    Diversity of plant growth-promoting bacteria associated with sugarcane

    The sugarcane (Saccharum spp) presents economic importance, mainly for tropical regions, being an important Brazilian commodity. However, this crop is strongly dependent on fertilizers, mainly nitrogen (N). This study assessed the plant growth-promoting bacteria (PGPB) associated with sugarcane that could be used as a potential inoculant to the crop. We evaluated the genetic diversity of PGPB in the plant tissue of sugarcane varieties (RB 867515, RB 1011, and RB 92579). The primer BOX-A1R was used to differentiate the similar isolated and further sequencing 16S rRNA ribosomal gene. The 16S rRNA gene showed the presence of seven different genera distributed into four groups, the genus Bacillus, followed by Paenibacillus (20%), Burkholderia (14%), Herbaspirillum (6%), Pseudomonas (6%), Methylobacterium (6%), and Brevibacillus (3%). The molecular characterization of endophytic isolates from sugarcane revealed a diversity of bacteria colonizing this plant, with a possible biotechnological potential to be used as inoculant and biofertilizers

    Sexual dimorphism, female fertility, and diet of Physalaemus feioi (Anura: Leptodactylidae) from southeastern Brazil

    Physalaemus feioi Ă© uma rĂŁ da AmĂ©rica do Sul, endĂȘmica da Mata AtlĂąntica, registrada nos estados de Minas Gerais e SĂŁo Paulo. Avaliamos o dimorfismo sexual, a fertilidade nas fĂȘmeas e dieta dessa espĂ©cie em duas localidades do estado de Minas Gerais, incluindo a localidade-tipo da espĂ©cie. Coletamos 73 indivĂ­duos (16 fĂȘmeas adultas e 57 machos adultos) por meio de busca ativa. O nĂșmero de oĂłcitos encontrados para P. feioi (x̅ = 1385 ± 498,13) foi alto quando comparado com outras espĂ©cies do gĂȘnero como P. maculiventris, P. signifer, P. cuvieri e P. ephippifer mas este semelha o nĂșmero de oĂłcitos de P. centralis, P. kroyeri e P. marmoratus. As presas com maior Ă­ndice de importĂąncia foram Coleoptera (IIR = 24,26) e Formicidae (IIR = 16,46). NĂŁo houve diferenças entre machos e fĂȘmeas quanto ao nĂșmero de presas ingeridas e nem o volume do conteĂșdo estomacal. Em resumo, Physalaemus feioi teve semelhanças com outras espĂ©cies do gĂȘnero em relação ao dimorfismo sexual, fecundidade e hĂĄbitos alimentares.Physalaemus feioi is a South American frog endemic to the Brazilian Atlantic Forest that occurs in the states of Minas Gerais and SĂŁo Paulo. We evaluated sexual dimorphism, female fertility, and diet of P. feioi from two locations in the state of Minas Gerais, including the type locality of the species. We collected 73 individuals (16 adult females and 57 adult males). The number of oocytes of P. feioi (x̅ = 1385 ±498.13) was higher than in other species of the genus, such as P. maculiventris, P. signifer, P. cuvieri, and P. ephippifer, but it resembles that of P. centralis, P. kroyeri, and P. marmoratus. The prey items with the highest index of importance were Coleoptera (IIR = 24.26) and Formicidae (IIR = 16.46). We found no sex differences regarding the number of ingested prey and the volume of the stomach contents. In summary, Physalaemus feioi was similar to other species of the genus regarding sexual dimorphism, fecundity, and feeding habits

    Differential effects of food restriction and warming in the two-spotted goby: impaired reproductive performance and stressed offspring

    Climate change is a growing threat to marine organisms and ecosystems, and it is already modifying ocean properties by, for example, increasing temperature and decreasing pH. Increasing water temperature may also lead to an impairment of primary productivity and an overall depletion of available zooplankton. Understanding how the crossover between warming and zooplankton availability impacts fish populations has paramount implications for conservation and mitigation strategies. Through a cross factorial design to test the effects of ocean temperature and food availability in a temperate marine teleost, Pomatochistus flavescens, we showed that hindered feeding impacted sheltering and avoidance behaviour. Also, low food availability impaired fish reproduction, particularly male reproduction, as the expression of cyp11b1, a gene with a pivotal role in the synthesis of the most important fish androgen, 11-ketotestosterone, was significantly reduced under a low food regime. In contrast, temperature alone did not affect reproductive success, but offspring showed increased saturated fatty acid content (embryos) and increased lipid peroxidation (larvae). Altogether, food availability had a stronger effect on fitness, showing that coping with elevated temperatures, an ability that may be expected in shallow-water fish, can be indirectly impacted, or even overwhelmed, by the effects of ocean warming on primary productivity and downstream ecological processes.info:eu-repo/semantics/publishedVersio

    Effect of Sheep Grazing, Stocking Rates and Dolomitic Limestone Application on the Floristic Composition of a Permanent Dryland Pasture, in the Montado Agroforestry System of Southern Portugal

