    The Illumination and Growth of CRL 2688: An Analysis of New & Archival HST Observations

    We present four-color images of CRL 2688 obtained in 2009 using the Wide-Field Camera 3 on HST. The F606W image is compared with archival images in very similar filters to monitor the proper motions of nebular structure. We find that the bright N-S lobes have expanded uniformly by 2.5% and that the ensemble of rings has translated radially by 0.07 in 6.65 y. The rings were ejected every 100y for ~4 millennia until the lobes formed 250y ago. Starlight scattered from the edges of the dark E-W dust lane is coincident with extant H2 images and leading tips of eight pairs of CO outflows. We interpret this as evidence that fingers lie within geometrically opposite cones of opening angles {\approx} 30{\circ} like those in CRL618. By combining our results of the rings with 12CO absorption from the extended AGB wind we ascertain that the rings were ejected at ~18 km s-1 with very little variation and that the distance to CRL2688, v_{exp}/ / {\dot\theta}_exp$, is 300 - 350 pc. Our 2009 imaging program included filters that span 0.6 to 1.6{\mu}m. We constructed a two-dimensional dust scattering model of stellar radiation through CRL2688 that successfully reproduces the details of the nebular geometry, its integrated spectral energy distribution, and nearly all of its color variations. The model implies that the optical opacity of the lobes >~ 1, the dust particle density in the rings decreases as radius^{-3} and that the mass and momentum of the AGB winds and their rings have increased over time.Comment: (51 pages, 6 figures; accepted by ApJ

    Insight into the OH polarimetric structure of OH 26.5+0.6

    We present the first view of the magnetic field structure in the OH shell of the extreme OH/IR star OH 26.5+0.6. MERLIN interferometric observations of this object were obtained in December 1993 in full polarisation, at 1612, 1665 and 1667 MHz. The maser spots show a spheroidal distribution both at 1612 and 1667 MHz, while at 1665 MHz emission from the blue-shifted maser peak is concentrated on the stellar position, and the red-shifted peak emission exhibits a filamentary structure oriented on a SE-NW axis. The linear polarisation in both main lines is rather faint, ranging from 9 to 20% at 1665 MHz and from 0 to 30% at 1667 MHz. At 1612 MHz most maser spots exhibit a similar range of linear polarisation although those in the outermost parts of the envelope reach values as high as 66%. This is particularly apparent in the southern part of the shell. The detailed distribution of the polarisation vectors could only be obtained at 1612 MHz. The polarisation vectors show a highly structured distribution indicative of a poloidal magnetic field inclined by 40-60^\circ to the line of sight. The velocity distribution of the maser spots with respect to the radial distance is well explained by an isotropic outflow at constant velocity in the case of a prolate shaped spheroid envelope, also tilted about 45-65^\circ to the line of sight.Comment: 20 pages, 16 figures, accepted for publication in MNRA

    Phase II Trial of IL-12 Plasmid Transfection and PD-1 Blockade in Immunologically Quiescent Melanoma.

    PurposeTumors with low frequencies of checkpoint positive tumor-infiltrating lymphocytes (cpTIL) have a low likelihood of response to PD-1 blockade. We conducted a prospective multicenter phase II trial of intratumoral plasmid IL-12 (tavokinogene telseplasmid; "tavo") electroporation combined with pembrolizumab in patients with advanced melanoma with low frequencies of checkpoint positive cytotoxic lymphocytes (cpCTL).Patients and methodsTavo was administered intratumorally days 1, 5, and 8 every 6 weeks while pembrolizumab (200 mg, i.v.) was administered every 3 weeks. The primary endpoint was objective response rate (ORR) by RECIST, secondary endpoints included duration of response, overall survival and progression-free survival. Toxicity was evaluated by the CTCAE v4. Extensive correlative analysis was done.ResultsThe combination of tavo and pembrolizumab was well tolerated with adverse events similar to those previously reported with pembrolizumab alone. Patients had a 41% ORR (n = 22, RECIST 1.1) with 36% complete responses. Correlative analysis showed that the combination enhanced immune infiltration and sustained the IL-12/IFNγ feed-forward cycle, driving intratumoral cross-presenting dendritic cell subsets with increased TILs, emerging T cell receptor clones and, ultimately, systemic cellular immune responses.ConclusionsThe combination of tavo and pembrolizumab was associated with a higher than expected response rate in this poorly immunogenic population. No new or unexpected toxicities were observed. Correlative analysis showed T cell infiltration with enhanced immunity paralleling the clinical activity in low cpCTL tumors

