    Caractérisation structurale et immunologique d'allergènes alimentaires: les protéines de transfert de lipides de fruits

    Les protéines de transfert de lipides possèdent, à leur surface, des régions responsables des interactions avec les molécules du système immunitaire. Ainsi, la liaison de ces régions appelées " épitopes ", avec les IgE de patients allergiques représente une étape importante dans l'étude des bases de la réaction allergique. Nous avons prédit et caractérisé ces régions épitopiques grâce à la combinaison d'outils de bioinformatique d'une part et, d'autre part, une étude biochimique et immunologique complétée par une étude de biologie moléculaire. La carte épitopique de cette famille d'allergènes de fruits a permis de mettre en évidence des zones consensus, certainement impliquées dans des réactions croisées entre les fruits. Des dosages de ces allergènes dans les fruits comestibles ont également été réalisés, ainsi que l'étude fine de leur localisation cellulaire. Des allergènes recombinants ont été produits; ils devraient permettre d'envisager le démarrage dans un futur proche de protocoles de désensibilisation des patients allergiques.Non-specific Lipid Transfer Proteins (nsLTP) share, on their molecular surface, some IgE-binding areas responsible for their allergenicity. Analyzing the conformation of these epitopes is an important step for understanding the molecular basis of the allergic reaction. IgE-binding epitopes of nsLTP from plants were predicted using a combination of predictive tools and subsequently characterized by biochemical and immunological approaches using IgE from allergic patients. Consensus epitopic regions responsible for some IgE-binding cross-reactivity among different Rosaceae fruits were identified by epitope mapping and conformational analysis. The localization and distribution of nsLTP allergens in the skin and pulp of different fruits has been investigated. nsLTP essentially concentrate as surface allergens in the pericarp of the fruits whereas the pulp contains lower amounts of allergens. nsLTP from apple and peach were produced as correctly-folded and reactive recombinant allergens, usable as standardized allergens for diagnosis and immunotherapy purposes

    The Gaia satellite: a tool for Emission Line Stars and Hot Stars

    The Gaia satellite will be launched at the end of 2011. It will observe at least 1 billion stars, and among them several million emission line stars and hot stars. Gaia will provide parallaxes for each star and spectra for stars till V magnitude equal to 17. After a general description of Gaia, we present the codes and methods, which are currently developed by our team.     The ambient temperature travertine deposits of the calcareous Hoyoux River (Modave, Belgium) and several tributaries are organized and promoted by the filamentous cyanobacterium identified by its morphotype and ecological properties as Phormidium cf. incrustatum. A combination of techniques was used to study this biotope: physico-chemical parameters and CO2 measurements, Scanning and Transmission Electron Microscopy, RAMAN microspectroscopy. A molecular diversity study with pyrosequencing of the cyanobacterial 16S rRNA is in progress.     Carbon dynamics and CO2 and CH4 exchange in the mangrove dominated Guayas river delta, Ecuador

    Although estuaries are considered important pathways in the global carbon cycle, carbon dynamics in tropical estuaries is relatively understudied. Here, the tidal, seasonal and spatial variability of particulate organic carbon (POC), dissolved inorganic carbon (DIC), carbon dioxide (CO2) and methane (CH4), among other biogeochemical variables related to carbon cycling, were studied in the Guayas river delta (Ecuador) to document the sources, processing and fluxes of these carbon forms. All variables were studied during a semi-diurnal (13 hour) tidal cycle and along river transects at low and high tides, all carried out during one dry and rainy season. POC and total suspended matter (TSM) strongly covaried and peaked at high tidal flow velocities during a tidal cycle and at high river discharge during the rainy season, suggesting that resuspension of bottom sediments and/or surface erosion in the river catchment were a dominant source of particulate matter in the water column. The δ13C of POC, (from ~-22‰ to ~-27‰) showed an increasing contribution of marine phytoplankton to the POC pool as moving downstream along the delta during the dry season. Upstream DIC concentrations (~1200 μmol L-1) were high in the Guayas river delta as compared to other tropical estuarine systems, and the δ13C of DIC revealed a shift from a more phytoplankton dominated source in the dry season and downstream (~-4‰) to a relatively more terrestrial source in the rainy season and upstream (~-12.5‰). Both DIC and its δ13C showed slight but consistent deviations from conservative mixing that hint at inputs of 13C depleted DIC from mineralization along the delta. High values of the partial pressure of CO2 (pCO2) observed upstream and in the rainy season (~5250 μatm), associated with O2 undersaturation (~60%) and low δ13CDIC, suggest a strongly heterotrophic system, and resulted in high CO2 efflux to the atmosphere. CH4 concentrations were also higher during the rainy than dry season (93.5±62.5 vs. 61.3±39.5 nmol L-1), but unlike pCO2, showed tidal variations similar to TSM and POC, thus alluding to potential CH4 release from sediments during resuspension events at high tidal flow velocities. This explorative survey revealed complex drivers and biogeochemical processes acting upon various spatio-temporal scales which are necessary to consider for a complete understanding of the carbon biogeochemistry in estuarine systems. Similar surveys on estuarine carbon in data scarce regions are encouraged to constrain uncertainties in coastal zone carbon budgets

