    The mushroom, Cordyceps cicadae, ameliorates renal interstitial fibrosis via TLR2-mediated pathways

    Purpose: To evaluate the mushroom, Cordyceps cicadae, for its ability to suppress tissue fibrosis and Toll-like receptors 2 (TLR 2) pathway activation in a mouse model of renal interstitial fibrosis (RIF).Methods: Cordyceps cicadae powder was obtained from BioAsia Group (Shanghai, China). RIF was induced via unilateral ureteral obstruction (UUO) in male C57Bl/6 mice. Animals were treated via the intragastric administration of Cordyceps cicadae powder (0.1g, 0.3 g/ml/100 g/day), beginning 24 h prior to UUO, and the treatment was continued for the following 14 days. Changes in tissue histology were then assessed via hematoxylin and eosin, and Sirius red stainings. Tissue macrophages were characterized based upon their expression of inducible nitric oxide synthase (iNOS) and interleukin-10 (IL-10), while Western blotting technique was used to measure the levels of TLR2, Myeloid differentiation factor 88 (MyD88), and nuclear factor-κB (NF-κB)/p-NF-κB in samples from these animals.Results: Treatment with Cordyceps cicadae powder is associated with a shift in macrophage phenotype that in turn decreased the production of extracellular matrix and alleviated RIF occurrence in mice model.Conclusion: This mechanistic study highlights the novel potential approach for treating and preventing RIF using Cordyceps cicadae powder.     Open-Set Object Detection Using Classification-free Object Proposal and Instance-level Contrastive Learning with Appendix

    Detecting both known and unknown objects is a fundamental skill for robot manipulation in unstructured environments. Open-set object detection (OSOD) is a promising direction to handle the problem consisting of two subtasks: objects and background separation, and open-set object classification. In this paper, we present Openset RCNN to address the challenging OSOD. To disambiguate unknown objects and background in the first subtask, we propose to use classification-free region proposal network (CF-RPN) which estimates the objectness score of each region purely using cues from object's location and shape preventing overfitting to the training categories. To identify unknown objects in the second subtask, we propose to represent them using the complementary region of known categories in a latent space which is accomplished by a prototype learning network (PLN). PLN performs instance-level contrastive learning to encode proposals to a latent space and builds a compact region centering with a prototype for each known category. Further, we note that the detection performance of unknown objects can not be unbiasedly evaluated on the situation that commonly used object detection datasets are not fully annotated. Thus, a new benchmark is introduced by reorganizing GraspNet-1billion, a robotic grasp pose detection dataset with complete annotation. Extensive experiments demonstrate the merits of our method. We finally show that our Openset RCNN can endow the robot with an open-set perception ability to support robotic rearrangement tasks in cluttered environments.     User Response Learning for Directly Optimizing Campaign Performance in Display Advertising

    Learning and predicting user responses, such as clicks and conversions, are crucial for many Internet-based businesses including web search, e-commerce, and online advertising. Typically, a user response model is established by optimizing the prediction accuracy, e.g., minimizing the error between the prediction and the ground truth user response. However, in many practical cases, predicting user responses is only part of a rather larger predictive or optimization task, where on one hand, the accuracy of a user response prediction determines the final (expected) utility to be optimized, but on the other hand, its learning may also be influenced from the follow-up stochastic process. It is, thus, of great interest to optimize the entire process as a whole rather than treat them independently or sequentially. In this paper, we take real-time display advertising as an example, where the predicted user's ad click-through rate (CTR) is employed to calculate a bid for an ad impression in the second price auction. We reformulate a common logistic regression CTR model by putting it back into its subsequent bidding context: rather than minimizing the prediction error, the model parameters are learned directly by optimizing campaign profit. The gradient update resulted from our formulations naturally fine-tunes the cases where the market competition is high, leading to a more cost-effective bidding. Our experiments demonstrate that, while maintaining comparable CTR prediction accuracy, our proposed user response learning leads to campaign profit gains as much as 78.2% for offline test and 25.5% for online A/B test over strong baselines

    N6-(2-Hydroxyethyl) Adenosine From Cordyceps cicadae Ameliorates Renal Interstitial Fibrosis and Prevents Inflammation via TGF-β1/Smad and NF-κB Signaling Pathway

