    This study employs the research and development model of Borg & Gall and Dick & Carey involving students of SMPN 4 Kota Cirebon. This study aims to develop Cirebonese language learning media for the junior high school of the eleventh-grade students. To find out the results of the content of media, instructional design and instructional media experts validation were conducted. In addition, individual, group, and field try out were conducted.The data regarding the quality of the product were collected through questionnaire then analyzed by using descriptive analysis of qualitative and quantitative descriptive analysis. The try-out process involved the experts of Cirebonese language media and instructional design. It also involved six students an individual (one to one) try out, fifteen students for group and thirty-four students for the field try out.The results showed, first, because t, 3,0569E-18 or = 3.0569 x 10-18 or = .000000000000030596 smaller than t table at a significance level (α) amounted to 2.034515 0.05, then it rejects Ho and accepts Ha. It can be concluded that the Cirebonese language learning using developed instructional media is effective in improving students' learning outcomes. Second, the mean scores after using the media of Cirebonese language learning showed higher (better) than before using learning media of Cirebonese language. It is shown in the table Paired Sample Statistics, where the mean score before using media language learning Cirebon amounted 50.44117647 while the mean score after using learning media of Cirebonese language amounted 65.88235294.     A Comparative Molecular Dynamics Study of Methylation State Specificity of JMJD2A

    Histone modifications have great importance in epigenetic regulation. JMJD2A is a histone demethylase which is selective for di- and trimethyl forms of residues Lys9 and Lys36 of Histone 3 tail (H3K9 and H3K36). We present a molecular dynamics simulations of mono-, di- and trimethylated histone tails in complex with JMJD2A catalytic domain to gain insight into how JMJD2A discriminates between the methylation states of H3K9. The methyl groups are located at specific distances and orientations with respect to Fe(II) in methylammonium binding pocket. For the trimethyllysine the mechanism which provides the effectual orientation of methyl groups is the symmetry, whereas for the dimethyllysine case the determining factors are the interactions between methyllysine head and its environment and subsequently the restriction on angular motion. The occurrence frequency of methyl groups in a certain proximity of Fe(II) comes out as the explanation of the enzyme activity difference on di- and tri-methylated peptides. Energy analysis suggests that recognition is mostly driven by van der Waals and followed by Coulombic interactions in the enzyme-substrate interface. The number (mono, di or tri) and orientations of methyl groups and water molecules significantly affect the extent of van der Waals interaction strengths. Hydrogen bonding analysis suggests that the interaction between JMJD2A and its substrates mainly comes from main chain-side chain interactions. Binding free energy analysis points out Arg8 as an important residue forming an intra-substrate hydrogen bond with tri and dimethylated Lys9 of the H3 chain. Our study provides new insights into how JMJD2A discriminates between its substrates from both a structural and dynamical point of view

    Corticosteroid effects on ventilator-induced diaphragm dysfunction in anesthetized rats depend on the dose administered

    <p>Abstract</p> <p>Background</p> <p>High dose of corticosteroids has been previously shown to protect against controlled mechanical ventilation (CMV)-induced diaphragmatic dysfunction while inhibiting calpain activation. Because literature suggests that the calpain inhibiting effect of corticosteroid depends on the dose administered, we determined whether lower doses of corticosteroids would also provide protection of the diaphragm during CMV. This may be important for patients undergoing mechanical ventilation and receiving corticosteroids.</p> <p>Methods</p> <p>Rats were assigned to controls or to 24 hours of CMV while being treated at the start of mechanical ventilation with a single intramuscular administration of either saline, or 5 mg/kg (low MP) or 30 mg/kg (high MP) of methylprednisolone.</p> <p>Results</p> <p>Diaphragmatic force was decreased after CMV and this was exacerbated in the low MP group while high MP rescued this diaphragmatic dysfunction. Atrophy was more severe in the low MP group than after CMV while no atrophy was observed in the high MP group. A significant and similar increase in calpain activity was observed in both the low MP and CMV groups whereas the high dose prevented calpain activation. Expression of calpastatin, the endogenous inhibitor of calpain, was decreased in the CMV and low MP groups but its level was preserved to controls in the high MP group. Caspase-3 activity increased in all CMV groups but to a lesser extent in the low and high MP groups. The 20S proteasome activity was increased in CMV only.</p> <p>Conclusions</p> <p>Administration of 30 mg/kg methylprednisolone during CMV protected against CMV-induced diaphragm dysfunction while 5 mg/kg was more deleterious. The protective effect is due mainly to an inhibition of the calpain system through preservation of calpastatin levels and to a lesser extent to a caspase-3 inhibition.</p

    Observation of two new Ξb\Xi_b^- baryon resonances

    Two structures are observed close to the kinematic threshold in the Ξb0π\Xi_b^0 \pi^- mass spectrum in a sample of proton-proton collision data, corresponding to an integrated luminosity of 3.0 fb1^{-1} recorded by the LHCb experiment. In the quark model, two baryonic resonances with quark content bdsbds are expected in this mass region: the spin-parity JP=12+J^P = \frac{1}{2}^+ and JP=32+J^P=\frac{3}{2}^+ states, denoted Ξb\Xi_b^{\prime -} and Ξb\Xi_b^{*-}. Interpreting the structures as these resonances, we measure the mass differences and the width of the heavier state to be m(Ξb)m(Ξb0)m(π)=3.653±0.018±0.006m(\Xi_b^{\prime -}) - m(\Xi_b^0) - m(\pi^{-}) = 3.653 \pm 0.018 \pm 0.006 MeV/c2/c^2, m(Ξb)m(Ξb0)m(π)=23.96±0.12±0.06m(\Xi_b^{*-}) - m(\Xi_b^0) - m(\pi^{-}) = 23.96 \pm 0.12 \pm 0.06 MeV/c2/c^2, Γ(Ξb)=1.65±0.31±0.10\Gamma(\Xi_b^{*-}) = 1.65 \pm 0.31 \pm 0.10 MeV, where the first and second uncertainties are statistical and systematic, respectively. The width of the lighter state is consistent with zero, and we place an upper limit of Γ(Ξb)<0.08\Gamma(\Xi_b^{\prime -}) < 0.08 MeV at 95% confidence level.     Search for CP violation in D+→ϕπ+ and D+s→K0Sπ+ decays

    A search for CP violation in D + → ϕπ + decays is performed using data collected in 2011 by the LHCb experiment corresponding to an integrated luminosity of 1.0 fb−1 at a centre of mass energy of 7 TeV. The CP -violating asymmetry is measured to be (−0.04 ± 0.14 ± 0.14)% for candidates with K − K + mass within 20 MeV/c 2 of the ϕ meson mass. A search for a CP -violating asymmetry that varies across the ϕ mass region of the D + → K − K + π + Dalitz plot is also performed, and no evidence for CP violation is found. In addition, the CP asymmetry in the D+s→K0Sπ+ decay is measured to be (0.61 ± 0.83 ± 0.14)%