    Polarization properties of OH masers in AGB and post-AGB stars

    Context: Ground-state OH maser emission from late-type stars is usually polarized and remains a powerful probe of the magnetic field structure in the outer regions of circumstellar envelopes if observed with high angular and spectral resolutions. Observations in all four Stokes parameters are quite sparse and this is the most thorough, systematic study published to date. Aims: We aim to determine polarization properties of OH masers in an extensive sample of stars that show copious mass loss and search for candidate objects that are well-suited for high angular resolution studies. Methods: Full-polarization observations of the OH 1612 and 1667 MHz maser transitions were carried out for a sample of 117 AGB and post-AGB stars. Several targets were also observed in the 1665 MHz line. Results: Polarized features occur in more than 75% of the sources in the complete sample and there is no intrinsic difference in the occurrence of polarized emission between the three classes of objects of different infrared characteristics. The highest fractional polarization occurs for the post-AGB+PN and the Mira+SR classes at 1612 and 1667 MHz, respectively. Differences in the fractional polarization between the sources at different evolutionary stages appear to be related to depolarization caused by blending. The alignment of the polarization angles at the extreme sides of the shell implies a regular structure of the magnetic field of a strength of 0.3-2.3 mG. Conclusions: Polarized OH maser features are widespread in AGB and post-AGB stars. The relationship between the circular and linear fractional polarizations for a representative sample are consistent with the standard models of polarization for the Zeeman splitting higher than the Doppler line width, whereas the polarized features are the sigma components.Comment: Accepted for publication in Astronomy and Astrophysics.     Light echoes reveal an unexpectedly cool Eta Carinae during its 19th-century Great Eruption

    Eta Carinae (Eta Car) is one of the most massive binary stars in the Milky Way. It became the second-brightest star in the sky during its mid-19th century "Great Eruption," but then faded from view (with only naked-eye estimates of brightness). Its eruption is unique among known astronomical transients in that it exceeded the Eddington luminosity limit for 10 years. Because it is only 2.3 kpc away, spatially resolved studies of the nebula have constrained the ejected mass and velocity, indicating that in its 19th century eruption, Eta Car ejected more than 10 M_solar in an event that had 10% of the energy of a typical core-collapse supernova without destroying the star. Here we report the discovery of light echoes of Eta Carinae which appear to be from the 1838-1858 Great Eruption. Spectra of these light echoes show only absorption lines, which are blueshifted by -210 km/s, in good agreement with predicted expansion speeds. The light-echo spectra correlate best with those of G2-G5 supergiant spectra, which have effective temperatures of ~5000 K. In contrast to the class of extragalactic outbursts assumed to be analogs of Eta Car's Great Eruption, the effective temperature of its outburst is significantly cooler than allowed by standard opaque wind models. This indicates that other physical mechanisms like an energetic blast wave may have triggered and influenced the eruption.Comment: Accepted for publication by Nature; 4 pages, 4 figures, SI: 6 pages, 3 figures, 5 table

