    Oxidative and non-oxidative degradation of a TDI-based polyurethane foam : volatile product and condensed phase characterisation by FTIR and solid state 13C NMR spectroscopy

    The oxidative and non-oxidative degradation behaviour of a flexible polyurethane foam, synthesised from toluene diisocyanate and a polyether polyol, is reported. Both toluene diisocyanate and diaminotoluene were identified as major products under non-oxidative conditions, which indicates that the urethane linkages are degrading by two competing degradation mechanisms. Degradation of the urethane linkage by a depolymerisation reaction to yield toluene diisocyanate and polyol is proposed to occur initially. In addition, the atmospheric pressure conditions favour the degradation of the urethane linkages via a six-membered ring transition state reaction to form diaminotoluene, carbon dioxide and alkene terminated polyol chains. Solid-state 13C NMR spectroscopy and elemental analysis of the residues indicates that at temperatures above 300°C ring fusion of the aromatic components within the foam occurs, and this leads to a nitrogen-containing carbonaceous char which has a complex aromatic structure. It is proposed that under the confined conditions of the degradation the aromatic nitrogen-containing species, such as toluene diisocyanate and diaminotoluene, undergo secondary reactions and ring fusion to yield a complex char structureUnder oxidative conditions, degradation, including ring fusion, occurs at a lower temperature than under non-oxidative conditions. Neither toluene diisocyanate nor diaminotoluene were observed as major degradation products. The polyol is observed to undergo thermo-oxidative degradation at much lower temperatures than purely thermal degradation. As a consequence, the depolymerisation reaction via the six-membered ring transition state is limited in extent and diaminotoluene is not evolved. The absence of toluene diisocyanate is proposed to be a result of this species undergoing oxidative degradation reactions which lead to it being incorporated into the char

    Ms2lda.org: web-based topic modelling for substructure discovery in mass spectrometry

    Motivation: We recently published MS2LDA, a method for the decomposition of sets of molecular fragment data derived from large metabolomics experiments. To make the method more widely available to the community, here we present ms2lda.org, a web application that allows users to upload their data, run MS2LDA analyses and explore the results through interactive visualisations. Results: Ms2lda.org takes tandem mass spectrometry data in many standard formats and allows the user to infer the sets of fragment and neutral loss features that co-occur together (Mass2Motifs). As an alternative workflow, the user can also decompose a dataset onto predefined Mass2Motifs. This is accomplished through the web interface or programmatically from our web service

    In silico optimization of mass spectrometry fragmentation strategies in metabolomics

    Liquid chromatography (LC) coupled to tandem mass spectrometry (MS/MS) is widely used in identifying small molecules in untargeted metabolomics. Various strategies exist to acquire MS/MS fragmentation spectra; however, the development of new acquisition strategies is hampered by the lack of simulators that let researchers prototype, compare, and optimize strategies before validations on real machines. We introduce Virtual Metabolomics Mass Spectrometer (ViMMS), a metabolomics LC-MS/MS simulator framework that allows for scan-level control of the MS2 acquisition process in silico. ViMMS can generate new LC-MS/MS data based on empirical data or virtually re-run a previous LC-MS/MS analysis using pre-existing data to allow the testing of different fragmentation strategies. To demonstrate its utility, we show how ViMMS can be used to optimize N for Top-N data-dependent acquisition (DDA) acquisition, giving results comparable to modifying N on the mass spectrometer. We expect that ViMMS will save method development time by allowing for offline evaluation of novel fragmentation strategies and optimization of the fragmentation strategy for a particular experiment

    Synthesis and characterisation of an extended series of biodegradable cycloaliphatic polyesters

    We report the synthesis and characterisation of an extended series of biodegradable cycloaliphatic polyesters for medical purposes. These polymers are based on cis- and trans-1,4-cyclohexanedicarboxylic acid, cis- and trans-1,4-cyclohexanediol and straight chain aliphatic diols and diacids with different chain lengths. The results demonstrate the possibility of controlling polymer morphology through the cis/trans composition of the 1,4-cyclohexane moiety. The trans isomer increases the regularity of the polymer chain and hence modifies the crystalline morphology, the most obvious indication of which is an increase in melting point

    Influence of Bovine Viral Diarrhea Virus Infection on Artificial Insemination Conception and Breeding Season Pregnancy Success in Vaccinated Beef Herds

