    Effects of a soluble dietary fibre NUTRIOSE® on colonic fermentation and excretion rates in rats

    The resistant dextrin NUTRIOSE®, developed from starch, is expected to act as a prebiotic. The aim of this study was to determine the effects of NUTRIOSE® on cecal parameters, short-chain fatty acid (SCFA) concentrations, and fecal excretion in rats. In an initial experiment, twenty-four male Fischer F344 rats were randomly assigned to one of the following four treatments for 14 days: G0 (control diet), G2.5 (control diet + 2.5% of dextrin), G5 (control diet + 5% of dextrin), and G10 (control diet + 10% of dextrin). After 14 days, total cecal weight, cecal content, and cecal wall weight were significantly increased in G5 and G10 compared to G0. At the same time, cecal pH was significantly lower in G10 compared to G0. Total SCFA concentration was significantly higher in G10 than in G5, G2.5, and G0, and significantly higher in G5 than in G0. Acetate, butyrate, and propionate concentrations were significantly increased in G5 and G10 compared to the controls. In a second trial based on a similar design, eighteen male Fischer F344 rats were treated with a control diet supplemented with 5% of dextrin or 5% of fructo-oligosaccharide. The results obtained with NUTRIOSE® were similar to those obtained with the fructo-oligosaccharide. In a third experiment, two groups of 5 Fischer F344 rats were orally treated with 100 and 1,000 mg/kg NUTRIOSE®, respectively, and from 18% to 25% of the dextrin was excreted in the feces. The results of these three studies show that the consumption of NUTRIOSE®, by its effects on total cecal weight, cecal content, cecal wall weight, pH, and SCFA production, could induce healthy benefits since these effects are reported to be prebiotic effects

    V diplomski nalogi je obravnavano varovanje gradbene jame za objekt parkirno garažne hiše v Ljubljani. Varovalna konstrukcija je bila izvedena s sidranimi slopi po tehnologiji injektiranja pod visokimi pritiski (jet grouting). Sidranje smo izvedli z začasnimi geotehničnimi sidri v enem ali dveh nivojih. V nalogi je podan pregled postopkov za izvedbo jet grouting slopov in začasnih geotehničnih sider po veljavnih standardih. Pozornost je posvečena načinom izvedbe, ki so uveljavljeni pri nas, in problemom, ki se pojavljajo zaradi odstopanja od zhatev tehnične regulative ali nezmožnosti upoštevanja le-te. To je delno posledica zastarele opreme in utečenih postopkov izvajanja del. Dodatno pa na to vplivajo tudi zahteve po čim cenejši izvedbi gradbenih del. V nalogi so predstavljene izkušnje pridobljene med izvedbo varovalne konstrukcije gradbene jame, rezultati kontrolnih preiskav in problemi, ki so se pojavili med izvedbo del, ter rešitve teh problemov. V fazi izvedbe varovalne konstrukcije iz jet grouting slopov smo največ pozornosti posvetili tehnologiji enofaznega postopka injektiranja in njeni učinkovitosti v danih razmerah. Pri izvedbi geotehničnih sider pa smo največ pozornosti posvetili vgradnji in napenjanju preskusnih sider. Na gradbišču nastali problemi so lahko tudi posledica nepopolne gradbene dokumentacije, vključno s starimi in pomanjkljivimi načrti sosednjih objektov. V nalogi je prav tako obravnavano reševanje problemov, povezanih z nepričakovano sestavo temeljnih tal. Ta je pomembno vplivala na težave in zamude pri izvedbi in nemoteno napredovanje del.The subject of this thesis is the execution construction pit for multi-storey car-park building in Ljubljana. The supporting structure was made with high pressure grouting (e.q jet grouting), and anchored with temporary ground anchors in one or two levels. An overview of jet grouting and temporary ground anchors procedures accoding to current standards is shown. A special attention is put on the currently valid execution of works in our country. We primarily examined the problems occurring due to the deviation from technical regulations or inability to stick to them, which may be the consequence of either old equipment or sticking to generally accepted procedures of the execution, as well as requirements for a cheaper execution. The thesis presents the description of experiences gained during the execution works, the results of control examinations, the problems that occurred during the execution and appropriate solutions. In the phase of the jet-grouting-piles execution, which were used as a supporting structure, the attention was on the technology of the single fluid system and its effectiveness in the ground, while with ground anchors the most attention was paid to ground anchor installation and. The problems occurring on the construction site are often result of an incomplete project documentation, often involving the old and incomplete documentation of neighbouring buildings. The solution of issues in relation to the unexpected ground conditions, which significantly delayed the execution of works, is examined

    Research Note: Relative bioavailability of zinc in zinc hydroxychloride for chicks

