    Survey of Microbial Contamination Status in Terminal Equipment Direct Drinking Water Sampled Yantai City

    To investigate terminal equipment direct drinking water quality and sanitation status that sampled from community, school and home, and to provide evidence for the setting of standard, department supervision and consumer consumption. 232 samples were randomly sampled from community, school and home during June to November of 2019. The aerobic plate count, coliforms and Pseudomonas aeruginosa were detected in accordance with standard operating procedure provided by manual of China national food contamination and harmful factors risk monitoring 2019. The positive detection rates of aerobic plate count. Coliforms and Pseudomonas aeruginosa in 232 samples were 84.05%, 3.02% and 7.33%, respectively, the aerobic plate count was main contamination factor of fine drinking water, while Pseudomonas aeruginosa was the main pathogenic bacteria. The overall exceeded the standard rate of terminal equipment fine drinking water was 9.48%. The exceeded the standard rate ranked in order of high was the family, schools and community, compared the sampling link, which was 12.68%、8.97% and 7.23%, respectively, and was not statistically significant between the groups ( χ2=1.36 , P>0.05). There was not seasonal variation regularity of the detection rate to the aerobic plate count, coliforms and Pseudomonas aeruginosa. The was different of exceeded the standard rate for every months, which was November > June > September > October > August > July, in order from higher to low, the exceeded the standard rate was 15.00%, 13.89%, 11.76%, 6.25%, 5.88% and 5.00%, respectively. There was not statistically significant between the groups( χ2=4.47, P>0.05). There were multiple contaminations of the aerobic plate count, coliforms and Pseudomonas aeruginosa in a single sample.     Improving yield and water use efficiency of apple trees through intercrop-mulch of crown vetch (Coronilla varia L.) combined with different fertilizer treatments in the Loess Plateau

    Improving water use efficiency (WUE) and soil fertility is relevant for apple production in drylands. The effects of intercrop-mulch (IM) of crown vetch (Coronilla varia L.) combined with different fertilizer treatments on WUE of apple trees and soil fertility of apple orchards were assessed over three years (2011, 2013 and 2014). A split-plot design was adopted, in which the main treatments were IM and no intercrop-mulch (NIM). Five sub-treatments were established: no fertilization (CK); nitrogen and phosphorus fertilizer (NP); manure (M); N, P and potassium fertilizer (NPK); and NPK fertilizer combined with manure (NPKM). Due to mowing and mulching each month during July–September, the evapotranspiration for IM was 17.3% lower than that of NIM in the dry year of 2013. Additionally, the soil water storage of NPKM treatment was higher than that of CK during the experimental period. Thus, single fruit weight and fruit number per tree increased with IM and NPKM application. Moreover, applying NPKM with IM resulted in the highest yield (on average of three years), which was 73.25% and 130.51% greater than that of CK in IM and NIM, respectively. The WUE of NPKM combined with IM was also the highest in 2013 and 2014 (47.69 and 56.95% greater than applying IM alone). In addition, due to application of IM combined with NPKM, soil organic matter was increased by 25.8% compared with that of CK (in NIM). Additionally, application of IM combined with NPKM obtained more economic net return, compared to other combinations. Therefore, applying NPKM with IM is recommended for improving apple production in this rain-fed agricultural area

    Optimizing Methanol Blending Performance of Electronically Controlled Diesel Engines through Fuzzy Analysis

    This paper presents a comprehensive optimization approach for enhancing the performance of a methanol/diesel Exhaust Gas Recirculation (EGR) engine. Initially, a hybrid fuel engine combustion chamber model was developed using AVL-FIRE software, and the simulated results were compared with the values obtained from bench tests. An orthogonal experimental design was employed to optimize five key factors, namely methanol blending ratio, EGR rate, injection advance angle, intake pressure, and intake temperature. Evaluation indexes were established, with indicated power and NO emissions assigned weights of 0.35 and 0.65, respectively. The optimal parameter combinations were determined as follows: methanol blending ratio (a1=20%), EGR rate (a2=12.5%), injection advance angle (a3=16.6°CA), intake temperature (a4 = 315.15 K), and intake pressure (a5=0.173 MPa). The indicated power of the optimized configuration reached 47.8 kW, slightly lower than the original 55 kW, while the NO emission mass fraction decreased to 1.9×10-4%, representing a significant reduction of 77.6% compared to the original value of 8.5×10-4%. This optimization methodology demonstrates the effective reduction of NO emissions without compromising power performance in methanol/diesel EGR engines

    Cis-Effects Condition the Induction of a Major Unfolded Protein Response Factor, ZmbZIP60, in Response to Heat Stress in Maize

