    El presente trabajo tuvo como objetivo llevar a cabo la propuesta de implementación de un plan de mantenimiento preventivo en una empresa de estacionamiento aplicando la metodología RCM para la mejora de la disponibilidad de los equipos en Lima, 2021 mediante la determinación de la influencia de la metodología RCM aplicado en la disponibilidad de los equipos en una empresa de estacionamiento, el diseño de la propuesta de un plan de mantenimiento preventivo aplicando la metodología RCM en la empresa de estacionamiento contemplando el sistema AMFE, la medida del impacto en el indicador de disponibilidad de los equipos luego de la implementación de la propuesta de mantenimiento y el cálculo de la viabilidad económica de la propuesta de implementación de mantenimiento preventivo bajo la metodología RCM en la empresa de estacionamiento. Dentro de su marco, se caracterizó por contar con un enfoque cuantitativo, aplicado y experimental, con el cual se logró concluir que se puede aumentar la disponibilidad de los equipos de un 33% hasta 98%.The objective of this work was to carry out the proposal for the implementation of preventive maintenance in a parking company applying the RCM methodology to improve the availability of parking equipment in Lima, 2021 by determining the influence of the RCM methodology applied in the availability of equipment in a parking company, the design of the proposal of a preventive maintenance plan applying the RCM methodology in the parking company contemplating the FMEA system, the measurement of the impact on the equipment availability indicator after the implementation of the maintenance proposal and the calculation of the economic feasibility of the proposal for the implementation of preventive maintenance under the RCM methodology in the parking company. Within its framework, it was characterized by having a quantitative, applied and experimental approach, with which it was possible to conclude that the availability of the equipment can be increased from 33% to 98%

    The subject of this research work was the burnishing process of a cylindrical opening of a multicuspid mandrel (push burnishing broach). It was examined by dividing the change in mesh texture of the surface layer of metal fibers in a section of a cylindrical opening, which coincides with the axis of the mandrel opening of the assembled bush. Processing of the distortion of the grid and comparing the results with its initial parameters allow to establish the nature and depth of distribution of the elastoplastic deformation.Исследовался процесс дорнования цилиндрического отверстия многозубым дорном. Характер изменения текстуры волокон металла поверхностного слоя в диаметральном продольном сечении цилиндрического отверстия сборной втулки изучался при помощи метода делительных сеток. Полученное искажение делительной сетки и сравнивание с исходными ее параметрами позволило установить характер и глубину упругопластических деформаций

    ACtivE:Assembly and CRISPR-targeted in vivo editing for yeast genome engineering using minimum reagents and time

    [Image: see text] Thanks to its sophistication, the CRISPR/Cas system has been a widely used yeast genome editing method. However, CRISPR methods generally rely on preassembled DNAs and extra cloning steps to deliver gRNA, Cas protein, and donor DNA. These laborious steps might hinder its usefulness. Here, we propose an alternative method, Assembly and CRISPR-targeted in vivo Editing (ACtivE), that only relies on in vivo assembly of linear DNA fragments for plasmid and donor DNA construction. Thus, depending on the user’s need, these parts can be easily selected and combined from a repository, serving as a toolkit for rapid genome editing without any expensive reagent. The toolkit contains verified linear DNA fragments, which are easy to store, share, and transport at room temperature, drastically reducing expensive shipping costs and assembly time. After optimizing this technique, eight loci proximal to autonomously replicating sequences (ARS) in the yeast genome were also characterized in terms of integration and gene expression efficiencies and the impacts of the disruptions of these regions on cell fitness. The flexibility and multiplexing capacity of the ACtivE were shown by constructing a β-carotene pathway. In only a few days, >80% integration efficiency for single gene integration and >50% integration efficiency for triplex integration were achieved on Saccharomyces cerevisiae BY4741 from scratch without using in vitro DNA assembly methods, restriction enzymes, or extra cloning steps. This study presents a standardizable method to be readily employed to accelerate yeast genome engineering and provides well-defined genomic location alternatives for yeast synthetic biology and metabolic engineering purposes

    The impact that catatonic surfactants have on the soft matter world

    Catanionic surfactants are a class of amphiphile which result from the equimolar mixture of a cationic and an anionic surfactant, where the salt formed by the counterions is removed. In our group, for the first time, we have synthetized the unique catanionic surfactant: benzyl-n-hexadecyldimethylammonium 1,4-bis-2-ethylhexylsulfosuccinate (BHD-AOT), which has a tremendous impact in the soft matter field. This came out because it can form reverse micelles (RMs) or spontaneous large unilamellar vesicles, depending on the solvent used. RMs are supramolecular assemblies of surfactants formed in nonpolar solvents, in which the polar head groups of the surfactants point inward and the hydrocarbon chains point toward to the nonpolar medium. Vesicles are spherical aggregates formed by some amphiphilic compounds in water, in which the bilayer surrounds an aqueous void volume that can be ?loaded? with a wide variety of water-soluble marker molecules. In this review, we will show the synthesis, characterization, and properties of the different organized media created by BHD-AOT and, exciting applications since, as we proved, it is non-toxic and results especially interesting for drug delivery system.Fil: Villa, Cristian C..     Enhanced production of taxadiene in <i>Saccharomyces cerevisiae</i>

    BackgroundCost-effective production of the highly effective anti-cancer drug, paclitaxel (Taxol®), remains limited despite growing global demands. Low yields of the critical taxadiene precursor remains a key bottleneck in microbial production. In this study, the key challenge of poor taxadiene synthase (TASY) solubility in S. cerevisiae was revealed, and the strains were strategically engineered to relieve this bottleneck.ResultsMulti-copy chromosomal integration of TASY harbouring a selection of fusion solubility tags improved taxadiene titres 22-fold, up to 57 ± 3 mg/L at 30 °C at microscale, compared to expressing a single episomal copy of TASY. The scalability of the process was highlighted through achieving similar titres during scale up to 25 mL and 250 mL in shake flask and bioreactor cultivations, respectively at 20 and 30 °C. Maximum taxadiene titres of 129 ± 15 mg/L and 127 mg/L were achieved through shake flask and bioreactor cultivations, respectively, of the optimal strain at a reduced temperature of 20 °C.ConclusionsThe results of this study highlight the benefit of employing a combination of molecular biology and bioprocess tools during synthetic pathway development, with which TASY activity was successfully improved by 6.5-fold compared to the highest literature titre in S. cerevisiae cell factories

    Elliptic flow of muons from heavy-flavour hadron decays at forward rapidity in Pb-Pb collisions at root s(NN)=2.76TeV

    The elliptic flow, v(2), of muons from heavy-flavour hadron decays at forward rapidity (2.5 <y <4) is measured in Pb-Pb collisions at root s(NN)= 2.76TeVwith the ALICE detector at the LHC. The scalar product, two- and four-particle Q cumulants and Lee-Yang zeros methods are used. The dependence of the v(2) of muons from heavy-flavour hadron decays on the collision centrality, in the range 0-40%, and on transverse momentum, p(T), is studied in the interval 3 <p(T)<10 GeV/c. A positive v(2) is observed with the scalar product and two-particle Q cumulants in semi-central collisions (10-20% and 20-40% centrality classes) for the p(T) interval from 3 to about 5GeV/c with a significance larger than 3 sigma, based on the combination of statistical and systematic uncertainties. The v(2) magnitude tends to decrease towards more central collisions and with increasing pT. It becomes compatible with zero in the interval 6 <p(T)<10 GeV/c. The results are compared to models describing the interaction of heavy quarks and open heavy-flavour hadrons with the high-density medium formed in high-energy heavy-ion collisions.
