    Upwind responses of Anopheles stephensi to carbon dioxide and L-lactic acid: an olfactometer study.

    Excretion of carbon dioxide and L-lactic acid through exhalation and perspiration provides olfactory signals to mosquitoes which allow them to find and bite humans; however, mosquito species differ in this regard. This study investigated upwind responses of Anopheles stephensi, mysorensis form, an important malaria vector in Asia, to carbon dioxide and L-lactic acid under laboratory conditions. While a minimal dose of carbon dioxide (90 ppm) activated the mosquitoes, 10 times this amount suppressed them. L-lactic acid alone did not produce a significant effect by itself, but addition of 6 microg/min of L-lactic acid to a range of 90 to 410 ppm carbon dioxide resulted in attraction. The results provide further support for the hypothesis that CO2 plays an important role in the host-seeking behaviour of zoophilic mosquitoes, and suggests that L-lactic acid might play a more critical role than CO2 in the attraction of An. stephensi

    Microsporidium infecting Anopheles supepictus (Diptera: Culicidae) larvae

    Background: Microsporidia are known to infect a wide variety of animals including mosquitoes (Diptera: Culicidae). In a recent study on the mosquito fauna of Chahar Mahal and Bakhtiari Province, at the central western part of Iran, a few larvae of Anopheles superpictus were infected with a microsporidium-resembled microorganism. Current investigation deals with the identification of the responsible microorganism at the genus level. Methods: Fresh infected larvae were collected from the field. After determining the species identity they were dissected to extract their infective contents. Wet preparations were checked for general appearance and the size of the pathogenic microorganism. Fixed preparations were stained with Geimsa and Ryan-Blue modified Trichrome techniques to visualize further morphological characters. The obtained light microscopy data were used in the identification process. Results: The infected larvae were bulged by a whitish material filling the involved segments corresponding to a microsporidium infection. Bottle-shaped semioval spores ranged 4.33±0.19×2.67±0.12 and 4.18±0.43×2.45±0.33 micron in wet and fixed preparations, respectively. They were mostly arranged in globular structures comprised of 8 spores. These data was in favor of a species from the genus Parathelohania in the family Ambliosporidae. Conclusion: This is the first report of a microsporidium infection in An. superpictus. The causative agent is diagnosed as a member of the genus Parathelohania. Further identification down to the species level needs to determine its ultrastructural characteristics and the comparative analysis of ss rRNA sequence data. It is also necessary to understand the detail of the components of the transmission cycle

    Vertical Distribution, Biodiversity, and Some Selective Aspects of the Physicochemical Characteristics of the Larval Habitats of Mosquitoes (Diptera: Culicidae) in Chaharmahal and Bakhtiari Province, Iran

    Background and aims: Mosquitoes (Diptera: Culicidae) are still a focus of research because of their role in the transmission of diseases and annoying biting behavior. Source reduction is an effective measure to control mosquito populations, which is based on good knowledge of larval habitats. This study was conducted to obtain that basic knowledge in Chaharmahal and Bakhtiari province. Methods: This study was carried out in 2011 and 2012. Geographical coordinates, altitude, pH, temperature, and the dissolved oxygen level of larval habitats were recorded by relevant devices, followed by documenting physical attributes by direct observation. In addition, the indices of biodiversity were calculated to analyze the vertical biodiversity of species. Finally, the affinity index was calculated to elucidate species co-occurrence. Results: Eighteen species were recovered from 92 larval habitats. Low- (≤ 1400 m), mid- (1401–2000 m), and high- (≥ 2001 m) altitudes lodged 7, 17, and 14 species, respectively. Further, the indices of the species richness and biodiversity for these altitudinal categories were 0.93, 1.94, and 1.58, as well as 1.54, 2.13, and 1.96, respectively. Larval habitats were mostly natural, temporary, with standing but clear water, muddy substrate, sunlit, and with vegetation. Other physicochemical characteristics and affinity of species were described and discussed as well. Conclusion: To the best of our knowledge, this is the first report of vertical distribution and biodiversity of mosquito larvae in Iran. The relative uniformity of physicochemical characteristics of larval habitats was attributed to prevailing water resources in the studied area and sampling design.     Fauna and the distribution of mosquitoes (Diptera: Culicidae) in Chahar Mahal and Bakhtiari province, 2011-2012