    The Montado is a complex agroforestry–pastoral ecosystem due to the interactions between soil–pasture–trees–animals and climate. The typical Montado soil has an acidic pH and manganese toxicity, which affect the pastureÊčs productivity and pasture floristic composition (PFC). The PFC, on the other hand, can also be influenced by the type and intensity of grazing, which can lead to significant decreases in the amount of biomass produced and the biodiversity of species in the pasture. The objective of this study was to evaluate the effect of grazing type, by sheep, and different stocking rates on the PFC throughout the vegetative pasture cycle in areas with and without dolomitic limestone application. Thus, four treatments (P1UC to P4TC) were constituted: P1UC—without limestone application (U) and continuous grazing (CG); P2UD—U and deferred grazing (DG); P3TD—with the application of limestone (T) and DG; P4TC—T and CG. In DG plots, the placement and removal of the animals were carried out as a function of the average height of the pasture (placement—10 cm; removal—3 to 5 cm). The PFC was characterized in winter, at the peak of spring and in late spring. The PFC data were subjected to a multilevel pattern analysis (ISA). The combination of rainfall and temperature influenced the pasture growth rates and consequently the height of the pasture at different times of the year. Therefore, with the different growth rates of the pasture throughout the year, the sheep remain for different periods of time in the deferred grazing treatments. In the four treatments, 103 plant species were identified. The most representative botanical families in the four treatments were Asteraceae, Fabaceae and Poaceae. ISA identified 14 bioindicator species: eight for the winter period, three for the late spring vegetative period and three for the TC treatment

    GABA uptake transporters support dopamine release in dorsal striatum with maladaptive downregulation in a parkinsonism model

    Striatal dopamine (DA) is critical for action and learning. Recent data show that DA release is under tonic inhibition by striatal GABA. Ambient striatal GABA tone on striatal projection neurons can be determined by plasma membrane GABA uptake transporters (GATs) located on astrocytes and neurons. However, whether striatal GATs and astrocytes determine DA output are unknown. We reveal that DA release in mouse dorsolateral striatum, but not nucleus accumbens core, is governed by GAT-1 and GAT-3. These GATs are partly localized to astrocytes, and are enriched in dorsolateral striatum compared to accumbens core. In a mouse model of early parkinsonism, GATs are downregulated, tonic GABAergic inhibition of DA release augmented, and nigrostriatal GABA co-release attenuated. These data define previously unappreciated and important roles for GATs and astrocytes in supporting DA release in striatum, and reveal a maladaptive plasticity in early parkinsonism that impairs DA output in vulnerable striatal regions

    Xylella fastidiosa gene expression analysis by DNA microarrays

    Xylella fastidiosa genome sequencing has generated valuable data by identifying genes acting either on metabolic pathways or in associated pathogenicity and virulence. Based on available information on these genes, new strategies for studying their expression patterns, such as microarray technology, were employed. A total of 2,600 primer pairs were synthesized and then used to generate fragments using the PCR technique. The arrays were hybridized against cDNAs labeled during reverse transcription reactions and which were obtained from bacteria grown under two different conditions (liquid XDM2 and liquid BCYE). All data were statistically analyzed to verify which genes were differentially expressed. In addition to exploring conditions for X. fastidiosa genome-wide transcriptome analysis, the present work observed the differential expression of several classes of genes (energy, protein, amino acid and nucleotide metabolism, transport, degradation of substances, toxins and hypothetical proteins, among others). The understanding of expressed genes in these two different media will be useful in comprehending the metabolic characteristics of X. fastidiosa, and in evaluating how important certain genes are for the functioning and survival of these bacteria in plants

    The role of natural science collections in the biomonitoring of environmental contaminants in apex predators in support of the EU's zero pollution ambition

    The chemical industry is the leading sector in the EU in terms of added value. However, contaminants pose a major threat and significant costs to the environment and human health. While EU legislation and international conventions aim to reduce this threat, regulators struggle to assess and manage chemical risks, given the vast number of substances involved and the lack of data on exposure and hazards. The European Green Deal sets a 'zero pollution ambition for a toxic free environment' by 2050 and the EU Chemicals Strategy calls for increased monitoring of chemicals in the environment. Monitoring of contaminants in biota can, inter alia: provide regulators with early warning of bioaccumulation problems with chemicals of emerging concern; trigger risk assessment of persistent, bioaccumulative and toxic substances; enable risk assessment of chemical mixtures in biota; enable risk assessment of mixtures; and enable assessment of the effectiveness of risk management measures and of chemicals regulations overall. A number of these purposes are to be addressed under the recently launched European Partnership for Risk Assessment of Chemicals (PARC). Apex predators are of particular value to biomonitoring. Securing sufficient data at European scale implies large-scale, long-term monitoring and a steady supply of large numbers of fresh apex predator tissue samples from across Europe. Natural science collections are very well-placed to supply these. Pan-European monitoring requires effective coordination among field organisations, collections and analytical laboratories for the flow of required specimens, processing and storage of specimens and tissue samples, contaminant analyses delivering pan-European data sets, and provision of specimen and population contextual data. Collections are well-placed to coordinate this. The COST Action European Raptor Biomonitoring Facility provides a well-developed model showing how this can work, integrating a European Raptor Biomonitoring Scheme, Specimen Bank and Sampling Programme. Simultaneously, the EU-funded LIFE APEX has demonstrated a range of regulatory applications using cutting-edge analytical techniques. PARC plans to make best use of such sampling and biomonitoring programmes. Collections are poised to play a critical role in supporting PARC objectives and thereby contribute to delivery of the EU's zero-pollution ambition.Non peer reviewe