    Adverse environmental conditions such as heat and salt stress create endoplasmic reticulum (ER) stress in maize and set off the unfolded protein response (UPR). A key feature of the UPR is the upregulation of ZmbZIP60 and the splicing of its messenger RNA. We conducted an association analysis of a recombinant inbred line (RIL) derived from a cross of a tropical founder line, CML52 with a standard temperate line, B73. We found a major QTL conditioning heat-induced ZmbZIP60 expression located cis to the gene. Based on the premise that the QTL might be associated with the ZmbZIP60 promoter, we evaluated various maize inbred lines for their ability to upregulate the expression of ZmbZIP60 in response to heat stress. In general, tropical lines with promoter regions similar to CML52 were more robust in upregulating ZmbZIP60 in response to heat stress. This finding was confirmed by comparing the strength of the B73 and CML52 ZmbZIP60 promoters in transient maize protoplast assays. We concluded that the upstream region of ZmbZIP60 is important in conditioning the response to heat stress and was under selection in maize when adapted to different environments.Summary: Heat stress has large negative effects on maize grain yield. Heat stress creates ER stress in maize and sets off the UPR. We searched for factors conditioning heat induction of the UPR in maize seedlings by conducting an association analysis based on the upregulation of unspliced and spliced forms of ZmbZIP60 mRNA (ZmbZIP60u and ZmbZIP60s, respectively). ZmbZIP60u was upregulated more robustly by heat stress in the tropical maize line, CML52, than in B73, and a major QTL derived from the analysis of RILs from a cross of these two lines mapped in the vicinity of ZmbZIP60. We conducted a cis/trans test to determine whether the QTL was acting as a cis regulatory element or in trans, as might be expected for a transcription factor. We found that the QTL was acting in cis, likely involving the ZmbZIP60 promoter. ZmbZIP60 promoters in other temperate and tropical lines similar to CML52 showed enhanced expression of ZmbZIP60u by heat. The contribution of the CML52 promoter to heat induction of ZmbZIP60 was confirmed by analyzing the CML52 and B73 promoters linked to a luciferase reporter and assayed in heat-treated maize protoplasts

    MicroRNA-197 Promotes Metastasis of Hepatocellular Carcinoma by Activating Wnt/β-Catenin Signaling

    Background/Aims: MicroRNA-197 (miR-197) has been shown to play roles in epithelialmesenchymal transition (EMT) and metastasis. The Wnt/β-catenin pathway is associated with EMT, but whether miR-197 regulatesWnt/β-catenin remains unclear. This study was to demonstrate the role of miR-197 on the Wnt/β-catenin pathway in hepatocellular carcinoma (HCC). Methods: Quantitative reverse transcription polymerase chain reaction (qRT-PCR) was used to detect the expression of miR-197 in 105 HCC specimens and 15 HCC cell lines. We tested the predicted target gene of miR-197 using a genetic report system. The role of miR-197 in HCC cell invasion and migration (wound healingand cell invasion and migrationby Transwell assays) and in an HCC xenograft modelwas analyzed. Results: Using a miRNA microarray analysis of HCC specimens and compared with non-metastatic HCC, miR-197 was identified as one of the most upregulated miRNAs in metastatic HCC. miR-197 expression was positively associated with the invasiveness of HCC cell lines. Metastatic HCC cells with high miR-197 expression had Wnt/β-catenin signaling activation. High levels of miR-197 expression also promoted EMT and invasionHCC cells in vitro and in vivo. miR-197 directly targeted Axin-2, Naked cuticle 1 (NKD1), and Dickkopf-related protein 2 (DKK2), leading to inhibition of Wnt/β-catenin signaling. High miR-197 expression was found in HCC specimens from patients with portal vein metastasis;high miR-197 expression correlated to the expression of Axin2, NKD1, and DKK2. Conclusion: miR-197 promotes HCC invasion and metastasis by activating Wnt/β-catenin signaling. miR-197 could possibly be used as a prognostic marker and therapeutic target for HCC

    Cytotoxic T Lymphocyte Trafficking and Survival in an Augmented Fibrin Matrix Carrier

    Cell-based therapies have intriguing potential for the treatment of a variety of neurological disorders. One such example is genetically engineered cytotoxic T lymphocytes (CTLs) that are being investigated in brain tumor clinical trials. The development of methods for CTL delivery is critical to their use in the laboratory and clinical setting. In our study, we determined whether CTLs can migrate through fibrin matrices and if their migration, survival, and function could be modulated by adding chemokines to the matrix. Our results indicated that CTLs can freely migrate through fibrin matrices. As expected, the addition of the monocyte chemotactic protein-1 (MCP-1), also known as chemokine C-C motif ligand 2 (CCL2), to the surrounding media increased egress of the CTLs out of the fibrin clot. Interleukin (IL) -2 and/or IL-15 embedded in the matrix enhanced T cell survival and further promoted T cell migration. The interleukin-13 receptor alpha 2 specific (IL-13R alpha2) T cells that traveled out of the fibrin clot retained the capacity to kill U251 glioma cells. In summary, CTLs can survive and migrate robustly in fibrin matrices. These processes can be influenced by modification of matrix constituents. We conclude that fibrin matrices may be suitable T cell carriers and can be used to facilitate understanding of T cell interaction with the surrounding microenvironment

    Differential cross section measurements for the production of a W boson in association with jets in proton–proton collisions at √s = 7 TeV

    Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript −1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States.     Juxtaposing BTE and ATE – on the role of the European insurance industry in funding civil litigation

    One of the ways in which legal services are financed, and indeed shaped, is through private insurance arrangement. Two contrasting types of legal expenses insurance contracts (LEI) seem to dominate in Europe: before the event (BTE) and after the event (ATE) legal expenses insurance. Notwithstanding institutional differences between different legal systems, BTE and ATE insurance arrangements may be instrumental if government policy is geared towards strengthening a market-oriented system of financing access to justice for individuals and business. At the same time, emphasizing the role of a private industry as a keeper of the gates to justice raises issues of accountability and transparency, not readily reconcilable with demands of competition. Moreover, multiple actors (clients, lawyers, courts, insurers) are involved, causing behavioural dynamics which are not easily predicted or influenced. Against this background, this paper looks into BTE and ATE arrangements by analysing the particularities of BTE and ATE arrangements currently available in some European jurisdictions and by painting a picture of their respective markets and legal contexts. This allows for some reflection on the performance of BTE and ATE providers as both financiers and keepers. Two issues emerge from the analysis that are worthy of some further reflection. Firstly, there is the problematic long-term sustainability of some ATE products. Secondly, the challenges faced by policymakers that would like to nudge consumers into voluntarily taking out BTE LEI
