    Parenting and child behaviour as predictors of toothbrushing difficulties in young children

    Background: Oral disease is one of the most prevalent chronic health conditions affecting children. Twice-daily toothbrushing is recommended to promote good oral health; however, a large proportion of Australian families are not meeting this recommendation. Aim: This study aimed to identify important barriers to regular toothbrushing for young children. Design: In this study, 239 parents of 0- to 4-year-old children completed an online survey that investigated child, family, and parent factors associated with child toothbrushing. Hierarchical linear regression was used to identify predictors of toothbrushing frequency in children and perceived difficulty of the task by parents. Results: We found that parent factors, specifically oral health knowledge, were the most significant predictors of toothbrushing frequency. Conversely, parent factors did not contribute significantly to the prediction of perceived difficulty of toothbrushing once family and child factors were taken into account. Oral health knowledge and use of routines were identified as the most important predictors of toothbrushing frequency, whereas resistant child behaviour and household organisation were found to be the most important predictors of perceived difficulty of regular toothbrushing. Conclusions: The findings of the study have implications for behavioural interventions to support parents, as well as directions for future research

    The shape of warmth: temperature profiles in living rooms

    The most commonly used family of models in representing the UK building stock, BREDEM (the Building Research Establishment Domestic Energy Models), assume that all homes exhibit the same heating pattern and hence can be expected to have similar temperature profiles over the course of a day. The presented research shows that homes differ significantly in their respective temperature profile over the course of the day. A cluster analysis performed on temperature data from 275 living rooms in English homes over three winter months resulted in four different clusters of temperature profiles. The clusters differ significantly in their shape, as revealed by visual inspection, and supported by significant differences in minimum and maximum temperatures and temperature variability across the day. About 40% of homes showed a bimodal temperature pattern as assumed under BREDEM. However, the remaining 60% showed very different profiles. These findings challenge the assumption that one standard pattern fits all homes. Different temperature demand profiles have important implications for future peak power demands, particularly if domestic space heating is switched to electricity.     Empirical variation in 24-h profiles of delivered power for a sample of UK dwellings: Implications for evaluating energy savings

    Improved methods for quantifying energy savings in buildings need to be supported by empirical measures rather than modeled estimates of future annual energy demand. This paper uses power temperature gradient (PTG, W/K), or the slope of power demand in response to changes in external air temperature; first, to categorise dwelling energy performance from daily energy data (when 0–15 °C outside); second, to investigate variations in 24-h profiles of delivered power. Estimates of PTG were obtained from 567 UK dwellings with 118,000 days of gas and electricity data. From a multivariable regression model, PTG was predicted by dwelling characteristics (number of bedrooms, number of floors, dwelling type, and dwelling age category (all p < 0.001)) but not by number of occupants. When dwellings were grouped into quintiles of PTG, mean PTG had threefold increase from the first to fifth quintile (188 to 563 W/K, respectively). This was reflected in 24-h profiles of delivered power (30 min intervals): at 0 °C, each 100 W/K decline in PTG corresponded to ∌2.5 kW decline in mean morning and evening peak power.     Direct Reprogramming of Mouse Fibroblasts into Functional Skeletal Muscle Progenitors

    Skeletal muscle harbors quiescent stem cells termed satellite cells and proliferative progenitors termed myoblasts, which play pivotal roles during muscle regeneration. However, current technology does not allow permanent capture of these cell populations in vitro. Here, we show that ectopic expression of the myogenic transcription factor MyoD, combined with exposure to small molecules, reprograms mouse fibroblasts into expandable induced myogenic progenitor cells (iMPCs). iMPCs express key skeletal muscle stem and progenitor cell markers including Pax7 and Myf5 and give rise to dystrophin-expressing myofibers upon transplantation in vivo. Notably, a subset of transplanted iMPCs maintain Pax7 expression and sustain serial regenerative responses. Similar to satellite cells, iMPCs originate from Pax7+ cells and require Pax7 itself for maintenance. Finally, we show that myogenic progenitor cell lines can be established from muscle tissue following small-molecule exposure alone. This study thus reports on a robust approach to derive expandable myogenic stem/progenitor-like cells from multiple cell types

