Probing the Star Formation Main Sequence down to M at
We investigate the star formation main sequence (MS) (SFR-M) down
to 10 using a sample of 34,061 newly-discovered
ultra-faint ( mag) galaxies at detected in
the GOODS-N field. Virtually these galaxies are not contained in previous
public catalogs, effectively doubling the number of known sources in the field.
The sample was constructed by stacking the optical broad-band observations
taken by the HST/GOODS-CANDELS surveys as well as the 25 ultra-deep medium-band
images gathered by the GTC/SHARDS project. Our sources are faint (average
observed magnitudes mag, mag), blue (UV-slope
), star-forming (rest-frame colors mag,
mag) galaxies. These observational characteristics are
identified with young (mass-weighted age Gyr)
stellar populations subject to low attenuations (
mag). Our sample allows us to probe the MS down to
at and at , around 0.6 dex deeper than
previous analysis. In the low-mass galaxy regime, we find an average value for
the slope of 0.97 at and 1.12 at . Nearly 60% of our
sample presents stellar masses in the range M between
. If the slope of the MS remained constant in this regime, the sources

Generation and characterization of two immortalized human osteoblastic cell lines useful for epigenetic studies
Different model systems using osteoblastic cell lines have been developed to help understand the process of bone formation. Here, we report the establishment of two human osteoblastic cell lines obtained from primary cultures upon transduction of immortalizing genes. The resulting cell lines had no major differences to their parental lines in their gene expression profiles. Similar to primary osteoblastic cells, osteocalcin transcription increased following 1,25-dihydroxyvitamin D3 treatment and the immortalized cells formed a mineralized matrix, as detected by Alizarin Red staining. Moreover, these human cell lines responded by upregulating ALPL gene expression after treatment with the demethylating agent 5-aza-2 Œ-deoxycytidine (AzadC), as shown before for primary osteoblasts. We further demonstrate that these cell lines can differentiate in vivo, using a hydroxyapatite/tricalcium phosphate composite as a scaffold, to produce bone matrix. More importantly, we show that these cells respond to demethylating treatment, as shown by the increase in SOST mRNA levels, the gene encoding sclerostin, upon treatment of the recipient mice with AzadC. This also confirms, in vivo, the role of DNA methylation in the regulation of SOST expression previously shown in vitro. Altogether our results show that these immortalized cell lines constitute a particularly useful model system to obtain further insight into bone homeostasis, and particularly into the epigenetic mechanisms regulating sclerostin production
The synovial and blood monocyte DNA methylomes mirror prognosis, evolution and treatment in early arthritis
Identifying predictive biomarkers at early stages of early inflammatory arthritis is crucial for starting appropriate therapies to avoid poor outcomes. Monocytes and macrophages, largely associated with arthritis, are contributors and sensors of inflammation through epigenetic modifications. In this study, we investigated associations between clinical features and DNA methylation in blood and synovial fluid (SF) monocytes in a prospective cohort of early inflammatory arthritis patients. Undifferentiated arthritis (UA) blood monocyte DNA methylation profiles exhibited significant alterations in comparison with those from healthy donors. We identified additional differences both in blood and SF monocytes after comparing UA patients grouped by their future outcomes, good versus poor. Patient profiles in subsequent visits revealed a reversion towards a healthy level in both groups, those requiring disease-modifying antirheumatic drugs (DMARDs) and those that remitted spontaneously. Changes in disease activity between visits also impacted DNA methylation, partially concomitant in the SF of UA and in blood monocytes of rheumatoid arthritis patients. Epigenetic similarities between arthritis types allow a common prediction of disease activity. Our results constitute a resource of DNA methylation-based biomarkers of poor prognosis, disease activity and treatment efficacy in early untreated UA patients for the personalized clinical management of early inflammatory arthritis patients
FERO: Finding Extreme Relativistic Objects. I. Statistics of Relativistic Fe Kalpha lines in Radio-Quiet Type 1 AGN
Accretion models predict that fluorescence lines broadened by relativistic
effects should arise from reflection of X-ray emission onto the inner region of
the accretion disc surrounding the central black hole of active galactic nuclei
(AGN). The theory behind the origin of relativistic lines is well established,
and observational evidence from a moderate number of sources seems to support
the existence of these lines. The aim of this work is to establish the fraction