    On the reliability of mass-loss-rate estimates for AGB stars

    In the recent literature there has been some doubt as to the reliability of CO multi-transitional line observations as a mass-loss-rate estimator for AGB stars. Mass-loss rates for 10 intermediate- to high-mass-loss-rate AGB stars are derived using a detailed non-LTE, non-local radiative transfer code based on the Monte-Carlo method to model the CO radio line intensities. The circumstellar envelopes are assumed to be spherically symmetric and formed by constant mass-loss rates. The energy balance is solved self-consistently and the effects of dust on the radiation field and thermal balance are included. An independent estimate of the mass-loss rate is also obtained from the combination of dust radiative transfer modelling with a dynamical model of the gas and dust particles. We find that the CO radio line intensities and shapes are successfully reproduced for the majority of our objects assuming a constant mass-loss rate. Moreover, the CO line intensities are only weakly dependent on the adopted micro-turbulent velocity, in contrast to recent claims in the literature. The two methods used in the present work to derive mass-loss-rates are consistent within a factor of ~3 for intermediate- to high-mass-loss-rate objects, indicating that this is a lower limit to the uncertainty in present mass-loss-rate estimates. We find a tentative trend with chemistry. Mass-loss rates from the dust/dynamical model are systematically higher than those from the CO model for the carbon stars and vice versa for the M-type stars. This could be ascribed to a discrepancy in the adopted CO/H_2-abundance ratio, but we caution that the sample is small and systematic errors cannot be excluded.Comment: 18 pages, 17 figures, accepted for publication in A&

    Annual variation in the levels of transcripts of sex-specific genes in the mantle of the common mussel, Mytilus edulis

    Mytilus species are used as sentinels for the assessment of environmental health but sex or stage in the reproduction cycle is rarely considered even though both parameters are likely to influence responses to pollution. We have validated the use of a qPCR assay for sex identification and related the levels of transcripts to the reproductive cycle. A temporal study of mantle of Mytilus edulis found transcripts of male-specific vitelline coat lysin (VCL) and female-specific vitelline envelope receptor for lysin (VERL) could identify sex over a complete year. The levels of VCL/VERL were proportional to the numbers of sperm/ova and are indicative of the stage of the reproductive cycle. Maximal levels of VCL and VERL were found in February 2009 declining to minima between July - August before increasing and re-attaining a peak in February 2010. Water temperature may influence these transitions since they coincide with minimal water temperature in February and maximal temperature in August. An identical pattern of variation was found for a cryptic female-specific transcript (H5) but a very different pattern was observed for oestrogen receptor 2 (ER2). ER2 varied in a sex-specific way with male > female for most of the cycle, with a female maxima in July and a male maxima in December. Using artificially spawned animals, the transcripts for VCL, VERL and H5 were shown to be present in gametes and thus their disappearance from mantle is indicative of spawning. VCL and VERL are present at equivalent levels in February and July-August but during gametogenesis (August to January) and spawning (March to June) VCL is present at lower relative amounts than VERL. This may indicate sex-specific control mechanisms for these processes and highlight a potential pressure point leading to reduced reproductive output if environmental factors cause asynchrony to gamete maturation or release

    香港的老年人口增長迅速,從1988年至2018年間,老年人口由46萬增加到127萬(政府經濟顧問辦公室, 2019)。隨著人口老化,長期護理服務需求也相對增加。資助安老院舍的平均等待時間長達20個月(香港政府, 2021)。在2017至2018年間,有40,000人在輪候資助安老院舍,其中6,600人在等候期間去世(香港政府, 2018)。安老服務業一直面臨人力短缺的問題,2017年的相關職位空缺率為15-20%(香港政府, 2017)。2013年的研究統計發現超過一半的安老服務前線員工超過50歲(勞工及福利局, 2019)。隨著退休潮的來臨,人力短缺的問題會進一步加劇。因此,科技與安老服務的結合被視爲減輕人手壓力的方向。結合科技和院舍服務不僅可以減輕工作量,還能提高服務質量和效率。社交媒體和視訊會議平台可以減少在訊息傳遞上的時間和人力成本。樂齡科技產品,諸如可穿戴設備和遠程監控傳感器,可以讓護理人員更快捷和系統化地掌握服務對象的狀態,從而減少親自監測的次數(Chen, 2020)。因此,自2017年起,香港政府與各部門積極推廣樂齡科技,並於2018年設立樂齡及康復創科應用基金,幫助合資格的安老及康復服務單位購置、租借和試用科技產品。香港政府於2022年更將此計劃擴展至其他非資助的私營或自負盈虧的安老院舍及殘疾人士院舍。在過去三年的新冠疫情影響下,大部分安老院舍因為防疫工作量的增加而面臨更嚴重的人手短缺,不少院舍開始將科技融入服務,以應對疫情衍生的需求和改善現有服務質素。然而,香港院舍對於如何應用社交媒體、會議平台和樂齡科技產品的研究甚少。在此背景下,本研究旨在瞭解院舍應用相關科技的情況,並提供應對有關障礙的政策建議