    Renal interstitial fibrosis is characterized by inflammation and an excessive accumulation of extracellular matrix, which leads to end-stage renal failure. Our previous studies have shown that a natural product from Cordyceps cicadae can ameliorate chronic kidney diseases. N6-(2-Hydroxyethyl) adenosine (HEA), a physiologically active compound in C. cicadae, has been identified as a Ca2+ antagonist and an anti-inflammatory agent in pharmacological tests. However, its role in renal interstitial fibrosis and the underlying mechanism remains unclear. Here, unilateral ureteral obstruction (UUO) was used to induce renal interstitial fibrosis in male C57BL/6 mice. Different doses of HEA (2.5, 5, and 7.5 mg/kg) were given by intraperitoneal injection 24 h before UUO, and the treatment was continued for 14 days post-operatively. Histologic changes were examined by hematoxylin & eosin, Masson’s trichrome, and picrosirius red stain. Quantitative real-time PCR analysis, enzyme-linked immunosorbent assays, immunohistochemistry, and western blot analysis were used to evaluate proteins levels. And the results showed that HEA significantly decreased UUO-induced renal tubular injury and fibrosis. In vivo, HEA apparently decreased UUO-induced inflammation and renal fibroblast activation by suppression of the NF-κB and TGF-β1/Smad signaling pathway. In vitro, HEA also obviously decreased lipopolysaccharide-induced inflammatory cytokine level in RAW 264.7 cells and TGF-β1-induced fibroblast activation in NRK-49F cells by modulating NF-κB and TGF-β1/Smad signaling. In general, our findings indicate that HEA has a beneficial effect on UUO-induced tubulointerstitial fibrosis by suppression of inflammatory and renal fibroblast activation, which may be a potential therapy in chronic conditions such as renal interstitial fibrosis

    Effects of aroma quality on the biotransformation of natural 2-phenylethanol produced using ascorbic acid

    Background: Natural 2-phenylethanol (2-PE) is an important flavoring that emits the aroma of roses. During biotransformation, the aroma quality of natural 2-PE is affected by its main by-products, which include butanol, isobutyric acid, butyric acid, and isovaleric acid. Thus, controlling undesirable by-product formation can reduce the effect of odor on 2-PE aroma quality. Results: 2-PE was produced through biotransformation using L-phenylalanine as a substrate and glucose as a carbon source. Ascorbic acid was added to the system to improve the redox reaction and suppress the generation of by-products. Principal component analysis of the aroma quality of 2-PE was performed using an electronic nose. Similarity analysis revealed that the effects of four by-products on 2-PE aroma quality may be ranked in the following order: isovaleric acid < butyric acid < isobutyric acid < butanol. The sample that exhibited the best similarity to the standard 2-PE sample (99.19%) was the sample to which ascorbic acid had been added during glucose metabolism. Conclusions: 2-PE produced through the addition of ascorbic acid exhibited the closest aroma similarity to the standard 2-PE sample

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.     Juxtaposing BTE and ATE – on the role of the European insurance industry in funding civil litigation

    One of the ways in which legal services are financed, and indeed shaped, is through private insurance arrangement. Two contrasting types of legal expenses insurance contracts (LEI) seem to dominate in Europe: before the event (BTE) and after the event (ATE) legal expenses insurance. Notwithstanding institutional differences between different legal systems, BTE and ATE insurance arrangements may be instrumental if government policy is geared towards strengthening a market-oriented system of financing access to justice for individuals and business. At the same time, emphasizing the role of a private industry as a keeper of the gates to justice raises issues of accountability and transparency, not readily reconcilable with demands of competition. Moreover, multiple actors (clients, lawyers, courts, insurers) are involved, causing behavioural dynamics which are not easily predicted or influenced. Against this background, this paper looks into BTE and ATE arrangements by analysing the particularities of BTE and ATE arrangements currently available in some European jurisdictions and by painting a picture of their respective markets and legal contexts. This allows for some reflection on the performance of BTE and ATE providers as both financiers and keepers. Two issues emerge from the analysis that are worthy of some further reflection. Firstly, there is the problematic long-term sustainability of some ATE products. Secondly, the challenges faced by policymakers that would like to nudge consumers into voluntarily taking out BTE LEI
