    Invasive Techniken in der Notfallmedizin: IV. Koniotomie im Notarztdienst

    Zusammenfassung: Die Koniotomie im Notarztdienst ist eine sehr invasive Methode zur Atemwegssicherung. Sie ist aber unabdingbar, wenn andere weniger invasive Verfahren der Atemwegssicherung misslingen oder sich situationsabhängig als nicht zweckmäßig erweisen. Für die Durchführung wurden verschiedene Verfahren beschrieben, die in anatomisch-chirurgische Präpariertechniken und Punktionsverfahren unterteilt werden können. Das im jeweiligen Notarztsystem bevorzugte Verfahren orientiert sich meist an abteilungsüblichen Standards, die vom klinischen Vorgehen abgeleitet werden. Ausbildungs- und Trainingsmöglichkeiten für die einzelnen Verfahren bestehen in der Intensivmedizin, am Leichenpräparat und am Phantom. In dieser Arbeit werden verschiedene Methoden der Koniotomie besprochen sowie eine anatomische Präpariertechnik und 2Punktionsverfahren im Detail dargestell

    Evaluation of an Active LF Tracking System and Data Processing Methods for Livestock Precision Farming in the Poultry Sector

    Tracking technologies offer a way to monitor movement of many individuals over long time periods with minimal disturbances and could become a helpful tool for a variety of uses in animal agriculture, including health monitoring or selection of breeding traits that benefit welfare within intensive cage-free poultry farming. Herein, we present an active, low-frequency tracking system that distinguishes between five predefined zones within a commercial aviary. We aimed to evaluate both the processed and unprocessed datasets against a “ground truth” based on video observations. The two data processing methods aimed to filter false registrations, one with a simple deterministic approach and one with a tree-based classifier. We found the unprocessed data accurately determined birds’ presence/absence in each zone with an accuracy of 99% but overestimated the number of transitions taken by birds per zone, explaining only 23% of the actual variation. However, the two processed datasets were found to be suitable to monitor the number of transitions per individual, accounting for 91% and 99% of the actual variation, respectively. To further evaluate the tracking system, we estimated the error rate of registrations (by applying the classifier) in relation to three factors, which suggested a higher number of false registrations towards specific areas, periods with reduced humidity, and periods with reduced temperature. We concluded that the presented tracking system is well suited for commercial aviaries to measure individuals’ transitions and individuals’ presence/absence in predefined zones. Nonetheless, under these settings, data processing remains a necessary step in obtaining reliable data. For future work, we recommend the use of automatic calibration to improve the system’s performance and to envision finer movements

    Similarity in Temporal Movement Patterns in Laying Hens Increases with Time and Social Association.

    We explored the relationship between social associations and individual activity patterns in domestic hens. Out of 1420 laying hens, 421 hens were equipped with RFID tags attached to RFID-specific leg bands (leg bands from Company Roxan, Selkirk, Scotland) to continuously track their change in location across four different areas (one indoor and three outdoor areas). Using a combination of social network analysis for quantifying social relationships and dynamic time warping for characterizing the movement patterns of hens, we found that hens were consistent in their individual variation in temporal activity and maintained stable social relationships in terms of preferred association partners. In addition to being consistent, social associations correlated with movement patterns and this correlation strengthened over the period of observation, suggesting that the animals aligned their activity patterns with those of their social affiliates. These results demonstrate the importance of social relationships when considering the expression of individual behaviour. Notably, differences in temporal patterns emerge despite rather homogeneous rearing conditions, same environment, and low genetic diversity. Thus, while variation in behavioural phenotypes can be observed across isolated individuals, this study shows that the social environment within a group can shape and enhance variation in general movement patterns of individual animals

    Reliability of common mouse behavioural tests of anxiety: A systematic review and meta-analysis on the effects of anxiolytics.

    The validity of widely used rodent behavioural tests of anxiety has been questioned, as they often fail to produce consistent results across independent replicate studies. In this study, we assessed the sensitivity of common behavioural tests of anxiety in mice to detect anxiolytic effects of drugs prescribed to treat anxiety in humans. We conducted a pre-registered systematic review of 814 studies reporting effects of 25 anxiolytic compounds using common behavioural tests for anxiety. Meta-analyses of effect sizes of treatments showed that only two out of 17 commonly used test measures reliably detected effects of anxiolytic compounds. We report considerable between-study variation in size and even direction of effects of most anxiolytics on most outcome variables. Our findings indicate a general lack of sensitivity of those behavioural tests and cast serious doubt on both construct and predictive validity of most of these tests. In view of scientifically valid and ethically responsible research, we call for a revision of behavioural tests of anxiety in mice and the development of more predictive tests

    The rearing environment persistently modulates mouse phenotypes from the molecular to the behavioural level.

