    Selective phosphodiesterase-5 inhibition reduces neointimal hyperplasia in rat carotid arteries after surgical endarterectomy.

    OBJECTIVE: Long-term results of surgical vessel reconstruction are compromised by restenosis caused by neointimal hyperplasia. Recent studies suggest that reduced cyclic guanosine monophosphate signaling is associated with neointima formation. In a rat model of endarterectomy, we investigated the effect of pharmacologic inhibition of cyclic guanosine monophosphate degradation on neointima formation by using the selective phosphodiesterase-5 inhibitor vardenafil. METHODS: Carotid endarterectomy was performed in male Sprague-Dawley rats by means of incision of the right common carotid artery with removal of intima. Four groups were studied: unoperated control rats (n = 4), sham-operated rats (n = 9), control rats with endarterectomy (n = 9), or endarterectomized rats treated with vardenafil (10 mg/kg/day) postoperatively (n = 9). After 3 weeks, vessel compartment areas were measured by means of conventional microscopy with hematoxylin and eosin staining. Immunohistochemical analysis was performed to confirm neointima formation and the local cyclic guanosine monophosphate content. Plasma levels of cyclic guanosine monophosphate were determined by means of enzyme immunoassay. Student's t test was used for statistical evaluation. RESULTS: Immunohistochemical analysis demonstrated intensive staining for transforming growth factor beta1 and alpha-smooth muscle actin in the control neointima. Vardenafil significantly reduced the stenosis grade (24.64% +/- 7.46% vs 54.12% +/- 10.30% in the control group, P < .05) and expression of transforming growth factor beta1, as well as alpha-smooth muscle actin, in the neointima. The immunohistochemical score for cyclic guanosine monophosphate was higher in the treated neointima (4.80 +/- 0.76 vs 2.84 +/- 0.40 in the control group, P < .05), and increased plasma cyclic guanosine monophosphate levels were found by means of enzyme immunoassay as well (84.65 +/- 12.77 pmol/mL vs 43.50 +/- 3.30 pmol/mL in the control group, P < .05). CONCLUSIONS: Treatment with vardenafil can be considered a new possibility to prevent neointimal hyperplasia after endarterectomy

    Education for Disaster Prevention in England : Analysis of Secondary Geography Textbooks

    The purpose of this study is to clarify the aims, contents and activities of education for disaster prevention on geographical education in England, by analysis of secondary textbooks. Thereby, we want to contribute the improvement of Japanese geographical education. As a result, we clarified the following: 1) when students study a natural disaster in UK, they learn a plurality of cases including domestic and foreign regions, 2) all textbooks contain activities that students can form attitudes and decide what to do for disaster prevention. On the basis of these results, we suggest improvements of education for disaster prevention. The first is to develop a clear understanding of a natural disaster with multi-area including Japan. The second is to place contents and activities for forming a value relevant to awareness of disaster prevention into Japanese geography textbook. The third is to set an activity to learn practical strategies for disaster prevention in the real world

    Characteristics of the stone monuments in Saijo, Higashi-Hiroshima City, southwest Japan, and their significance for teaching materials of social studies.

    本稿の目的は,広島県東広島市旧西条町に分布する石碑の特徴を明らかにすると共に,小学校や中学校社会科の地域学習における石碑の教材的意義を検討することである。調査の結果,202基の石碑が確認され,寺社や公共施設,溜池などに石碑の立地が多くみられた。石碑の建立年との関連を検討すると,第二次世界大戦の終戦を境に,石碑の主な建立目的が地域社会に貢献した人物や行為の顕彰から,寺社への寄附に関連したものへと変化していったことが明らかになった。社会科教材としての意義については,1)学習指導要領への適合と2)石碑の情報量という観点から検討を行い,石碑のもつ教材的意義を3つに分類した。各分類において,具体的な活用方法を考察し想定される学習の展開を提示した。本稿は,「地域学習」の不振の要因のひとつである,地域に即した資料の不足の改善の一助になると期待される。An objective of this study is to clarify the characteristics and distribution of the stone monuments in Saijo, Higashi-Hiroshima City, Hiroshima Prefecture, and discuss their significance for elementary and junior high school social studies teaching materials. Two hundred two monuments were recognized in this area. They were mostly located in temples, shrines, public institutions, roadsides, and irrigation ponds. The purpose of erecting monuments has changed over 150 years from the Meiji era to the present day, which reflects the transition of the history of the local community or local people’s consciousness about their community. All monuments were classified into three levels considering their significance for elementary and junior high school social studies teaching materials: i.e., A. Useful and sufficient information; B. Useful but insufficient information; and C. Others. As a result, we defined 6 monuments as level A, 71 as level B, and 125 as level C. We propose an example of a social studies teaching plan including information about the monuments according to each level. The monuments directly provided information about the history of the local community and the location of historical events; therefore, the teaching plan using information about the monuments has possibility to improve for social studies