    Bovine Viral Diarrhea Virus (BVDV) causes reproductive economic losses in cattle. The objective of this study was to evaluate the influence of BVDV infection on reproductive success. Vaccinated cows (n = 370) and heifers (n = 528) from nine different herds were synchronized using the 7-day CO-Synch + CIDR protocol and were bred using fixed-time artificial insemination (FTAI). On d 28 following insemination, blood samples were collected and pregnancy status was determined. Non-pregnant animals were resynchronized and FTAI occurred a second time. In six herds, bulls were comingled with females beginning 10-15 d after the second AI. Final pregnancy status was determined 33-80 d following the first pregnancy diagnosis. Blood samples were tested for the presence of BVDV antigen using the IDEXX BVDV PI X2 Kit. Animals that tested positive were considered infected with BVDV at the time of blood collection. Herds were determined to be BVDV infected by the presence of at least one animal having a positive test for antigen (n = 4 infected herds, n = 5 non-infected herds). Statistical analyses were performed using the GLIMMIX procedure of SAS with herd as a random variable. Herds that had evidence of BVDV infection at d 28 following insemination had significantly decreased (P \u3c 0.01) first service AI conception rates compared to herds that had no evidence of infection (34 ± 2.3% vs. 54 ± 2.3%, respectively). Additionally, breeding season pregnancy rates were decreased (P \u3c 0.01) in BVDV infected herds compared to non-infected herds (69 ± 3.4% vs. 80 ± 3.6%, respectively). There was no significant effect of BVDV infection status on embryonic loss (P = 0.42) or percentage of animals which lost a pregnancy and rebred by the end of the breeding season (P = 0.63). In conclusion, BVDV infection in well vaccinated herds had a significant negative impact on both first service AI conception rate and overall breeding season pregnancy success

    Rapid development of improved data-dependent acquisition strategies

    Tandem mass spectrometry (LC-MS/MS) is widely used to identify unknown ions in untargeted metabolomics. Data-dependent acquisition (DDA) chooses which ions to fragment based upon intensities observed in MS1 survey scans and typically only fragments a small subset of the ions present. Despite this inefficiency, relatively little work has addressed the development of new DDA methods, partly due to the high overhead associated with running the many extracts necessary to optimize approaches in busy MS facilities. In this work, we first provide theoretical results that show how much improvement is possible over current DDA strategies. We then describe an in silico framework for fast and cost-efficient development of new DDA strategies using a previously developed virtual metabolomics mass spectrometer (ViMMS). Additional functionality is added to ViMMS to allow methods to be used both in simulation and on real samples via an Instrument Application Programming Interface (IAPI). We demonstrate this framework through the development and optimization of two new DDA methods that introduce new advanced ion prioritization strategies. Upon application of these developed methods to two complex metabolite mixtures, our results show that they are able to fragment more unique ions than standard DDA strategies

    Observational Constraints on Chaplygin Quartessence: Background Results

    We derive the constraints set by several experiments on the quartessence Chaplygin model (QCM). In this scenario, a single fluid component drives the Universe from a nonrelativistic matter-dominated phase to an accelerated expansion phase behaving, first, like dark matter and in a more recent epoch like dark energy. We consider current data from SNIa experiments, statistics of gravitational lensing, FR IIb radio galaxies, and x-ray gas mass fraction in galaxy clusters. We investigate the constraints from this data set on flat Chaplygin quartessence cosmologies. The observables considered here are dependent essentially on the background geometry, and not on the specific form of the QCM fluctuations. We obtain the confidence region on the two parameters of the model from a combined analysis of all the above tests. We find that the best-fit occurs close to the Λ\LambdaCDM limit (α=0\alpha=0). The standard Chaplygin quartessence (α=1\alpha=1) is also allowed by the data, but only at the 2σ\sim2\sigma level.Comment: Replaced to match the published version, references update

    Measurement of the polarisation of W bosons produced with large transverse momentum in pp collisions at sqrt(s) = 7 TeV with the ATLAS experiment

    This paper describes an analysis of the angular distribution of W->enu and W->munu decays, using data from pp collisions at sqrt(s) = 7 TeV recorded with the ATLAS detector at the LHC in 2010, corresponding to an integrated luminosity of about 35 pb^-1. Using the decay lepton transverse momentum and the missing transverse energy, the W decay angular distribution projected onto the transverse plane is obtained and analysed in terms of helicity fractions f0, fL and fR over two ranges of W transverse momentum (ptw): 35 < ptw < 50 GeV and ptw > 50 GeV. Good agreement is found with theoretical predictions. For ptw > 50 GeV, the values of f0 and fL-fR, averaged over charge and lepton flavour, are measured to be : f0 = 0.127 +/- 0.030 +/- 0.108 and fL-fR = 0.252 +/- 0.017 +/- 0.030, where the first uncertainties are statistical, and the second include all systematic effects.Comment: 19 pages plus author list (34 pages total), 9 figures, 11 tables, revised author list, matches European Journal of Physics C versio

    Observation of a new chi_b state in radiative transitions to Upsilon(1S) and Upsilon(2S) at ATLAS

    The chi_b(nP) quarkonium states are produced in proton-proton collisions at the Large Hadron Collider (LHC) at sqrt(s) = 7 TeV and recorded by the ATLAS detector. Using a data sample corresponding to an integrated luminosity of 4.4 fb^-1, these states are reconstructed through their radiative decays to Upsilon(1S,2S) with Upsilon->mu+mu-. In addition to the mass peaks corresponding to the decay modes chi_b(1P,2P)->Upsilon(1S)gamma, a new structure centered at a mass of 10.530+/-0.005 (stat.)+/-0.009 (syst.) GeV is also observed, in both the Upsilon(1S)gamma and Upsilon(2S)gamma decay modes.
