    Food habits of the Cougar Puma concolor (Carnivora: Felidae) in the Central Andes of the Colombian Coffee Region

    The feeding habits of the cougar have been scarcely studied in Colombia, despite its importance in the ecosystems it inhabits. In this study, we analysed the diet of the cougar using 53 feces samples collected from several sites in the Central Andes of Colombia’s Coffee Growing Region. To identify prey species, we examined and compared bone fragments and hairs found in each sample with descriptions published in relevant literature and with specimens from the Natural History Museum of the Universidad de Caldas, Colombia. We found that rabbits (Sylvilagus sp.) provided half of the relative biomass consumed by the cougar (50%). Other preys identified belong to representatives of medium and large mammals such as marsupials (Didelphis: 10%), sloths (Choloepus: 9%), rodents (Coendou: 10%, Cuniculus: 9%), and deer (Mazama: 9%). Of the preys, Coendou rufescens has not, to our knowledge, been previously included in cougar diet literature. The curve of accumulation of species indicated that the number of samples obtained in this work fully describe the diet of this feline. Neither domestic species were recorded in this work, nor were there any relationships between the dry or rainy seasons and the consumption of prey species. The comparison of the information reported in this study and others carried out in the Andes may indicate the general nature of this feline’s diet and its adaptability to the prey species found in each area, since these vary in each one of them. It is probable that the obtained trophic niche value (0.33) is due to the abundance of rabbits present in the study area. It is also probable that the cougar has not consumed domestic species since, although it is close to cattle ranches, it has enough of wild prey in the area to supply its needs. The results of this study may help to improve our understanding of the feeding habits of this predator and its interaction with other species in this important region of Colombia. This will, in turn, promote major comprehension of the role of cougars in the Colombian Cordilleras and lead actions for their conservation

    The miRNome function transitions from regulating developmental genes to transposable elements during pollen maturation

    Animal and plant microRNAs (miRNAs) are essential for the spatio-temporal regulation of development. Together with this role, plant miRNAs have been proposed to target transposable elements (TEs) and stimulate the production of epigenetically active small interfering RNAs. This activity is evident in the plant male gamete containing structure, the male gametophyte or pollen grain. How the dual role of plant miRNAs, regulating both genes and TEs, is integrated during pollen development and which mRNAs are regulated by miRNAs in this cell type at a genome-wide scale are unknown. Here, we provide a detailed analysis of miRNA dynamics and activity during pollen development in Arabidopsis thaliana using small RNA and degradome parallel analysis of RNA end high-throughput sequencing. Furthermore, we uncover miRNAs loaded into the two main active Argonaute (AGO) proteins in the uninuclear and mature pollen grain, AGO1 and AGO5. Our results indicate that the developmental progression from microspore to mature pollen grain is characterized by a transition from miRNAs targeting developmental genes to miRNAs regulating TE activity.sRNA, PARE, and AGO-IP sequencing uncovered the role of miRNAs during pollen development, showing that miRNAs transition from regulating genes involved in development to transposable elements

    Vigilancia tecnológica y análisis del ciclo de vida de la tecnología: Aplicación en productos generados a partir del café

