    Analytical Evaluation of RBS Moment Connection for Indian Profiles Under Seismic Behavior

    The effects of Northridge (1994) and Kobe (1995) earthquake, intensive analysis and testing efforts are currently being sought for higher ways of designing seismic resistant steel connections. A connection with RBS (Reduced beam Section) is an acceptable solution to emphasizing a moment connection and can be the most reliable type of connection. Considering the benefits of RBS connection and the lack of information on resources in Indian standards, an analytical study of RBS connections of Indian profiles is performed. This study elaborates on Indian profiles to understand the beam-column connectionñ€ℱs overall performance characteristics during the cyclic response. FEA study is carried out to focus on improving the plastic deformation potential of the connection. The moment capacity of the connection is strengthened by the inclusion of RBS region in the beam. The seven specimens which includes Specimen without RBS, Flange cut RBS and Flange hole RBS have been designed and evaluated using the Finite element ABAQUS program. The findings of this study revealed that specimen without RBS exhibits low Seismic activity efficiency. While RBS specimens performed well, in all instances, Flange holes specimens demonstrate good performance

    Effect of isospin dependent cross-section on fragment production in the collision of charge asymmetric nuclei

    To understand the role of isospin effects on fragmentation due to the collisions of charge asymmetric nuclei, we have performed a complete systematical study using isospin dependent quantum molecular dynamics model. Here simulations have been carried out for 124Xn+124Xn^{124}X_{n}+ ^{124}X_{n}, where n varies from 47 to 59 and for 40Ym+40Ym^{40}Y_{m}+ ^{40}Y_{m}, where m varies from 14 to 23. Our study shows that isospin dependent cross-section shows its influence on fragmentation in the collision of neutron rich nuclei

    Professional Development for Early Career DBER Scholars through In-Person and Virtual Career Panel Workshops

    In discipline-based education research (DBER), early career scholars, such as graduate students and postdoctoral researchers, observe a slew of possible career pathways. Yet, there is a lack of opportunities to learn about such pathways, particularly when transitioning from traditional science, technology, engineering, or math (STEM) disciplinary training into a DBER position. Thus, the DBER Scholars-in-Training Professional Development subcommittee was created within the Society for the Advancement of Biology Education Research (SABER) community to develop a collection of workshops that would serve the greatest professional development needs of early career scholars entering DBER. Through a series of surveys disseminated over multiple years, early career scholars expressed interest in better navigating their career options, which led to the development of the career panel workshop, held during the 2019 and 2020 SABER Annual National Conferences. In this report, we explore the development, implementation, and results of two career panel workshops and compare and contrast the 2019 in-person workshop with the 2020 virtual workshop. We also offer our insights on the value of the career workshop, discuss the next steps, and explore valuable resources for those planning on organizing similar events

    SARS-CoV-2 neutralizing antibodies : longevity, breadth, and evasion by emerging viral variants

    The Severe Acute Respiratory Syndrome Coronavirus 2 (SAU ARS-CoV-2) antibody neutralization response and its evasion by emerging viral variants and variant of concern (VOC) are unknown, but critical to understand reinfection risk and breakthrough infection following vaccination. Antibody immunoreactivity against SARS-CoV-2 antigens and Spike variants, inhibition of Spike-driven virus–cell fusion, and infectious SARS-CoV-2 neutralization were characterized in 807 serial samples from 233 reverse transcription polymerase chain reaction (RT-PCR)–confirmed Coronavirus Disease 2019 (COVID-19) individuals with detailed demographics and followed up to 7 months. A broad and sustained polyantigenic immunoreactivity against SARS-CoV-2 Spike, Membrane, and Nucleocapsid proteins, along with high viral neutralization, was associated with COVID-19 severity. A subgroup of “high responders” maintained high neutralizing responses over time, representing ideal convalescent plasma donors. Antibodies generated against SARS-CoV-2 during the first COVID-19 wave had reduced immunoreactivity and neutralization potency to emerging Spike variants and VOC. Accurate monitoring of SARS-CoV-2 antibody responses would be essential for selection of optimal responders and vaccine monitoring and design

    BCL6-mediated repression of p53 is critical for leukemia stem cell survival in chronic myeloid leukemia

    Chronic myeloid leukemia (CML) is induced by the oncogenic BCR-ABL1 tyrosine kinase and can be effectively treated for many years with tyrosine kinase inhibitors (TKIs). However, unless CML patients receive life-long TKI treatment, leukemia will eventually recur; this is attributed to the failure of TKI treatment to eradicate leukemia-initiating cells (LICs). Recent work demonstrated that FoxO factors are critical for maintenance of CML-initiating cells; however, the mechanism of FoxO-dependent leukemia initiation remained elusive. Here, we identified the BCL6 protooncogene as a critical effector downstream of FoxO in self-renewal signaling of CML-initiating cells. BCL6 represses Arf and p53 in CML cells and is required for colony formation and initiation of leukemia. Importantly, peptide inhibition of BCL6 in human CML cells compromises colony formation and leukemia initiation in transplant recipients and selectively eradicates CD34+ CD38− LICs in patient-derived CML samples. These findings suggest that pharmacological inhibition of BCL6 may represent a novel strategy to eradicate LICs in CML. Clinical validation of this concept could limit the duration of TKI treatment in CML patients, which is currently life-long, and substantially decrease the risk of blast crisis transformation

    Our growing awareness of the microbial world's importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable bacterial and archaeal ribosomal RNA gene sequences to be followed across multiple studies and allow us to explore patterns of diversity at an unprecedented scale. The result is both a reference database giving global context to DNA sequence data and a framework for incorporating data from future studies, fostering increasingly complete characterization of Earth's microbial diversity.Peer reviewe

    SCRUB TYPHUS INFECTION AMONG PUO CASES IN THE PEDIATRIC POPULATION IN A TERTIARY CARE HOSPITAL

    Objectives: Scrub typhus is an acute, febrile, exanthematous illness. This disease is underdiagnosed in India due to its non-specific and varied clinical presentation, low index of suspicion among clinicians, and lack of diagnostic facilities. This study was carried out to know the seroprevalence of scrub typhus in children with PUO and to compare a rapid test with IgM enzyme-linked immunosorbent assay (ELISA) for the diagnosis of scrub typhus. Methods: This cross-sectional analytical study was conducted for a period of 1 year. The study population comprised mainly 280 young children attending pediatric OP and in patients admitted to a tertiary care teaching hospital with fever and related symptoms. The serum samples were tested for Weil-Felix reaction, IgM ELISA, and rapid card test. Results: The mean age group of the study population was 7–9 years, of which 20 cases were positive. The major predisposing factor for scrub typhus infection was vegetation around houses. The sensitivity and specificity of both, card test and IgM ELISA, were 100%. Conclusion: In this study, 7.1% of febrile children were positive for scrub typhus. Leptospirosis, dengue, and typhoid were the common coinfections found in scrub typhus, positive children. Early identification of cases and treatment at the earliest will prevent complications
