    Isolation and Characterization of Eight Polymorphic Microsatellites for the Spotted Spiny Lobster, Panulirus guttatus

    Microsatellite sequences were isolated from enriched genomic libraries of the spotted spiny lobster, Panulirus guttatus using 454 pyrosequencing. Twenty-nine previously developed polymerase chain reaction primer pairs of Panulirus argus microsatellite loci were also tested for cross-species amplification in Panulirus guttatus. In total, eight consistently amplifying, and polymorphic loci were characterized for 57 individuals collected in the Florida Keys and Bermuda. The number of alleles per locus ranged from 8 to 20 and observed heterozygosities ranged from 0.409 to 0.958. Significant deviations from Hardy–Weinberg equilibrium were found in one locus from Florida and three loci from Bermuda. Quality control testing indicated that all loci were easy to score, highly polymorphic and showed no evidence of linkage disequilibrium. Null alleles were detected in three loci with moderate frequencies ranging from (20% to 22%).     Oceanographic features and limited dispersal shape the population genetic structure of the vase sponge Ircinia campana in the Greater Caribbean

    Understanding population genetic structure can help us to infer dispersal patterns, predict population resilience and design effective management strategies. For sessile species with limited dispersal, this is especially pertinent because genetic diversity and connectivity are key aspects of their resilience to environmental stressors. Here, we describe the population structure of Ircinia campana, a common Caribbean sponge subject to mass mortalities and disease. Microsatellites were used to genotype 440 individuals from 19 sites throughout the Greater Caribbean. We found strong genetic structure across the region, and significant isolation by distance across the Lesser Antilles, highlighting the influence of limited larval dispersal. We also observed spatial genetic structure patterns congruent with oceanography. This includes evidence of connectivity between sponges in the Florida Keys and the southeast coast of the United States (>700 km away) where the oceanographic environment is dominated by the strong Florida Current. Conversely, the population in southern Belize was strongly differentiated from all other sites, consistent with the presence of dispersal-limiting oceanographic features, including the Gulf of Honduras gyre. At smaller spatial scales (<100 km), sites showed heterogeneous patterns of low-level but significant genetic differentiation (chaotic genetic patchiness), indicative of temporal variability in recruitment or local selective pressures. Genetic diversity was similar across sites, but there was evidence of a genetic bottleneck at one site in Florida where past mass mortalities have occurred. These findings underscore the relationship between regional oceanography and weak larval dispersal in explaining population genetic patterns, and could inform conservation management of the species

    Using genetics to inform restoration and predict resilience in declining populations of a keystone marine sponge

    Genetic tools can have a key role in informing conservation management of declining populations. Genetic diversity is an important determinant of population ftness and resilience, and can require careful management to ensure sufcient variation is present. In addition, population genetics data reveal patterns of connectivity and gene fow between locations, enabling mangers to predict recovery and resilience, identify areas of local adaptation, and generate restoration plans. Here, we demonstrate a conservation genetics approach to inform restoration and management of the loggerhead sponge (Spheciospongia vesparium) in the Florida Keys, USA. This species is a dominant, habitat-forming component of marine ecosystems in the Caribbean region, but in Florida has sufered numerous mass mortality events. We developed microsatellite markers and used them to genotype sponges from 14 locations in Florida and a site each in The Bahamas, Belize and Barbuda. We found that genetic diversity levels were similar across all sites, but inbreeding and bottleneck signatures were present in Florida. Populations are highly structured at the regional scale, whilst within Florida connectivity is present in a weak isolation by distance pattern, coupled with chaotic genetic patchiness. Evidence of a weak barrier to gene fow was found in Florida among sites situated on opposite sides of the islands in the Middle Keys. Loggerhead sponge populations in Florida are vulnerable in the face of mass mortalities due to low connectivity with other areas in the region, as well as distance-limited and unpredictable local connectivity patterns. However, our discovery of Florida’s high genetic diversity increases hope for resilience to future perturbations.     Ethnic mirrors: self-representations in the Welsh and Mennonite museums in Argentina and Paraguay

