    A Transition State Theory for Calculating Hopping Times and Diffusion in Highly Confined Fluids

    Monte Carlo simulation is used to study the dynamical crossover from single file diffusion to normal diffusion in fluids confined to narrow channels. We show that the long time diffusion coefficients for a series of systems involving hard and soft interaction potentials can be described in terms of a hopping time that measures the time it takes for a particle to escape the cage formed by its neighbors in the pore. Free energy barriers for the particle hopping process are calculated and used to show that transition state theory effectively describes the hopping time for all the systems studied, over a range of pore diameters. Our work suggests that the combination of hopping times and transition state theory offers a useful and general framework to describe the dynamics of these highly confined fluids.Comment: 6 figure

    Indigenous Food Practices of the Dambana Vedda Community in Sri Lanka: Past and Present Scenario

    Veddas are considered to be the indigenous community of Sri Lanka. A century ago, the Veddas had scattered across the Eastern Province, North-Central and Uva Provinces. Presently, their main settlement is confined to Dambana in the Badulla district. They inherit unique indigenous knowledge from their ancestors to hunt animals, gather forest goods, collect honey and dig yams to fulfill their food needs as ‘Forest dwellers’. Therefore, they had simple and efficient indigenous techniques for gathering and processing food. The main objective of the present study was to identify and document the past indigenous food practices and current food practices of the Dambana Vedda community. Focus group discussions were carried out with the different age categories of the Vedda community at Dambana to collect information. The indigenous lifestyle of the Vedda community was affected by modernization and civilization. Wild Veddas who lived in the forest have transmitted to village Veddas in colonies with modified food culture. Their indigenous food culture is mixed with the neighboring Sinhalese and Tamil communities. However, with forest reserves restricted, the Veddas are still engaging in a questionable struggle to protect their indigenous tradition and culture. Consequently, they have consumed indigenous foods rarely and novel food varieties become the daily diets of Veddas.     Optimisation of enzymatic hydrolysis for concentration of squalene in palm fatty acid distillate

    BACKGROUND: Squalene was concentrated from palm fatty acid distillate (PFAD) in this study using commercial immobilised Candida antarctica lipase (Novozyme 435®). The PFAD was neutralised (NPFAD) using an alkali to liberate the free fatty acids and then hydrolysed at 65 ± 1 °C. The enzymatic hydrolysis on NPFAD was optimised using response surface methodology (RSM) before being neutralised again to obtain a concentrated squalene fraction. RESULTS: A five-level, three-factor central composite rotatable design was adopted to evaluate the effects of the enzymatic hydrolysis parameters reaction time (4-12 h), water content (50-70% w/w) and enzyme concentration (1.5-3.5% w/w) on the percentage yield of squalene concentration. The optimal reaction parameters for maximum yield of squalene concentration were identified from the respective contour plots. The optimal enzymatic hydrolysis conditions were a reaction time of 7.05 h, a water content of 61.40% w/w and an enzyme concentration of 2.23% w/w. CONCLUSION: RSM was used to determine the optimal conditions for enzymatic hydrolysis of NPFAD with C. antarctica lipase for maximum recovery of squalene which could be implemented on an industrial scale

    The role of salt bridges, charge density, and subunit flexibility in determining disassembly routes of protein complexes

    Mass spectrometry can be used to characterize multiprotein complexes, defining their subunit stoichiometry and composition following solution disruption and collision-induced dissociation (CID). While CID of protein complexes in the gas phase typically results in the dissociation of unfolded subunits, a second atypical route is possible wherein compact subunits or subcomplexes are ejected without unfolding. Because tertiary structure and subunit interactions may be retained, this is the preferred route for structural investigations. How can we influence which pathway is adopted? By studying properties of a series of homomeric and heteromeric protein complexes and varying their overall charge in solution, we found that low subunit flexibility, higher charge densities, fewer salt bridges, and smaller interfaces are likely to be involved in promoting dissociation routes without unfolding. Manipulating the charge on a protein complex therefore enables us to direct dissociation through structurally informative pathways that mimic those followed in solution

    Managing housing needs in post conflict housing reconstruction in Sri Lanka: gaps versus recommendations

    Addressing housing needs in post conflict housing reconstruction leads to successful housing reconstruction. As part of a study of investigating how the housing needs can be effectively addressed in post conflict housing reconstruction, this paper identifies the gaps in managing housing needs in post conflict housing reconstruction within the context of Sri Lanka and presents the recommendations to minimise such gaps. Data was collected through un-structured interviews conducted with 37 participants, comprising policy makers, practitioners, academics and beneficiaries who engaged in post conflict housing reconstruction in Sri Lanka. Gaps were mainly found in conflict sensitivity, measures related to physical housing, performance of implementing agencies, policy and practice issues. On the job training, application of ‘do no harm’ principles, enhanced beneficiary participation, enhanced accountability, effective monitoring, enhanced knowledge sharing, adequate drinking water facilities, irrigation development and initiatives for material manufacturing were suggested as recommendations to minimise these gaps. Identification of gaps in managing housing needs in post conflict housing reconstruction and recommendations to minimise them inform policy makers to address the housing needs effectively through incorporating these aspects into the related policies. This in turn enhances the sustainability in housing development after conflicts

