    Characterization of phytoplankton by pigment analysis and the detection of toxic cyanobacteria in reservoirs with aquaculture production

No. of bitstreams: 0 Previous issue date: 2018-01-01The intensified use of water bodies and reservoirs for aquaculture production has increased the need for monitoring and early warning of toxins from cyanobacteria. To minimize effects from toxins, simple and fast analytical monitoring methods are crucial. Here, the content of pigments and microcystins in 14 different strains of cyanobacteria cultured under different growth conditions was investigated to determine the influence of light and nutrient starvation on pigment/ chlorophyll a (chl a) ratios. The obtained pigment/chl a ratios were applied in the software CHEMTAX to calculate the biomass of toxic cyanobacteria, as well as other phytoplankton groups. CHEMTAX ratios from the laboratory cultures were applied to water samples collected during 4 sampling periods at 6 fish farms in different reservoirs in São Paulo State, Brazil. Cyanobacteria generally dominated in all reservoirs in all sampling periods and constituted on average 44 to 66% of the average phytoplankton biomass. The concentrations of microcystins were significantly correlated with the chl a concentrations of cyanobacteria and showed that the pigment method can be used to detect microcystin-producing cyanobacteria in these Brazilian reservoirs. When the concentration of cyanobacteria in the reservoirs was above 4 μg chl a l-1, microcystins were always detected.     Osteosynthesis metal plate system for bone fixation using bicortical screws: numerical–experimental characterization

    This study reports the numerical and experimental characterization of a standard immobilization system currently being used to treat simple oblique bone fractures of femoral diaphyses. The procedure focuses on the assessment of the mechanical behavior of a bone stabilized with a dynamic compression plate (DCP) in a neutralization function, associated to a lag screw, fastened with surgical screws. The non-linear behavior of cortical bone tissue was revealed through four-point bending tests, from which damage initiation and propagation occurred. Since screw loosening was visible during the loading process, damage parameters were measured experimentally in independent pull-out tests. A realistic numerical model of the DCP-femur setup was constructed, combining the evaluated damage parameters and contact pairs. A mixed-mode (I+II) trapezoidal damage law was employed to mimic the mechanical behavior of both the screw–bone interface and bone fractures.     The knowledge of anatomical and phytochemical structures of many species has drawn the attention of researchers in several areas, because these species are characterized by the production of chemical compounds, mainly fixed and essential oils, which are of great industrial interest. The “pequiá” tree is a majestic tree from primary forest and represents huge economic potential. This work aimed to study the anatomical aspects, extraction and characterization of the fixed oil present in the fruit and the seed of Caryocar villosum. There were used fifty fruits of pequiá collected from the municipality of Tartarugalzinho (Amapá). The identification of species was made by comparison with exsiccates available in the Amapá Herbarium – HAMAB. For anatomical and phytochemical analyses, conventional methodology was used. The result in the macerate of the mesocarp corresponds to a yellow mass impregnated by lipids; in the endocarp there were registered trichomes which secret these lipids, forming an arc in all its extension. In the solvent-based phytochemical analyses of the mass of pericarp, mesocarp and fixed oil seed, favorable and satisfactory oil yields were obtained. The analyses of the acidity, saponification, ester and peroxide indexes are parameters that are related to the quality of the oil, therefore, the values obtained meet the Anvisa/2015 Resolution. It is thus concluded that the fixed oil obtained from the mesocarp/seed of C. villosum is indicated for alimentary purposes, with potential to reduce total cholesterol and LDL cholesterol, as well as in cosmetic industry.     Cationic dyes immobilized on cellulose acetate surface modified with titanium dioxide: factorial design and an application as sensor for NADH

    The electrochemical properties of meldola blue and toluidine blue cationic dyes immobilized on cellulose acetate surface modified with titanium dioxide were investigated by cyclic voltammetry. The materials synthesized were employed as carbon paste electrodes. The redox mediator properties of the meldola blue and toluidine blue chemically modified electrodes were optimized using a factorial design, consisting of two levels and four factors with two pseudo-central points (n= 20 experiments). The factorial analysis was carried out by searching for better reversibility of the redox process, such as the lowest separation between anodic and cathodic potential peaks and a current ratio near unity. The factors that presented significant effects on the overall optimization of the system to achieve the best conditions of the reversibility of electron transfer were the main factors scan rate and type of electrode (meldola blue or toluidine blue), besides the interaction factors KCl concentration × type of electrode (B×D) and the pH × [KCl] concentration × scan rate (A×B×C) interaction. The best electrochemical reversibility conditions obtained were: using the CA-TiO2-MB electrode, 1.0 mol L-1 KCl as supporting electrolyte, at scan rate of 10.0 mV s-1. Afterwards, the CA-TiO2-MB modified electrode was tested as an amperometric sensor for the determination of NADH, with a detection limit of 0.1 µmol L-1.As propriedades eletroquímicas dos corantes catiônicos, azul de meldola e azul de toluidina, imobilizados na superfície do acetato de celulose modificado com dióxido de titânio foram investigadas por voltametria cíclica. Os materiais sintetizados foram empregados como eletrodos de pasta de carbono. As propriedades mediadoras redox dos eletrodos modificados quimicamente com azul de meldola e azul de toluidina foram otimizadas utilizando um planejamento fatorial consistindo de dois níveis e quatro fatores com dois pseudo-pontos centrais (n=20 experimentos). A análise fatorial foi realizada para investigar as melhores condições de reversibilidade do processo redox tais como uma diminuição na separação entre os picos de potenciais anódico e catódico e razão de corrente próxima da unidade. Os fatores que apresentaram efeitos significantes na otimização global do sistema e que permitiram alcançar as melhores condições de reversibilidade de transferência de elétrons foram os fatores principais velocidade de varredura e tipo de eletrodo (azul de meldola ou azul de toluidina) além dos fatores de interação concentração do KCl × tipo de eletrodo (B×D) e a interação pH × [KCl] × velocidade de varredura (A×B×C). As melhores condições de reversibilidade eletroquímica obtidas foram: o eletrodo CA-TiO2-MB, KCl 1,0 mol L-1 como eletrólito suporte e velocidade de varredura de 10,0 mV s-1.     ESTABLISHING THE REFERENCE RANGE FOR T LYMPHOCYTES SUBPOPULATIONS IN ADULTS AND CHILDREN FROM BRAZIL