    Soil water-holding capacity and monodominance in Southern Amazon tropical forests

    Background and aims: We explored the hypothesis that low soil water-holding capacity is the main factor driving the monodominance of Brosimum rubescens in a monodominant forest in Southern Amazonia. Tropical monodominant forests are rare ecosystems with low diversity and high dominance of a single tree species. The causes of this atypical condition are still poorly understood. Some studies have shown a relationship between monodominance and waterlogging or soil attributes, while others have concluded that edaphic factors have little or no explanatory value, but none has accounted for soil-moisture variation other than waterlogging. This study is the first to explicitly explore how low soil water-holding capacity influences the monodominance of tropical forests. Methods: We conducted in situ measurements of vertical soil moisture using electrical resistance collected over 1 year at 0–5; 35–40 and 75–80 cm depths in a B. rubescens monodominant forest and in an adjacent mixed-species forest in the Amazon-Cerrado transition zone, Brazil. Minimum leaf water potential (ιmin) of the seven most common species, including B. rubescens, and soil water-holding capacity for both forests were determined. Results: The vertical soil moisture decay pattern was similar in both forests for all depths. However, the slightly higher water availability in the monodominant forest and ιmin similarity between B. rubescens and nearby mixed forest species indicate that low water-availability does not cause the monodominance. Conclusions: We reject the hypothesis that monodominance of B. rubescens is primarily determined by low soil water-holding capacity, reinforcing the idea that monodominance in tropical forests is not determined by a single factor

    A relação entre a ĂĄrea foliar e o crescimento dos frutos Ă© um tema que freqĂŒentemente recebe a atenção dos pesquisadores, por influenciar diretamente na produtividade das plantas e na qualidade dos frutos. Neste trabalho, avaliou-se o efeito da ĂĄrea foliar sobre o crescimento dos frutos da laranjeira 'Salustiana'. Foram utilizados ramos com 12 meses de idade e com apenas um fruto terminal. Os ramos foram anelados visando a manter diversas relaçÔes de folhas/fruto. Avaliaram-se, semanalmente, o crescimento dos frutos e os teores de amido presentes nas folhas durante um perĂ­odo de 42 dias. O crescimento dos frutos, avaliado na "fase de crescimento II", dependeu da ĂĄrea foliar disponĂ­vel por fruto, sendo que 30 folhas foram suficientes para garantir o seu crescimento. As reservas de amido nas folhas dependeram da ĂĄrea foliar disponĂ­vel por fruto e reduziram Ă  medida que os frutos apresentaram aumentos no diĂąmetro e nas massas fresca e seca.The relationship between the foliar area and the fruit growth is an important theme because affects the tree productivity and fruits quality. In this work was evaluated the effect of the foliar area on the growth of the Salustiana's sweet orange fruits. Girdled shoots of 12 months were used with a single terminal fruit and several leaves-fruit ratios. It was evaluated, weekly, the fruit growth and the leaves starch contents during 42 days. The fruit growth, evaluated in the stage II, depended on the available leaf area per fruit, provided that 30 leaves were enough to guarantee its growth. The starch reserves in the leaves depended on the available leaf area per fruit and they reduced with the increase in the diameter and dry and fresh weight of the fruits

    The question whether taxonomic descriptions naming new animal species without type specimen(s) deposited in collections should be accepted for publication by scientific journals and allowed by the Code has already been discussed in Zootaxa (Dubois & NemĂ©sio 2007; Donegan 2008, 2009; NemĂ©sio 2009a–b; Dubois 2009; Gentile & Snell 2009; Minelli 2009; Cianferoni & Bartolozzi 2016; Amorim et al. 2016). This question was again raised in a letter supported by 35 signatories published in the journal Nature (Pape et al. 2016) on 15 September 2016. On 25 September 2016, the following rebuttal (strictly limited to 300 words as per the editorial rules of Nature) was submitted to Nature, which on 18 October 2016 refused to publish it. As we think this problem is a very important one for zoological taxonomy, this text is published here exactly as submitted to Nature, followed by the list of the 493 taxonomists and collection-based researchers who signed it in the short time span from 20 September to 6 October 2016
