    Estudio de la estabilidad química de la curcumina en medio acuoso: formación de compuestos supramoleculares con β-ciclodextrina

    En este trabajo se estudia el impacto de diferentes variables experimentales sobre la estabilidad de la curcumina evaluando el impacto que tienen sobre la respuesta de espectrofotométrica y voltamperométrica. A partir de esta información se hace un estudio de estabilidad obteniendo valores de porcentaje de degradación así como valores de velocidad de degradación de la curcumina en medio acuoso a diferentes valores de pH bajo dos condiciones experimentales: a) sin control experimental y b) bajo condiciones controladas. Siendo este último definido por: la protección de las disoluciones de trabajo de la luz incidente, ausencia de O2(g) mediante el burbujeo con gas N2(g) así como una atmósfera inerte con este mismo gas y una temperatura constante (21.0 ± 0.1) °C. Conociendo esta información, se propone un método para calcular las constantes de acidez de la curcumina en medio acuoso. Adicionalmente se exploran dos estrategias para disminuir la velocidad de degradación de la curcumina en medio acuoso mediante: 1) la formación de compuestos de coordinación entre la curcumina y el zinc y 2) la formación de compuestos supramoleculares entre la curcumina y la β-ciclodextrina siendo esta última la que mejores resultados ofrece. A partir de esto se propone el diseño de un método analítico de cuantificación en medio acuoso bajo condiciones experimentales controladas en medio amortiguado de β-ciclodextrina y pH

    Mechanical properties of dense cordierite discs sintered by solar radiation heating

    Consolidation of cordierite disc specimens was undertaken under concentrated solar beam in a solar furnace at PSA (Plataforma Solar de Almerıá). Satisfactory extent of densification was achieved by the present solar-sintering experiment. The mechanical properties measured for the solar-sintered cordierite test pieces were; density ¼ 2:45 0:02 g/cm3, Vickers microhardness HV ¼ 7:31 0:29 GPa, Young’s modulus E ¼ 97 5 GPa, shear modulus G ¼ 38 2 GPa, Poisson ratio ¼ 0:27 0:01, fracture toughness KIC ¼ 1:50 0:15 MPa m1=2 and modulus of rupture evaluated by ring-on-ring test MORROR ¼ 57:8 13:7 MPa which were comparable to those of the counterparts sintered by conventional industrial gas furnace

    Heterogeneity along the Height in Disc Specimens of Graphite/Tungsten Powder Mixtures with Sub-Stoichiometric Carbon Atom Ratios Heated by Concentrated Solar Beam to 1600 C

    Compositional heterogeneity along the thickness of compacted disc specimens of graphite/tungsten powder mixtures with substoichiometric carbon atom ratios (0.35, 0.50 and 1.00) heated by concentrated solar beam to 1600 C was characterised by X-ray diffraction. Top surface of any examined test piece was consisted purely of mono-carbide WC while the bottom surface showed different constitution depending on the net initial C/W ratio of the test piece; almost pure metallicWfor the C/W ¼ 0:35 specimen, dominant metallicWassociated with small proportion ofW2C for the C/W ¼ 0:50 and dominantW2C with trivial proportion of WC for the C/W ¼ 1:0. In the intermediate zone between the top and the bottom layers, the constitution held virtually constant depending on the nominal C/W ratio of the starting material: co-existing metallicWandW2C for the C/W ¼ 0:35, pureW2C for the C/W ¼ 0:50 and predominant WC with traceW2C for the C/W ¼ 1:00. Unlike for the top surface of the C/W ¼ 0:35 test piece heated in a solar furnace to 1900 C reported earlier, no evidence of formation of nano-meter scale WC whisker was detected for the top surface in any sample heated to 1600 C in the present work

    A one-pot synthesis of benzo[f]indole-4,9-diones from 1,4-naphthoquinone with α-aminoacetals has been developed. This method provides a straightforward synthesis of benzo[f]indole-4,9-diones via intramolecular nucleophilic attack of aminoquinones to aldehydes under mild reaction conditions.     Tumor regression and resistance mechanisms upon CDK4 and RAF1 inactivation in KRAS/P53 mutant lung adenocarcinomas.

    KRAS mutant lung adenocarcinomas remain intractable for targeted therapies. Genetic interrogation of KRAS downstream effectors, including the MAPK pathway and the interphase CDKs, identified CDK4 and RAF1 as the only targets whose genetic inactivation induces therapeutic responses without causing unacceptable toxicities. Concomitant CDK4 inactivation and RAF1 ablation prevented tumor progression and induced complete regression in 25% of KRAS/p53-driven advanced lung tumors, yet a significant percentage of those tumors that underwent partial regression retained a population of CDK4/RAF1-resistant cells. Characterization of these cells revealed two independent resistance mechanisms implicating hypermethylation of several tumor suppressors and increased PI3K activity. Importantly, these CDK4/RAF1-resistant cells can be pharmacologically controlled. These studies open the door to new therapeutic strategies to treat KRAS mutant lung cancer, including resistant tumors.We thank S. Ortega for the generation of the Cdk4FxKD mouse model; and M.     Biophysical and lipidomic biomarkers of cardiac remodeling post-myocardial infarction in humans

    Few studies have analyzed the potential of biophysical parameters as markers of cardiac remodeling post-myocardial infarction (MI), particularly in human hearts. Fourier transform infrared spectroscopy (FTIR) illustrates the overall changes in proteins, nucleic acids and lipids in a single signature. The aim of this work was to define the FTIR and lipidomic pattern for human left ventricular remodeling post-MI. A total of nine explanted hearts from ischemic cardiomyopathy patients were collected. Samples from the right ventricle (RV), left ventricle (LV) and infarcted left ventricle (LV INF) were subjected to biophysical (FTIR and differential scanning calorimetry, DSC) and lipidomic (liquid chromatography–high-resolution mass spectrometry, LC–HRMS) studies. FTIR evidenced deep alterations in the myofibers, extracellular matrix proteins, and the hydric response of the LV INF compared to the RV or LV from the same subject. The lipid and esterified lipid FTIR bands were enhanced in LV INF, and both lipid indicators were tightly and positively correlated with remodeling markers such as collagen, lactate, polysaccharides, and glycogen in these samples. Lipidomic analysis revealed an increase in several species of sphingomyelin (SM), hexosylceramide (HexCer), and cholesteryl esters combined with a decrease in glycerophospholipids in the infarcted tissue. Our results validate FTIR indicators and several species of lipids as useful markers of left ventricular remodeling post-MI in humans

    The ocean sampling day consortium

    Ocean Sampling Day was initiated by the EU-funded Micro B3 (Marine Microbial Biodiversity, Bioinformatics, Biotechnology) project to obtain a snapshot of the marine microbial biodiversity and function of the world’s oceans. It is a simultaneous global mega-sequencing campaign aiming to generate the largest standardized microbial data set in a single day. This will be achievable only through the coordinated efforts of an Ocean Sampling Day Consortium, supportive partnerships and networks between sites. This commentary outlines the establishment, function and aims of the Consortium and describes our vision for a sustainable study of marine microbial communities and their embedded functional traits