    SUMMARY In Brazil, the existing reference values for T-lymphocytes subsets are based on data originated in other countries. There is no local information on normal variation for these parameters in Brazilian adults and children. We evaluated the normal variation found in blood donors from five large Brazilian cities, in different regions, and in children living in Salvador, and Rio de Janeiro. All samples were processed by flow cytometry. The results were analyzed according to region, gender, and lifestyle of blood donors. A total of 641 adults (63% males), and 280 children (58% males) were involved in the study. The absolute CD3+, and CD4+ cells count were significantly higher for females (adults and children). Higher CD4+ cell count in adults was associated with smoking, while higher CD8+ count was found among female children. Higher counts, for all T-cells subsets, were detected in blood donors from southeast / south regions while those living in the northern region had the lowest values. Individuals from midwestern and northeastern regions had an intermediate count for all these cells subsets. However, these differences did not reach statistical significance. In Brazil, gender and smoking, were the main determinants of differences in T-lymphocytes reference values.RESUMO Os valores de referências de linfócitos T existentes no Brasil são baseados em dados originados de outros países. Não existem dados locais da variação normal para estes parâmetros em adultos e crianças brasileiras. Avaliamos a variação normal encontrada em doadores de sangue de cinco grandes cidades brasileiras em diferentes regiões e em crianças residentes em Salvador e Rio de Janeiro. Todas as amostras foram processadas por citometria de fluxo. Os resultados foram analisados de acordo com região, gênero e estilo de vida dos doadores. Um total de 641 adultos (63% homens) e 280 crianças (58% meninos) participaram do estudo. Valores absolutos de CD3+ e CD4+ foram significantemente maiores no gênero feminino (adultos e crianças). Maiores valores de CD4+ em adultos foram associados com tabagismo, enquanto que maiores valores de CD8+ foram encontrados entre crianças do sexo feminino. Adultos das regiões sul e sudeste apresentaram maiores valores absolutos para todas as células T enquanto que adultos da região norte, apresentaram menores valores. Indivíduos residentes no nordeste e centro-oeste obtiveram contagens intermediárias para todas as populações de células T. Entretanto, estas diferenças entre as regiões, não demonstraram diferença estatística.     Search for the standard model Higgs boson in the H to ZZ to 2l 2nu channel in pp collisions at sqrt(s) = 7 TeV

    A search for the standard model Higgs boson in the H to ZZ to 2l 2nu decay channel, where l = e or mu, in pp collisions at a center-of-mass energy of 7 TeV is presented. The data were collected at the LHC, with the CMS detector, and correspond to an integrated luminosity of 4.6 inverse femtobarns. No significant excess is observed above the background expectation, and upper limits are set on the Higgs boson production cross section. The presence of the standard model Higgs boson with a mass in the 270-440 GeV range is excluded at 95% confidence level.Comment: Submitted to JHE

    Measurement of the t t-bar production cross section in the dilepton channel in pp collisions at sqrt(s) = 7 TeV

    The t t-bar production cross section (sigma[t t-bar]) is measured in proton-proton collisions at sqrt(s) = 7 TeV in data collected by the CMS experiment, corresponding to an integrated luminosity of 2.3 inverse femtobarns. The measurement is performed in events with two leptons (electrons or muons) in the final state, at least two jets identified as jets originating from b quarks, and the presence of an imbalance in transverse momentum. The measured value of sigma[t t-bar] for a top-quark mass of 172.5 GeV is 161.9 +/- 2.5 (stat.) +5.1/-5.0 (syst.) +/- 3.6(lumi.) pb, consistent with the prediction of the standard model.Comment: Replaced with published version.     Search for anomalous t t-bar production in the highly-boosted all-hadronic final state

    A search is presented for a massive particle, generically referred to as a Z', decaying into a t t-bar pair. The search focuses on Z' resonances that are sufficiently massive to produce highly Lorentz-boosted top quarks, which yield collimated decay products that are partially or fully merged into single jets. The analysis uses new methods to analyze jet substructure, providing suppression of the non-top multijet backgrounds. The analysis is based on a data sample of proton-proton collisions at a center-of-mass energy of 7 TeV, corresponding to an integrated luminosity of 5 inverse femtobarns. Upper limits in the range of 1 pb are set on the product of the production cross section and branching fraction for a topcolor Z' modeled for several widths, as well as for a Randall--Sundrum Kaluza--Klein gluon.
