    Reclaiming heritage: colourization, culture wars and the politics of nostalgia

    This article considers the discursive continuities between a specifically liberal defence of cultural patrimony, evident in the debate over film colourization, and the culture war critique associated with neo-conservatism. It examines how a rhetoric of nostalgia, linked to particular ideas of authenticity,canonicity and tradition,has been mobilized by the right and the left in attempts to stabilize the confguration and perceived transmission of American cultural identity. While different in scale, colourization and multiculturalism were seen to create respective (postmodern) barbarisms against which defenders of culture, heritage and good taste could unite. I argue that in its defence of the ‘classic’ work of art, together with principles of aesthetic distinction and the value of cultural inheritance,the anti-colourization lobby helped enrich and legitimize a discourse of tradition that, at the end of the 1980s, was beginning to reverberate powerfully in the conservative challenge to a ‘crisis’ within higher education and the humanities. This article attempts to complicate the contemporary politics of nostalgia, showing how a defence of cultural patrimony has distinguished major and minor culture wars, engaging left and right quite differently but with similar presuppositions

    Effect of synbiotic supplementation in children and adolescents with cystic fibrosis: a randomized controlled clinical trial

    BACKGROUND/OBJECTIVES:Cystic fibrosis (CF) is characterized by excessive activation of immune processes. The aim of this study was to evaluate the effect of synbiotic supplementation on the inflammatory response in children/adolescents with CF. SUBJECTS/METHODS:A randomized, placebo-controlled, double-blind, clinical-trial was conducted with control group (CG, n = 17), placebo-CF-group (PCFG, n = 19), synbiotic CF-group (SCFG, n = 22), PCFG negative (n = 8) and positive (n = 11) bacteriology, and SCFG negative (n = 12) and positive (n = 10) bacteriology. Markers of lung function (FEV1), nutritional status [body mass index-for age (BMI/A), height-for-age (H/A), weight-for-age (W/A), upper-arm fat area (UFA), upper-arm muscle area (UMA), body fat (%BF)], and inflammation [interleukin (IL)-12, tumor necrosis factor-alpha (TNF-α), IL-10, IL-6, IL-1β, IL-8, myeloperoxidase (MPO), nitric oxide metabolites (NOx)] were evaluated before and after 90-day of supplementation with a synbiotic. RESULTS:No significance difference was found between the baseline and end evaluations of FEV1 and nutricional status markers. A significant interaction (time vs. group) was found for IL-12 (p = 0.010) and myeloperoxidase (p = 0.036) between PCFG and SCFG, however, the difference was not maintained after assessing the groups individually. NOx diminished significantly after supplementation in the SCFG (p = 0.030). In the SCFG with positive bacteriology, reductions were found in IL-6 (p = 0.033) and IL-8 (p = 0.009) after supplementation. CONCLUSIONS: Synbiotic supplementation shown promise at diminishing the pro-inflammatory markers IL-6, IL-8 in the SCFG with positive bacteriology and NOx in the SCFG in children/adolescents with CF

    Liposome-Mediated Cellular Delivery of Active gp91phox

    International audienceBACKGROUND: Gp91(phox) is a transmembrane protein and the catalytic core of the NADPH oxidase complex of neutrophils. Lack of this protein causes chronic granulomatous disease (CGD), a rare genetic disorder characterized by severe and recurrent infections due to the incapacity of phagocytes to kill microorganisms. METHODOLOGY: Here we optimize a prokaryotic cell-free expression system to produce integral mammalian membrane proteins. CONCLUSIONS: Using this system, we over-express truncated forms of the gp91(phox) protein under soluble form in the presence of detergents or lipids resulting in active proteins with a "native-like" conformation. All the proteins exhibit diaphorase activity in the presence of cytosolic factors (p67(phox), p47(phox), p40(phox) and Rac) and arachidonic acid. We also produce proteoliposomes containing gp91(phox) protein and demonstrate that these proteins exhibit activities similar to their cellular counterpart. The proteoliposomes induce rapid cellular delivery and relocation of recombinant gp91(phox) proteins to the plasma membrane. Our data support the concept of cell-free expression technology for producing recombinant proteoliposomes and their use for functional and structural studies or protein therapy by complementing deficient cells in gp91(phox) protein