    زمینه و هدف: پشه‌ها خانواده‌ی بزرگی از دوبالان را شامل می‌شوند که به لحاظ توانایی انتقال انواع عوامل بیماری‌زا در پزشکی و بهداشت اهمیت فراوانی دارند. این مطالعه به منظور تکمیل اطلاعات موجود در زمینه‌ی تنوع گونه‌ای و انتشار پشه‌های بومی استان چهارمحال و بختیاری انجام گرفته است. روش بررسی: این پژوهش توصیفی و مقطعی در طول تابستان 1390 و 1391 در تمامی شهرستان‌های استان چهارمحال و بختیاری انجام گردید. نمونه‌ها به روش ملاقه زنی جمع آوری و با استفاده از کلیدهای معتبر شناسایی شدند. داده‌های تعیین هویت و مختصات جغرافیایی نقاط نمونه برداری در نرم افزار ArcGIS 10 وارد گردید تا نقشه‌ی پراکندگی هر یک از گونه‌ها به دست آید. یافته ها: در این مطالعه، 8335 لارو از 92 زیستگاه لاروی جمع آوری گردید که به 18 گونه شامل کولیستا لانجی آرئولاتا، کولیستا ساب اوکریا، اوکلروتاتوس کاسپیوس سنسولاتو، کولکس تیلری، کولکس تریتانس، کولکس هورتنزیس، کولکس پیپینس، کولکس پرگزیگوس، کولکس میمتیکوس، کولکس ترای تنیورینکوس، کولکس اربیینی، کولکس لاتیسینکتوس، آنوفل ماکولیپنیس سنسولاتو، آنوفل سوپرپیکتوس، آنوفل دتالی، آنوفل مارتری، آنوفل کلاویژر و آنوفل تورکودی تعلق داشتند. کولکس تریتانس به عنوان یک گونه‌ی جدید برای استان چهارمحال و بختیاری گزارش می‌گردد. کولکس تیلری و آنوفل سوپرپیکتوس بیش ترین فراوانی و گسترده ترین انتشار را در استان نشان دادند. نتیجه گیری: این مطالعه نشان می‌دهد که برخی از مهم ترین پشه‌های ناقل بیماری‌ها در سطح استان چهارمحال و بختیاری از تنوع و پراکندگی وسیعی برخوردار هستند. برای تعیین توانایی پشه‌های بومی در انتقال عوامل بیماری زا در استان به مطالعات بیشتری نیاز است

    Differential Responses of Anopheles stephensi (Diptera: Culicidae) to Skin Emanations of a Man, a Cow, and a Guinea Pig in the Olfactometer

    Background: Biting habit of mosquitoes plays an important role in the epidemiology of mosquito-borne diseases. Mosquitoes use a set of elaborate sensory modalities to find their preferred hosts by exploiting cues emanating from a nearby host. It has been suggested that the chemical profile of skin can provide further support for anthropophilic mosquito species to find their suitable hosts. This study aimed at revealing the value of skin emanation for a zoophilic species like Anopheles stephensi as a model. Methods: Skin emanations of a man, a cow and a Guinea pig were collected by ethanol soaked cottons. Upwind responses of mosquitoes to 100 and 200 mu l of filtered skin materials were non-competitively explored in a dual-choice olfactometer. L-lactic acid and other chemical content of the skin samples were identified by an enzymatic kit and GC-MS, respectively. Results: Unexpectedly, only human skin emanation was resulted in the statistically significant activation and attraction responses of An. stephensi in the wind tunnel. L-lactic acid content of this skin sample was 10 and 29 times more than the cow and the Guinea pig, respectively. The possible role of lactic acid and a few other identified compounds have been discussed here. Conclusion: Anopheles stephensi showed higher and more specific upwind responses to human skin emanation in the olfactometer. Undoubtedly, the thorough explanation of this unexpected finding needs further investigation. But, if new data verify this result, then, it may be necessary to reconsider the role of skin emanation and thence the human blood index and vectorial capacity of this zoophilic mosquito

