    Home-based art therapy in families with seriously ill children : a heuristic inquiry

    This heuristic study is an exploration of an art therapy student's practicum experience working with ill children and their siblings in their homes. Pragmatic issues include isolation, therapist safety and challenges of travelling, scheduling, and providing art materials. Therapeutic issues are difficulties collecting information from busy parents, acclimating to the overwhelming physical and emotional space, and tolerating chaos in the therapy environment. The emotional effect of seeing medical paraphernalia in the home, as well as issues surrounding privacy, instruction, and discipline are also explored, as are the therapeutic frame, accepting food and gifts, and the role of language in the therapy. Balancing family focus with client focus, interacting with parents, countertransference reactions to the home environment, client secrecy, and termination issues were also relevant. Finally, the author describes in detail her emotional and philosophical reaction to the death of a client, and explores the meaning of home

    Synthesis of Modified Peptidoglycan Precursor Analogues for the Inhibition of Glycosyltransferase.

    The peptidoglycan glycosyltransferases (GTs) are essential enzymes that catalyze the polymerization of glycan chains of the bacterial cell wall from lipid II and thus constitute a validated antibacterial target. Their enzymatic cavity is composed of a donor site for the growing glycan chain (where the inhibitor moenomycin binds) and an acceptor site for lipid II substrate. In order to find lead inhibitors able to fill this large active site, we have synthesized a series of substrate analogues of lipid I and lipid II with variations in the lipid, the pyrophosphate, and the peptide moieties and evaluated their biological effect on the GT activity of E. coli PBP1b and their antibacterial potential. We found several compounds able to inhibit the GT activity in vitro and cause growth defect in Bacillus subtilis . The more active was C16-phosphoglycerate-MurNAc-(l-Ala-d-Glu)-GlcNAc, which also showed antibacterial activity. These molecules are promising leads for the design of new antibacterial GT inhibitors

    Enriched interactions which support speech and language development: short- and medium-term effects (6 months) of parental guidance in logopedics

    peer reviewedThe aim of parent based language intervention is to help parents of children with language disorders to play a main part in their child’s linguistic development by adjusting the speech addressed to them. This study focuses on the efficiency of our intervention methods and of indirect therapy. We analyze four dyads filmed at three different points in time: 1) before the intervention;2) directly after the sessions with the speech therapists; 3) six months later. Each film was transcribed and analyzed. The results confirm that parental based language intervention induces long-term changes in the parents’ behavior.La guidance logopédique parentale vise à aider des parents d’enfants en difficulté langagière à tenir un rôle d’acteur principal dans le développement langagier de leurs enfants, en leur fournissant un langage plus adapté à ses difficultés. Cette étude s’intéresse à l’efficacité de cette méthode d’intervention en analysant quatre dyades parent/enfant filmées à trois reprises : avant la prise en charge, directement après les sessions de formation et 6 mois plus tard. Chaque film a été transcrit et analysé. Les résultats confirment que la guidance parentale induit des changements de comportement des parents dont l’effet persiste à long terme

    Subdivision of the helix-turn-helix GntR family of bacterial regulators in the FadR, HutC, MocR, and YtrA subfamilies

    Haydon and Guest (Haydon, D. J, and Guest, J. R. (1991) FEMS Microbiol Lett. 63, 291-295) first described the helix-turn-helix GntR family of bacterial regulators. They presented them as transcription factors sharing a similar N-terminal DNA-binding (D-b) domain, but they observed near-maximal divergence in the C-terminal effector-binding and oligomerization (E-b/O) domain. To elucidate this C-terminal heterogeneity, structural, phylogenetic, and functional analyses were performed on a family that now comprises about 270 members. Our comparative study first focused on the C-terminal E-b/O domains and next on DNA-binding domains and palindromic operator sequences, has classified the GntR members into four subfamilies that we called FadR, HutC, MocR, and YtrA. Among these subfamilies a degree of similarity of about 55% was observed throughout the entire sequence. Structure/function associations were highlighted although they were not absolutely stringent. The consensus sequences deduced for the DNA-binding domain were slightly different for each subfamily, suggesting that fusion between the D-b and E-b/O domains have occurred separately, with each subfamily having its own D-b domain ancestor. Moreover, the compilation of the known or predicted palindromic cis-acting elements has highlighted different operator sequences according to our subfamily subdivision. The observed C-terminal E-b/O domain heterogeneity was therefore reflected on the DNA-binding domain and on the cis-acting elements, suggesting the existence of a tight link between the three regions involved in the regulating process.Peer reviewe

    Long-term and large-scale multispecies dataset tracking population changes of common European breeding birds

    Around fifteen thousand fieldworkers annually count breeding birds using standardized protocols in 28 European countries. The observations are collected by using country-specific and standardized protocols, validated, summarized and finally used for the production of continent-wide annual and long-term indices of population size changes of 170 species. Here, we present the database and provide a detailed summary of the methodology used for fieldwork and calculation of the relative population size change estimates. We also provide a brief overview of how the data are used in research, conservation and policy. We believe this unique database, based on decades of bird monitoring alongside the comprehensive summary of its methodology, will facilitate and encourage further use of the Pan-European Common Bird Monitoring Scheme results.publishedVersio

    Regulation of antibiotic production in Actinobacteria: new perspectives from the post-genomic era

    The antimicrobial activity of many of their natural products has brought prominence to the Streptomycetaceae, a family of Gram-positive bacteria that inhabit both soil and aquatic sediments. In the natural environment, antimicrobial compounds are likely to limit the growth of competitors, thereby offering a selective advantage to the producer, in particular when nutrients become limited and the developmental programme leading to spores commences. The study of the control of this secondary metabolism continues to offer insights into its integration with a complex lifecycle that takes multiple cues from the environment and primary metabolism. Such information can then be harnessed to devise laboratory screening conditions to discover compounds with new or improved clinical value. Here we provide an update of the review we published in NPR in 2011. Besides providing the essential background, we focus on recent developments in our understanding of the underlying regulatory networks, ecological triggers of natural product biosynthesis, contributions from comparative genomics and approaches to awaken the biosynthesis of otherwise silent or cryptic natural products. In addition, we highlight recent discoveries on the control of antibiotic production in other Actinobacteria, which have gained considerable attention since the start of the genomics revolution. New technologies that have the potential to produce a step change in our understanding of the regulation of secondary metabolism are also described

    Peptidoglycan glycosyltransferase substrate mimics as templates for the design of new antibacterial drugs.

    Peptidoglycan (PG) is an essential net-like macromolecule that surrounds bacteria, gives them their shape, and protects them against their own high osmotic pressure. PG synthesis inhibition leads to bacterial cell lysis, making it an important target for many antibiotics. The final two reactions in PG synthesis are performed by penicillin-binding proteins (PBPs). Their glycosyltransferase (GT) activity uses the lipid II precursor to synthesize glycan chains and their transpeptidase (TP) activity catalyzes the cross-linking of two glycan chains via the peptide side chains. Inhibition of either of these two reactions leads to bacterial cell death. beta-lactam antibiotics target the transpeptidation reaction while antibiotic therapy based on inhibition of the GTs remains to be developed. Ongoing research is trying to fill this gap by studying the interactions of GTs with inhibitors and substrate mimics and utilizing the latter as templates for the design of new antibiotics. In this review we present an updated overview on the GTs and describe the structure-activity relationship of recently developed synthetic ligands
