    A Prototype National Drought Alert Strategic Information System for Australia

    Defining and categorizing drought in a quantitative and scientific manner are important national issues for Australian state and Commonwealth governments, landholders, and agribusiness. The challenge for modelers of Australia’s grasslands is to integrate biological models, geographic information systems (GIS), satellite imagery, economics, climatology, and visual high-performance computing into an Internet-deliverable application that can provide easily understood monitoring and prediction advice in near real-time— a national drought alert strategic information system. Although NOAA satellite-derived imagery has been somewhat useful in the broad-scale spatial assessment of green cover, especially the spatial response of vegetation to rainfall events (Smith, 1994; Dudgeon et al., 1990; Filet et al., 1990), it has inherent limitations in providing a total solution for drought and rangeland monitoring; biomass relationships are not good, tree cover confounds the signal, and a future projection of the current situation is not inherent. Also, the interpretation of the imagery does not usually consider the effects of soil type, vegetation structure, or rangeland “condition.” Similarly, rainfall analyses alone do not necessarily reflect the quantity and quality of pasture available on the ground. In the recent 1991–95 record-breaking drought in Queensland, rainfall analyses did not map the drought-declared southwestern areas of the state as droughted, and, conversely, coastal areas of the state were classed as droughted by rainfall analyses, when there was no community push for their declaration. Measures of rainfall effectiveness expressed as measures of plant biomass are required for drought definition. Improved assessments of the quantity and quality of biomass are needed, as well as consideration of herbivore densities and future climatic scenarios

    A review of practical tools for rapid monitoring of membrane bioreactors

    The production of high quality effluent from membrane bioreactors (MBRs) arguably requires less supervision than conventional activated sludge (CAS) processes. Nevertheless, the use of membranes brings additional issues of activated sludge filterability, cake layer formation and membrane fouling. From a practical standpoint, process engineers and operators require simple tools which offer timely information about the biological health and filterability of the mixed liquor as well as risks of membrane fouling. To this end, a range of analytical tools and biological assays are critically reviewed from this perspective. This review recommends that Capillary Suction Time (CST) analysis along with Total Suspended and Volatile Solids (TSS/VSS) analysis is used daily. For broad characterisation, total carbon and nitrogen analysis offer significant advantages over the commonly used chemical and biological oxygen demand (COD/BOD) analyses. Of the technologies for determining the vitality of the microbial biomass the most robust and reproducible, are the second generation adenosine-5'-triphosphate (ATP) test kits. Extracellular polymer concentrations are best monitored by measurement of turbidity after centrifugation. Taken collectively these tools can be used routinely to ensure timely intervention and smoother operation of MBR systems. © 2016 Elsevier Ltd

    Reading Dickens’s characters: employing psycholinguistic methods to investigate the cognitive reality of patterns in texts

    This article reports the findings of an empirical study that uses eye-tracking and follow-up interviews as methods to investigate how participants read body language clusters in novels by Charles Dickens. The study builds on previous corpus stylistic work that has identified patterns of body language presentation as techniques of characterisation in Dickens (Mahlberg, 2013). The article focuses on the reading of ‘clusters’, that is, repeated sequences of words. It is set in a research context that brings together observations from both corpus linguistics and psycholinguistics on the processing of repeated patterns. The results show that the body language clusters are read significantly faster than the overall sample extracts which suggests that the clusters are stored as units in the brain. This finding is complemented by the results of the follow-up questions which indicate that readers do not seem to refer to the clusters when talking about character information, although they are able to refer to clusters when biased prompts are used to elicit information. Beyond the specific results of the study, this article makes a contribution to the development of complementary methods in literary stylistics and it points to directions for further subclassifications of clusters that could not be achieved on the basis of corpus data alone

    An in-vitro screening assay for the detection of inhibitors of proinflammatory cytokine synthesis: a useful tool for the development of new antiarthritic and disease modifying drugs

    AbstractObjective This work targets the development of a new tool to help develop new anticytokine drugs that prevent or reduce the progression of arthritic diseases. The specific aim of our study was to establish a fast and reliable in vitro screening assay of cytokine synthesis inhibitors (TNFα, IL-1β) which shows better correlation with enzyme assays than previously reported in vitro assays. The test system should be able to detect p38-MAP kinase inhibitors.Material and methods Human peripheral blood mononuclear cells (PBMCs) were isolated by Ficoll density gradient centrifugation from human EDTA-potassium whole blood. Cells were adjusted at 1×106 cells/ml. PBMCs were stimulated with lipopolysaccharide (LPS; E. coli serotype 026:B6: 1μg/ml) in the presence of test compound (10−5–10−8M) for 4h at 37°C in a 5% CO2-incubator. Induced TNFα and IL-1β protein were measured by ELISA.Results The following are representative examples of inhibitors which effect cytokine synthesis. Corticoid Dexamethasone inhibits IL-1β and TNFα synthesis at IC50 of 38nM and 25nM, respectively. ERK1/ERK2 inhibitor U0126 effects cytokine synthesis at IC50 of 0.34μM for IL-1β production and 0.26μM for TNFα synthesis.p38-MAP kinase inhibitor SB 203580 inhibits IL-1β- and TNF-α-synthesis (IC50sof 0.052μM and 0.46μM) in the same degree as p38-MAP kinase activity (IC50: 0.34μM). Same results could be shown for SB 210313, which had same efficacy on IL-1β and TNFα biosynthesis (IC50's: 1.88μM and 1.01μM) and on p38-MAP kinase (IC50: 6.85μM). Also for SB 202190 this correlation in inhibition of IL-1β and TNFα synthesis (IC50's: 0.055μM and 1.01μM) and p38-MAP kinase inhibition (IC50: 0.088μM) could be shown.Conclusion This study shows the screening assay using PBMCs stimulated with LPS for IL-1β and TNFα synthesis is a reliable test system for the quantification of the effectiveness of new drugs modulating IL-1β and TNFα synthesis which is mainly mediated by p38-MAP Kinase. These assay allows fast detection of IL-1β and TNFα synthesis inhibitors with different modes of action, including p38-MAP kinase inhibitors. The results obtained with our in-vitro screening assay show good correlation with results from enzyme assays. Copyright 2002 OsteoArthritis Research Society International.     Genesis of active sand-filled polygons in lower and central Beacon Valley, Antarctica