    Role of mprF1 and mprF2 in the Pathogenicity of Enterococcus faecalis

    Aujourd hui, Enterococcus faecalis est considĂ©rĂ© comme l un des plus importants agents pathogĂšnes causant des maladies nosocomiales. En raison de sa rĂ©sistance innĂ©e et acquise aux antibiotiques, l identification de nouvelles cibles pour le traitement de cette bactĂ©rie est une grande prioritĂ©. Le facteur Multiple Peptide RĂ©sistance (MprF), qui a Ă©tĂ© dĂ©crit en premier chez Staphylococcus aureus, modifie le phosphatidylglycĂ©rol avec de la lysine et rĂ©duit ainsi la charge nĂ©gative de l enveloppe cellulaire. Ceci a comme consĂ©quence d augmenter la rĂ©sistance aux peptides antimicrobiens cationiques (PAC). Deux gĂšnes paralogues putatifs (mprF1 et mprF2) ont Ă©tĂ© identifiĂ©s chez E. faecalis par recherche BLAST en utilisant le gĂšne dĂ©crit chez S. aureus. Une caractĂ©risation de ces deux gĂšnes d E. faecalis ainsi que des mĂ©canismes conduisant Ă  une rĂ©sistance aux PAC, pourrait aider Ă  dĂ©velopper des nouvelles stratĂ©gies thĂ©rapeutiques contre ce pathogĂšne. Deux mutants de dĂ©lĂ©tion et un double mutant ont Ă©tĂ© construits par recombinaison homologue chez E. faecalis. L analyse des phospholipides des membranes cytoplasmiques des deux mutants mprF1 et mprF2 par chromatographie sur couche mince a montrĂ© que seule l inactivation de mprF2 inhibe la synthĂšse de trois amino-phosphatidlyglycĂ©rol distincts (comme la Lysine-PG, l Alanine-PG et l Arginine-PG). De plus, le mutant mprF2 est Ă©galement plus sensible aux PAC que la souche sauvage. La capacitĂ© de formation d un biofilm est gĂ©nĂ©ralement considĂ©rĂ©e comme un facteur important de virulence, ce qui est Ă©galement le cas pour les entĂ©rocoques. Le mutant mprF2 montre une capacitĂ© accrue dans ce phĂ©nomĂšne. Ceci semble ĂȘtre du Ă  une augmentation de la concentration d ADN extracellulaire dans le biofilm formĂ© par ce mutant. Curieusement, cette augmentation est indĂ©pendante d une autolyse. Le mutant mprF2 est Ă©galement plus rĂ©sistant Ă  l opsonophagocytose. Cependant, le gĂšne mprF2 ne joue aucun rĂŽle dans les bactĂ©riĂ©mies de souris et les endocardites de rats.En revanche, aucun phĂ©notype n a Ă©tĂ© trouvĂ© pour un mutant mprF1 jusqu Ă  prĂ©sent. Cette mutation ne modifie ni la synthĂšse de l aminoacyl-PG en condition de laboratoire ni la rĂ©sistance aux PAC et Ă  l opsonophagocytose. Par consĂ©quent, il semble que mprF2 soit le seul gĂšne mprF fonctionnel chez E. faecalis. NĂ©anmoins, contrairement Ă  d autres bactĂ©ries, mprF2 ne semble pas ĂȘtre un facteur de virulence majeur pour cette espĂšce.Enterococcus faecalis is regarded nowadays as one of the most important nosocomial pathogens. Due to its innate and acquired resistance to antibiotics, identification of new targets for antimicrobial treatment of E. faecalis is a high priority. The multiple peptides resistance factor (MprF), which was first described in Staphylococcus aureus, modifies phosphatidylglycerol with lysine and reduces the negative charge of the membrane, thus increasing resistance to cationic antimicrobial peptides (CAMPs). Two putative mprF paralogs (mprF1 and mprF2) were identified in E. faecalis by Blast search using the well-described S. aureus gene as a lead. A better understanding of these two genes and mechanisms leads to enterococcal resistance to CAMPs might help designing therapeutic strategies against this bacteria. Two single deletion mutants and double mutant in E. faecalis were created by homologues recombination. Analysis of cell membrane phospholipids from both mutants by thin-layer chromatography showed that inactivation of mprF2 abolished the synthesis of three distinct amino-phosphatidylglycerol (mostly likely Lysin-PG, Alanine-PG and Argine-PG). The CAMPs testing assay demonstrated that the deletion mutant of mprF2 was more susceptible to CAMPs than the wild type. Biofilm formation is usually regarded as a virulence factor which provides an important way for enterococci to cause infections. Inactivation of mprF2 led to increase the biofilm formation which we showed that it was due to the accumulation of eDNA in the biofilm, but the release of eDNA is independent from autolysis. The mprF2 mutant was resistance to killing by opsonophagocytosis more than wild type. However, the mprF2 gene plays no role in bacteremia in mice and rat endocarditis. Our results showed that non polar effect mprF1 mutant does not affect in the synthesis of aminoacyl-PG in the laboratory condition. It also has no effect on susceptible to CAMPs, opsonic killing and autolysis. Therefore, it seems that mprF2 is the only functional mprF gene in E. faecalis in the laboratory condition.     HIT family genes: FHIT but not PKCI-1/HINT produces altered transcripts in colorectal cancer