    The generality of the GUGA MRCI approach in COLUMBUS for treating complex quantum chemistry

    The core part of the program system COLUMBUS allows highly efficient calculations using variational multireference (MR) methods in the framework of configuration interaction with single and double excitations (MR-CISD) and averaged quadratic coupled-cluster calculations (MR-AQCC), based on uncontracted sets of configurations and the graphical unitary group approach (GUGA). The availability of analytic MR-CISD and MR-AQCC energy gradients and analytic nonadiabatic couplings for MR-CISD enables exciting applications including, e.g., investigations of π-conjugated biradicaloid compounds, calculations of multitudes of excited states, development of diabatization procedures, and furnishing the electronic structure information for on-the-fly surface nonadiabatic dynamics. With fully variational uncontracted spin-orbit MRCI, COLUMBUS provides a unique possibility of performing high-level calculations on compounds containing heavy atoms up to lanthanides and actinides. Crucial for carrying out all of these calculations effectively is the availability of an efficient parallel code for the CI step. Configuration spaces of several billion in size now can be treated quite routinely on standard parallel computer clusters. Emerging developments in COLUMBUS, including the all configuration mean energy multiconfiguration self-consistent field method and the graphically contracted function method, promise to allow practically unlimited configuration space dimensions. Spin density based on the GUGA approach, analytic spin-orbit energy gradients, possibilities for local electron correlation MR calculations, development of general interfaces for nonadiabatic dynamics, and MRCI linear vibronic coupling models conclude this overview

    ARIA digital anamorphosis : Digital transformation of health and care in airway diseases from research to practice

    Digital anamorphosis is used to define a distorted image of health and care that may be viewed correctly using digital tools and strategies. MASK digital anamorphosis represents the process used by MASK to develop the digital transformation of health and care in rhinitis. It strengthens the ARIA change management strategy in the prevention and management of airway disease. The MASK strategy is based on validated digital tools. Using the MASK digital tool and the CARAT online enhanced clinical framework, solutions for practical steps of digital enhancement of care are proposed.Peer reviewe

    EU-wide exposure data of 11 chemical substance groups from the HBM4EU Aligned Studies (2014–2021)

Further details on funding for all the participating studies can be found in the Supplemental Material, Table S12.As one of the core elements of the European Human Biomonitoring Initiative (HBM4EU) a human biomonitoring (HBM) survey was conducted in 23 countries to generate EU-wide comparable HBM data. This survey has built on existing HBM capacity in Europe by aligning national or regional HBM studies, referred to as the HBM4EU Aligned Studies. The HBM4EU Aligned Studies included a total of 10,795 participants of three age groups: (i) 3,576 children aged 6–12 years, (ii) 3,117 teenagers aged 12–18 years and (iii) 4,102 young adults aged 20–39 years. The participants were recruited between 2014 and 2021 in 11–12 countries per age group, geographically distributed across Europe. Depending on the age group, internal exposure to phthalates and the substitute DINCH, halogenated and organophosphorus flame retardants, per- and polyfluoroalkyl substances (PFASs), cadmium, bisphenols, polycyclic aromatic hydrocarbons (PAHs), arsenic species, acrylamide, mycotoxins (deoxynivalenol (total DON)), benzophenones and selected pesticides was assessed by measuring substance specific biomarkers subjected to stringent quality control programs for chemical analysis. For substance groups analyzed in different age groups higher average exposure levels were observed in the youngest age group, i.e., phthalates/DINCH in children versus teenagers, acrylamide and pesticides in children versus adults, benzophenones in teenagers versus adults. Many biomarkers in teenagers and adults varied significantly according to educational attainment, with higher exposure levels of bisphenols, phthalates, benzophenones, PAHs and acrylamide in participants (from households) with lower educational attainment, while teenagers from households with higher educational attainment have higher exposure levels for PFASs and arsenic. In children, a social gradient was only observed for the non-specific pyrethroid metabolite 3-PBA and di-isodecyl phthalate (DiDP), with higher levels in children from households with higher educational attainment. Geographical variations were seen for all exposure biomarkers. For 15 biomarkers, the available health-based HBM guidance values were exceeded with highest exceedance rates for toxicologically relevant arsenic in teenagers (40%), 3-PBA in children (36%), and between 11 and 14% for total DON, Σ (PFOA + PFNA + PFHxS + PFOS), bisphenol S and cadmium. The infrastructure and harmonized approach succeeded in obtaining comparable European wide internal exposure data for a prioritized set of 11 chemical groups. These data serve as a reference for comparison at the global level, provide a baseline to compare the efficacy of the European Commission's chemical strategy for sustainability and will give leverage to national policy makers for the implementation of targeted measures.publishersversionpublishe
