    In-Situ Star Formation in the Outskirts of the Large Magellanic Cloud: Gaia DR2 Confirmation

    We explore the Gaia DR2 proper motions of six young, main-sequence stars, members of the Large Magellanic Cloud (LMC) reported by Moni Bidin et al. (2017). These stars are located in the outskirts of the disk, between 7 and 13 degrees from the LMC's center where there is very low H I content. Gaia DR2 proper motions confirm that four stars formed locally, in situ, while two are consistent with being expelled via dynamical interactions from inner, more gas-rich regions of the LMC. This finding establishes that recent star formation occurred in the periphery of the LMC, where thus far only old populations are known.Comment: Accepted for publication in Astrophysical Journal Letter

    The Origins of Young Stars in the Direction of the Leading Arm of the Magellanic Stream: Abundances, Kinematics, and Orbits

    We explore the origins of the young B-type stars found by Casetti-Dinescu et al.(2014) at the outskirts of the Milky-Way disk in the sky region of Leading Arm of the Magellanic Stream. High-resolution spectroscopic observations made with the MIKE instrument on the Magellan Clay 6.5m telescope for nine stars are added to the previous sample analyzed by Zhang et al. (2017). We compile a sample of fifteen young stars with well-determined stellar types, ages, abundances and kinematics. With proper motions from Gaia DR2 we also derive orbits in a realistic Milky-Way potential. We find that our previous radial-velocity selected LA candidates have substantial orbital angular momentum. The substantial amount of rotational component for these stars is in contrast with the near-polar Magellanic orbit, thus rendering these stars unlikely members of the LA. There are four large orbital-energy stars in our sample. The highest orbital-energy one has an age shorter than the time to disk crossing, with a birthplace z=2.5z=2.5~kpc and RGC28R_{\rm GC}\sim 28~kpc. Therefore, the origin of this star is uncertain. The remaining three stars have disk runaway origin with birthplaces between 12 and 25 kpc from the Galactic center. Also, the most energetic stars are more metal poor ([Mg/H] =0.50±0.07-0.50\pm0.07) and with larger He scatter (σ[He/H]=0.72\sigma_{\rm [He/H]} = 0.72) than the inner disk ones ([Mg/H] =0.12±0.36=0.12\pm0.36, σ[He/H]=0.15\sigma_{\rm [He/H]} = 0.15). While the former group's abundance is compatible with that of the Large Magellanic Cloud, it could also reflect the metallicity gradient of the MW disk and their runaway status via different runaway mechanisms.Comment: 58 pages, 17 figures, 7 tables.     Targeting the PAI-1 Mechanism with a Small Peptide Increases the Efficacy of Alteplase in a Rabbit Model of Chronic Empyema

    The incidence of empyema is increasing and associated with a mortality rate of 20% in patients older than 65 years. Since 30% of patients with advanced empyema have contraindications to surgical treatment, novel, low-dose, pharmacological treatments are needed. A Streptococcus pneumoniae-induced rabbit model of chronic empyema recapitulates the progression, loculation, fibrotic repair, and pleural thickening of human disease. Treatment with single chain (sc) urokinase (scuPA) or tissue type (sctPA) plasminogen activators in doses 1.0–4.0 mg/kg were only partially effective in this model. Docking Site Peptide (DSP; 8.0 mg/kg), which decreased the dose of sctPA for successful fibrinolytic therapy in acute empyema model did not improve efficacy in combination with 2.0 mg/kg scuPA or sctPA. However, a two-fold increase in either sctPA or DSP (4.0 and 8.0 mg/kg or 2.0 and 16.0 mg/kg sctPA and DSP, respectively) resulted in 100% effective outcome. Thus, DSP-based Plasminogen Activator Inhibitor 1-Targeted Fibrinolytic Therapy (PAI-1-TFT) of chronic infectious pleural injury in rabbits increases the efficacy of alteplase rendering ineffective doses of sctPA effective. PAI-1-TFT represents a novel, well-tolerated treatment of empyema that is amenable to clinical introduction. The chronic empyema model recapitulates increased resistance of advanced human empyema to fibrinolytic therapy, thus allowing for studies of muti-injection treatments

    Plasma CC16 levels are associated with development of ALI/ARDS in patients with ventilator-associated pneumonia: a retrospective observational study