    This article reports the results of an exercise of technological surveillance and life cycle analysis of a technology for the Agroindustrial sector, with the purpose of determining the state of the technology within its life cycle in order to reduce the uncertainty in the decisions making with a point of reference or inflection point. Search equations were constructed for articles and patents that were used in specialized databases. The technology selected was coffee (Agroindustrial utilization, active ingredients and co-products generated from coffee). All of the above was the input to apply the models that offer the specialized software for curves in Sigmaplot S with the objective of finding the inflection points, which are the ones that indicate the moment in phase where the technology is. The model with the best fit is selected according to the established statistical parameters: value t: greater than 2 and less than -2; P-value: less than 0.005; Durbin Watson: between 1.8 and 2.2; Adjusted R2: that store to 1. In conclusion the inflection point was generated above the year 2023 for technology in relation to articles and for patents its point between 2008 and 2013. Finally, he performed the analysis of the technology.Este artículo reporta los resultados de un ejercicio de vigilancia tecnológica y análisis del ciclo de vida de una tecnología para el sector Agroindustrial, con el propósito de determinar el estado de la tecnología dentro de su ciclo de vida para de esta manera disminuir la incertidumbre en la toma de decisiones con un punto de referencia o punto de inflexión. Se construyeron ecuaciones de búsqueda para artículos y patentes que fueron utilizadas en bases de datos especializadas. La tecnología seleccionada fue café (Aprovechamiento agroindustrial, principios activos y co-productos generados a partir del café). Todo lo anterior fue el insumo para aplicar trece modelos que ofrece el software especializado para curvas en S Sigmaplot con el objetivo de encontrar los puntos de inflexión, que son aquellos que indican el momento o fase en el que se encuentra la tecnología. Se selecciona el modelo que presenta mejor ajuste según los parámetros estadísticos establecidos: valor t: mayor que 2 y menor que -2; valor p: menor que 0.005; Durbin Watson: entre 1.8 y 2.2; R2 Ajustado: que tienda a 1. Como conclusión el punto de inflexión se generó por encima del año 2023 para la tecnología en relación con artículos y para patentes su punto fue entre el 2008 y el 2013.     Blockade of IL-33R/ST2 Signaling Attenuates Toxoplasma gondii Ileitis Depending on IL-22 Expression

    Oral T. gondii infection (30 cysts of 76K strain) induces acute lethal ileitis in sensitive C57BL/6 (B6) mice with increased expression of IL-33 and its receptor ST2 in the ileum. Here we show that IL-33 is involved in ileitis, since absence of IL-33R/ST2 attenuated neutrophilic inflammation and Th1 cytokines upon T. gondii infection with enhanced survival. Blockade of ST2 by neutralizing ST2 antibody in B6 mice conferred partial protection, while rmIL-33 aggravated ileitis. Since IL-22 expression further increased in absence of ST2, we blocked IL-22 by neutralizing antibody, which abrogated protection from acute ileitis in ST2 deficient mice. In conclusion, severe lethal ileitis induced by oral T. gondii infection is attenuated by blockade of ST2 signaling and may be mediated in part by endogenous IL-22

    Dental Health and Mortality in People With End-Stage Kidney Disease Treated With Hemodialysis: A Multinational Cohort Study

    Background Dental disease is more extensive in adults with chronic kidney disease, but whether dental health and behaviors are associated with survival in the setting of hemodialysis is unknown. Study Design Prospective multinational cohort. Setting & Participants 4,205 adults treated with long-term hemodialysis, 2010 to 2012 (Oral Diseases in Hemodialysis [ORAL-D] Study). Predictors Dental health as assessed by a standardized dental examination using World Health Organization guidelines and personal oral care, including edentulousness; decayed, missing, and filled teeth index; teeth brushing and flossing; and dental health consultation. Outcomes All-cause and cardiovascular mortality at 12 months after dental assessment. Measurements Multivariable-adjusted Cox proportional hazards regression models fitted with shared frailty to account for clustering of mortality risk within countries. Results During a mean follow-up of 22.1 months, 942 deaths occurred, including 477 cardiovascular deaths. Edentulousness (adjusted HR, 1.29; 95% CI, 1.10-1.51) and decayed, missing, or filled teeth score ≥ 14 (adjusted HR, 1.70; 95% CI, 1.33-2.17) were associated with early all-cause mortality, while dental flossing, using mouthwash, brushing teeth daily, spending at least 2 minutes on oral hygiene daily, changing a toothbrush at least every 3 months, and visiting a dentist within the past 6 months (adjusted HRs of 0.52 [95% CI, 0.32-0.85], 0.79 [95% CI, 0.64-0.97], 0.76 [95% CI, 0.58-0.99], 0.84 [95% CI, 0.71-0.99], 0.79 [95% CI, 0.65-0.95], and 0.79 [95% CI, 0.65-0.96], respectively) were associated with better survival. Results for cardiovascular mortality were similar. Limitations Convenience sample of clinics. Conclusions In adults treated with hemodialysis, poorer dental health was associated with early death, whereas preventive dental health practices were associated with longer survival