    Hypoxia induces dilated cardiomyopathy in the chick embryo: mechanism, intervention, and long-term consequences

    Background: Intrauterine growth restriction is associated with an increased future risk for developing cardiovascular diseases. Hypoxia in utero is a common clinical cause of fetal growth restriction. We have previously shown that chronic hypoxia alters cardiovascular development in chick embryos. The aim of this study was to further characterize cardiac disease in hypoxic chick embryos. Methods: Chick embryos were exposed to hypoxia and cardiac structure was examined by histological methods one day prior to hatching (E20) and at adulthood. Cardiac function was assessed in vivo by echocardiography and ex vivo by contractility measurements in isolated heart muscle bundles and isolated cardiomyocytes. Chick embryos were exposed to vascular endothelial growth factor (VEGF) and its scavenger soluble VEGF receptor-1 (sFlt-1) to investigate the potential role of this hypoxia-regulated cytokine. Principal Findings: Growth restricted hypoxic chick embryos showed cardiomyopathy as evidenced by left ventricular (LV) dilatation, reduced ventricular wall mass and increased apoptosis. Hypoxic hearts displayed pump dysfunction with decreased LV ejection fractions, accompanied by signs of diastolic dysfunction. Cardiomyopathy caused by hypoxia persisted into adulthood. Hypoxic embryonic hearts showed increases in VEGF expression. Systemic administration of rhVEGF165 to normoxic chick embryos resulted in LV dilatation and a dose-dependent loss of LV wall mass. Lowering VEGF levels in hypoxic embryonic chick hearts by systemic administration of sFlt-1 yielded an almost complete normalization of the phenotype. Conclusions/Significance: Our data show that hypoxia causes a decreased cardiac performance and cardiomyopathy in chick embryos, involving a significant VEGF-mediated component. This cardiomyopathy persists into adulthood

    Protective effect of stromal Dickkopf-3 in prostate cancer: opposing roles for TGFBI and ECM-1

    Aberrant transforming growth factor–β (TGF-β) signaling is a hallmark of the stromal microenvironment in cancer. Dickkopf-3 (Dkk-3), shown to inhibit TGF-β signaling, is downregulated in prostate cancer and upregulated in the stroma in benign prostatic hyperplasia, but the function of stromal Dkk-3 is unclear. Here we show that DKK3 silencing in WPMY-1 prostate stromal cells increases TGF-β signaling activity and that stromal cellconditioned media inhibit prostate cancer cell invasion in a Dkk-3-dependent manner. DKK3 silencing increased the level of the cell-adhesion regulator TGF-β–induced protein (TGFBI) in stromal and epithelial cell-conditioned media, and recombinant TGFBI increased prostate cancer cell invasion. Reduced expression of Dkk-3 in patient tumors was associated with increased expression of TGFBI. DKK3 silencing reduced the level of extracellular matrix protein-1 (ECM-1) in prostate stromal cell-conditioned media but increased it in epithelial cell-conditioned media, and recombinant ECM-1 inhibited TGFBI-induced prostate cancer cell invasion. Increased ECM1 and DKK3 mRNA expression in prostate tumors was associated with increased relapse-free survival. These observations are consistent with a model in which the loss of Dkk-3 in prostate cancer leads to increased secretion of TGFBI and ECM-1, which have tumor-promoting and tumor-protective roles, respectively. Determining how the balance between the opposing roles of extracellular factors influences prostate carcinogenesis will be key to developing therapies that target the tumor microenvironment
