Exploring the clothing overconsumption of young adults:An experimental study with communication interventions
The increase in clothing consumption per capita in the last decades presents substantial environmental and societal challenges. Young adults, heavily influenced by advertisements and new trends via social media, emerge as substantial contributors to the escalating issue of clothing overconsumption. This research aims at better understanding the drivers of clothing consumption for young adults and the policy interventions that can be designed to change clothing overconsumption behaviour. This study employs a survey experiment with communication interventions using different framing strategies. In doing so, the study focuses on the potential impact of communication interventions on the clothing consumption rates of young adults. The study showed that a communication intervention can motivate young adults to purchase less clothing and gives an initial insight into how to implement this type of intervention. Moreover, it provides initial evidence that intervention strategies inspired by sufficiency can be effective. This research calls for more transformative policies to stimulate sustainable consumption that go beyond promoting sustainable alternatives

Interactive screw conveyor learning
Treball desenvolupat en el marc del programa "European Project Semester".This report documents the development process and outcomes of the "Interactive Screw Conveyor Learning" project, conducted as part of the European Project Semester at Avans University of Applied Sciences. The initiative aimed to engage students in the technical field through an interactive hands-on game, fostering knowledge about screw conveying technology. The project collaborates with Van Beek Schroeftransport, a company dedicated to innovation in screw conveyor systems. Challenges faced by the company, such as a limited scope in educational activities, led to the development of an interactive game targeting students aged 10-20. The game, incorporating screw technology, industrial processes, and basic engineering principles, serves the dual purpose of informing and engaging the younger generation in the world of engineering. This abstract summarizes the efforts, challenges, and achievements encountered throughout the project

Testing the current paradigm for space weather prediction with heliospheric imagers
Predictions of the arrival of four coronal mass ejections (CMEs) in geospace are produced through use of three CME geometric models combined with CME drag modeling, constraining these models with the available Coronagraph and Heliospheric Imager data. The efficacy of these predications is assessed by comparison with the Space Weather Prediction Center (SWPC) numerical MHD forecasts of these same events. It is found that such a prediction technique cannot outperform the standard SWPC forecast at a statistically meaningful level. We test the Harmonic Mean, Self-Similar Expansion, and Ellipse Evolution geometric models, and find that, for these events at least, the differences between the models are smaller than the observational errors. We present a new method of characterizing CME fronts in the Heliospheric Imager field of view, utilizing the analysis of citizen scientists working with the Solar Stormwatch project, and we demonstrate that this provides a more accurate representation of the CME front than is obtained by experts analyzing elongation time maps for the studied events. Comparison of the CME kinematics estimated independently from the STEREO-A and STEREO-B Heliospheric Imager data reveals inconsistencies that cannot be explained within the observational errors and model assumptions. We argue that these observations imply that the assumptions of the CME geometric models are routinely invalidated and question their utility in a space weather forecasting context. These results argue for the continuing development of more advanced techniques to better exploit the Heliospheric Imager observations for space weather forecasting
The National Lung Matrix Trial: translating the biology of stratification in advanced non-small-cell lung cancer
© The Author 2015.Background: The management of NSCLC has been transformed by stratified medicine. The National Lung Matrix Trial (NLMT) is a UK-wide study exploring the activity of rationally selected biomarker/targeted therapy combinations. Patients and methods: The Cancer Research UK (CRUK) Stratified Medicine Programme 2 is undertaking the large volume national molecular pre-screening which integrates with the NLMT. At study initiation, there are eight drugs being used to target 18 molecular cohorts. The aim is to determine whether there is sufficient signal of activity in any drug-biomarker combination to warrant further investigation. A Bayesian adaptive design that gives a more realistic approach to decision making and flexibility to make conclusions without fixing the sample size was chosen. The screening platform is an adaptable 28-gene Nextera next-generation sequencing platform designed by Illumina, covering the range of molecular abnormalities being targeted. The adaptive design allows new biomarker-drug combination cohorts to be incorporated by substantial amendment. The pre-clinical justification for each biomarker-drug combination has been rigorously assessed creating molecular exclusion rules and a trumping strategy in patients harbouring concomitant actionable genetic abnormalities. Discrete routes of pathway activation or inactivation determined by cancer genome aberrations are treated as separate cohorts. Key translational analyses include the deep genomic analysis of pre- and post-treatment biopsies, the establishment of patient-derived xenograft models and longitudinal ctDNA collection, in order to define predictive biomarkers, mechanisms of resistance and early markers of response and relapse. Conclusion: The SMP2 platform will provide large scale genetic screening to inform entry into the NLMT, a trial explicitly aimed at discovering novel actionable cohorts in NSCLC
