    Characterization of Developmental- and Stress-Mediated Expression of Cinnamoyl-CoA Reductase in Kenaf ( Hibiscus cannabinus

    Cinnamoyl-CoA reductase (CCR) is an important enzyme for lignin biosynthesis as it catalyzes the first specific committed step in monolignol biosynthesis. We have cloned a full length coding sequence of CCR from kenaf (Hibiscus cannabinus L.), which contains a 1,020-bp open reading frame (ORF), encoding 339 amino acids of 37.37 kDa, with an isoelectric point (pI) of 6.27 (JX524276, HcCCR2). BLAST result found that it has high homology with other plant CCR orthologs. Multiple alignment with other plant CCR sequences showed that it contains two highly conserved motifs: NAD(P) binding domain (VTGAGGFIASWMVKLLLEKGY) at N-terminal and probable catalytic domain (NWYCYGK). According to phylogenetic analysis, it was closely related to CCR sequences of Gossypium hirsutum (ACQ59094) and Populus trichocarpa (CAC07424). HcCCR2 showed ubiquitous expression in various kenaf tissues and the highest expression was detected in mature flower. HcCCR2 was expressed differentially in response to various stresses, and the highest expression was observed by drought and NaCl treatments

    Iron removal, energy consumption and operating cost of electrocoagulation of drinking water using a new flow column reactor

    The goal of this project was to remove iron from drinking water using a new electrocoagulation (EC) cell. In this research, a flow column has been employed in the designing of a new electrocoagulation reactor (FCER) to achieve the planned target. Where, the water being treated flows through the perforated disc electrodes, thereby effectively mixing and aerating the water being treated. As a result, the stirring and aerating devices that until now have been widely used in the electrocoagulation reactors are unnecessary. The obtained results indicated that FCER reduced the iron concentration from 20 to 0.3 mg/L within 20 min of electrolysis at initial pH of 6, inter-electrode distance (ID) of 5 mm, current density (CD) of 1.5 mA/cm2, and minimum operating cost of 0.22 US $/m3. Additionally, it was found that FCER produces H2 gas enough to generate energy of 10.14 kW/m3. Statistically, it was found that the relationship between iron removal and operating parameters could be modelled with R2 of 0.86, and the influence of operating parameters on iron removal followed the order: C0>t>CD>pH. Finally, the SEM (scanning electron microscopy) images showed a large number of irregularities on the surface of anode due to the generation of aluminium hydroxides

    This article reviews the dynamics of global food prices since the food crisis of2007–08, the extent to which international prices have influenced national prices and poverty and wellbeing outcomes, and considers whether this exceptional period represents an anomaly or likely signals future episodes of food price volatility. It finds that although some factors that contributed to recent events have eased considerably, some significant drivers remain structural threats to future food security. There is little reason to be confident that recent reductions in food prices and volatilities augur well for the food security or wellbeing of those living on low and precarious incomes in the future

    Abstract We have cloned a full-length gene, putatively encoding for caffeoyl-coenzyme A 3-O-methyltransferase (CCoAOMT), an important enzyme involved in lignin biosynthesis, from kenaf (Hibiscus cannabinus L.). Herein, we investigated the expression pattern of a CCoAOMT orthologue from various tissues and organs during development, and in response to different environmental cues. The full-length CCoAOMT orthologue of kenaf consists of a 744 bp open reading frame (ORF), encoding for 247 amino acids of 27.91 kDa and an isoelectric point (pI) of 5.43. The deduced amino acids of CCoAOMT evidenced a high degree of identity (up to 84%) with other plant CCoAOMT sequences. Phylogenetic analysis demonstrated its close relationship with the CCoAOMT of Gossypium hirsutum (ACQ59096). Kenaf CCoAOMT harbors eight highly conserved motifs: A, B, and C are putative S-adenosylmethioine (SAM)-binding motifs and D, E, F, G, and H are CCoAOMT signature motifs. According to quantitative real-time reverse transcriptase polymerase chain reaction (q-PCR) analysis, the kenaf CCoAOMT transcript was detected in all plant tissues and organs, whereas the highest expression was noted in mature flower tissues, which indicates that it might be involved in the flower development or in the biosynthesis of flower specific compound. All the treatments highly induced the expression of CCoAOMT transcripts in the stems of 3-week-old kenaf, which indicates that it might have a role in stress regulatory pathway. Among the treatments, the cold and H 2 O 2 -treated samples evidenced the highest levels of expression at 6 and 24 h after treatment, respectively, whereas the wounded and NaCl-treated samples evidenced lower expression levels, which suggest that different signaling networks are involved for stress mediated up regulation of HcCCoAOMT transcripts. The highest transcript level of CCoAOMT was detected at either early (within 12 h of treatments) or intermediate (24 h after treatments) time points of treatments, except drought treated sample. Early induction was observed in the case of H 2 O 2 and SA (salicylic acid), and intermediate induction occurring as the result of wounding, NaCl, cold and ABA (abscisic acid). Whereas drought treated sample showed highest expression at seven days after treatment. MeJA (methyl jasmonic acid) treatment showed a complex biphasic expression which is different from others.     Defluoridation of drinking water using a new flow column-electrocoagulation reactor (FCER) - Experimental, statistical, and economic approach.