    Zebrafish ProVEGF-C Expression, Proteolytic Processing and Inhibitory Effect of Unprocessed ProVEGF-C during Fin Regeneration

    BACKGROUND: In zebrafish, vascular endothelial growth factor-C precursor (proVEGF-C) processing occurs within the dibasic motif HSIIRR(214) suggesting the involvement of one or more basic amino acid-specific proprotein convertases (PCs) in this process. In the present study, we examined zebrafish proVEGF-C expression and processing and the effect of unprocessed proVEGF-C on caudal fin regeneration. METHODOLOGY/PRINCIPAL FINDINGS: Cell transfection assays revealed that the cleavage of proVEGF-C, mainly mediated by the proprotein convertases Furin and PC5 and to a less degree by PACE4 and PC7, is abolished by PCs inhibitors or by mutation of its cleavage site (HSIIRR(214) into HSIISS(214)). In vitro, unprocessed proVEGF-C failed to activate its signaling proteins Akt and ERK and to induce cell proliferation. In vivo, following caudal fin amputation, the induction of VEGF-C, Furin and PC5 expression occurs as early as 2 days post-amputation (dpa) with a maximum levels at 4-7 dpa. Using immunofluorescence staining we localized high expression of VEGF-C and the convertases Furin and PC5 surrounding the apical growth zone of the regenerating fin. While expression of wild-type proVEGF-C in this area had no effect, unprocessed proVEGF-C inhibited fin regeneration. CONCLUSIONS/SIGNIFICANCES: Taken together, these data indicate that zebrafish fin regeneration is associated with up-regulation of VEGF-C and the convertases Furin and PC5 and highlight the inhibitory effect of unprocessed proVEGF-C on fin regeneration

    The Global Burden of Diseases, Injuries and Risk Factors 2017 includes a comprehensive assessment of incidence, prevalence, and years lived with disability (YLDs) for 354 causes in 195 countries and territories from 1990 to 2017. Previous GBD studies have shown how the decline of mortality rates from 1990 to 2016 has led to an increase in life expectancy, an ageing global population, and an expansion of the non-fatal burden of disease and injury. These studies have also shown how a substantial portion of the world's population experiences non-fatal health loss with considerable heterogeneity among different causes, locations, ages, and sexes. Ongoing objectives of the GBD study include increasing the level of estimation detail, improving analytical strategies, and increasing the amount of high-quality data. METHODS: We estimated incidence and prevalence for 354 diseases and injuries and 3484 sequelae. We used an updated and extensive body of literature studies, survey data, surveillance data, inpatient admission records, outpatient visit records, and health insurance claims, and additionally used results from cause of death models to inform estimates using a total of 68 781 data sources. Newly available clinical data from India, Iran, Japan, Jordan, Nepal, China, Brazil, Norway, and Italy were incorporated, as well as updated claims data from the USA and new claims data from Taiwan (province of China) and Singapore. We used DisMod-MR 2.1, a Bayesian meta-regression tool, as the main method of estimation, ensuring consistency between rates of incidence, prevalence, remission, and cause of death for each condition. YLDs were estimated as the product of a prevalence estimate and a disability weight for health states of each mutually exclusive sequela, adjusted for comorbidity. We updated the Socio-demographic Index (SDI), a summary development indicator of income per capita, years of schooling, and total fertility rate. Additionally, we calculated differences between male and female YLDs to identify divergent trends across sexes. Globally, for females, the causes with the greatest age-standardised prevalence were oral disorders, headache disorders, and haemoglobinopathies and haemolytic anaemias in both 1990 and 2017. For males, the causes with the greatest age-standardised prevalence were oral disorders, headache disorders, and tuberculosis including latent tuberculosis infection in both 1990 and 2017. In terms of YLDs, low back pain, headache disorders, and dietary iron deficiency were the leading Level 3 causes of YLD counts in 1990, whereas low back pain, headache disorders, and depressive disorders were the leading causes in 2017 for both sexes combined. All-cause age-standardised YLD rates decreased by 3·9% (95% uncertainty interval [UI] 3·1-4·6) from 1990 to 2017; however, the all-age YLD rate increased by 7·2% (6·0-8·4) while the total sum of global YLDs increased from 562 million (421-723) to 853 million (642-1100). The increases for males and females were similar, with increases in all-age YLD rates of 7·9% (6·6-9·2) for males and 6·5% (5·4-7·7) for females. We found significant differences between males and females in terms of age-standardised prevalence estimates for multiple causes. The causes with the greatest relative differences between sexes in 2017 included substance use disorders (3018 cases [95% UI 2782-3252] per 100 000 in males vs s1400 [1279-1524] per 100 000 in females), transport injuries (3322 [3082-3583] vs 2336 [2154-2535]), and self-harm and interpersonal violence (3265 [2943-3630] vs 5643 [5057-6302]). Global all-cause age-standardised YLD rates have improved only slightly over a period spanning nearly three decades. However, the magnitude of the non-fatal disease burden has expanded globally, with increasing numbers of people who have a wide spectrum of conditions. A subset of conditions has remained globally pervasive since 1990, whereas other conditions have displayed more dynamic trends, with different ages, sexes, and geographies across the globe experiencing varying burdens and trends of health loss. This study emphasises how global improvements in premature mortality for select conditions have led to older populations with complex and potentially expensive diseases, yet also highlights global achievements in certain domains of disease and injury