CD8+ T cells from a novel T cell receptor transgenic mouse induce liver-stage immunity that can be boosted by blood-stage infection in rodent malaria
To follow the fate of CD8+ T cells responsive to Plasmodium berghei ANKA (PbA) infection, we generated an MHC I-restricted TCR transgenic mouse line against this pathogen. T cells from this line, termed PbT-I T cells, were able to respond to blood-stage infection by PbA and two other rodent malaria species, P. yoelii XNL and P. chabaudi AS. These PbT-I T cells were also able to respond to sporozoites and to protect mice from liver-stage infection. Examination of the requirements for priming after intravenous administration of irradiated sporozoites, an effective vaccination approach, showed that the spleen rather than the liver was the main site of priming and that responses depended on CD8α+ dendritic cells. Importantly, sequential exposure to irradiated sporozoites followed two days later by blood-stage infection led to augmented PbT-I T cell expansion. These findings indicate that PbT-I T cells are a highly versatile tool for studying multiple stages and species of rodent malaria and suggest that cross-stage reactive CD8+ T cells may be utilized in liver-stage vaccine design to enable boosting by blood-stage infections
Food versus fuel? Going beyond biofuels
In less than a decade, biofuels transitioned from being a socially and politically acceptable alternative to conventional transport fuels to a deeply contested solution. Claims of land grabs, forest loss and food riots emerged to undermine the sustainability rationale that originally motivated their adoption. One of the early controversies to hit biofuels was that of food versus fuel. This framing drew attention not only to the competing uses of land i.e. for food or for fuel, but also to the impacts of consumption on marginalised people, particularly in the global South. While the debate has provided a useful hook on which to hang criticisms of increased demand for biofuels, it also masks a more complex reality. In particular, the multifaceted and global linkages between the stewardship of land, the food sector, and global energy policies. In this paper, we use the debate on food versus fuel as a lens to examine the interdependencies between the multiple end-uses of feedstocks and the multifunctionality of land. Revealing a more nuanced understanding of the realities of agricultural networks, land use conflicts and the values of the people managing land, we argue that the simplification achieved by food versus fuel, although effective in generating public resonance that has filtered into political response, has failed to capture much that is at the heart of the issue
Autoantibodies neutralizing type I IFNs are present in ~4% of uninfected individuals over 70 years old and account for ~20% of COVID-19 deaths
Publisher Copyright: © 2021 The Authors, some rights reserved.Circulating autoantibodies (auto-Abs) neutralizing high concentrations (10 ng/ml; in plasma diluted 1:10) of IFN-alpha and/or IFN-omega are found in about 10% of patients with critical COVID-19 (coronavirus disease 2019) pneumonia but not in individuals with asymptomatic infections. We detect auto-Abs neutralizing 100-fold lower, more physiological, concentrations of IFN-alpha and/or IFN-omega (100 pg/ml; in 1:10 dilutions of plasma) in 13.6% of 3595 patients with critical COVID-19, including 21% of 374 patients >80 years, and 6.5% of 522 patients with severe COVID-19. These antibodies are also detected in 18% of the 1124 deceased patients (aged 20 days to 99 years; mean: 70 years). Moreover, another 1.3% of patients with critical COVID-19 and 0.9% of the deceased patients have auto-Abs neutralizing high concentrations of IFN-beta. We also show, in a sample of 34,159 uninfected individuals from the general population, that auto-Abs neutralizing high concentrations of IFN-alpha and/or IFN-omega are present in 0.18% of individuals between 18 and 69 years, 1.1% between 70 and 79 years, and 3.4% >80 years. Moreover, the proportion of individuals carrying auto-Abs neutralizing lower concentrations is greater in a subsample of 10,778 uninfected individuals: 1% of individuals 80 years. By contrast, auto-Abs neutralizing IFN-beta do not become more frequent with age. Auto-Abs neutralizing type I IFNs predate SARS-CoV-2 infection and sharply increase in prevalence after the age of 70 years. They account for about 20% of both critical COVID-19 cases in the over 80s and total fatal COVID-19 cases.Peer reviewe
Flow cytometric immunobead assay for fast and easy detection of PML-RARA fusion proteins for the diagnosis of acute promyelocytic leukemia
The PML-RARA fusion protein is found in approximately 97% of patients with acute promyelocytic leukemia (APL). APL can be associated with life-threatening bleeding complications when undiagnosed and not treated expeditiously. The PML-RARA fusion protein arrests maturation of myeloid cells at the promyelocytic stage, leading to the accumulation of neoplastic promyelocytes. Complete remission can be obtained by treatment with all-trans-retinoic acid (ATRA) in combination with chemotherapy. Diagnosis of APL is based on the detection of t(15;17) by karyotyping, fluorescence in situ hybridization or PCR. These techniques are laborious and demand specialized laboratories. We developed a fast (performed within 4-5 h) and sensitive (detection of at least 10% malignant cells in normal background) flow cytometric immunobead assay for the detection of PML-RARA fusion proteins in cell lysates using a bead-bound anti-RARA capture antibody and a phycoerythrin-conjugated anti-PML detection antibody. Testing of 163 newly diagnosed patients (including 46 APL cases) with the PML-RARA immunobead assay showed full concordance with the PML-RARA PCR results. As the applied antibodies recognize outer domains of the fusion protein, the assay appeared to work independently of the PML gene break point region. Importantly, the assay can be used in parallel with routine immunophenotyping for fast and easy diagnosis of APL