    According to some scholars and philosophers, ethnic identities are the best political, social, economic, ethic (and even aesthetic) alternative to State centralism, which is incapable of dealing with cultural diversity. Ethnic communitarism is then defined as a more authentic, humane, democratic and inclusive form of organization. The Welsh colonies of Chubut (Argentine) and the established Mennonite colonies of the Chaco Region (Paraguay) are two ethnic groups with forms of community life that have been thoroughly studied from different perspectives. However, neither has been analyzed their point of view of alterity or their relation with those who do not belong to the community. In their museums the history of the community is represented, self-images and other people's images are constructed and spread. The interesting part of these stories is not what they say but what they do, the form in which contents are expressed. These communitarian historical museums tell about the past but they mainly have an impact on the present. Like national or even imperial museums, Welsh and Mennonite museums tend to naturalize a particular self-centered, prejudicial and evolutionist point of view that often excludes other perspectives, especially those elaborated by the neighboring indigenous communities. In contrast, we believe it is necessary to take a stance for democratic, horizontal relations between communities and more polyphonic and responsible historical representations.Alguns filósofos e acadêmicos assinalam que as identidades étnicas são a melhor alternativa política, social, econômica, ética (e mesmo estética) ao centralismo estatal, que é negligente ao lidar com a diversidade cultural. O comunitarismo étnico é definido como uma forma de organização mais autêntica, humana, democrática e inclusiva. As colônias galesas de Chubut (Argentina) e as colônias rurais dos menonitas no Chaco (Paraguay) são dois grupos étnicos cujas vidas comunitárias têm sido muito estudadas desde diversas perspectivas, mas o seu ponto de vista acerca da alteridade ou sua relação com os atores extracomunitários nunca foi levado em conta. A historia comunitária é representada nos museus galeses e menonitas das respectivas regiões e aí são construídas e difundidas não só as autoimagens mas também as representações dos outros. O aspecto relevante dessas histórias não é o que elas dizem, mas o que fazem, a forma como os conteúdos são expressos. Esses museus históricos comunitários falam sobre o passado, mas seu maior impacto recai sobre o presente. Como os museus nacionais ou imperiais, os museus galeses e menonitas tentam naturalizar pontos de vista particulares, auto-centrados, preconceituosos e evolucionistas que geralmente excluem as perspectivas elaboradas pelas comunidades indígenas vizinhas.     The development and validation of a scoring tool to predict the operative duration of elective laparoscopic cholecystectomy

    Background: The ability to accurately predict operative duration has the potential to optimise theatre efficiency and utilisation, thus reducing costs and increasing staff and patient satisfaction. With laparoscopic cholecystectomy being one of the most commonly performed procedures worldwide, a tool to predict operative duration could be extremely beneficial to healthcare organisations. Methods: Data collected from the CholeS study on patients undergoing cholecystectomy in UK and Irish hospitals between 04/2014 and 05/2014 were used to study operative duration. A multivariable binary logistic regression model was produced in order to identify significant independent predictors of long (> 90 min) operations. The resulting model was converted to a risk score, which was subsequently validated on second cohort of patients using ROC curves. Results: After exclusions, data were available for 7227 patients in the derivation (CholeS) cohort. The median operative duration was 60 min (interquartile range 45–85), with 17.7% of operations lasting longer than 90 min. Ten factors were found to be significant independent predictors of operative durations > 90 min, including ASA, age, previous surgical admissions, BMI, gallbladder wall thickness and CBD diameter. A risk score was then produced from these factors, and applied to a cohort of 2405 patients from a tertiary centre for external validation. This returned an area under the ROC curve of 0.708 (SE = 0.013, p  90 min increasing more than eightfold from 5.1 to 41.8% in the extremes of the score. Conclusion: The scoring tool produced in this study was found to be significantly predictive of long operative durations on validation in an external cohort. As such, the tool may have the potential to enable organisations to better organise theatre lists and deliver greater efficiencies in care

