Regulación de la citotoxicidad inducida por oleuropeína en células humanas de osteosarcoma: papel de la vía apoptótica Bax/Bcl-2
Tesis por compendio de publicacionesLa dieta mediterránea es ampliamente consumida y aceptada en nuestra sociedad. La oleuropeína, uno de los principales componentes del aceite de oliva, ha sido objeto de estudio en numerosos trabajos, destacando aquellos que tratan sobre sus propiedades anticancerígenas. La agresividad de esta patología y el crecimiento descontrolado que produce en las células hace que exista un gran interés biomédico que permita buscar nuevas estrategias para abordar esta enfermedad. En la primera parte de este estudio, se evaluó la actividad antiproliferativa de la oleuropeína en dos líneas celulares humanas de osteosarcoma (MG-63 y Saos-2). El análisis estadístico de IC50 por el método de regresión Probit sugirió que la oleuropeína tuvo efectos tóxicos similares en ambas líneas celulares (rango de IC50 247.4-475.0 μM en MG-63 y 359.9-798.7 μM en Saos-2). Posteriormente, se estudió la vía apoptótica Bax/Bcl-2 en las células humanas de osteosarcoma MG-63 carentes de p-53, tras ser expuestas a la oleuropeína. La actividad de caspasa-3 se midió mediante ensayo colorimétrico. Además, la expresión del mRNA de Bax, Bcl-2 y p-53 fue determinada por PCR en tiempo real después de 24 horas de exposición. Tanto la relación entre la expresión del mRNA de Bax/Bcl-2 como la actividad de la caspasa-3 aumentaron en las células expuestas a la oleuropeína en comparación con las del control no expuesto (P<0.05). Como se esperaba, no se detectó ningún mRNA de p-53 en las células MG-63. Nuestros resultados sugieren que la oleuropeína induce muerte compatible con apoptosis independiente de p-53 en células de osteosarcoma humano MG-63.Mediterranean diet is widely consumed and accepted in our society. Oleuropein, one of the main components of olive oil, has been investigated in numerous studies, especially those dealing with its anti-cancer properties. The severity of this pathology and the uncontrolled growth it produces in the cells lead to a great biomedical interest in exploring new strategies to efficiently confront this disease.
This study evaluated the antiproliferative activity of oleuropein in two human osteosarcoma cell lines (MG-63 and Saos-2). IC50 statistical analysis by Probit regression method showed that oleuropein had similar toxic effects on both cell lines (IC50 ranges from 247.4-475.0 μM for MG-63 cells and from 359.9-798.7 μM for Saos-2 cells). Thereafter, the p-53-null MG-63 human osteosarcoma cells were exposed to oleuropein and the Bax/Bcl-2 apoptotic pathway was investigated. Caspase-3 activity was determined by a colorimetric assay. Furthermore, Bax, Bcl-2 and p-53 mRNA expression was assessed by real-time PCR following 24 hours of exposure. Both the ratio of Bax/Bcl-2 mRNA expression and caspase-3 activity were enhanced in cells exposed to oleuropein when compared to unexposed control cells (P < 0.05). Predictably, no p-53 mRNA expression was observed in MG-63 cells. These results show that oleuropein induces a p-53-independent apoptosis-like mechanism in human MG-63 osteosarcoma cells
El Efecto De La Implantación Temprana En Niños Con Hipoacusia
Hearing loss in pediatric population is a major health concern, taking into account the immediate repercussions on the cognitive, emotional and language development (serious difficulties in communication and language development). Therefore, it is of crucial importance its early diagnosis and implantation. The objective of this study is to know the effect of cochlear implant on the development of oral language in implanted children in the Province of Santa Cruz de Tenerife (Canary Islands). In order to carry out this research, the psycholinguistic profiles of children who were implanted between 2011 and 2014 were studied, through the use of two standardized tests, Peabody Picture Vocabulary Test and The Illinois Test of Psycholinguistic Abilities (ITPA). As regards the age of implantation, we could observe a significant tendency to use the visual channel in the communication of those children who were implanted at a later age and a tendency to use the auditory channel at an early age of implantation. Children who were implanted at a later age showed a nonfunctional use of the implant and a limited development of their speaking skills
REFLEXIONES CONCEPTUALES SOBRE LA EVALUACIÓN DEL APRENDIZAJE
In this essay we are presenting the conceptual reflections on learning assessment based on the following research questions: What is currently being investigated about learning assessment?, What are the theoretical approaches that predominate in this research?, What are the main contributions?, and, What theoretical considerations can be made to conceptualize learning assessment?. The study is part of a research project of educational evaluation at the University of Camaguey, Cuba and is ascribed to the qualitative and interpretative approach. Documentary analysis is used, the triangulation of information and literature review, considering this last one in the period from 2009 to 2013. As a result of this study, the factors that influence the implementation of formative assessment and the theoretical premises that should be considered for development in the educational practice are specified. Finally it is assumed, conceptually, the learning assessment as a directional function of the teaching-learning process.