    Forty-five colorectal adenocarcinomas were examined for alterations in the HIT family genes FHIT and PKCI-1/HINT by a combination of reverse transcriptase polymerase chain reaction and DNA sequencing. In all cases a single transcript corresponding to the reported sequence was detected using primers specific for the PKCI-1/HINT gene. In contrast multiple transcripts were detected using primers specific for the FHIT gene transcript. 6% (3/45) of tumours evinced no detectable expression of any FHIT transcript and a further 12% (6/45) produced only the normal full length transcripts. Ninety-six aberrant transcripts were characterized from the remaining tumours. Deviations from the normal full length sequence characterized included deletions, insertions of novel sequences, a point mutation as well as the usage of a putative alternate splice site in exon 10. Message variants were detected with approximately equal frequency in all tumour stages with the exception that templates with insertions were found solely in Dukes’ stage B tumours (P < 0.001). With the exception of the putative alternate splice site, aberrant transcripts were not detected in matched normal mucosa. These results suggest that members of the HIT family of genes are only selectively involved in tumorigenesis and that perturbation of FHIT gene expression is an early event in colorectal tumorigenesis. © 1999 Cancer Research Campaig

    Dynamics of pneumococcal nasopharyngeal carriage in healthy children attending a day care center in northern Spain. influence of detection techniques on the results