    ABSTRACT: A chick assay was conducted to determine the effects of Zn source on performance and to establish a Zn relative bioavailability value (RBV) for a new source of Zn hydroxychloride. In the assay, 8-day-old chicks were fed a Zn-deficient soy protein concentrate diet supplemented with 0, 7, and 15 mg Zn/kg from feed grade ZnSO4 monohydrate for 14 d to establish a standard response curve. The same basal diet was supplemented with 3, 7, and 10 mg Zn/kg from a new Zn hydroxychloride (SAMZn). A second source of Zn hydroxychloride (IBZn) was supplemented at 10 mg Zn/kg as a direct comparison to the highest level of SAMZn. Weight gain increased (P 0.05) from chicks fed 15 mg Zn/kg from ZnSO4. Weight gain was not different (P > 0.05) when comparing the 2 sources of Zn hydroxychloride supplemented at 10 mg Zn/kg. Tibia ash Zn and total tibia Zn were increased (P 0.05) for chicks fed 10 mg Zn/kg from either source of Zn hydroxychloride. Multiple linear regression of total tibia Zn on supplemental Zn intake (R2 = 0.95) resulted in a RBV of 115% for SAMZn compared with ZnSO4 (set at 100%). The RBV of SAMZn was higher (P < 0.05) than ZnSO4. In conclusion, relative bioavailability of Zn (based on tibia Zn) in Zn hydroxychloride from SAMZn was higher than feed grade ZnSO4 based on multiple regression slope-ratio analysis and was similar to that of IBZn Zn hydroxychloride based on tibia Zn responses to 10 mg/kg supplemental dietary Zn

    Molecular ecological analysis of porcine ileal microbiota responses to antimicrobial growth promoters

    Cultivation-independent microbial molecular ecology approaches were used to examine the effects of antibiotic growth promoters on the pig ileal microbiota. Five-week-old barrows were fitted with a simple T-cannula at the distal ileum. Three diets meeting or exceeding the minimum nutrient requirements were fed for 5 wk and supplemented as follows: 1) negative control (no antibiotic; n = 5), 2) continuous tylosin administration (n = 5), and 3) an antibiotic rotation sequence (wk 1, chlorotetracycline sulfathiazole penicillin; wk 2, bacitracin and roxarsone; wk 3, lincomycin; wk 4, carbadox; wk 5, virginiamycin; n = 5). Ileal luminal contents were collected for DNA isolation at the end of each of the 5 wk of the testing period. The V3 region of 16S rDNA was amplified by PCR and analyzed via denaturing gradient gel electrophoresis (DGGE) and quantitative polymerase chain reaction (qPCR). Resulting PCR-DGGE band numbers (bacterial species) were counted, and the banding patterns analyzed by calculating Sorenson's pairwise similarity coefficients (C(S)), an index measuring bacterial species in common among samples. Band numbers and total bacterial DNA concentrations decreased (P < 0.05) temporally in antibiotic-treated pigs compared with controls. Comparisons between treatments yielded low intertreatment C(S) indices, indicating treatment-dependent alterations in banding patterns, whereas intratreatment comparisons revealed increased homogeneity in antibiotic-treated vs. control pigs. Sequence analysis of treatment-specific bands identified three Lactobacillus, one Streptococcus, and one Bacillus species that were diminished with antibiotic rotation treatment, whereas tylosin selected for the presence of L. gasseri. Lactobacillus-specific qPCR was performed and analyzed as a percentage of total bacteria to further evaluate the effects of antibiotic administration on this genus. Total bacteria were decreased (P < 0.05) by tylosin and rotation treatments, whereas the percentage of lactobacilli increased (P < 0.05) by d 14 and through d 28 in tylosin-treated pigs. The decrease in total bacteria by antibiotics may reduce host-related intestinal or immune responses, which would divert energy that could otherwise be used for growth. Conversely, the ability of tylosin to improve animal growth may relate to its apparent selection for lactobacilli, commensals known to competitively exclude potentially pathogenic species from colonizing the intestine

    Native Lantang and commercial Duroc pigs were used as animal models to evaluate the differences existing in dietary fiber utilization ability between breeds. Animals were fed the same diet from weaning (4 weeks) to 4 months of age. Neutral detergent fiber (NDF) from wheat bran (as substrate) and fecal samples from the two breeds (as inoculum) were used in an in vitro gas production trial. Results showed that cumulative and maximum gas productions were higher in inocula from Lantang than those from the Duroc breed (P < 0.05). The degradation capacity of NDF for microbiome from Lantang fecal samples were significantly higher compared to Duroc (P < 0.01). The total quantity of short-chain fatty acids and its constituents from the fermentation liquors were different between breeds, suggesting that the dynamic characteristics of fermentation differed between the two breeds. The PCR denaturing gradient gel electrophoresis fingerprint and cluster analysis demonstrated that microbial communities of the two breeds were separated into two clusters and the bacterial community structure of large intestine among the two breed of pigs was different. Our results concluded that Lantang had higher dietary fiber degradation capacity than Duroc pigs, and the higher degradation capacity for the former breed was due to differences in the inherent microbial community in their respective large intestines