En el presente ensayo se presentan las reflexiones conceptuales, sobre la evaluación del aprendizaje, que toma como base las siguientes preguntas de investigación: ¿Qué se está investigando, en la actualidad, sobre la evaluación del aprendizaje?, ¿Cuáles son los enfoques teóricos que predominan en estas investigaciones?, ¿Cuáles son sus principales aportaciones? y ¿Qué reflexiones teóricas se pueden hacer para conceptualizar la evaluación del aprendizaje?. El estudio forma parte de un proyecto de investigación de evaluación educativa de la Universidad de Camagüey, Cuba y se adscribe al enfoque cualitativo e interpretativo. Se hace uso del análisis documental, la triangulación de la información y la revisión de literatura, considerando esta última en el período del 2009 al 2013. Como resultado del estudio se especifican los factores que influyen en la implementación de la evaluación formativa y las premisas teóricas que se deben tener en cuenta para su desarrollo en la práctica educativa. Finalmente se asume, conceptualmente, a la evaluación del aprendizaje como una función del sistema de dirección del proceso de enseñanza aprendizaje
Evaluation of the dietary intake of cadmium, lead and mercury and its relationship with bone health among postmenopausal women in Spain
ANTECEDENTES: Los metales pesados, como el plomo, el cadmio y el mercurio, se absorben a través de fuentes de alimentos contaminados y agua. Pocos estudios han investigado hasta qué punto los metales pesados de la dieta están asociados con una baja densidad mineral ósea.
OBJETIVOS: Nuestro objetivo fue caracterizar la ingesta alimentaria de metales pesados, plomo, cadmio y mercurio entre mujeres posmenopáusicas sanas y no fumadoras en España. Además, buscamos establecer una relación supuesta entre la salud ósea y la ingesta de estos metales pesados en esta población.
Diseño del estudio: La ingesta diaria de los metales pesados considerados para los diferentes grupos de alimentos se calculó tomando en cuenta el contenido y el consumo de alimentos en 281 mujeres posmenopáusicas. Las mediciones óseas se realizaron utilizando un ultrasonido óseo cuantitativo (QUS), una absorciometría de rayos X de energía dual (DXA) y una tomografía computada cuantitativa periférica (pQCT).
RESULTADOS: El promedio estimado de exposición a cadmio en la dieta entre las 281 mujeres estudiadas fue de 29.87 μg / día (20.41–41.04) y 3.03 μg / kg de peso corporal (b.w .; 2.17–4.40). La exposición al plomo en la dieta fue de 46.24 μg / día (38.11–54.77) y 4.87 μg / kg b.w. (4.00–6.14). La exposición estimada al mercurio en la dieta fue de 11.64 μg / día y 1.19 μg / kg b.w. (0.82–1.76). Los participantes se clasificaron según su ingesta de metales pesados (por encima o por debajo de las medianas respectivas). Después de un ajuste adicional para los factores de confusión potenciales, no se encontraron diferencias significativas en todos los parámetros medidos (p> 0.05).
CONCLUSIONES: No encontramos asociaciones entre la salud ósea y la ingesta dietética de tres metales pesados en mujeres posmenopáusicas. La ingesta dietética de los metales pesados medidos estuvo dentro de los valores recomendados.BACKGROUND: Heavy metals, such as lead, cadmium, and mercury, are absorbed through contaminated food sources and water. Few studies have investigated the extent to which dietary heavy metals are associated with low bone mineral density.
AIMS: We aimed to characterize the dietary intake of the heavy metals lead, cadmium and mercury among healthy, non-smoking postmenopausal women in Spain. Furthermore, we sought to establish a putative relationship between bone health and the intake of these heavy metals in this population.
STUDY DESIGN: The daily intake of the heavy metals considered for the different food groups was calculated by accounting for food content and consumption in 281 postmenopausal women. Bone measurements were performed using a Quantitative Bone Ultrasound (QUS), a Dual-Energy X-ray Absorptiometry (DXA) and a Peripheral Quantitative Computed Tomography (pQCT).
