Estimation of musculotendon parameters for scaled and subject specific musculoskeletal models using an optimization technique.
A challenging aspect of subject specific musculoskeletal modeling is the estimation of muscle parameters, especially optimal fiber length and tendon slack length. In this study, the method for scaling musculotendon parameters published by Winby et al. (2008), J. Biomech. 41, 1682-1688, has been reformulated, generalized and applied to two cases of practical interest: 1) the adjustment of muscle parameters in the entire lower limb following linear scaling of a generic model and 2) their estimation "from scratch" in a subject specific model of the hip joint created from medical images. In the first case, the procedure maintained the muscles׳ operating range between models with mean errors below 2.3% of the reference model normalized fiber length value. In the second case, a subject specific model of the hip joint was created using segmented bone geometries and muscle volumes publicly available for a cadaveric specimen from the Living Human Digital Library (LHDL). Estimated optimal fiber lengths were found to be consistent with those of a previously published dataset for all 27 considered muscle bundles except gracilis. However, computed tendon slack lengths differed from tendon lengths measured in the LHDL cadaver, suggesting that tendon slack length should be determined via optimization in subject-specific applications. Overall, the presented methodology could adjust the parameters of a scaled model and enabled the estimation of muscle parameters in newly created subject specific models. Biofeedback for gait retraining based on real-time estimation of tibiofemoral joint contact forces
Biofeedback assisted rehabilitation and intervention technologies have the potential to modify clinically relevant biomechanics. Gait retraining has been used to reduce the knee adduction moment, a surrogate of medial tibiofemoral joint loading often used in knee osteoarthritis research. In this study we present an electromyogram-driven neuromusculoskeletal model of the lower-limb to estimate, in real-time, the tibiofemoral joint loads. The model included 34 musculotendon units spanning the hip, knee, and ankle joints. Full-body inverse kinematics, inverse dynamics, and musculotendon kinematics were solved in real-time from motion capture and force plate data to estimate the knee medial tibiofemoral contact force (MTFF). We analyzed 5 healthy subjects while they were walking on an instrumented treadmill with visual biofeedback of their MTFF. Each subject was asked to modify their gait in order to vary the magnitude of their MTFF. All subjects were able to increase their MTFF, whereas only 3 subjects could decrease it, and only after receiving verbal suggestions about possible gait modification strategies. Results indicate the important role of knee muscle activation patterns in modulating the MTFF. While this study focused on the knee, the technology can be extended to examine the musculoskeletal tissue loads at different sites of the human body

MARKERLESS ANALYSIS OF SWIMMERS’ MOTION: A PILOT STUDY
Regular laboratory-based motion analysis with skin surface markers is not always feasible. In particular, when studying swimmers kinematics, traditional motion capture techniques cannot be adopted. Although video recordings from swimmers often exist, current methods for biomechanical analysis of these are inadequate. They usually rely on manual digitization of joints’ position on a single sagittal view of the subject. Therefore, in this study a method for three dimensional (3D) markerless motion capture of swimmers is presented. Validation of a model-based inverse kinematics approach based on wearable inertial sensors
Wearable inertial measurement units (IMUs) are a promising solution to human motion estimation. Using IMUs 3D orientations, a model-driven inverse kinematics methodology to estimate joint angles is presented. Estimated joint angles were validated against encoder-measured kinematics (robot) and against marker-based kinematics (passive mechanism). Results are promising, with RMS angular errors respectively lower than 3 and 6 deg over a minimum range of motion of 50 deg (robot) and 160 deg (passive mechanism). Moreover, a noise robustness analysis revealed that the model-driven approach reduces the effects of experimental noises, making the proposed technique particularly suitable for application in human motion analysis
Electrostatic polarization fields trigger glioblastoma stem cell differentiation
Over the last few years it has been understood that the interface between living cells and the underlying materials can be a powerful tool to manipulate cell functions. In this study, we explore the hypothesis that the electrical cell/material interface can regulate the differentiation of cancer stem-like cells (CSCs). Electrospun polymer fibres, either polyamide 66 or poly(lactic acid), with embedded graphene nanoplatelets (GnPs), have been fabricated as CSC scaffolds, providing both the 3D microenvironment and a suitable electrical environment favorable for CSCs adhesion, growth and differentiation. We have investigated the impact of these scaffolds on the morphological, immunostaining and electrophysiological properties of CSCs extracted from human glioblastoma multiform (GBM) tumor cell line. Our data provide evidence in favor of the ability of GnP-incorporating scaffolds to promote CSC differentiation to the glial phenotype. Numerical simulations support the hypothesis that the electrical interface promotes the hyperpolarization of the cell membrane potential, thus triggering the CSC differentiation. We propose that the electrical cell/material interface can regulate endogenous bioelectrical cues, through the membrane potential manipulation, resulting in the differentiation of CSCs. Material-induced differentiation of stem cells and particularly of CSCs, can open new horizons in tissue engineering and new approaches to cancer treatment, especially GBM
Crowd-sourced amputee gait data : a feasibility study using YouTube videos of unilateral trans-femoral gait
Collecting large datasets of amputee gait data is notoriously difficult. Additionally, collecting data on less prevalent amputations or on gait activities other than level walking and running on hard surfaces is rarely attempted. However, with the wealth of user-generated content on the Internet, the scope for collecting amputee gait data from alternative sources other than traditional gait labs is intriguing. Here we investigate the potential of YouTube videos to provide gait data on amputee walking. We use an example dataset of trans-femoral amputees level walking at self-selected speeds to collect temporal gait parameters and calculate gait asymmetry. We compare our YouTube data with typical literature values, and show that our methodology produces results that are highly comparable to data collected in a traditional manner. The similarity between the results of our novel methodology and literature values lends confidence to our technique. Nevertheless, clear challenges with the collection and interpretation of crowd-sourced gait data remain, including long term access to datasets, and a lack of validity and reliability studies in this area
Single view silhouette fitting techniques for estimating tennis racket position
Stereo camera systems have been used to track markers attached to a racket, allowing its position to be obtained in three-dimensional (3D) space. Typically, markers are manually selected on the image plane, but this can be time-consuming. A markerless system based on one stationary camera estimating 3D racket position data is desirable for research and play. The markerless method presented in this paper relies on a set of racket silhouette views in a common reference frame captured with a calibrated camera and a silhouette of a racket captured with a camera whose relative pose is outside the common reference frame. The aim of this paper is to provide validation of these single view fitting techniques to estimate the pose of a tennis racket. This includes the development of a calibration method to provide the relative pose of a stationary camera with respect to a racket. Mean static racket position was reconstructed to within ±2 mm. Computer generated camera poses and silhouette views of a full size racket model were used to demonstrate the potential of the method to estimate 3D racket position during a simplified serve scenario. From a camera distance of 14 m, 3D racket position was estimated providing a spatial accuracy of 1.9 ± 0.14 mm, similar to recent 3D video marker tracking studies of tennis