of AGN with relativistic Fe Kalpha lines, and study possible correlations with
source physical properties. An XMM-Newton collection of 149 radio-quiet Type 1
AGN has been systematically and uniformly analyzed in order to search for
significant evidence of a relativistically broadened Fe Kalpha line. To enable
statistical studies, an almost complete, flux-limited subsample of 31 sources
has been defined. The 2-10 keV spectra of the FERO sources have been compared
with a complex model including most of the physical components observed in the
X-ray spectra of Seyfert galaxies: a power law primary continuum modified by
non-relativistic Compton reflection and warm absorption, plus a series of
narrow Fe line reflection features. The observed fraction of sources in the
flux-limited sample that show significant evidence of a relativistic Fe Kalpha
line is 36%. The average line Equivalent Width (EW) is of the order of 100 eV,
while the average disc inclination angle is 28+/-5 deg and the average
power-law index of the radial disc emissivity law is 2.4+/-0.4. The spin value
is well constrained only in 2 cases (MCG-6-30-15 and MRK509), and in the rest
of the cases, whenever a constraint can be placed, it always implies the
rejection of the static black hole solution. The Fe Kalpha line EW does not
correlate with disc parameters or with system physical properties, such as
black hole mass, accretion rate and hard X-ray luminosity.Comment: 22 pages, 7 figures. Accepted for publication in A&
An e-health driven laboratory information system to support HIV treatment in Peru: E-quity for laboratory personnel, health providers and people living with HIV
<p>Abstract</p> <p>Background</p> <p>Peru has a concentrated HIV epidemic with an estimated 76,000 people living with HIV (PLHIV). Access to highly active antiretroviral therapy (HAART) expanded between 2004-2006 and the Peruvian National Institute of Health was named by the Ministry of Health as the institution responsible for carrying out testing to monitor the effectiveness of HAART. However, a national public health laboratory information system did not exist. We describe the design and implementation of an e-health driven, web-based laboratory information system - NETLAB - to communicate laboratory results for monitoring HAART to laboratory personnel, health providers and PLHIV.</p> <p>Methods</p> <p>We carried out a needs assessment of the existing public health laboratory system, which included the generation and subsequent review of flowcharts of laboratory testing processes to generate better, more efficient streamlined processes, improving them and eliminating duplications. Next, we designed NETLAB as a modular system, integrating key security functions. The system was implemented and evaluated.</p> <p>Results</p> <p>The three main components of the NETLAB system, registration, reporting and education, began operating in early 2007. The number of PLHIV with recorded CD4 counts and viral loads increased by 1.5 times, to reach 18,907. Publication of test results with NETLAB took an average of 1 day, compared to a pre-NETLAB average of 60 days. NETLAB reached 2,037 users, including 944 PLHIV and 1,093 health providers, during its first year and a half. The percentage of overall PLHIV and health providers who were aware of NETLAB and had a NETLAB password has also increased substantially.</p> <p>Conclusion</p> <p>NETLAB is an effective laboratory management tool since it is directly integrated into the national laboratory system and streamlined existing processes at the local, regional and national levels. The system also represents the best possible source of timely laboratory information for health providers and PLHIV, allowing patients to access their own results and other helpful information about their health, extending the scope of HIV treatment beyond the health facility and providing a model for other countries to follow. The NETLAB system now includes 100 diseases of public health importance for which the Peruvian National Institute of Health and the network of public health laboratories provide testing and results.</p
A Variable-Density Absorption Event in NGC 3227 mapped with Suzaku and Swift
The morphology of the circumnuclear gas accreting onto supermassive black
holes in Seyfert galaxies remains a topic of much debate. As the innermost
regions of Active Galactic Nuclei (AGN) are spatially unresolved, X-ray
spectroscopy, and in particular line-of-sight absorption variability, is a key
diagnostic to map out the distribution of gas. Observations of variable X-ray
absorption in multiple Seyferts and over a wide range of timescales indicate
the presence of clumps/clouds of gas within the circumnuclear material. Eclipse
events by clumps transiting the line of sight allow us to explore the
properties of the clumps over a wide range of radial distances from the
optical/UV Broad Line Region (BLR) to beyond the dust sublimation radius.