RESULTS: The average estimated dietary cadmium exposure among the 281 women studied was 29.87 μg/day (20.41–41.04) and 3.03 μg/kg body weight (b.w.; 2.17–4.40). Dietary lead exposure was 46.24 μg/day (38.11–54.77) and 4.87 μg/kg b.w. (4.00–6.14). The estimated dietary mercury exposure was 11.64 μg/day and 1.19 μg/kg b.w. (0.82–1.76). Participants were classified according to their heavy metal intake (above or below the respective medians). After further adjustment for potential confounding factors, no significant differences were found in all the measured parameters (p > 0.05).
CONCLUSIONS: We did not find associations between bone health and the dietary intake of three heavy metals in postmenopausal women. Route Analysis Through Filial Generations of Modern Varieties of Tomato (Solanum lycopersicum L.)
In Mexico, to reduce production costs, small producers use tomato seeds of advanced generations such as F2 and F3. The objectives of this study were to measure the efficiency of using advanced populations of some commercial tomato hybrids and to identify the most important yield components associated with yield. Seven saladette-type hybrids of indeterminate growth were evaluated, as well as their respective generations F2s and F3s under greenhouse conditions. The evaluation of the three generations was carried out during the 2013 agricultural cycle. Three harvests were made at 82, 94 and 136 days after transplanting. The experiment was carried out under a randomized complete block experimental design, with 4 replications and 10 individuals per repletion. The obtained results indicated that there were significant differences for few traits in F2 and F3. Path analysis showed that the total number of fruits had the highest direct and indirect effects on yield through generations. Based on our analysis, the advanced generations of ‘LORETO’, ‘CUAUHTÉMOC’ and ‘ESPARTACO’ could be used by the small growers since low values of inbreeding depression was observed in F2 and F3.Objective: To measure the efficiency of using advanced generations of some commercial tomato hybrids for small farmers and to identify the most important yield components associated with yield.

Limitations of the study/implications: The usefulness of advanced generations of tomato commercial hybrids would depend on the genetic background of the parental lines that take part in such as hybrids, as some hybrids would present high inbreeding depression depending on the genetic composition of their progenitors.
Findings/Conclusions: Advanced generations of ‘LORETO’, ‘CUAUHTÉMOC’ and ‘ESPARTACO’ could be used by the small growers since low values of inbreeding depression were observed in F2 and F3 familie

La investigación busca describir los significados psicológicos en relación con la felicidad, partiendo del sujeto común no experto en la materia. Para ello se utilizará la técnica de Redes Semánticas Naturales Modificadas (RSNM). Esto en el contexto de la UNAD Zona Sur, específicamente con el cuerpo docente de las diferentes escuelas académicas, excepto los que pertenecen al programa de psicología. Esta última decisión en razón a prevenir alguna influencia en los significados derivada de la formación disciplinar de tales docentes, téngase en cuenta que el tema de la felicidad ha sido objeto de estudio desde la psicología.
Este es un estudio sobre los significados psicológicos acerca de la felicidad en docentes de la UNAD Zona Sur. Esto a través de la técnica de RSNM, la cual se orienta justamente a dicho propósito. Esto en articulación con el proyecto “Felicidad: un estudio mixto desde la perspectiva de docentes y estudiantes de psicología de dos universidades de Latinoamérica”, en el que vienen trabajando docentes de la UNAD en convenio con la UNAM (Universidad Nacional Autónoma de México)
All-cause mortality in the cohorts of the Spanish AIDS Research Network (RIS) compared with the general population: 1997Ł2010
Abstract Background: Combination antiretroviral therapy (cART) has produced significant changes in mortality of HIVinfected persons. Our objective was to estimate mortality rates, standardized mortality ratios and excess mortality rates of cohorts of the AIDS Research Network (RIS) (CoRIS-MD and CoRIS) compared to the general population. Methods: We analysed data of CoRIS-MD and CoRIS cohorts from 1997 to 2010. We calculated: (i) all-cause mortality rates, (ii) standardized mortality ratio (SMR) and (iii) excess mortality rates for both cohort for 100 personyears (py) of follow-up, comparing all-cause mortality with that of the general population of similar age and gender. Results: Between 1997 and 2010, 8,214 HIV positive subjects were included, 2,453 (29.9%) in CoRIS-MD and 5,761 (70.1%) in CoRIS and 294 deaths were registered. All-cause mortality rate was 1.02 (95% CI 0.91-1.15) per 100 py, SMR was 6.8 (95% CI 5.9-7.9) and excess mortality rate was 0.8 (95% CI 0.7-0.9) per 100 py. Mortality was higher in patients with AIDS, hepatitis C virus (HCV) co-infection, and those from CoRIS-MD cohort (1997. Conclusion: Mortality among HIV-positive persons remains higher than that of the general population of similar age and sex, with significant differences depending on the history of AIDS or HCV coinfection
Immunoglobulin, glucocorticoid, or combination therapy for multisystem inflammatory syndrome in children: a propensity-weighted cohort study.
