    Kwanu Local – A High Yielding Traditional Maize Cultivar of Jaunsar Tribal Region of Uttarakhand and a Promising Genetic Resource for Maize Improvement

    164-169Globally maize is the second most important crop in terms of acreage where as in India it ranks third after wheat and rice. Maize productivity has been breaking unprecedented barriers owing chiefly to wide scale cultivation of high yielding hybrids. Sustaining high yields of hybrids necessitates continued efforts for creation of novel gene assemblages and/or discovery of such naturally existing constellations. Traditional local cultivars are an important source of the latter. Kwanu Local is an example of such a traditional cultivar that contributes significantly towards sustaining food, fodder and fuel requirement of Jaunsari tribal community of Kwanu cluster in Dehradun district of Uttarakhand. It is a tall, high yielding, medium duration, semi-dent yellow bold-grained cultivar and owes its characteristic present form to multiple infusions from diverse populations that have taken place over the long history of its cultivation in the region. Its cultivation over a vast contiguous expanse and the selection practices followed by the farming community ensure maintenance of high heterozygosity in the population, assuring sustained high yields. The cultivar possesses many desirable features that make it a potential genetic resource for a variety of traits of agronomic importance (cob length, cob girth, number of kernels/row and kernel size). Its use for increasing kernel size has been well demonstrated. Kernel size in the backcross progenies involving inbreds VQL 1 (255 g) and V 400 (215 g) as recipients and Kwanu Local (343 g) as donor exhibited kernel size range of 260-293 g (VQL 1 x Kwanu Local) and 228-245 g (V 400 x Kwanu Local), showing increase of 6-14 and 2-15%, respectively, in the two crosses. With its local adaptability and high yield coupled with other desirable traits, Kwanu Local holds potential as a promising genetic resource for maize improvement

    Dosimetric effect of intra-fractional and inter-fractional target motion in lung cancer radiotherapy techniques

    Purpose: The purpose of present study was to experimentally evaluate the dosimetric uncertainties in 3-dimensional conformal radiotherapy (3DCRT), dynamic intensity modulated radiotherapy (D-IMRT), step-shoot (SS-IMRT), and volumetric modulated arc therapy (VMAT) treatment delivery techniques due to intra- and inter-fractional target motion. Methods: A previously treated lung patient was selected for this study and was replanned for 60 Gy in 30 fractions using four techniques (3DCRT, D-IMRT, SS-IMRT, and VMAT). These plans were delivered in a clinical linear accelerator equipped with HexaPOD™ evo RT System. The target dose of static QUASAR phantom was calculated that served as reference dose to the target. The QUASAR respiratory body phantom along with patients breathing wave form and HexaPOD™ evo RT System was used to simulate the intra-fraction and inter-fraction motions. Dose measurements were done by applying the intra-fractional and inter-fractional motions in all the four treatment delivery techniques.Results: The maximum percentage deviation in a single field was -4.3%, 10.4%, and -12.2% for 3DCRT, D-IMRT and SS-IMRT deliveries, respectively. Similarly, the deviation for a single fraction was -1.51%, -1.88%, -2.22%, and -3.03% for 3DCRT, D-IMRT, SS-IMRT and VMAT deliveries, respectively. Conclusion: The impact of inter-fractional and intra-fractional uncertainties calculated as deviation between dynamic and static condition dose was large in some fractions, however average deviation calculated for thirty fractions was well within 0.5% in all the four techniques. Therefore, inter- and intra-fractional uncertainties could be concern in fewer fraction treatments such as stereotactic body radiation therapy, and should be used in conjunction with intra- and inter-fractional motion management techniques

    The Impact of Antiretroviral Therapy in a Cohort of HIV Infected Patients Going in and out of the San Francisco County Jail

    BackgroundJails are an important venue of HIV care and a place for identification, treatment and referral for care. HIV infected inmates in the San Francisco County jail are offered antiretroviral treatment (ART), which many take only while in jail. We evaluated the effect of ART administration in a cohort of jail inmates going in and out of jail over a nine year period.Methodology/Principal FindingsIn this retrospective study, we examined inmates with HIV going in and out of jail. Inmates were categorized by patterns of ART use: continuous ART - ART both in and out of jail, [...] more likely to have higher VL than inmates on continuous ART. Furthermore, Inmates on intermittent ART and never-on ART lost 1.60; 95%CI (1.06, 2.13) and 1.97; 95%CI (0.96, 3.00) more CD4 cells per month, respectively, compared to continuously treated inmates. The continuous ART inmates gained 0.67CD4 cells/month.Conclusions/SignificanceContinuous ART therapy in jail inmate's benefits CD4 cell counts and control of VL especially compared to those who never took ART. Although jail inmates on intermittent ART were more likely to lose CD4 cells and experience higher VL over time than those on continuous ART, CD4 cell loss was slower in these inmates as compared to inmates never on ART. Further studies are needed to evaluate whether or not intermittent ART provides some benefit in outcome if continuous ART is not possible or likely

    The Spatial Heterogeneity between Japanese Encephalitis Incidence Distribution and Environmental Variables in Nepal