    <p>Abstract</p> <p>Background</p> <p>Pneumococcal nasopharyngeal carriage precedes invasive infection and is the source for dissemination of the disease. Differences in sampling methodology, isolation or identification techniques, as well as the period (pre -or post-vaccination) when the study was performed, can influence the reported rates of colonization and the distribution of serotypes carried.</p> <p>Objectives</p> <p>To evaluate the prevalence and dynamics of pneumococcal nasopharyngeal colonization in healthy children aged 6-34 months attending a day care center with a high level of hygiene and no overcrowding. The study was performed 3-4 years after the 7-valent pneumococcal vaccine was introduced, using multiple methodologies to detect and characterize the isolates.</p> <p>Methods</p> <p>Over 12 months, 25 children were sampled three times, 53 children twice and 27 children once. Three <it>Streptococcus pneumoniae </it>typing techniques were used: Quellung, Pneumotest-Latex-kit and multiplex-polymerase chain reaction (PCR). The similarity of isolates of the same serotype was established by pulsed field gel electrophoresis (PFGE) and occasionally the multilocus sequence type (ST) was also determined.</p> <p>Results</p> <p>Overall pneumococcal carriage and multiple colonization rates were 89.5% (94/105) and 39%, respectively. Among 218 pneumococci detected, 21 different serotypes and 13 non-typeable isolates were found. The most prevalent serotypes were 19A, 16F and 15B. Serotypes 15B, 19A and 21 were mainly found as single carriage; in contrast serotypes 6B, 11A and 20, as well as infrequent serotypes, were isolated mainly as part of multiple carriage. Most 19A isolates were ST193 but most serotypes showed high genetic heterogeneity. Changes in the pneumococci colonizing each child were frequent and the same serotype detected on two occasions frequently showed a different genotype. By multiplex-PCR, 100% of pneumococci could be detected and 94% could be serotyped versus 80.3% by the Quellung reaction and Pneumotest-Latex in combination (p < 0.001).</p> <p>Conclusions</p> <p>Rates of <it>S. pneumoniae </it>carriage and multiple colonization were very high. Prevalent serotypes differed from those found in similar studies in the pre-vaccination period. In the same child, clearance of a pneumococcal strain and acquisition of a new one was frequent in a short period of time. The most effective technique for detecting pneumococcal nasopharyngeal carriers was multiplex-PCR.</p

    Search for new phenomena in final states with an energetic jet and large missing transverse momentum in pp collisions at √ s = 8 TeV with the ATLAS detector

    Results of a search for new phenomena in final states with an energetic jet and large missing transverse momentum are reported. The search uses 20.3 fb−1 of √ s = 8 TeV data collected in 2012 with the ATLAS detector at the LHC. Events are required to have at least one jet with pT > 120 GeV and no leptons. Nine signal regions are considered with increasing missing transverse momentum requirements between Emiss T > 150 GeV and Emiss T > 700 GeV. Good agreement is observed between the number of events in data and Standard Model expectations. The results are translated into exclusion limits on models with either large extra spatial dimensions, pair production of weakly interacting dark matter candidates, or production of very light gravitinos in a gauge-mediated supersymmetric model.     The German Young Olympic Athletes' Lifestyle and Health Management Study (GOAL Study): design of a mixed-method study

    <p>Abstract</p> <p>Background</p> <p>In order to perform at top levels, elite athletes have to both protect and risk their health at the same time. Adolescent elite athletes have the additional challenge of coping with substantial physical, psychological and social transformations. The contradictory phenomenon of protecting and risking the adolescent athletes' health in sports challenges the development of health promotion and protection strategies. The GOAL Study (German Young Olympic Athletes' Lifestyle and Health Management Study) analyzes the individual and organizational management of health in adolescent elite sports.</p> <p>Methods/design</p> <p>We combine quantitative and qualitative approaches in a mixed-method study. This allows us to gather a broad range of representative information on squad athletes from all Olympic disciplines as well as in-depth information on four selected Olympic disciplines (artistic gymnastics, biathlon, handball and wrestling). Within the quantitative section we attempt to identify the young athletes' health and nutrition behavior, their subjective health state and their lay health representations, health-related social networks, and structures of medical attendance. 1138 national team level athletes born between 1992 and 1995 from 51 Olympic disciplines responded to the questionnaire (response rate: 61,75%). The qualitative section investigates the meaning and relevance of health and nutrition within the athletes' sports specific surroundings, the impact of biographic backgrounds on individual health behavior, and sports specific cultures of health, nutrition and risk. We interviewed 24 athletes and 28 coaching and medical experts, and carried out 14 multi-day participant observations at training sessions and competitions.</p> <p>Conclusions</p> <p>The studies' results will serve as the basis for developing tailored health promotion strategies to be in cooperation with German elite sports associations.</p
