    Immunogenicity and safety of a quadrivalent high-dose inactivated influenza vaccine compared with a standard-dose quadrivalent influenza vaccine in healthy people aged 60 years or older: a randomized Phase III trial

    A quadrivalent high-dose inactivated influenza vaccine (IIV4-HD) is licensed for adults 6565 y of age based on immunogenicity and efficacy studies. However, IIV4-HD has not been evaluated in adults aged 60\u201364 y. This study compared immunogenicity and safety of IIV4-HD with a standard-dose quadrivalent influenza vaccine (IIV4-SD) in adults aged 6560 y. This Phase III, randomized, modified double-blind, active-controlled study enrolled 1,528 participants aged 6560 y, randomized 1:1 to a single injection of IIV4-HD or IIV4-SD. Hemagglutination inhibition (HAI) geometric mean titers (GMTs) were measured at baseline and D 28 and seroconversion assessed. Safety was described for 180 d after vaccination. The primary immunogenicity objective was superiority of IIV4-HD versus IIV4-SD, for all four influenza strains 28 d post vaccination in participants aged 60\u201364 and 6565 y. IIV4-HD induced a superior immune response versus IIV4-SD in terms of GMTs in participants aged 60\u201364 y and those aged 6565 y for all four influenza strains. IIV4-HD induced higher GMTs in those aged 60\u201364 y than those aged 6565 y. Seroconversion rates were higher for IIV4-HD versus IIV4-SD in each age-group for all influenza strains. Both vaccines were well tolerated in participants 6560 y of age, with no safety concerns identified. More solicited reactions were reported with IIV4-HD than with IIV4-SD. IIV4-HD provided superior immunogenicity versus IIV4-SD and was well tolerated in adults aged 6560 y. IIV4-HD is assumed to offer improved protection against influenza compared with IIV4-SD in adults aged 6560 y, as was previously assessed for adults aged 6565 y

    A multi-metric approach to investigate the effects of weather conditions on the demographic of a terrestrial mammal, the European badger (Meles meles)

    Models capturing the full effects of weather conditions on animal populations are scarce. Here we decompose yearly temperature and rainfall into mean trends, yearly amplitude of change and residual variation, using daily records. We establish from multi-model inference procedures, based on 1125 life histories (from 1987 to 2008), that European badger (Meles meles) annual mortality and recruitment rates respond to changes in mean trends and to variability in proximate weather components. Variation in mean rainfall was by far the most influential predictor in our analysis. Juvenile survival and recruitment rates were highest at intermediate levels of mean rainfall, whereas low adult survival rates were associated with only the driest, and not the wettest, years. Both juvenile and adult survival rates also exhibited a range of tolerance for residual standard deviation around daily predicted temperature values, beyond which survival rates declined. Life-history parameters, annual routines and adaptive behavioural responses, which define the badgers’ climatic niche, thus appear to be predicated upon a bounded range of climatic conditions, which support optimal survival and recruitment dynamics. That variability in weather conditions is influential, in combination with mean climatic trends, on the vital rates of a generalist, wide ranging and K-selected medium-sized carnivore, has major implications for evolutionary ecology and conservation

    Ovarian damage from chemotherapy and current approaches to its protection

    BACKGROUND: Anti-cancer therapy is often a cause of premature ovarian insufficiency and infertility since the ovarian follicle reserve is extremely sensitive to the effects of chemotherapy and radiotherapy. While oocyte, embryo and ovarian cortex cryopreservation can help some women with cancer-induced infertility achieve pregnancy, the development of effective methods to protect ovarian function during chemotherapy would be a significant advantage.OBJECTIVE AND RATIONALE: This paper critically discusses the different damaging effects of the most common chemotherapeutic compounds on the ovary, in particular, the ovarian follicles and the molecular pathways that lead to that damage. The mechanisms through which fertility-protective agents might prevent chemotherapy drug-induced follicle loss are then reviewed.SEARCH METHODS: Articles published in English were searched on PubMed up to March 2019 using the following terms: ovary, fertility preservation, chemotherapy, follicle death, adjuvant therapy, cyclophosphamide, cisplatin, doxorubicin. Inclusion and exclusion criteria were applied to the analysis of the protective agents.OUTCOMES: Recent studies reveal how chemotherapeutic drugs can affect the different cellular components of the ovary, causing rapid depletion of the ovarian follicular reserve. The three most commonly used drugs, cyclophosphamide, cisplatin and doxorubicin, cause premature ovarian insufficiency by inducing death and/or accelerated activation of primordial follicles and increased atresia of growing follicles. They also cause an increase in damage to blood vessels and the stromal compartment and increment inflammation. In the past 20 years, many compounds have been investigated as potential protective agents to counteract these adverse effects. The interactions of recently described fertility-protective agents with these damage pathways are discussed.WIDER IMPLICATIONS: Understanding the mechanisms underlying the action of chemotherapy compounds on the various components of the ovary is essential for the development of efficient and targeted pharmacological therapies that could protect and prolong female fertility. While there are increasing preclinical investigations of potential fertility preserving adjuvants, there remains a lack of approaches that are being developed and tested clinically

    Assessing motivations for carbon offsetting by environmentally certified tourism enterprises

    This paper presents survey results on tourism operator preferences and motivations for carbon offset options by environmentally certified tourism enterprises in Queensland, Australia. It found that less than a third are involved in carbon offset programmes, whereas just over a third plan to implement offsetting in the near future. Preferred offset projects are regional tree planting or renewable energy. Motivations for offsetting are related to environmental concern, commercial benefits of climate-friendly tourism, and financially supporting offset projects. This study found motivations for offsetting by larger tourism enterprises aligning with the more general business motivations and drivers for adopting corporate social and environmental responsibility practices. However, the extra cost and uncertainty about carbon issues limited the uptake of offsetting by smaller tourism operators

Open access repositories may make research available even prior to a peer-reviewed printed publication, as well as unpublished material. Here lies the challenge to discern between refereed (post-print) and non-refereed (or preprint) material

    Drivers of foot and mouth disease in cattle at wild/domestic interface: insights from farmers, buffalo and lions

    Humans live increasingly in the proximity of natural areas, leading to increased interactions between people, their livestock and wildlife. Aim: We explored the role of these interactions in the risk of disease transmission (foot-and-mouth disease (FMD)) between cattle and the African buffalo (the maintenance host) and how a top predator, the lion, may modulate these interactions. Location: The interface of Hwange National Park (HNP) and surrounding communal lands, Zimbabwe Method: We combined a longitudinal serological cattle survey for FMD, GPS-collar data and cattle owners’ interviews during 4 seasons in 2010-11. Results: Overall FMD incidence in cattle was low, but showed a peak during the rainy season. The incidence dynamic was significantly explained by cattle incursion into the protected area ( i.e. buffer zone of 3km inside HNP) and not by contacts with buffalo or contacts among cattle. These results suggest that FMD virus either survives in the environment or is transmitted by other ungulate groups or species. The analysis of incursion frequency in the buffer suggests that (i) buffalo and cattle are avoiding each other up to two months after one species track and that (ii) lions make frequent incursions in the buffer few days to few weeks after buffalo had used it, whereas buffalo did not use areas occupied by lions. Lions could thus reduce the spatio-temporal overlap between cattle and buffalo in the interface, which could contribute to the low level of FMD incidence. Main conclusions: During the rainy season, traditional herding practices push cattle away from growing crops near villages into the HNP but not during the dry season, suggesting that cattle owners may decide to rely on lower quality resources in the communal land in the dry season to avoid the risks of infection and/or predation in the HNP.