    Fabrication of an olfactometer for mosquito behavioural studies

    Background & objectives: Olfaction is the major sensory modality involved in the resource searching behaviour of insects including vector mosquitoes (Diptera: Culicidae). To date, our current country-wide knowledge on the host-seeking behaviour of Iranian mosquitoes is mainly confined to host preference which has exclusively come from field studies. Olfactometer is a scientific tool by which more naive aspects of man-vector contact can be clarified under controlled and less biased conditions. Methods: The wind tunnel and stimulus delivery system was constructed from acrylic materials based on previously introduced models with some modifications. Air supply and required light were ensured by a powerful compressor and incandescent bulbs, respectively. Desired level of temperature was maintained by controllable heating radiators. For humidity production a unique in-built piezo system was devised in the course of the air flow. Fine regulators facilitated the continuous generatation of the humidity at a preset level. Results: Titanium tetrachloride smoke plus monitoring of the wind speed revealed that the flow of air was proper and invariable. A desired level of humidity and temperature could be set up in just 10 and 15-45 min, respectively. These physical parameters varied only +/-2% (humidity) and +/-0.15 degrees C (temperature) in a typical 20 min duration. Conclusion: The first sophisticated olfactometer in the field of medical entomology in Iran is reported here. Fast set up and stability of physical parameters are its salient features. It is expected that with the aid of this olfactometer further information on the physiological principles of the host-seeking behaviour of mosquitoes become available soon

    Inflammaging is driven by upregulation of innate immune receptors and systemic interferon signaling and is ameliorated by dietary restriction

    Aging is characterized by a chronic low-grade inflammation known as inflammaging in multiple tissues, representing a risk factor for age-related diseases. Dietary restriction (DR) is the best-known non-invasive method to ameliorate aging in many organisms. However, the molecular mechanism and the signaling pathways that drive inflammaging across different tissues and how they are modulated by DR are not yet understood. Here we identify a multi-tissue gene network regulating inflammaging. This network is characterized by chromatin opening and upregulation in the transcription of innate immune system receptors and by activation of interferon signaling through interferon regulatory factors, inflammatory cytokines, and Stat1-mediated transcription. DR ameliorates aging-induced alterations of chromatin accessibility and RNA transcription of the inflammaging gene network while failing to rescue those alterations on the rest of the genome. Our results present a comprehensive understanding of the molecular network regulating inflammation in aging and DR and provide anti-inflammaging therapeutic targets

    The Global Burden of Diseases, Injuries and Risk Factors 2017 includes a comprehensive assessment of incidence, prevalence, and years lived with disability (YLDs) for 354 causes in 195 countries and territories from 1990 to 2017. Previous GBD studies have shown how the decline of mortality rates from 1990 to 2016 has led to an increase in life expectancy, an ageing global population, and an expansion of the non-fatal burden of disease and injury. These studies have also shown how a substantial portion of the world's population experiences non-fatal health loss with considerable heterogeneity among different causes, locations, ages, and sexes. Ongoing objectives of the GBD study include increasing the level of estimation detail, improving analytical strategies, and increasing the amount of high-quality data. METHODS: We estimated incidence and prevalence for 354 diseases and injuries and 3484 sequelae. We used an updated and extensive body of literature studies, survey data, surveillance data, inpatient admission records, outpatient visit records, and health insurance claims, and additionally used results from cause of death models to inform estimates using a total of 68 781 data sources. Newly available clinical data from India, Iran, Japan, Jordan, Nepal, China, Brazil, Norway, and Italy were incorporated, as well as updated claims data from the USA and new claims data from Taiwan (province of China) and Singapore. We used DisMod-MR 2.1, a Bayesian meta-regression tool, as the main method of estimation, ensuring consistency between rates of incidence, prevalence, remission, and cause of death for each condition. YLDs were estimated as the product of a prevalence estimate and a disability weight for health states of each mutually exclusive sequela, adjusted for comorbidity. We updated the Socio-demographic Index (SDI), a summary development indicator of income per capita, years of schooling, and total fertility rate. Additionally, we calculated differences between male and female YLDs to identify divergent trends across sexes. Globally, for females, the causes with the greatest age-standardised prevalence were oral disorders, headache disorders, and haemoglobinopathies and haemolytic anaemias in both 1990 and 2017. For males, the causes with the greatest age-standardised prevalence were oral disorders, headache disorders, and tuberculosis including latent tuberculosis infection in both 1990 and 2017. In terms of YLDs, low back pain, headache disorders, and dietary iron deficiency were the leading Level 3 causes of YLD counts in 1990, whereas low back pain, headache disorders, and depressive disorders were the leading causes in 2017 for both sexes combined. All-cause age-standardised YLD rates decreased by 3·9% (95% uncertainty interval [UI] 3·1-4·6) from 1990 to 2017; however, the all-age YLD rate increased by 7·2% (6·0-8·4) while the total sum of global YLDs increased from 562 million (421-723) to 853 million (642-1100). The increases for males and females were similar, with increases in all-age YLD rates of 7·9% (6·6-9·2) for males and 6·5% (5·4-7·7) for females. We found significant differences between males and females in terms of age-standardised prevalence estimates for multiple causes. The causes with the greatest relative differences between sexes in 2017 included substance use disorders (3018 cases [95% UI 2782-3252] per 100 000 in males vs s1400 [1279-1524] per 100 000 in females), transport injuries (3322 [3082-3583] vs 2336 [2154-2535]), and self-harm and interpersonal violence (3265 [2943-3630] vs 5643 [5057-6302]). Global all-cause age-standardised YLD rates have improved only slightly over a period spanning nearly three decades. However, the magnitude of the non-fatal disease burden has expanded globally, with increasing numbers of people who have a wide spectrum of conditions. A subset of conditions has remained globally pervasive since 1990, whereas other conditions have displayed more dynamic trends, with different ages, sexes, and geographies across the globe experiencing varying burdens and trends of health loss. This study emphasises how global improvements in premature mortality for select conditions have led to older populations with complex and potentially expensive diseases, yet also highlights global achievements in certain domains of disease and injury