    The phenotype of an organism results from its genotype and the influence of the environment throughout development. Even when using animals of the same genotype, independent studies may test animals of different phenotypes, resulting in poor replicability due to genotype-by-environment interactions. Thus, genetically defined strains of mice may respond differently to experimental treatments depending on their rearing environment. However, the extent of such phenotypic plasticity and its implications for the replicability of research findings have remained unknown. Here, we examined the extent to which common environmental differences between animal facilities modulate the phenotype of genetically homogeneous (inbred) mice. We conducted a comprehensive multicentre study, whereby inbred C57BL/6J mice from a single breeding cohort were allocated to and reared in 5 different animal facilities throughout early life and adolescence, before being transported to a single test laboratory. We found persistent effects of the rearing facility on the composition and heterogeneity of the gut microbial community. These effects were paralleled by persistent differences in body weight and in the behavioural phenotype of the mice. Furthermore, we show that environmental variation among animal facilities is strong enough to influence epigenetic patterns in neurons at the level of chromatin organisation. We detected changes in chromatin organisation in the regulatory regions of genes involved in nucleosome assembly, neuronal differentiation, synaptic plasticity, and regulation of behaviour. Our findings demonstrate that common environmental differences between animal facilities may produce facility-specific phenotypes, from the molecular to the behavioural level. Furthermore, they highlight an important limitation of inferences from single-laboratory studies and thus argue that study designs should take environmental background into account to increase the robustness and replicability of findings

    IFN-γ-Based ELISpot as a New Tool to Detect Human Infections with Borna Disease Virus 1 (BoDV-1): A Pilot Study

    More than 40 human infections with the zoonotic Borna disease virus 1 (BoDV-1) have been reported to German health authorities from endemic regions in southern and eastern Germany. Diagnosis of a confirmed case is based on the detection of BoDV-1 RNA or BoDV-1 antigen. In parallel, serological assays such as ELISA, immunoblots, and indirect immunofluorescence are in use to detect the seroconversion of Borna virus-reactive IgG in serum or cerebrospinal fluid (CSF). As immunopathogenesis in BoDV-1 encephalitis appears to be driven by T cells, we addressed the question of whether an IFN-γ-based ELISpot may further corroborate the diagnosis. For three of seven BoDV-1-infected patients, peripheral blood mononuclear cells (PBMC) with sufficient quantity and viability were retrieved. For all three patients, counts in the range from 12 to 20 spot forming units (SFU) per 250,000 cells were detected upon the stimulation of PBMC with a peptide pool covering the nucleocapsid protein of BoDV-1. Additionally, individual patients had elevated SFU upon stimulation with a peptide pool covering X or phosphoprotein. Healthy blood donors (n = 30) and transplant recipients (n = 27) were used as a control and validation cohort, respectively. In this pilot study, the BoDV-1 ELISpot detected cellular immune responses in human patients with BoDV-1 infection. Its role as a helpful diagnostic tool needs further investigation in patients with BoDV-1 encephalitis

    Plant Species Loss Affects Life-History Traits of Aphids and Their Parasitoids

    The consequences of plant species loss are rarely assessed in a multi-trophic context and especially effects on life-history traits of organisms at higher trophic levels have remained largely unstudied. We used a grassland biodiversity experiment and measured the effects of two components of plant diversity, plant species richness and the presence of nitrogen-fixing legumes, on several life-history traits of naturally colonizing aphids and their primary and secondary parasitoids in the field. We found that, irrespective of aphid species identity, the proportion of winged aphid morphs decreased with increasing plant species richness, which was correlated with decreasing host plant biomass. Similarly, emergence proportions of parasitoids decreased with increasing plant species richness. Both, emergence proportions and proportions of female parasitoids were lower in plots with legumes, where host plants had increased nitrogen concentrations. This effect of legume presence could indicate that aphids were better defended against parasitoids in high-nitrogen environments. Body mass of emerged individuals of the two most abundant primary parasitoid species was, however, higher in plots with legumes, suggesting that once parasitoids could overcome aphid defenses, they could profit from larger or more nutritious hosts. Our study demonstrates that cascading effects of plant species loss on higher trophic levels such as aphids, parasitoids and secondary parasitoids begin with changed life-history traits of these insects. Thus, life-history traits of organisms at higher trophic levels may be useful indicators of bottom-up effects of plant diversity on the biodiversity of consumers

    The peroxisome: still a mysterious organelle

    More than half a century of research on peroxisomes has revealed unique features of this ubiquitous subcellular organelle, which have often been in disagreement with existing dogmas in cell biology. About 50 peroxisomal enzymes have so far been identified, which contribute to several crucial metabolic processes such as β-oxidation of fatty acids, biosynthesis of ether phospholipids and metabolism of reactive oxygen species, and render peroxisomes indispensable for human health and development. It became obvious that peroxisomes are highly dynamic organelles that rapidly assemble, multiply and degrade in response to metabolic needs. However, many aspects of peroxisome biology are still mysterious. This review addresses recent exciting discoveries on the biogenesis, formation and degradation of peroxisomes, on peroxisomal dynamics and division, as well as on the interaction and cross talk of peroxisomes with other subcellular compartments. Furthermore, recent advances on the role of peroxisomes in medicine and in the identification of novel peroxisomal proteins are discussed