    To identify potential environmental drivers of Japanese Encephalitis virus (JE) transmission in Nepal, we conducted an ecological study to determine the spatial association between 2005 Nepal JE incidence, and climate, agricultural, and land-cover variables at district level.District-level data on JE cases were examined using Local Indicators of Spatial Association (LISA) analysis to identify spatial clusters from 2004 to 2008 and 2005 data was used to fit a spatial lag regression model with climate, agriculture and land-cover variables.Prior to 2006, there was a single large cluster of JE cases located in the Far-West and Mid-West terai regions of Nepal. After 2005, the distribution of JE cases in Nepal shifted with clusters found in the central hill areas. JE incidence during the 2005 epidemic had a stronger association with May mean monthly temperature and April mean monthly total precipitation compared to mean annual temperature and precipitation. A parsimonious spatial lag regression model revealed, 1) a significant negative relationship between JE incidence and April precipitation, 2) a significant positive relationship between JE incidence and percentage of irrigated land 3) a non-significant negative relationship between JE incidence and percentage of grassland cover, and 4) a unimodal non-significant relationship between JE Incidence and pig-to-human ratio.JE cases clustered in the terai prior to 2006 where it seemed to shift to the Kathmandu region in subsequent years. The spatial pattern of JE cases during the 2005 epidemic in Nepal was significantly associated with low precipitation and the percentage of irrigated land. Despite the availability of an effective vaccine, it is still important to understand environmental drivers of JEV transmission since the enzootic cycle of JEV transmission is not likely to be totally interrupted. Understanding the spatial dynamics of JE risk factors may be useful in providing important information to the Nepal immunization program

    Quality and Reporting of Diagnostic Accuracy Studies in TB, HIV and Malaria: Evaluation Using QUADAS and STARD Standards

    BackgroundPoor methodological quality and reporting are known concerns with diagnostic accuracy studies. In 2003, the QUADAS tool and the STARD standards were published for evaluating the quality and improving the reporting of diagnostic studies, respectively. However, it is unclear whether these tools have been applied to diagnostic studies of infectious diseases. We performed a systematic review on the methodological and reporting quality of diagnostic studies in TB, malaria and HIV.MethodsWe identified diagnostic accuracy studies of commercial tests for TB, malaria and HIV through a systematic search of the literature using PubMed and EMBASE (2004–2006). Original studies that reported sensitivity and specificity data were included. Two reviewers independently extracted data on study characteristics and diagnostic accuracy, and used QUADAS and STARD to evaluate the quality of methods and reporting, respectively.FindingsNinety (38%) of 238 articles met inclusion criteria. All studies had design deficiencies. Study quality indicators that were met in less than 25% of the studies included adequate description of[...] and description of the team executing the test and management of indeterminate/outlier results (both 17%). The use of STARD was not explicitly mentioned in any study. Only 22% of 46 journals that published the studies included in this review required authors to use STARD

The European Respiratory Society (ERS), Health Effects Institute (HEI), and International Society for Environmental Epidemiology (ISEE) organised a joint meeting on May 24, 2023 in Brussels, Belgium, to review and critically evaluate the latest evidence on the health effects of air pollution and discuss ongoing revisions of the European Ambient Air Quality Directives (AAQDs).     Sphingolipid imbalance is the culprit in a variety of neurological diseases, some affecting the myelin sheath. We have used whole-exome sequencing in patients with undetermined leukoencephalopathies to uncover the endoplasmic reticulum lipid desaturase DEGS1 as the causative gene in 19 patients from 13 unrelated families. Shared features among the cases include severe motor arrest, early nystagmus, dystonia, spasticity, and profound failure to thrive. MRI showed hypomyelination, thinning of the corpus callosum, and progressive thalamic and cerebellar atrophy, suggesting a critical role of DEGS1 in myelin development and maintenance. This enzyme converts dihydroceramide (DhCer) into ceramide (Cer) in the final step of the de novo biosynthesis pathway. We detected a marked increase of the substrate DhCer and DhCer/Cer ratios in patients' fibroblasts and muscle. Further, we used a knockdown approach for disease modeling in Danio rerio, followed by a preclinical test with the first-line treatment for multiple sclerosis, fingolimod (FTY720, Gilenya). The enzymatic inhibition of Cer synthase by fingolimod, 1 step prior to DEGS1 in the pathway, reduced the critical DhCer/Cer imbalance and the severe locomotor disability, increasing the number of myelinating oligodendrocytes in a zebrafish model. These proof-of-concept results pave the way to clinical translation

    Performance of the CMS Cathode Strip Chambers with Cosmic Rays

    The Cathode Strip Chambers (CSCs) constitute the primary muon tracking device in the CMS endcaps. Their performance has been evaluated using data taken during a cosmic ray run in fall 2008. Measured noise levels are low, with the number of noisy channels well below 1%. Coordinate resolution was measured for all types of chambers, and fall in the range 47 microns to 243 microns. The efficiencies for local charged track triggers, for hit and for segments reconstruction were measured, and are above 99%. The timing resolution per layer is approximately 5 ns

    Performance of CMS muon reconstruction in pp collision events at sqrt(s) = 7 TeV

    The performance of muon reconstruction, identification, and triggering in CMS has been studied using 40 inverse picobarns of data collected in pp collisions at sqrt(s) = 7 TeV at the LHC in 2010. A few benchmark sets of selection criteria covering a wide range of physics analysis needs have been examined. For all considered selections, the efficiency to reconstruct and identify a muon with a transverse momentum pT larger than a few GeV is above 95% over the whole region of pseudorapidity covered by the CMS muon system, abs(eta) < 2.4, while the probability to misidentify a hadron as a muon is well below 1%. The efficiency to trigger on single muons with pT above a few GeV is higher than 90% over the full eta range, and typically substantially better. The overall momentum scale is measured to a precision of 0.2% with muons from Z decays. The transverse momentum resolution varies from 1% to 6% depending on pseudorapidity for muons with pT below 100 GeV and, using cosmic rays, it is shown to be better than 10% in the central region up to pT = 1 TeV.