    A new batch, flow column electrocoagulation reactor (FCER) that utilises a perforated plate flow column as a mixer has been used to remove fluoride from drinking water. A comprehensive study has been carried out to assess its performance. The efficiency of fluoride removal (R%) as a function of key operational parameters such as initial pH, detention time (t), current density (CD), inter-electrode distance (ID) and initial concentration (C0) has been examined and an empirical model has been developed. A scanning electron microscopy (SEM) investigation of the influence of the EC process on morphology of the surface of the aluminium electrodes, showed the erosion caused by aluminium loss. A preliminary estimation of the reactor's operating cost is suggested, allowing for the energy from recycling of hydrogen gas hydrogen gas produced amount. The results obtained showed that 98% of fluoride was removed within 25 min of electrolysis at pH of 6, ID of 5 mm, and CD of 2 mA/cm(2). The general relationship between fluoride removal and operating parameters could be described by a linear model with R(2) of 0.823. The contribution of the operating parameters to the suggested model followed the order: t > CD > C0 > ID > pH. The SEM images obtained showed that, after the EC process, the surface of the anodes, became non-uniform with a large number of irregularities due to the generation of aluminium hydroxides. It is suggested that these do not materially affect the performance. A provisional estimate of the operating cost was 0.379 US $/m(3). Additionally, it has been found that 0.6 kW/m(3) is potentially recoverable from the H2 gas

    Resilience of refugees displaced in the developing world: a qualitative analysis of strengths and struggles of urban refugees in Nepal

    BACKGROUND: Mental health and psychosocial wellbeing are key concerns in displaced populations. Despite urban refugees constituting more than half of the world's refugees, minimal attention has been paid to their psychosocial wellbeing. The purpose of this study was to assess coping behaviour and aspects of resilience amongst refugees in Kathmandu, Nepal. METHODS: This study examined the experiences of 16 Pakistani and 8 Somali urban refugees in Kathmandu, Nepal through in-depth individual interviews, focus groups, and Photovoice methodology. Such qualitative approaches enabled us to broadly discuss themes such as personal experiences of being a refugee in Kathmandu, perceived causes of psychosocial distress, and strategies and resources for coping. Thematic network analysis was used in this study to systematically interpret and code the data. RESULTS: Our findings highlight that urban refugees' active coping efforts, notwithstanding significant adversity and resulting distress, are most frequently through primary relationships. Informed by Axel Honneth's theory on the struggle for recognition, findings suggest that coping is a function beyond the individual and involves the ability to negotiate recognition. This negotiation involves not only primary relationships, but also the legal order and other social networks such as family and friends. Honneth's work was used because of its emphasis on the importance of legal recognition and larger structural factors in facilitating daily coping. CONCLUSIONS: Understanding how urban refugees cope by negotiating access to various forms of recognition in the absence of legal-recognition will enable organisations working with them to leverage such strengths and develop relevant programmes. In particular, building on these existing resources will lead to culturally compelling and sustainable care for these populations

    Cluster-randomised controlled trial of community mobilisation in Mumbai slums to improve care during pregnancy, delivery, postpartum and for the newborn

    Background: The United Nations Millennium Development Goals look to substantial improvements in child and maternal survival. Morbidity and mortality during pregnancy, delivery and the postnatal period are prime obstacles to achieving these goals. Given the increasing importance of urban health to global prospects, Mumbai's City Initiative for Newborn Health aims to improve maternal and neonatal health in vulnerable urban slum communities, through a combination of health service quality improvement and community participation. The protocol describes a trial of community intervention aimed at improving prevention, care seeking and outcomes.Objective: To test an intervention that supports local women as facilitators in mobilising communities for better health care. Community women's groups will build an understanding of their potential to improve maternal and infant health, and develop and implement strategies to do so.Design: Cluster-randomized controlled trial.Methods: The intervention will employ local community-based female facilitators to convene groups and help them to explore maternal and neonatal health issues. Groups will meet fortnightly through a seven-phase process of sharing experiences, discussion of the issues raised, discovery of potential community strengths, building of a vision for action, design and implementation of community strategies, and evaluation.The unit of allocation will be an urban slum cluster of 1000-1500 households. 48 clusters have been randomly selected after stratification by ward. 24 clusters have been randomly allocated to receive the community intervention. 24 clusters will act as control groups, but will benefit from health service quality improvement. Indicators of effect will be measured through a surveillance system implemented by the project. Key distal outcome indicators will be neonatal mortality and maternal and neonatal morbidity. Key proximate outcome indicators will be home care practices, uptake of antenatal, delivery and postnatal care, and care for maternal and neonatal illness.Data will be collected through a vital registration system for births and deaths in the 48 study clusters. Structured interviews with families will be conducted at about 6 weeks after index deliveries. We will also collect both quantitative and qualitative data to support a process evaluation.Trial registration: Current controlled trials ISRCTN9625679