The definitive version was published in Permafrost and Periglacial Processes 20 (2009): 295-308, doi:10.1002/ppp.661.Nonsorted polygons with sand-filled wedges were investigated in lower and central Beacon Valley, Antarctica (77.82ºS, 160.67ºE) using field observations coupled with a 2-m resolution Digital Elevation Model and a high-resolution aerial photograph. A gasoline-powered concrete breaker was employed to expose the sediments of four representative polygon centers and six wedges from geomorphic surfaces containing tills of two different ages. The excavated polygons ranged from 9 to 16 m in diameter (average = 12 m); the sand-filled wedges ranged from 0.2 m to 2.5 m in width (average = 0.9 m). The top of ice-bonded permafrost ranged from 12 to 62 cm in depth (average = 33 cm) in the polygon centers and from 64 to >90 cm (average = >75 cm) in wedges. One active thermal contraction fissure generally was apparent at the surface, but excavations revealed numerous inactive fissures. The wedges contain sand laminations averaging 3 mm in width when viewed in cross section. Although most of the polygons were of the sandwedge type, some contained ice veins up to 1 cm in width and could be classed as composite wedges. Three stages of polygon development were observed, including strongly developed polygons on Taylor II surfaces (ca. 117 ka), moderately developed polygons on Taylor III surfaces (ca. 200 ka), and poorly developed polygons on Taylor IVa and older (ca. >1.1 Ma) surfaces. This retrogressive development may be due to sublimation of ice-bonded bonded permafrost following thermal cracking.     Refined localization of TSC1 by combined analysis of 9q34 and 16pl3 data in 14 tuberous sclerosis families

    Tuberous sclerosis (TSC) is a heterogeneous trait. Since 1990, linkage studies have yielded putative TSC loci on chromosomes 9, 11, 12 and 16. Our current analysis, performed on 14 Dutch and British families, reveals only evidence for loci on chromosome 9q34 (TSC1) and chromosome 16p13 (TSC2). We have found no indication for a third locus for TSC, linked or unlinked to either of these chromosomal regions. The majority of our families shows linkage to chromosome 9. We have refined the candidate region for TSC1 to a region of approximately 5 c M between ABL and ABO

    Offshore Geological Hazards: Charting the Course of Progress and Future Directions

    Offshore geological hazards can occur in any marine domain or environment and represent a serious threat to society, the economy, and the environment. Seismicity, slope sedimentary instabilities, submarine volcanism, fluid flow processes, and bottom currents are considered here because they are the most common hazardous processes; tsunamis are also examined because they are a secondary hazard generated mostly by earthquakes, slope instabilities, or volcanic eruptions. The hazards can co-occur and interact, inducing a cascading sequence of events, especially in certain contexts, such as tectonic indentations, volcanic islands, and canyon heads close to the coast. We analyze the key characteristics and main shortcomings of offshore geological hazards to identify their present and future directions for marine geoscience investigations of their identification and characterization. This review establishes that future research will rely on studies including a high level of multidisciplinarity. This approach, which also involves scientific and technological challenges, will require effective integration and interplay between multiscale analysis, mapping, direct deep-sea observations and testing, modelling, and linking offshore observations with onshore observationsVersión del edito

    Prioritization strategies in clinical practice guidelines development: a pilot study

    Objective: Few methodological studies address the prioritization of clinical topics for the development of Clinical Practice Guidelines (CPGs). The aim of this study was to validate a methodology for Priority Determination of Topics (PDT) of CPGs. Methods and results: Firstly, we developed an instrument for PDT with 41 criteria that were grouped under 10 domains, based on a comprehensive systematic search. Secondly, we performed a survey of stakeholders involved in CPGs development, and end users of guidelines, using the instrument. Thirdly, a pilot testing of the PDT procedure was performed in order to choose 10 guideline topics among 34 proposed projects; using a multicriteria analysis approach, we validated a mechanism that followed five stages: determination of the composition of groups, item/domain scoring, weights determination, quality of the information used to support judgments, and finally, topic selection. Participants first scored the importance of each domain, after which four different weighting procedures were calculated (including the survey results). The process of weighting was determined by correlating the data between them. We also reported the quality of evidence used for PDT. Finally, we provided a qualitative analysis of the process.     Laboratory Analysis of Tularemia in Wild-Trapped, Commercially Traded Prairie Dogs, Texas, 2002

    Oropharyngeal tularemia was identified as the cause of a die-off in captured wild prairie dogs at a commercial exotic animal facility in Texas. From this point source, Francisella tularensis–infected prairie dogs were traced to animals distributed to the Czech Republic and to a Texas pet shop. F. tularensis culture isolates were recovered tissue specimens from 63 prairie dogs, including one each from the secondary distribution sites. Molecular and biochemical subtyping indicated that all isolates were F. tularensis subsp. holarctica (Type B). Microagglutination assays detected antibodies against F. tularensis, with titers as great as 1:4,096 in some live animals. All seropositive animals remained culture positive, suggesting that prairie dogs may act as chronic carriers of F. tularensis. These findings demonstrate the need for additional studies of tularemia in prairie dogs, given the seriousness of the resulting disease, the fact that prairie dogs are sold commercially as pets, and the risk for pet-to-human transmission
