    SPOCK (Schnelle Produktentwicklung durch Optimierte Computerbasierte Kommunikation) ist eines von 19 Projekten im Rahmen der G7-GIBN Initiative. Das Ziel des Projektes ist die Einführung von CSCW (Computer Supported Collaborative Working) bei den Partnern von Airbus Industries. Im Airbus Industries-Konsortium arbeiten Ingenieure aus verschiedenen europäischen Ländern zusammen an einem Produkt. Dabei besteht ein großer Bedarf an Kommunikation und Datenaustausch, der bisher noch weitgehend ohne CSCW gehandhabt wird. Am Beispiel des aerodynamischen Tragflächen-Entwurfs, der aus den Teildisziplinen Strömungssimulation, Windkanal-Experiment und Flugversuch besteht, soll das Potential von CSCW-Technologien demonstriert werden. Die Deutsche Telekom und andere europäische Telekommunikationsgesellschaften haben mit einem auf ATM-Technologie basierendem Hochgeschwindigkeitsnetz die Grundlage für weltweit verteiltes kooperatives Arbeiten zur Verfügung gestellt

    On the Biological Importance of the 3-hydroxyanthranilic Acid: Anthranilic Acid Ratio

    Of the major components of the kynurenine pathway for the oxidative metabolism of tryptophan, most attention has focussed on the N-methyl-D-aspartate (NMDA) receptor agonist quinolinic acid, and the glutamate receptor blocker kynurenic acid. However, there is increasing evidence that the redox-active compound 3-hydroxyanthranilic acid may also have potent actions on cell function in the nervous and immune systems, and recent clinical data show marked changes in the levels of this compound, associated with changes in anthranilic acid levels, in patients with a range of neurological and other disorders including osteoporosis, chronic brain injury, Huntington’s disease, coronary heart disease, thoracic disease, stroke and depression. In most cases, there is a decrease in 3-hydroxyanthranilic acid levels and an increase in anthranilic acid levels. In this paper, we summarise the range of data obtained to date, and hypothesise that the levels of 3-hydroxyanthranilic acid or the ratio of 3-hydroxyanthranilic acid to anthranilic acid levels, may contribute to disorders with an inflammatory component, and may represent a novel marker for the assessment of inflammation and its progression. Data are presented which suggest that the ratio between these two compounds is not a simple determinant of neuronal viability. Finally, a hypothesis is presented to account for the development of the observed changes in 3-hydroxyanthranilic acid and anthranilate levels in inflammation and it is suggested that the change of the 3HAA:AA ratio, particularly in the brain, could possibly be a protective response to limit primary and secondary damage

    Nuclear actin polymerization rapidly mediates replication fork remodeling upon stress by limiting PrimPol activity

    Cells rapidly respond to replication stress actively slowing fork progression and inducing fork reversal. How replication fork plasticity is achieved in the context of nuclear organization is currently unknown. Using nuclear actin probes in living and fixed cells, we visualized nuclear actin filaments in unperturbed S phase and observed their rapid extension in number and length upon genotoxic treatments, frequently taking contact with replication factories. Chemically or genetically impairing nuclear actin polymerization shortly before these treatments prevents active fork slowing and abolishes fork reversal. Defective fork remodeling is linked to deregulated chromatin loading of PrimPol, which promotes unrestrained and discontinuous DNA synthesis and limits the recruitment of RAD51 and SMARCAL1 to nascent DNA. Moreover, defective nuclear actin polymerization upon mild replication interference induces chromosomal instability in a PRIMPOL-dependent manner. Hence, by limiting PrimPol activity, nuclear F-actin orchestrates replication fork plasticity and is a key molecular determinant in the rapid cellular response to genotoxic treatments

    Laufende und umfassende Informationen über den Zustand und die Entwicklung der Umwelt sind unabdingbare Voraussetzungen für einen vorsorglichen Umgang mit unseren natürlichen Lebensgrundlagen. In Baden-Württemberg wurden und werden im Bereich der Umweltverwaltung täglich eine große Menge von Meßdaten, Analysedaten, Verwaltungsdokumenten, statistischen Erhebungen und Berechnungen erzeugt. Diese Informationen zu sammeln, zu sichten, zu bewerten und bedarfsgerecht aufzuarbeiten, ist mit den herkömmlichen Methoden der Verwaltungsarbeit nicht mehr möglich, sondern erfordert neue Methoden insbesondere auch der Informationstechnik. In Baden-Württemberg wird auf Beschluß der Landesregierung seit 1986 (jetzt unter Federführung des Ministeriums für Umwelt und Verkehr - UVM) ein Konzept für ein ressortübergreifendes Umweltinformationssystem des Landes Baden-Württemberg entwickelt und umgesetzt