    Le croisement des sciences agronomiques, de l’écologie appliquée à la gestion des agroécosystèmes,et des sciences humaines et sociales, qu’implique la transition agroécologique, pose de nouveaux défis pour répondre aux enjeux agricoles: intégrer des connaissances de différentes disciplines et produites à différentes échelles d’organisation pour agir en situation; développer des cadres d’analyse et démarches intégrant la diversité de situations à gérer par les acteurs et permettant de construire des réponses adaptées à chaque situation; et concevoir et mettre en œuvre des pratiques d’enseignement et d’apprentissage, qui dotent les apprenants de capacités à penser leur action en contexte, en mobilisant des savoirs et savoir-faire multiples en termes de contenus disciplinaires et des savoir-être pour construire des solutions avec une diversité d’acteurs. Dans le champ de la formation, ces défis nécessitent dès lors de revisiter les contenus des enseignements dispensés, les modalités pédagogiques et les dispositifs de formation existants, de manière à appréhender au mieux la complexité des processus à l’œuvre. Pour autant, peu de travaux s’attardent sur les modalités pratiques de ce changement et de ses implications, alors même que de nombreuses initiatives en matière de pédagogie et d’agroécologie se développent ces dernières années. L’objectif de ce séminaire est de promouvoir une information partagée et l’échange d’expériences pour répondre aux enjeux posés par l’agroécologie dans la formation (transversalité, pluridisciplinarité, approche systémique, pédagogies actives). Ces enjeux peuvent se décliner suivant plusieurs entrées : les thématiques enseignées (agriculture, élevage, territoire, alimentation, ...); les pratiques et les dispositifs pédagogiques mis en œuvre pour aborder ces questions (enseignement numérique, dispositifs expérimentaux, projets professionnels, référentiels, ...);les publics d’apprenants: élèves, étudiants, professionnels, ..

    Mycobacterium tuberculosis Exploits Asparagine to Assimilate Nitrogen and Resist Acid Stress during Infection

    Mycobacterium tuberculosis is an intracellular pathogen. Within macrophages, M. tuberculosis thrives in a specialized membrane-bound vacuole, the phagosome, whose pH is slightly acidic, and where access to nutrients is limited. Understanding how the bacillus extracts and incorporates nutrients from its host may help develop novel strategies to combat tuberculosis. Here we show that M. tuberculosis employs the asparagine transporter AnsP2 and the secreted asparaginase AnsA to assimilate nitrogen and resist acid stress through asparagine hydrolysis and ammonia release. While the role of AnsP2 is partially spared by yet to be identified transporter(s), that of AnsA is crucial in both phagosome acidification arrest and intracellular replication, as an M. tuberculosis mutant lacking this asparaginase is ultimately attenuated in macrophages and in mice. Our study provides yet another example of the intimate link between physiology and virulence in the tubercle bacillus, and identifies a novel pathway to be targeted for therapeutic purposes. © 2014 Gouzy et al

    Molecular mechanisms of cell death: recommendations of the Nomenclature Committee on Cell Death 2018.

    Over the past decade, the Nomenclature Committee on Cell Death (NCCD) has formulated guidelines for the definition and interpretation of cell death from morphological, biochemical, and functional perspectives. Since the field continues to expand and novel mechanisms that orchestrate multiple cell death pathways are unveiled, we propose an updated classification of cell death subroutines focusing on mechanistic and essential (as opposed to correlative and dispensable) aspects of the process. As we provide molecularly oriented definitions of terms including intrinsic apoptosis, extrinsic apoptosis, mitochondrial permeability transition (MPT)-driven necrosis, necroptosis, ferroptosis, pyroptosis, parthanatos, entotic cell death, NETotic cell death, lysosome-dependent cell death, autophagy-dependent cell death, immunogenic cell death, cellular senescence, and mitotic catastrophe, we discuss the utility of neologisms that refer to highly specialized instances of these processes. The mission of the NCCD is to provide a widely accepted nomenclature on cell death in support of the continued development of the field

    Assumption without representation: the unacknowledged abstraction from communities and social goods

    We have not clearly acknowledged the abstraction from unpriceable “social goods” (derived from communities) which, different from private and public goods, simply disappear if it is attempted to market them. Separability from markets and economics has not been argued, much less established. Acknowledging communities would reinforce rather than undermine them, and thus facilitate the production of social goods. But it would also help economics by facilitating our understanding of – and response to – financial crises as well as environmental destruction and many social problems, and by reducing the alienation from economics often felt by students and the public

    According to André Malraux, the face-to-face of a work of art and the public is sufficient to bring about the miracle, which excludes the idea of, and even the need for, any form of mediation. As outlined here, we propose to relate this concept to the fact that the public policies regarding «culture» have mainly been policies about «art». Creation, and even more so the artists themselves, have been their beneficiaries to the detriment of actions intended to foster the «sharing of the sensible» and to diminish the inequalities in the relations to arts and to works of art.Pour André Malraux, la mise en présence de l'œuvre et du public suffit à l'accomplissement du miracle, ce qui exclut l'idée, voire la nécessité, de toute forme de médiation.     Les biosurfactants présentent un grand intérêt grâce à leur faible toxicité et à leur biodégradabilité. L'objectif de notre travail était l'obtention de nouveaux biosurfactants, lipoamino acides et lipopeptides, isoformes de la surfactine, lipoheptapeptide cyclique connu à ce jour comme le biosurfactant le plus efficace. Le criblage de souches de Bacillus pumilus et la synthèse enzymatique, ont permis l'obtention de lipoamino acides ayant des chaînes aliphatiques de tailles et de structures variables, dérivés de D-Asn et de L-Phe. Les lipo-D-Asn possèdent une activité surfactante modérée alors que les lipo-L-Phe, plus hydrophobes, sont de très bons surfactants. La synthèse d'isoformes de la surfactine, a été réalisée principalement en phase solide et leur cyclisation, en milieu liquide.