Time-resolved absorption events have been extremely rare so far, but suggest a
range of density profiles across Seyferts. We resolve a weeks-long absorption
event in the Seyfert NGC 3227. We examine six Suzaku and twelve Swift
observations from a 2008 campaign spanning 5 weeks. We use a model accounting
for the complex spectral interplay of three differently-ionized absorbers. We
perform time-resolved spectroscopy to discern the absorption variability
behavior. We also examine the IR-to-X-ray spectral energy distribution (SED) to
test for reddening by dust. The 2008 absorption event is due to
moderately-ionized () gas covering 90% of the line of
sight. We resolve the density profile to be highly irregular, in contrast to a
previous symmetric and centrally-peaked event mapped with RXTE in the same
object. The UV data do not show significant reddening, suggesting that the
cloud is dust-free. The 2008 campaign has revealed a transit by a filamentary,
moderately-ionized cloud of variable density that is likely located in the BLR,
and possibly part of a disk wind.Comment: Accepted for publication by A&
A simultaneous search for High-z LAEs and LBGs in the SHARDS survey
We have undertaken a comprehensive search for both Lyman Alpha Emitters (LAEs) and Lyman Break Galaxies (LBGs) in the SHARDS Survey of the GOODS-N field. SHARDS is a deep imaging survey, made with the 10.4 m Gran Telescopio Canarias (GTC), employing 25 medium band filters in the range from 500 to 941 nm. This is the first time that both LAEs and LBGs are surveyed simultaneously in a systematic way in a large field. We draw a sample of 1558 sources; 528 of them are LAEs. Most of the sources (1434) show rest-frame UV continua. A minority of them (124) are pure LAEs with virtually no continuum detected in SHARDS. We study these sources from z ∼ 3.35 up to z ∼ 6.8, well into the epoch of reionization. Note that surveys done with just one or two narrow band filters lack the possibility to spot the rest-frame UV continuum present in most of our LAEs. We derive redshifts, Star Formation Rates (SFRs), Lyα Equivalent Widths (EWs) and Luminosity Functions (LFs). Grouping within our sample is also studied, finding 92 pairs or groups of galaxies at the same redshift separated by less than 60 comoving kpc. In addition, we relate 87 and 55 UV-selected objects with two known overdensities at z = 4.05 and z = 5.198, respectively. Finally, we show that surveys made with broad band filters are prone to introduce many unwanted sources (∼20% interlopers), which means that previous studies may be overestimating the calculated LFs, specially at the faint end
A Proposed Approach to Chronic Airway Disease (CAD) Using Therapeutic Goals and Treatable Traits: A Look to the Future
© 2020 Pérez de Llano et al.Chronic airflow obstruction affects a wide range of airway diseases, the most frequent of which are asthma, COPD, and bronchiectasis; they are clearly identifiable in their extremes, but quite frequently overlap in some of their pathophysiological and clinical characteristics. This has generated the description of new mixed or overlapping disease phenotypes with no clear biological grounds. In this special article, a group of experts provides their perspective and proposes approaching the treatment of chronic airway disease (CAD) through the identification of a series of therapeutic goals (TG) linked to treatable traits (TT) – understood as clinical, physiological, or biological characteristics that are quantifiable using biomarkers. This therapeutic approach needs validating in a clinical trial with the strategy of identification of TG and treatment according to TT for each patient independently of their prior diagnosis