BACKGROUND: Multisystem inflammatory syndrome in children (MIS-C), a hyperinflammatory condition associated with SARS-CoV-2 infection, has emerged as a serious illness in children worldwide. Immunoglobulin or glucocorticoids, or both, are currently recommended treatments. METHODS: The Best Available Treatment Study evaluated immunomodulatory treatments for MIS-C in an international observational cohort. Analysis of the first 614 patients was previously reported. In this propensity-weighted cohort study, clinical and outcome data from children with suspected or proven MIS-C were collected onto a web-based Research Electronic Data Capture database. After excluding neonates and incomplete or duplicate records, inverse probability weighting was used to compare primary treatments with intravenous immunoglobulin, intravenous immunoglobulin plus glucocorticoids, or glucocorticoids alone, using intravenous immunoglobulin as the reference treatment. Primary outcomes were a composite of inotropic or ventilator support from the second day after treatment initiation, or death, and time to improvement on an ordinal clinical severity scale. Secondary outcomes included treatment escalation, clinical deterioration, fever, and coronary artery aneurysm occurrence and resolution. This study is registered with the ISRCTN registry, ISRCTN69546370. FINDINGS: We enrolled 2101 children (aged 0 months to 19 years) with clinically diagnosed MIS-C from 39 countries between June 14, 2020, and April 25, 2022, and, following exclusions, 2009 patients were included for analysis (median age 8·0 years [IQR 4·2-11·4], 1191 [59·3%] male and 818 [40·7%] female, and 825 [41·1%] White). 680 (33·8%) patients received primary treatment with intravenous immunoglobulin, 698 (34·7%) with intravenous immunoglobulin plus glucocorticoids, 487 (24·2%) with glucocorticoids alone; 59 (2·9%) patients received other combinations, including biologicals, and 85 (4·2%) patients received no immunomodulators. There were no significant differences between treatments for primary outcomes for the 1586 patients with complete baseline and outcome data that were considered for primary analysis. Adjusted odds ratios for ventilation, inotropic support, or death were 1·09 (95% CI 0·75-1·58; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids and 0·93 (0·58-1·47; corrected p value=1·00) for glucocorticoids alone, versus intravenous immunoglobulin alone. Adjusted average hazard ratios for time to improvement were 1·04 (95% CI 0·91-1·20; corrected p value=1·00) for intravenous immunoglobulin plus glucocorticoids, and 0·84 (0·70-1·00; corrected p value=0·22) for glucocorticoids alone, versus intravenous immunoglobulin alone. Treatment escalation was less frequent for intravenous immunoglobulin plus glucocorticoids (OR 0·15 [95% CI 0·11-0·20]; p<0·0001) and glucocorticoids alone (0·68 [0·50-0·93]; p=0·014) versus intravenous immunoglobulin alone. Persistent fever (from day 2 onward) was less common with intravenous immunoglobulin plus glucocorticoids compared with either intravenous immunoglobulin alone (OR 0·50 [95% CI 0·38-0·67]; p<0·0001) or glucocorticoids alone (0·63 [0·45-0·88]; p=0·0058). Coronary artery aneurysm occurrence and resolution did not differ significantly between treatment groups. INTERPRETATION: Recovery rates, including occurrence and resolution of coronary artery aneurysms, were similar for primary treatment with intravenous immunoglobulin when compared to glucocorticoids or intravenous immunoglobulin plus glucocorticoids. Initial treatment with glucocorticoids appears to be a safe alternative to immunoglobulin or combined therapy, and might be advantageous in view of the cost and limited availability of intravenous immunoglobulin in many countries. Taking the pulse of Earth's tropical forests using networks of highly distributed plots
Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science.