    <p>Abstract</p> <p>Background</p> <p>Despite consensus criteria, diagnosing acute lung injury, or its more severe form acute respiratory distress syndrome (ALI/ARDS) remains challenging. Adding objective measures, such as plasma levels of biological markers could facilitate recognition of ALI/ARDS. This study was designed to assess and compare the diagnostic accuracy of biological markers for ALI/ARDS with ventilator-associated pneumonia (VAP).</p> <p>Methods</p> <p>We performed serial measurements of Clara cell protein (CC16), soluble receptor for advanced glycation end products (sRAGE), surfactant protein D (SP-D) and Krebs von den Lungen (KL-6) in plasma of patients with VAP and mechanically ventilated control patients without VAP. ALI/ARDS was diagnosed using the criteria of the North-American European consensus conference.</p> <p>Results</p> <p>Thirty-seven patients were enrolled - 22 patients with VAP and 15 control patients. Ten patients with pneumonia met the ALI/ARDS consensus criteria. Control patients never met these criteria. Plasma CC16 had a good diagnostic capacity for ALI/ARDS as shown by the receiver operating characteristic curve with an area under the curve of 0.91 (95% confidence interval (CI) 0.79 - 1.00; <it>p </it>< 0.001). Identification of ALI/ARDS patients by sudden increases in plasma CC16 of 30% or more yielded a sensitivity of 90% and a specificity of 92%. Of note, levels of CC16 increased 2 days before ALI/ARDS diagnosis. A cut-off level of 50 ng/ml SP-D yielded a specificity of 100% while the sensitivity was 70%. The area under the curve for SP-D was 0.80 (95% CI 0.58 - 1.00; <it>p </it>= 0.02). The diagnostic accuracies of KL-6 and sRAGE were low.</p> <p>Conclusion</p> <p>Plasma CC16 seems a potential biological marker for ALI/ARDS in patients with VAP. Plasma levels of sRAGE, SP-D and KL-6 have limited discriminative power for diagnosing ALI/ARDS in VAP.</p

    Preparation of selective and segmentally labeled single-stranded DNA for NMR by self-primed PCR and asymmetrical endonuclease double digestion

    We demonstrate a new, efficient and easy-to-use method for enzymatic synthesis of (stereo-)specific and segmental 13C/15N/2H isotope-labeled single-stranded DNA in amounts sufficient for NMR, based on the highly efficient self-primed PCR. To achieve this, new approaches are introduced and combined. (i) Asymmetric endonuclease double digestion of tandem-repeated PCR product. (ii) T4 DNA ligase mediated ligation of two ssDNA segments. (iii) In vitro dNTP synthesis, consisting of in vitro rNTP synthesis followed by enzymatic stereo-selective reduction of the C2′ of the rNTP, and a one-pot add-up synthesis of dTTP from dUTP. The method is demonstrated on two ssDNAs: (i) a 36-nt three-way junction, selectively 13C9/15N3/2H(1′,2″,3′,4′,5′,5″)-dC labeled and (ii) a 39-nt triple-repeat three-way junction, selectively 13C9/15N3/2H(1′,2″,3′,4′,5′,5″)-dC and 13C9/15N2/2H(1′,2″,3′,4′,5′,5″)-dT labeled in segment C20-C39. Their NMR spectra show the spectral simplification, while the stereo-selective 2H-labeling in the deoxyribose of the dC-residues, straightforwardly provided assignment of their C1′–H2′ and C2′–H2′ resonances. The labeling protocols can be extended to larger ssDNA molecules and to more than two segments

    The James Webb Space Telescope Mission

    Twenty-six years ago a small committee report, building on earlier studies, expounded a compelling and poetic vision for the future of astronomy, calling for an infrared-optimized space telescope with an aperture of at least 4m4m. With the support of their governments in the US, Europe, and Canada, 20,000 people realized that vision as the 6.5m6.5m James Webb Space Telescope. A generation of astronomers will celebrate their accomplishments for the life of the mission, potentially as long as 20 years, and beyond. This report and the scientific discoveries that follow are extended thank-you notes to the 20,000 team members. The telescope is working perfectly, with much better image quality than expected. In this and accompanying papers, we give a brief history, describe the observatory, outline its objectives and current observing program, and discuss the inventions and people who made it possible.