    Genetic architecture of subcortical brain structures in 38,851 individuals

    Subcortical brain structures are integral to motion, consciousness, emotions and learning. We identified common genetic variation related to the volumes of the nucleus accumbens, amygdala, brainstem, caudate nucleus, globus pallidus, putamen and thalamus, using genome-wide association analyses in almost 40,000 individuals from CHARGE, ENIGMA and UK Biobank. We show that variability in subcortical volumes is heritable, and identify 48 significantly associated loci (40 novel at the time of analysis). Annotation of these loci by utilizing gene expression, methylation and neuropathological data identified 199 genes putatively implicated in neurodevelopment, synaptic signaling, axonal transport, apoptosis, inflammation/infection and susceptibility to neurological disorders. This set of genes is significantly enriched for Drosophila orthologs associated with neurodevelopmental phenotypes, suggesting evolutionarily conserved mechanisms. Our findings uncover novel biology and potential drug targets underlying brain development and disease

    Vaccine breakthrough hypoxemic COVID-19 pneumonia in patients with auto-Abs neutralizing type I IFNs

    Life-threatening `breakthrough' cases of critical COVID-19 are attributed to poor or waning antibody response to the SARS- CoV-2 vaccine in individuals already at risk. Pre-existing autoantibodies (auto-Abs) neutralizing type I IFNs underlie at least 15% of critical COVID-19 pneumonia cases in unvaccinated individuals; however, their contribution to hypoxemic breakthrough cases in vaccinated people remains unknown. Here, we studied a cohort of 48 individuals ( age 20-86 years) who received 2 doses of an mRNA vaccine and developed a breakthrough infection with hypoxemic COVID-19 pneumonia 2 weeks to 4 months later. Antibody levels to the vaccine, neutralization of the virus, and auto- Abs to type I IFNs were measured in the plasma. Forty-two individuals had no known deficiency of B cell immunity and a normal antibody response to the vaccine. Among them, ten (24%) had auto-Abs neutralizing type I IFNs (aged 43-86 years). Eight of these ten patients had auto-Abs neutralizing both IFN-a2 and IFN-., while two neutralized IFN-omega only. No patient neutralized IFN-ss. Seven neutralized 10 ng/mL of type I IFNs, and three 100 pg/mL only. Seven patients neutralized SARS-CoV-2 D614G and the Delta variant (B.1.617.2) efficiently, while one patient neutralized Delta slightly less efficiently. Two of the three patients neutralizing only 100 pg/mL of type I IFNs neutralized both D61G and Delta less efficiently. Despite two mRNA vaccine inoculations and the presence of circulating antibodies capable of neutralizing SARS-CoV-2, auto-Abs neutralizing type I IFNs may underlie a significant proportion of hypoxemic COVID-19 pneumonia cases, highlighting the importance of this particularly vulnerable population

    The genetic architecture of the human cerebral cortex

    The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity. Variation in cortical structure is genetically correlated with cognitive function, Parkinson's disease, insomnia, depression, neuroticism, and attention deficit hyperactivity disorder

    The large-scale structure of the halo of the Andromeda galaxy II. Hierarchical structure in the Pan-Andromeda Archaeological Survey

    The Pan-Andromeda Archaeological Survey is a survey of >400>400 square degrees centered on the Andromeda (M31) and Triangulum (M33) galaxies that has provided the most extensive panorama of a LL_\star galaxy group to large projected galactocentric radii. Here, we collate and summarise the current status of our knowledge of the substructures in the stellar halo of M31, and discuss connections between these features. We estimate that the 13 most distinctive substructures were produced by at least 5 different accretion events, all in the last 3 or 4 Gyrs. We suggest that a few of the substructures furthest from M31 may be shells from a single accretion event. We calculate the luminosities of some prominent substructures for which previous estimates were not available, and we estimate the stellar mass budget of the outer halo of M31. We revisit the problem of quantifying the properties of a highly structured dataset; specifically, we use the OPTICS clustering algorithm to quantify the hierarchical structure of M31's stellar halo, and identify three new faint structures. M31's halo, in projection, appears to be dominated by two `mega-structures', that can be considered as the two most significant branches of a merger tree produced by breaking M31's stellar halo into smaller and smaller structures based on the stellar spatial clustering. We conclude that OPTICS is a powerful algorithm that could be used in any astronomical application involving the hierarchical clustering of points.