    Rapeseed napin and cruciferin are readily digested by poultry

    Rapeseed proteins have been considered as being poorly digestible in the gut of non-ruminants. The aim of the study was to assess the digestibility of napin and cruciferin in ileal digesta of broiler chickens, testing sixteen samples of rapeseed co-products with protein levels ranging from 293 g/kg to 560 g/kg dry matter. Each sample was included into a semi-synthetic diet at a rate of 500 g/kg and evaluated with broiler chickens in a randomised design. Dietary and ileal digesta proteins were extracted and identified by gel-based liquid chromatography–tandem mass spectrometry (LC-MS/MS). Three isomers of napin (a 2S albumin) and nine cruciferins (an 11S globulin) were identified in the rapeseed co-products, whereas six endogenous enzymes such as trypsin (I-P1, II-P29), chymotrypsin (elastase and precursor), carboxypeptidase B and α-amylase were found in the ileal digesta. It is concluded that as none of the rapeseed proteins were detected in the ileal digesta, rapeseed proteins can be readily digested by broiler chickens, irrespective of the protein content in the diet

    Alfalfa and flax sprouts supplementation enriches the content of bioactive compounds and lowers the cholesterol in hen egg

    Abstract The effect of dietary supplementation with flax and alfalfa sprouts (40 g/d) on bioactive compounds and cholesterol contents of hen's egg was examined. Thirty White Leghorn hens, 26 weeks of age, were fed, for 66 days, three diets that included control (standard diet – C), standard diet + alfalfa sprouts (A), and standard diet + flax sprouts (F). Productive performance of hens was recorded daily. The cholesterol content of plasma and yolk, and the presence of bioactive compounds in the egg, were also analysed. Supplementation of flax and alfalfa sprouts reduced plasma and egg cholesterol probably due to the synergy between different compounds of the sprouts (polyunsaturated fatty acids - PUFAs, lignans, isoflavones and sterols). Eggs from A and F groups also had higher contents of n-3 PUFA, vitamins (α-tocopherol, α-, γ-tocotrienol, retinol), carotenes (β-carotene, lutein, zeaxanthin) and phytoestrogens (daidzein, equol, isolariciresinol) than eggs from the C group

    Determination of Sinapic Acid Derivatives in Canola Extracts Using High-Performance Liquid Chromatography

    A high-performance liquid chromatographic (HPLC) method with diode array detection (DAD) was used to determine the total phenolics, including sinapic acid derivatives in canola. Ten Western Canadian canola seeds, six other commodity canola seeds, their corresponding press cakes and meals were analyzed. Seeds of European 00 rapeseed and Brassica Juncea (Indian mustard) were included for comparison. Phenolic compounds were separated using a gradient elution system of water–methanol-ο-phosphoric acid solution with a flow rate of 0.8 ml/min. In addition to sinapine (SP) and sinapic acid (SA), sinapoyl glucose (SG) is reported in the methanolic extracts. The detection and quantification limits of these compounds were 0.20–0.40 and 0.50–0.80 μg/ml, respectively with recovery values over 98.0%. The content of total phenolics, SP, SA and SG in canola extracts ranged from 9.16 to 16.13, 6.39 to 12.28, 0.11 to 0.59 and 1.36 to 7.50 mg/g, respectively with significant differences among varieties

    Gamma Radiation Effects on Peanut Skin Antioxidants

    Peanut skin, which is removed in the peanut blanching process, is rich in bioactive compounds with antioxidant properties. The aims of this study were to measure bioactive compounds in peanut skins and evaluate the effect of gamma radiation on their antioxidant activity. Peanut skin samples were treated with 0.0, 5.0, 7.5, or 10.0 kGy gamma rays. Total phenolics, condensed tannins, total flavonoids, and antioxidant activity were evaluated. Extracts obtained from the peanut skins were added to refined-bleached-deodorized (RBD) soybean oil. The oxidative stability of the oil samples was determined using the Oil Stability Index method and compared to a control and synthetic antioxidants (100 mg/kg BHT and 200 mg/kg TBHQ). Gamma radiation changed total phenolic content, total condensed tannins, total flavonoid content, and the antioxidant activity. All extracts, gamma irradiated or not, presented increasing induction period (h), measured by the Oil Stability Index method, when compared with the control. Antioxidant activity of the peanut skins was higher than BHT. The present study confirmed that gamma radiation did not affect the peanut skin extracts’ antioxidative properties when added to soybean oil
