Influence of naturally-occurring 5′-pyrophosphate-linked substituents on the binding of adenylic inhibitors to ribonuclease a: An X-ray crystallographic study
Ribonuclease A is the archetype of a functionally diverse superfamily of vertebrate-specific ribonucleases. Inhibitors of its action have potential use in the elucidation of the in vivo roles of these enzymes and in the treatment of pathologies associated therewith. Derivatives of adenosine 5′-pyrophosphate are the most potent nucleotide-based inhibitors known. Here, we use X-ray crystallography to visualize the binding of four naturally-occurring derivatives that contain 5′-pyrophosphate-linked extensions. 5′-ATP binds with the adenine occupying the B2 subsite in the manner of an RNA substrate but with the γ-phosphate at the P1 subsite. Diadenosine triphosphate (Ap3A) binds with the adenine in syn conformation, the β-phosphate as the principal P1 subsite ligand and without order beyond the γ-phosphate. NADPH and NADP+ bind with the adenine stacked against an alternative rotamer of His119, the 2′-phosphate at the P1 subsite, and without order beyond the 5′-α-phosphate. We also present the structure of the complex formed with pyrophosphate ion. The structural data enable existing kinetic data on the binding of these compounds to a variety of ribonucleases to be rationalized and suggest that as the complexity of the 5′-linked extension increases, the need to avoid unfavorable contacts places limitations on the number of possible binding modes. © 2009 Wiley Periodicals, Inc. Thalamic neuromodulation and its implications for executive networks
The thalamus is a key structure that controls the routing of information in the brain. Understanding modulation at the thalamic level is critical to understanding the flow of information to brain regions involved in cognitive functions, such as the neocortex, the hippocampus, and the basal ganglia. Modulators contribute the majority of synapses that thalamic cells receive, and the highest fraction of modulator synapses is found in thalamic nuclei interconnected with higher order cortical regions. In addition, disruption of modulators often translates into disabling disorders of executive behavior. However, modulation in thalamic nuclei such as the midline and intralaminar groups, which are interconnected with forebrain executive regions, has received little attention compared to sensory nuclei. Thalamic modulators are heterogeneous in regards to their origin, the neurotransmitter they use, and the effect on thalamic cells. Modulators also share some features, such as having small terminal boutons and activating metabotropic receptors on the cells they contact. I will review anatomical and physiological data on thalamic modulators with these goals: first, determine to what extent the evidence supports similar modulator functions across thalamic nuclei; and second, discuss the current evidence on modulation in the midline and intralaminar nuclei in relation to their role in executive function
Plastral shape isometry in Western Hermann's tortoise (Testudo hermanni hermanni)
Elliptical Fourier Analysis is a good technique for characterizing the shape of complex biological and non-biological morphologies. The current investigation aimed to study plastral pigmentation contour (black areas of the ventral shell) changes, according to body size (plastral length), in Western Hermann's tortoise (Testudo hermanni hermanni), using Elliptical Fourier Analysis. For this goal, 52 domestic pure tortoises from authorized private breeders were selected, ventral pictures were individually taken and their contour automatically digitized and straight-line plastron length obtained. Straight-line plastron length ranged from 83.6 to 150.6 mm (for males) and from 78.9 to 171.8 mm (for females).. A regression was performed for second and third harmonics as dependent variables, against plastral length (log transformed) as independent variable. Based on this sample, it is demonstrated that plastral pigmentation design does not change along animals’ life-history. Therefore, a plastral pigmentation shape isometry can be supposed, at least for this subspecies
CaracterÃsticas da obstrução das pequenas vias aéreas na DPOC
Resumo: Foram estudados 159 doentes, estratificados pelos 5 graus de gravidade do GOLD, provindo de 2 grupos: um grupo de doentes submetidos a ressecção de tumores pulmonares periféricos (100 doentes) e outro (59 doentes), mais grave, que participou no National Emphysema Treatment Trial (NETT). Para além do FEV1, as pequenas vias aéreas (PVA) inferiores a 2 mm foram estudadas em termos de: área luminal máxima, conteúdo luminal, espessura da parede do brônquio, número de vias aéreas inflamadas fundamentalmente verificada no estádio IV (33 no total de 43 doentes). O número de PVA analisadas foi igual em todos os grupos, assim o comprimento da membrana basal. Foi encontrada uma correlação significativa entre o estadiamento GOLD (FEV1) e o conteúdo luminal, mas essa correlação foi ainda mais significativa com a espessura brônquica total (e também com os compartimentos epitélio, lâmina própria, músculo liso e adventÃcia, sem diferenças entre eles). O número de vias aéreas com infiltrados inflamatórios e o número de cada tipo de célula inflamatória aumentou com a gravidade por estádio. No entanto, nos estádios III e IV houve um número muito mais elevado de linfócitos B, CD8 e de folÃculos linfóides. A análise multivariável revelou que a espessura da parede brônquica foi a que teve a correlação mais forte com a progressão da DPOC. Na discussão são referidos os seguintes aspectos: - a espessura da parede brônquica é a caracterÃstica patológica mais associada à progressão da doença, seja por um processo de reparação ou de remodelação - o conteúdo luminal (exsudado inflamatório), o nº de PVA inflamadas e o número de células teve uma associação significativa, mas mais fraca, com a progressão da doença e suas caracterÃsticas (infiltração de neutrófilos, macrófagos, eosinófilos e linfócitos B e CD4 e CD8) e número de cada grupo celular (e presença de folÃculos linfóides). A idade média foi de 63 a 67 anos, e 39 doentes estavam no estádio 0 e I, 22 no estádio II, 16 no III e 43 no IV. Em termos de exposição ao tabaco, os doentes tinham entre 40 e 67 UMA, e nos estádios III e IV não havia fumadores activos e tinham cessado há 9 anos. A exposição a corticóides pré-operatório foi â a presença de muco a ocluir as PVA revela que o processo de hipersecreção caracterÃstico da bronquite crónica também existe nas PVA. Mas dados de outros estudos apontam para que o processo inflamatório possa ser independente nas grandes e nas pequenas vias e para que a progressão da DPOC (FEV1) seja independente da presença de bronquite crónica. â a presença progressivamente aumentada sobretudo de linfócitos B e CD8 (e a associação com folÃculos linfóides) com a gravidade da doença (estádios III e IV), e não de outros tipos celulares, pode ter a ver com um maior tempo de cessação tabágica e maior exposição a corticóides. Estes aspectos celulares e a presença organizada de folÃculos linfóides pode estar associada à colonização e infecção bacteriana caracterÃstica dos graus mais avançados da DPOC. - o aumento da espessura mais superficial à camada muscular (epitelial e sub-epetilial) pode explicar por que é que a presença de hiperreactividade brônquica é um dos melhores preditores do declÃnio acelerado do FEV1. â a presença aumentada de tecido conjuntivo (fibrose) subepitelial e na adventÃcia pode contribuir para a obstrução fixa e impedir que as PVA abram adequadamente na inspiração
