantle seismic structure using data from recent seismograph networks stretching from northern Ontario to Nunavut (60–100∘W and 50–80∘N). The large size of our network requires careful mitigation of the influence of source side structure that contaminates our relative arrival time residuals. Our tomographic models reveal a complicated internal structure in the Archean Churchill plate. However, no seismic wave speed distinction is observed across the Snowbird Tectonic Zone, which bisects the Churchill. The mantle lithosphere in the central region of Hudson Bay is distinct from the THO, indicating potential boundaries of microcontinents and lithospheric blocks between the principal colliders. Slow wave speeds underlie southern Baffin Island, the leading edge of the generally high wave speed Churchill plate. This is interpreted to be Paleoproterozoic material underthrust beneath Baffin Island in a modern‐style subduction zone setting

    Competitive Asymmetries in the Use of Supplementary Food by the Endangered Iberian Lynx (Lynx pardinus)

    Background: As a conservation tool, supplementary feeding programs may be directed to specific individuals or sectors of the target population whose productivity or survival is thought to be limited by food scarcity. However, the use of supplemental food by different sex and age classes has received little attention. We studied individual variation in the access of the endangered Iberian lynx (Lynx pardinus) to supplementary food. Methodology/Principal Findings: From 5349 pictures taken with automatic cameras placed in 25 feeding stations, we identified 28 individuals whose sex and age were known. All individuals known to live in areas subjected to supplementation regularly visited feeding stations. Food consumption was not proportional to expected variations in energy demand within sex and age classes. Food consumption by males was higher than by females, and increased with age, in agreement with a despotic distribution. Food consumption also increased with lynx body mass, and this pattern held for individuals sharing the same breeding territories. The access of inferior competitors increased with the number of feeding stations available within lynx territories. Conclusions/Significance: All lynx exposed to food supplementation made a regular use of extra food but individuals predicted to be competitively dominant visited stations more frequently than subordinates of the same breeding territory. Our results suggest that insufficient provision of supplementary food could restrict the access of juveniles, or even adult females, to feeding stations. Limited consumption by these target individuals may compromise the efficiency of the supplementary feeding programme at the population level, in endangered species that, as the Iberian lynx, exhibit marked sexual dimorphism in body size.Peer reviewe