    Selective aspects of the epizootiology of Parathelohania iranica (Microsporidia: Amblyosporidae)

    Parathelohania is a genus of microsporidia that preferentially attacks Anopheline mosquitoes. This study explored some selective aspects of the epizootiology of Parathelohania iranica (Microsporidia: Amblyosporidae) in its malaria mosquito host Anopheles superpictus s.l. (Diptera: Culicidae). For this study, Sar-rok Village, a place adjacent to the type locality of the parasite, located at the central western part of Iran was visited twice a month to collect host larvae from mid-summer to mid-autumn of 2017-2021. Patent infections were detected by the whitish discoloration of the involved segments. Superficially uninfected larvae were reared up to 26 days in the insectary to elucidate hidden infections. Molecular investigation and laboratory trials were conducted to evaluate the possibility of secondary infections in subsequent days. Morphological characters were used to determine the sex of larvae and adults. Data were entered in SPSS 23.0 and analyzed with relevant statistical tests as needed. In total, 584 P. iranica infected larvae of An. superpictus s.l. were collected in the study years at day zero (D0). Extended observations in the insectary revealed that 849 larvae (84.2 %), 22 dead pupae (2.2 %), and 137 emerged adults (13.6 %) were also infected. In the first two years of the study, the mean infection rate for D0 and D0 + D1-D26 infections was 6.25 % and 15.6 %, respectively. Exposure experiments indicated that subsequent infections in larvae (D1-D26) were not affected by a possible source in the accompanying field water. Patent (D0) infections were seasonal and had about a month delay compared to the general population of larvae. Concealed infections of larvae (D1-D26) were significantly more frequent in late mosquito season (P < 0.01). It is proposed that the gradual decrease of ambient temperature and the shortening of day length postpones the growth and development of P. iranica in the affected larvae. Both sexes of larvae were involved and none of them survived beyond a couple of days. The frequency of infections in adult males (5.8 %) were significantly more than females (4 %) (P = 0.02). The infections of larvae were more common in the late mosquito season, and the infections of adults were more frequent in the early mosquito season. This suggests the relative importance of vertical and horizontal routes of transmission in early and late mosquito seasons, respectively. However, age-specific data revealed that only 26 % of hidden infections of larvae (D1-D26) were stemmed from I to II age group. This implies that the vertical route may be less efficient than the horizontal route in the transmission of P. iranica to the mosquito host. These inferences should be verified with further field and laboratory investigations