    The kynurenine pathway as a therapeutic target in cognitive and neurodegenerative disorders

    Understanding the neurochemical basis for cognitive function is one of the major goals of neuroscience, with a potential impact on the diagnosis, prevention and treatment of a range of psychiatric and neurological disorders. In this review, the focus will be on a biochemical pathway that remains under-recognised in its implications for brain function, even though it can be responsible for moderating the activity of two neurotransmitters fundamentally involved in cognition – glutamate and acetylcholine. Since this pathway – the kynurenine pathway of tryptophan metabolism - is induced by immunological activation and stress it also stands in an unique position to mediate the effects of environmental factors on cognition and behaviour. Targetting the pathway for new drug development could, therefore, be of value not only for the treatment of existing psychiatric conditions, but also for preventing the development of cognitive disorders in response to environmental pressures

    A trajectory-based strict semantics for program slicing

    We define a program semantics that is preserved by dependence-based slicing algorithms. It is a natural extension, to non-terminating programs, of the semantics introduced by Weiser (which only considered terminating ones) and, as such, is an accurate characterisation of the semantic relationship between a program and the slice produced by these algorithms. Unlike other approaches, apart from Weiser’s original one, it is based on strict standard semantics which models the ‘normal’ execution of programs on a von Neumann machine and, thus, has the advantage of being intuitive. This is essential since one of the main applications of slicing is program comprehension. Although our semantics handles non-termination, it is defined wholly in terms of finite trajectories, without having to resort to complex, counter-intuitive, non-standard models of computation. As well as being simpler, unlike other approaches to this problem, our semantics is substitutive. Substitutivity is an important property becauseit greatly enhances the ability to reason about correctness of meaning-preserving program transformations such as slicing

    Disposable Platform Provides Visual and Color-Based Point-of-Care Anemia Self-Testing

    Anemia, or low blood hemoglobin (Hgb) levels, afflicts 2 billion people worldwide. Currently, Hgb levels are typically measured from blood samples using hematology analyzers, which are housed in hospitals, clinics, or commercial laboratories and require skilled technicians to operate. A reliable, inexpensive point-of-care (POC) Hgb test would enable cost-effective anemia screening and chronically anemic patients to self-monitor their disease. We present a rapid, standalone, and disposable POC anemia test that, via a single drop of blood, outputs color-based visual results that correlate with Hgb levels. METHODS. We tested blood from 238 pediatric and adult patients with anemia of varying degrees and etiologies and compared hematology analyzer Hgb levels with POC Hgb levels, which were estimated via visual interpretation using a color scale and an optional smartphone app for automated analysis. RESULTS. POC Hgb levels correlated with hematology analyzer Hgb levels (r = 0.864 and r = 0.856 for visual interpretation and smartphone app, respectively), and both POC test methods yielded comparable sensitivity and specificity for detecting any anemia (n = 178) (/dl) (sensitivity: 90.2% and 91.1%, specificity: 83.7% and 79.2%, respectively) and severe anemia (n = 10) (/dl) (sensitivity: 90.0% and 100%, specificity: 94.6% and 93.9%, respectively). CONCLUSIONS. These results demonstrate the feasibility of this POC color-based diagnostic test for self-screening/self-monitoring of anemia. TRIAL REGISTRATION. Not applicable. FUNDING.     Abnormal Frontostriatal Activity During Unexpected Reward Receipt in Depression and Schizophrenia: Relationship to Anhedonia.

    Alterations in reward processes may underlie motivational and anhedonic symptoms in depression and schizophrenia. However it remains unclear whether these alterations are disorder-specific or shared, and whether they clearly relate to symptom generation or not. We studied brain responses to unexpected rewards during a simulated slot-machine game in 24 patients with depression, 21 patients with schizophrenia, and 21 healthy controls using functional magnetic resonance imaging. We investigated relationships between brain activation, task-related motivation, and questionnaire rated anhedonia. There was reduced activation in the orbitofrontal cortex, ventral striatum, inferior temporal gyrus, and occipital cortex in both depression and schizophrenia in comparison with healthy participants during receipt of unexpected reward. In the medial prefrontal cortex both patient groups showed reduced activation, with activation significantly more abnormal in schizophrenia than depression. Anterior cingulate and medial frontal cortical activation predicted task-related motivation, which in turn predicted anhedonia severity in schizophrenia.