    Control de Tecia solanivora (Lepidoptera: Gelechiidae) mediante su feromona para la interrupción del apareamiento

    El objetivo del trabajo fue evaluar la técnica de control de Tecia solanivora (Lepidoptera: Gelechiidae) por medio de la interrupción del apareamiento para su posible factibilidad en el cultivo de la papa. En un túnel de viento una mezcla antagónica de la feromona constituida por E3-12Ac, Z3-12Ac y 12Ac en la proporción 100:50:100, respectivamente (70 mg por dispensador) produjo una inhibición total en la atracción de machos hacia la feromona liberada por las hembras, en comparación con hembras liberando la feromona (69% de atracción). En campo esta mezcla a 10 hg/trampa inhibió significativamente la captura con dos machos/trampa, a diferencia de trampas control conteniendo E3-12Ac y Z3-12Ac (100:1 respectivamente) capturándose 50 machos/trampa. En 3 ha tratadas con la mezcla antagónica en dispensadores colocados en la base de plantas de papa a una densidad de 1.227 dispensadores/ha, (equivalente a 86 g de feromona/ha), se redujo significativamente la captura de machos en trampas para monitoreo en 94%, en 89% en trampas con hembras y la cópula de parejas confinadas se redujo en 67%, en comparación con un área testigo (1 ha sin tratar). Estos resultados indican el potencial de esta técnica para reducir poblaciones de la plaga

    Influence of chronic volume overload-induced atrial remodeling on electrophysiological responses to cholinergic receptor stimulation in the isolated rat atria

    Whereas molecular mechanisms of atrial fibrillation (AF) have been widely investigated, there is limited information regarding interrelation between chronic volume overload and parasympathetic nervous system in the pathophysiology of AF. In this study, we investigated the influence of abdominal aorto-venocaval shunt (AVS)-induced atrial remodeling on electrophysiological responses to cholinergic receptor stimulation in the isolated rat atria. Interstitial fibrosis, cardiomyocyte hypertrophy and atrial enlargement, known as structural arrhythmogenic substrates for AF, took place after one month of AVS operation. Carbachol at 0.1 and 1 μM shortened the effective refractory period, acting as functional arrhythmogenic substrates, but increased the conduction velocity both in the atria of the sham-operated and AVS rats. The extents of the electrophysiological responses to carbachol in the atria of the AVS rat were greater than those in the sham-operated ones. Also, the higher inducibility and longer duration of carbachol-mediated AF were detected in the AVS atria than those in the sham-operated ones. These results showed that chronic volume overload-induced atrial remodeling promoted electrophysiological responses to cholinergic receptor stimulation in the isolated atria of rats, suggesting possible synergistic actions between structural arrhythmogenic substrate in the remodeled atria and functional arrhythmogenic substrates modulated by parasympathetic nerve activity.
