    Thinning and drilling laser-assisted hatching in thawed embryo transfer : A randomized controlled trial

    Objective: In frozen and thawed embryos, the zona pellucida (ZP) can be damaged due to hardening. Laser-assisted hatching (LAH) of embryos can increase the pregnancy rate. This study compared thinning and drilling of the ZP before frozen embryo transfer (FET). Methods: Patients were randomly allocated into two groups for LAH using thinning or drilling on day 2 after thawing. Twenty-five percent of the ZP circumference and 50% of the ZP thickness was removed in the thinning group, and a hole 40 μm in diameter was made in the drilling group. Results: A total of 171 in vitro fertilization/intracytoplasmic sperm injection FET cycles, including 85 cycles with drilling LAH and 86 cycles with thinning LAH, were carried out. The thinning group had a similar β-human chorionic gonadotropin-positive rate (38.4% vs. 29.4%), implantation rate (16.5% vs. 14.4%), clinical pregnancy rate (36.0% vs. 25.9%), miscarriage rate (5.8% vs. 2.4%), ongoing pregnancy rate (30.2% vs. 23.5%), and multiple pregnancy rate (7.0% vs. 10.6%) to the drilling LAH group. There were no significant differences in pregnancy outcomes between subgroups defined based on age (older or younger than 35 years) or ZP thickness (greater or less than 17 μm) according to the LAH method. Conclusion: The present study demonstrated that partial ZP thinning or drilling resulted in similar outcomes in implantation and pregnancy rates using thawed embryos, irrespective of women's age or ZP thickness. © 2018.     Efficient and precise CRISPR/Cas9-mediated MECP2 modifications in human-induced pluripotent stem cells

    Patients with Rett syndrome (RTT) have severe mental and physical disabilities. The majority of RTT patients carry a heterozygous mutation in methyl-CpG binding protein 2 (MECP2), an X-linked gene encoding an epigenetic factor crucial for normal nerve cell function. No curative therapy for RTT syndrome exists, and cellular mechanisms are incompletely understood. Here, we developed a CRISPR/Cas9-mediated system that targets and corrects the disease relevant regions of the MECP2 exon 4 coding sequence. We achieved homologous recombination (HR) efficiencies of 20% to 30% in human cell lines and iPSCs. Furthermore, we successfully introduced a MECP2(R270X) mutation into the MECP2 gene in human induced pluripotent stem cells (iPSCs). Consequently, using CRISPR/Cas9, we were able to repair such mutations with high efficiency in human mutant iPSCs. In summary, we provide a new strategy for MECP2 gene targeting that can be potentially translated into gene therapy or for iPSCs-based disease modeling of RTT syndrome

    Granularity-induced gapless superconductivity in NbN films: evidence of thermal phase fluctuations

    Using a single coil mutual inductance technique, we measure the low temperature dependence of the magnetic penetration depth in superconducting NbN films prepared with similar critical temperatures around 16 K but with different microstructures. Only (100) epitaxial and weakly granular (100) textured films display the characteristic exponential dependence of conventional BCS s-wave superconductors. More granular (111) textured films exhibit a linear dependence, indicating a gapless state in spite of the s-wave gap. This result is quantitatively explained by a model of thermal phase fluctuations favored by the granular structure.Comment: 10 pages, 4 figures, to appear in Phys. Rev.

    Weak localization and interaction effects in acceptor graphite intercalation compounds

    The presented work is devoted to investigations of manifestation of quantum effects of weak localization and interaction of charge carriers in electrical conductivity of acceptor graphite intercalation compounds (CICs). As shown by studies intercalation leads to a decrease in the resistivity and to change the resistivity temperature coefficient from negative sign in the source graphite on a positive sign in intercalated graphite. At the low temperature for all GICs specimens the minimum in the temperature dependence of resistivity is observed. In terms of the model of charge carrier's weak localization and interaction for two-dimensional systems temperature dependence of phase relaxation time, localization radius and charge carriers screening constant for all GICs are estimated

    Nonlinear weakly curved rod by Γ-Convergence

    We present a nonlinear model of weakly curved rod, namely the type of curved rod where the curvature is of the order of the diameter of the cross-section. We use an approach analogous to the one for rods and curved rods and start from the strain energy functional of three dimensional nonlinear elasticity. We do not impose any constitutional behavior of the material and work in a general framework. To derive the model, by means of Γ-convergence, we need to set the order of strain energy (i.e., its relation to the thickness of the body h). We analyze the situation when the strain energy (divided by the order of volume) is of the order h 4. This is the same approach as the one used in Föppl-von Kármán model for plates and the analogous model for rods. The obtained model is analogous to Marguerre-von Kármán for shallow shells and its linearization is the linear shallow arch model which can be found in the literature

    Strange particle production in proton-proton collisions at s=0.9\sqrt{s}=0.9 TeV with ALICE at the LHC

    The production of mesons containing strange quarks (Ks0^0_s, ϕ\phi) and both singly and doubly strange baryons (Λ\Lambda, Anti-Λ\Lambda, and Ξ\Xi+Anti-Ξ\Xi) are measured at central rapidity in pp collisions at s\sqrt{s} = 0.9 TeV with the ALICE experiment at the LHC. The results are obtained from the analysis of about 250 k minimum bias events recorded in 2009. Measurements of yields (dN/dy) and transverse momentum spectra at central rapidities for inelastic pp collisions are presented. For mesons, we report yields () of 0.184 ±\pm 0.002 stat. ±\pm 0.006 syst. for Ks0^0_s and 0.021 ±\pm 0.004 stat. ±\pm 0.003 syst. for ϕ\phi. For baryons, we find = 0.048 ±\pm 0.001 stat. ±\pm 0.004 syst. for Λ\Lambda, 0.047 ±\pm 0.002 stat. ±\pm 0.005 syst. for Anti-Λ\Lambda and 0.0101 ±\pm 0.0020 stat. ±\pm 0.0009 syst. for Ξ\Xi+Anti-Ξ\Xi.     Particle-yield modification in jet-like azimuthal di-hadron correlations in Pb-Pb collisions at sNN\sqrt{s_{\rm NN}} = 2.76 TeV

    The yield of charged particles associated with high-pTp_{\rm T} trigger particles (8<pT<158 < p_{\rm T} < 15 GeV/cc) is measured with the ALICE detector in Pb-Pb collisions at sNN\sqrt{s_{\rm NN}} = 2.76 TeV relative to proton-proton collisions at the same energy. The conditional per-trigger yields are extracted from the narrow jet-like correlation peaks in azimuthal di-hadron correlations. In the 5% most central collisions, we observe that the yield of associated charged particles with transverse momenta pT>3p_{\rm T}> 3 GeV/cc on the away-side drops to about 60% of that observed in pp collisions, while on the near-side a moderate enhancement of 20-30% is found.Comment: 15 pages, 2 captioned figures, 1 table, authors from page 10, published version, figures at http://aliceinfo.cern.ch/ArtSubmission/node/350