    Development and Validation of a Risk Score for Chronic Kidney Disease in HIV Infection Using Prospective Cohort Data from the D:A:D Study

Development and implementation of a risk score model for CKD would allow comparison of the risks and benefits of adding potentially nephrotoxic antiretrovirals to a treatment regimen and would identify those at greatest risk of CKD. The aims of this study were to develop a simple, externally validated, and widely applicable long-term risk score model for CKD in HIV-positive individuals that can guide decision making in clinical practice. Methods and Findings A total of 17,954 HIV-positive individuals from the Data Collection on Adverse Events of Anti-HIV Drugs (D:A:D) study with >= 3 estimated glomerular filtration rate (eGFR) values after 1 January 2004 were included. Baseline was defined as the first eGFR > 60 ml/min/1.73 m2 after 1 January 2004; individuals with exposure to tenofovir, atazanavir, atazanavir/ritonavir, lopinavir/ritonavir, other boosted protease inhibitors before baseline were excluded. CKD was defined as confirmed (>3 mo apart) eGFR In the D:A:D study, 641 individuals developed CKD during 103,185 person-years of follow-up (PYFU; incidence 6.2/1,000 PYFU, 95% CI 5.7-6.7; median follow-up 6.1 y, range 0.3-9.1 y). Older age, intravenous drug use, hepatitis C coinfection, lower baseline eGFR, female gender, lower CD4 count nadir, hypertension, diabetes, and cardiovascular disease (CVD) predicted CKD. The adjusted incidence rate ratios of these nine categorical variables were scaled and summed to create the risk score. The median risk score at baseline was -2 (interquartile range -4 to 2). There was a 1: 393 chance of developing CKD in the next 5 y in the low risk group (risk score = 5, 505 events), respectively. Number needed to harm (NNTH) at 5 y when starting unboosted atazanavir or lopinavir/ritonavir among those with a low risk score was 1,702 (95% CI 1,166-3,367); NNTH was 202 (95% CI 159-278) and 21 (95% CI 19-23), respectively, for those with a medium and high risk score. NNTH was 739 (95% CI 506-1462), 88 (95% CI 69-121), and 9 (95% CI 8-10) for those with a low, medium, and high risk score, respectively, starting tenofovir, atazanavir/ritonavir, or another boosted protease inhibitor. The Royal Free Hospital Clinic Cohort included 2,548 individuals, of whom 94 individuals developed CKD (3.7%) during 18,376 PYFU (median follow-up 7.4 y, range 0.3-12.7 y). Of 2,013 individuals included from the SMART/ESPRIT control arms, 32 individuals developed CKD (1.6%) during 8,452 PYFU (median follow-up 4.1 y, range 0.6-8.1 y). External validation showed that the risk score predicted well in these cohorts. Limitations of this study included limited data on race and no information on proteinuria. Conclusions Both traditional and HIV-related risk factors were predictive of CKD. These factors were used to develop a risk score for CKD in HIV infection, externally validated, that has direct clinical relevance for patients and clinicians to weigh the benefits of certain antiretrovirals against the risk of CKD and to identify those at greatest risk of CKD.Peer reviewe

L’idée de l’art et le besoin d’en retracer l’histoire sont apparus ensemble; ils ont eu comme fonction de baliser, par objets interposés, la trajectoire et l’accomplissement de sujets humains (entendre européens, blancs et mâles). Le dispositif des grandes expositions universelles, mises en scènes spectaculaires de l’appropriation impérialiste des cultures de tous les peuples du globe, n’est pas étranger aux procédés de l’histoire de l’art. Celle-ci doit au développement de la photographie d’avoir aménagé son propre site virtuel d’exposition, gigantesque musée imaginaire (dont les musées existants réalisent en partie le projet) qu’elle réorganise à l’infini sans que soient vraiment remis en cause la visée et la fonction du système. L’histoire de l’art demeure liée à un modernisme raciste et phallocentrique, et ceci malgré les revendications de tous les postmodernismes critiques
